To identify molecular pathways and interrelated genes whose differential expression was associated with asthma control.
We performed gene set enrichment analyses of asthma control in 1,170 adults with asthma, each with gene expression data derived from either whole blood (WB) or unstimulated CD4
T lymphocytes (CD4), and a self-reported asthma control score representing either the preceding 6 months (chronic) or 7 days (acute). Our study comprised a discovery WB cohort (n = 245, chronic) and three independent, nonoverlapping replication cohorts: a second WB set (n = 448, acute) and two CD4 sets (n = 300, chronic; n = 77, acute).
In the WB discovery cohort, we found significant overrepresentation of genes associated with asthma control in 1,106 gene sets from the Molecular Signatures Database (false discovery rate, <5%). Of these, 583 (53%) replicated in at least one replication cohort (false discovery rate, <25%). Suboptimal control was associated with signatures of eosinophilic and granulocytic inflammatory signals, whereas optimal control signatures were enriched for immature lymphocytic patterns. These signatures included two related biologic processes related to activation by TREM-1 (triggering receptor expressed on myeloid cells 1) and lipopolysaccharide.
Together, these results demonstrate the existence of specific, reproducible transcriptomic components in blood that vary with degree of asthma control and implicate a novel biologic target (TREM-1).
Purpose:Mechanical and surface characteristics of the zirconia dental ceramics may be adversely impacted by temperature changes during the veneering firing cycles. A potential synergistic failure ...could be caused by ageing when exposed to the oral environment and heat treatment. The current research aimed to determine the influence of heat treatment and in-vitro ageing on mechanical properties and surface roughness of cold isostatic pressed zirconia. Materials and Methods:15 bar shaped specimens with dimensions 28mm×4mm×2mm are milled from two types of partially sintered Y-TZP ceramic blanks (IPS e.max ZirCAD (IZ),ZENO Zr (WZ)). Out of 15 samples five are used as control (C group), five are heat treated (H group) and five are heat treated and subsequently aged (HA group). Flexural strength and shear bond strength are measured using universal testing machine and surface roughness is measured using proliferometer. Results:Heat treatment on the ivoclar and weiland samples led to an increase in surface roughness and a loss in mechanical properties (flexural strength and shear bond strength), although the differences are not statistically significant (p>0.05). Yet, the combined ageing and heat treatment created a synergistic effect on surface roughness and mechanical characteristics. (p<0.001). More variations were seen in the weiland samples. Conclusion:Firing and ageing processes may have an impact on zirconia ceramics clinical performance, which raises the risk of failure.
Purpose: Purpose of this study to evaluate the effect of implant position on prosthesis movement of mandibular two implant-supported overdenture under simulating biting force. Materials and ...methodology: 30 experimental overdentures (N=30) were made for an edentulous mandibular test model, which included artificial mucosa. Based on implant position, 30 samples overdentures were divided into three groups (n=10) (i.e., A and E, B and D,in between B and D). For this study, the ball and O-ring attachment system was used. 100 N vertical loads were used on the left first molar region, 55N on the left canine, and 40N on the mid-anterior area. The loading points vertical displacement as well as the right distal edges horizontal and vertical displacements were measured.A 1-way analysis of variance test was used to statistically analyse the displacement values with the implant position as a factor, (p=.05) was used to statistically analyze the values of the vertical and horizontal displacement at the distal edge of the overdenture. Results:The mean horizontal displacement at right distal edge on incisor, canine and molar loading was highest in Group III with p value < 0.05. when compared in between groups with paired t tests there was no statistically significant difference on incisor loading between group II and Group III with p value >0.05. Mean vertical displacement at right distal edge on incisor loading was highest in group II and on canine and molar loading, it was highest in Group III. Conclusion:Change in implant position influences the displacement of implant supported overdenture.

Cytogenetic reports of chondroblastoma, a rare benign bone tumor, are few. Cytogenetic analysis of a benign and a malignant chondroblastoma in this study revealed the following abnormal chromosomal complements: 47,XY,+5,t(5;5)(p10;q10) and 45, XY,del(2)(p23),del(3)(q23q27),dup(8)(q12q21.), del(11) (q14q23), -13, add (17) (q25) x 2, respectively. Although a specific chromosomal abnormality has not yet emerged for chondroblastoma, abnormalities of chromosomes 5 and 8 have been reported previously in this neoplasm, suggesting preferential involvement of these two chromosomes.

As knowledgebases continue to play an increasingly important ...role in scientific research, a number of studies have evaluated their accuracy and revealed various errors. While some are curation errors, others are the result of incorrect information published in the scientific literature. By taking the example of sirtuin-5, a complex annotation case, we will describe the curation procedure of UniProtKB/Swiss-Prot and detail how we report conflicting information in the database. We will demonstrate the importance of collaboration between resources to ensure curation consistency and the value of contributions from the user community in helping maintain error-free resources. To report a case of an extraskeletal myxoid chondrosarcoma (EMC) arising from the jugular foramen. EMCs are tumors usually seen in the deep soft tissues of the extremities and are rarely seen within ...the intracranial cavity. The histological differential diagnosis includes chordoma, conventional chondrosarcoma and chordoid meningioma, among others. A distinguishing feature of EMC is their characteristic reciprocal translocation t(9;22)(q22;q12).
A 63-year-old man presented with progressive hearing loss and gait imbalance. Magnetic resonance imaging showed a heterogeneously enhancing 2.4 cm mass in the cerebellopontine angle. A right far lateral transcondylar skull base approach with gross total removal of the tumor was performed. Intraoperative findings showed that the mass appeared to arise from the glossopharyngeal nerve within the jugular foramen.
Histology, immunohistochemistry, and fluorescence in situ hybridization studies were performed.
Histological and immunohistochemical studies were compatible with the diagnosis of EMC. Fluorescence in situ hybridization studies showed disruption of the EWS gene locus at 22q12 and added further support to the diagnosis.
We report a rare case of EMC arising from the jugular foramen, and the diagnosis of EMC can be supported by confirmation of disruption of the EWS gene locus.

Precision attachment-supported over Dentures with two teeth retained in canine region is a predictable treatment option in edentulous mandible rather than conventional complete denture as it offers increased retention, stability, comfort, bone preservation, maintenance of proproception and patient compliance. This study was carried out to determine patient satisfaction with precision attachment-supported overdentures and conventional overdenture and its impact on the treatment outcome. Material & Methods: Twenty(20) edentulous patients were randomly divided into two groups to receive precision attachment -supported overdentures and conventional overdenture in mandibular arch. Questionnaires were given to the patient after treatment at 6 and 12months interval. Data collected were statistically analyzed. Results: Results obtained from this study show that patients with precision attachment -supported overdentures were more satisfied with the treatment outcome, and there was a significant difference in patient satisfaction between the two groups.
Cytogenetic analysis was performed on three specimens of clear cell sarcoma, a rare neoplasm of uncertain histogenesis. Chromosomal analysis of clear cell sarcoma has not been reported previously. ...Two of the specimens analyzed consisted of the primary foot lesion and subsequent lymph node metastasis in a 29-year-old male. The other specimen was a primary foot lesion in a 61-year-old male. Clonal abnormalities were detected in all three of the specimens. The significance of these results with regard to the origin of this uncommon neoplasm is discussed.
A number of concerns have recently been raised about whether or not antidepressant medications are associated with suicidal thoughts and behaviour in children and adolescents. These concerns are ...based largely on results of meta-analyses of randomized, controlled trials (RCTs). Controversy exists about generalizing evidence from short-term RCTs, designed primarily to test efficacy outcomes, to routine practice settings. Pharmacoepidemiological studies complement RCTs by using observational methods to examine safety and effectiveness of medications in the general population. This article reviews the contribution of pharmacoepidemiology to the controversy surrounding suicide risk in children and adolescents taking antidepressants, noting how variations in study design and adjustment for potential confounding factors influence outcome.

Synovial sarcoma is characterized cytogenetically by an X;18 translocation t (X;18) (p11;q11) that results in the fusion of the SYT gene from chromosome 18 to either of two highly homologous genes at ...Xp11, SSX1 or SSX2. Heterogeneity within the synovial sarcoma fusion junctions is rare. Examination of a primary monophasic synovial sarcoma for an SYT-SSX fusion transcript by reverse-transcription polymerase chain reaction revealed a smaller-than-expected product. Direct sequencing of the product disclosed a novel SYT-SSX1 fusion transcript that contained an additional 51 bp of normal SSX1 sequence but lost 135 bp of the SYT sequence. Detection of synovial sarcoma hybrid transcripts is useful for diagnosis and should include the recognition of distinct variants.
