Chimeric insulin/insulin-like growth factor-1 receptors and insulin receptor alpha-subunit point mutants were characterized
with respect to their binding properties for insulin and insulin-like ...growth factor-1 (IGF-1) and their ability to translate
ligand interaction into tyrosine kinase activation in intact cells. We found that replacement of the amino-terminal 137 amino
acids of the insulin receptor (IR) with the corresponding 131 amino acids of the IGF-1 receptor (IGF-1R) resulted in loss
of affinity for both ligands. Further replacement of the adjacent cysteine region with IGF-1R sequences fully reconstituted
affinity for IGF-1, but only marginally for insulin. Unexpectedly, replacement of the IR cysteine-rich domain alone by IGF-1R
sequences created a high affinity receptor for both insulin and IGF-1. The binding characteristics of all receptor chimeras
reflected the potential of both ligands to regulate the receptor tyrosine kinase activity in intact cells. Our chimeric receptor
data, in conjunction with IR amino-terminal domain point mutants, strongly suggest major contributions of structural determinants
in both amino- and carboxyl-terminal IR alpha-subunit regions for the formation of the insulin-binding pocket, whereas, surprisingly,
the residues defining IGF-1 binding are present predominantly in the cysteine-rich domain of the IGF-1R.

The CLEO-III silicon vertex detector is a 4-layer device with double-sided silicon sensors arranged in a barrel design, covering 93% of the solid angle. After initially meeting its design goals of ...signal-to-noise performance and spatial resolution, the signal efficiency deteriorated on the rφ sensor side in the two innermost layers due to radiation induced sensor effects. Conclusion. Tissue marking with soot-covered fine needles enables the optical coherence tomography (OCT) scanning plane to be localized within the histological specimen to an accuracy of ∼50 µm. ...Tissue water content is an especially important parameter for in vitro measurements. Dehydration tends to produce an increase in surface reflections and a reduction in imaging depth. Objectives. The aim of this in vitro study was to evaluate parameters relevant to the visualization and conservation process to allow optimal images to be generated for later differentiation between healthy and degenerated tissue in vivo. Materials and methods. Various methods of marking samples were applied in vitro to achieve accurate overlaps of the OCT scanning plane and the corresponding section of the histological specimen. The influence of temperature and tissue water content was investigated using both porcine and human tissue. Samples were marked using fine needles, ablation craters generated by laser application, and colour markers introduced into the tissue. Results. It was demonstrated that the water content of tissue exerts a direct influence on OCT imaging, whereas above 15°C temperature had no effect on image quality. With regard to the marking of samples, the best results were obtained using sooted fine needles.

CLEO III is the new experimental phase of the CLEO experiment at the CESR accelerator. Both the accelerator and the detector have recently been upgraded. A new charged particle tracking system with ...the addition of a ring imaging Cherenkov particle identification system has been installed. A major part of the tracking system upgrade was the construction of a new four-layer double-sided silicon tracker with 93% solid angle coverage and new readout electronics. The CLEO III upgrade was completed in February 2000 with the installation of the silicon detector. CLEO III has finished the commissioning phase and is now taking data. The design of the detector and first performance results are presented here.
The Monorail Piccolino coronary angioplasty balloon catheter (MBC) was evaluated on 118 patients at two centers. Technical success was achieved in 110 patients (93%). Time for catheter exchange and ...total fluoroscopy time were significantly lower for the Monorail catheter than with standard equipment (exchange time 97 vs. 170 seconds P less than .05 and fluoroscopy time 17 vs. 88 seconds P less than .001). The advantages of rapid exchange and the ability of utilize 2 Monorail balloon catheters through one 9F guiding catheter for simultaneous inflations allowed for maximal flexibility in treating patients with bifurcation lesions. The double wire approach utilizing one Monorail balloon catheter with a 7F guiding catheter was also technically successful. The Monorail Piccolino balloon catheter has unique features that allow for greater ease of operator use, rapid catheter exchange, and optimal angiographic visualization. It is felt that this catheter design provides distinct advantages over standard angioplasty equipment.
Peanut (Arachis hypogaea L.) in the United States is generally grown in conventionally tilled systems. However, interest in reduced tillage peanut production has increased. Five experiments were ...conducted in North Carolina to determine if cultivar selection and digging date affected peanut yield and economic value when peanut was seeded into conventionally tilled seedbeds compared with strip tillage into small-grain cover crop or stubble from the crop planted the previous summer. In separate experiments, peanut yield and economic value in these tillage systems were compared with peanut strip-tilled into beds prepared the previous fall (stale seedbeds). Cultivar selection and digging date did not affect pod yield or gross value when comparing tillage systems. Pod yield in conventional and stale seedbed systems was similar in all five experiments where these systems were compared, and yields in these tillage systems exceeded those of strip tillage into crop stubble in three of five experiments. Pod yield was similar among all three tillage systems in the other two experiments. In experiments where only conventional tillage and strip tillage systems were compared, pod yield was similar between the two tillage systems in four experiments, higher in conventional tillage compared with strip tillage in one experiment, and higher for strip tillage compared with conventional tillage in one experiment. In 16 of 17 comparisons, pod yield of peanut planted in conventional tillage systems equaled or exceeded that of peanut planted into stubble from the previous crop.
We previously reported the isolation of a 2.5 Mb tumor-suppressing subchromosomal transferable fragment (STF) from 11p15.5 and the identification of nine known genes and four novel genes within this ...STF. We now report the isolation of a fifth novel cDNA, tumor-suppressing STF cDNA 5, designated TSSC5, located within the STF. TSSC5 encodes a predicted protein of 424 amino acids. Sequence analysis suggests that TSSC5 is a membrane protein with 10 transmembrane segments, and it is located between two imprinted genes, p57KIP2 and TSSC3. Northern blot hybridization revealed a 1.6-kb transcript in multiple adult tissues and in fetal liver and kidney, consistent with a potential role in embryonal tumors. We also found that TSSC5 is imprinted with preferential expression from the maternal chromosome. Reverse transcription-PCR analysis of TSSC5 revealed frequent occurrence of aberrant RNA splicing, which deleted exons 4, 5, and 6 in Wilms' tumors. Mutational analysis of TSSC5 by direct DNA sequencing of exons revealed a base substitution of G1120A in a Wilms' tumor, matched normal kidney, and the patient's mother, changed Arg at codon 309 to Gln. The G1120A substitution thus represents either a rare polymorphism or a tumor-predisposing mutation, because the mutant allele was of maternal origin and preferentially expressed in the patient's tissue. A second base substitution, C892T, was found in a lung cancer, changing Ser at codon 233 to Phe. This substitution was absent from the matched normal tissue and thus represented a somatic mutation. We also found loss of heterozygosity in the lung cancer, suggesting that TSSC5 may be a conventional tumor suppressor gene in the adult human lung and an imprinted tumor suppressor gene in the fetal kidney.
