There is increasing interest in protecting, restoring and creating ‘blue carbon’ ecosystems (BCE; mangroves, tidal marshes and seagrasses) to sequester atmospheric CO2-C and thereby contribute to climate change mitigation. While a growing number of countries aspire to report greenhouse gas emission and carbon sequestration changes from these ecosystems under voluntary international reporting requirements, few countries have domestic policy frameworks that specifically support the quantification and financing of carbon emission abatement through BCE management.
Australia, as home to approximately 5–11% of global blue carbon stocks, has a substantial interest in the development of blue carbon policy. Here we assess the potential inclusion of blue carbon within Australia's Emissions Reduction Fund, emphasizing issues and approaches that have global relevance. We used a participatory workshop of scientific experts and carbon industry stakeholders to identify blue carbon management actions that would meet the requirements of the Fund. In total, twelve actions were assessed for their greenhouse gas emission abatement potential and the ability to measure abatement reliably, using a combination of available data and qualitative and quantitative methods, including expert knowledge.
We identify and discuss the five most relevant and promising activities, encompassing the protection, restoration and creation of mangroves, tidal marshes and seagrasses. On a per area basis, mean abatement intensity of organic carbon (Corg) was highest for the (re)introduction of tidal flow resulting in establishment of mangrove (13–15 Mg Corg ha−1 yr−1) and tidal marsh (6–8 Mg Corg ha−1 yr−1), followed by land use planning for sea-level rise for the creation of new mangrove habitat (8 Mg Corg ha−1 yr−1). The avoided disturbance of existing mangroves, tidal marshes and seagrasses has the twofold benefit of avoiding remineralisation of existing stocks, plus the future annual abatement associated with the net sequestration of atmospheric CO2-C as Corg with the continued functioning of these BCE. Our approach offers a template that uses best available information to identify options for carbon abatement through management of coastal landscapes, and details current knowledge gaps and important technical aspects that need to be considered for implementation in carbon crediting schemes.
Global climate cooled from the early Eocene hothouse (∼52–50 Ma) to the latest Eocene (∼34 Ma). At the same time, the tectonic evolution of the Southern Ocean was characterized by the opening and ...deepening of circum-Antarctic gateways, which affected both surface- and deep-ocean circulation. The Tasmanian Gateway played a key role in regulating ocean throughflow between Australia and Antarctica. Southern Ocean surface currents through and around the Tasmanian Gateway have left recognizable tracers in the spatiotemporal distribution of plankton fossils, including organic-walled dinoflagellate cysts. This spatiotemporal distribution depends on both the physicochemical properties of the water masses and the path of surface-ocean currents. The extent to which climate and tectonics have influenced the distribution and composition of surface currents and thus fossil assemblages has, however, remained unclear. In particular, the contribution of climate change to oceanographic changes, superimposed on long-term and gradual changes induced by tectonics, is still poorly understood. To disentangle the effects of tectonism and climate in the southwest Pacific Ocean, we target a climatic deviation from the long-term Eocene cooling trend: the Middle Eocene Climatic Optimum (MECO; ∼40 Ma). This 500 kyr phase of global warming was unrelated to regional tectonism, and thus provides a test case to investigate the ocean's physicochemical response to climate change alone. We reconstruct changes in surface-water circulation and temperature in and around the Tasmanian Gateway during the MECO through new palynological and organic geochemical records from the central Tasmanian Gateway (Ocean Drilling Program Site 1170), the Otway Basin (southeastern Australia), and the Hampden Beach section (New Zealand). Our results confirm that dinocyst communities track specific surface-ocean currents, yet the variability within the communities can be driven by superimposed temperature change. Together with published results from the east of the Tasmanian Gateway, our new results suggest a shift in surface-ocean circulation during the peak of MECO warmth. Simultaneous with high sea-surface temperatures in the Tasmanian Gateway area, pollen assemblages indicate warm temperate rainforests with paratropical elements along the southeastern margin of Australia. Finally, based on new age constraints, we suggest that a regional southeast Australian transgression might have been coincident with the MECO.
Kunzea (Myrtaceae) trees and shrubs, generally called kānuka, grow across most of Aotearoa/New Zealand (NZ). With the exception of K. sinclairii, an offshore island endemic, kānuka had been treated ...as an Australasian species K. ericoides. However, a 2014 taxonomic revision recognized ten species, all endemic to NZ. Kānuka chemistry is less studied than that of its closest relative in NZ, mānuka (Leptospermum scoparium), which shows very distinct regional foliage chemotypes. We have used a miniaturized method with GC and 1H NMR to analyze foliage chemistry of voucher specimens from across the geographic ranges of the ten NZ Kunzea species. We found common mono- and sesquiterpenes, with α-pinene dominant in all samples, but only traces of antimicrobial triketones. Two unusual flavanones, with unsubstituted B-rings and known bioactivity against Phytophthora, did distinguish some of the samples. 5,7-Dihydroxy-6,8-dimethyl flavanone was only found at high concentrations in the three K. sinclairii samples in this study's sample set, but this compound has separately been reported in K. robusta samples from a nearby region. Therefore none of the NZ Kunzea species was distinguished by the chemistry analyzed in this study, but there is a possibility of regional flavonoid chemotypes cutting across the species boundaries.

Platelet-derived growth factor (PDGF) has been directly implicated in developmental and physiological processes, as well as in human cancer, fibrotic diseases and arteriosclerosis. The PDGF family ...currently consists of at least three gene products, PDGF-A, PDGF-B and PDGF-C, which selectively signal through two PDGF receptors (PDGFRs) to regulate diverse cellular functions. After two decades of searching, PDGF-A and B were the only ligands identified for PDGFRs. Recently, however, database mining has resulted in the discovery of a third member of the PDGF family, PDGF-C, a functional analogue of PDGF-A that requires proteolytic activation. PDGF-A and PDGF-C selectively activate PDGFR-α, whereas PDGF-B activates both PDGFR-α and PDGFR-β. Here we identify and characterize a new member of the PDGF family, PDGF D, which also requires proteolytic activation. Recombinant, purified PDGF-D induces DNA synthesis and growth in cells expressing PDGFRs. In cells expressing individual PDGFRs, PDGF-D binds to and activates PDGFR-β but not PDGFR-α. However, in cells expressing both PDGFRs, PDGF-D activates both receptors. This indicates that PDGFR-α activation may result from PDGFR-α/β heterodimerization.

Abstract
Background
The frequency of asymptomatic severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infections is unclear and may be influenced by how symptoms are evaluated. In this ...study, we sought to determine the frequency of asymptomatic SARS-CoV-2 infections in a prospective cohort of health care workers (HCWs).
Methods
A prospective cohort of HCWs, confirmed negative for SARS-CoV-2 exposure upon enrollment, were evaluated for SARS-CoV-2 infection by monthly analysis of SARS-CoV-2 antibodies as well as referral for polymerase chain reaction testing whenever they exhibited symptoms of coronavirus disease 2019 (COVID-19). Participants completed the standardized and validated FLU-PRO Plus symptom questionnaire scoring viral respiratory disease symptom intensity and frequency at least twice monthly during baseline periods of health and each day they had any symptoms that were different from their baseline.
Results
Two hundred sixty-three participants were enrolled between August 25 and December 31, 2020. Through February 28, 2021, 12 participants were diagnosed with SARS-CoV-2 infection. Symptom analysis demonstrated that all 12 had at least mild symptoms of COVID-19, compared with baseline health, near or at time of infection.
Conclusions
These results suggest that asymptomatic SARS-CoV-2 infection in unvaccinated, immunocompetent adults is less common than previously reported. While infectious inoculum doses and patient factors may have played a role in the clinical manifestations of SARS-CoV-2 infections in this cohort, we suspect that the high rate of symptomatic disease was due primarily to participant attentiveness to symptoms and collection of symptoms in a standardized, prospective fashion. These results have implications for studies that estimate SARS-CoV-2 infection prevalence and for public health measures to control the spread of this virus.
We report an improved procedure for purification of the ω subunit of
Escherichia coli RNA polymerase. In contrast to the original procedure, the revised procedure: (i) allows purification of ω ...entirely from the soluble fraction, obviating the need for denaturation/renaturation, (ii) results in >99% pure ω in only two chromatographic steps, and (iii) improves the yield of purified ω by at least 5-fold. E. coli RNAP from ω purified by this procedure, as well as purified sigma and core RNAP lacking ω, produces active holoenzyme
in vitro, and co-overexpression of ω from a plasmid containing
rpoZ and an additional plasmid encoding the other RNAP core subunits results in production of active core enzyme
in vivo.
We sought to determine pre-infection correlates of protection against SARS-CoV-2 post-vaccine inzfections (PVI) acquired during the first Omicron wave in the United States.
Serum and saliva samples ...from 176 vaccinated adults were collected from October to December of 2021, immediately before the Omicron wave, and assessed for SARS-CoV-2 Spike-specific IgG and IgA binding antibodies (bAb). Sera were also assessed for bAb using commercial assays, and for neutralization activity against several SARS-CoV-2 variants. PVI duration and severity, as well as risk and precautionary behaviors, were assessed by questionnaires.
Serum anti-Spike IgG levels assessed by research assay, neutralization titers against Omicron subvariants, and low home risk scores correlated with protection against PVIs after multivariable regression analysis. Commercial assays did not perform as well as research assay, likely due to their lower dynamic range.
In the 32 participants that developed PVI, anti-Spike IgG bAbs correlated with lower disease severity and shorter duration of illness.
Parkinson's disease is the second most common neurodegenerative disorder after Alzheimer's disease. Most cases are sporadic, however familial cases do exist. We examined 12 families with familial ...Parkinson's disease ascertained at the Movement Disorder clinic at the Oregon Health Sciences University for genetic linkage to a number of candidate loci. These loci have been implicated in familial Parkinson's disease or in syndromes with a clinical presentation that overlaps with parkinsonism, as well as potentially in the pathogenesis of the disease.
The examined loci were PARK3, Parkin, DRD (dopa-responsive dystonia), FET1 (familial essential tremor), BDNF (brain-derived neurotrophic factor), GDNF (glial cell line-derived neurotrophic factor), Ret, DAT1 (the dopamine transporter), Nurr1 and Synphilin-1. Linkage to the alpha-synuclein gene and the Frontotemporal dementia with parkinsonism locus on chromosome 17 had previously been excluded in the families included in this study. Using Fastlink, Genehunter and Simwalk both parametric and model-free non-parametric linkage analyses were performed.
In the multipoint parametric linkage analysis lod scores were below -2 for all loci except FET1 and Synphilin-1 under an autosomal dominant model with incomplete penetrance. Using non-parametric linkage analysis there was no evidence for linkage, although linkage could not be excluded. A few families showed positive parametric and non-parametric lod scores indicating possible genetic heterogeneity between families, although these scores did not reach any degree of statistical significance.
We conclude that in these families there was no evidence for linkage to any of the loci tested, although we were unable to exclude linkage with both parametric and non-parametric methods.
