SCUBA-2 is a 10 000-bolometer submillimetre camera on the James Clerk Maxwell Telescope. The instrument commissioning was completed in 2011 September, and full science operations began in 2011 ...October. To harness the full potential of this powerful new astronomical tool, the instrument calibration must be accurate and well understood. To this end, the algorithms for calculating the line-of-sight opacity have been improved, and the derived atmospheric extinction relationships at both wavebands of the SCUBA-2 instrument are presented. The results from over 500 primary and secondary calibrator observations have allowed accurate determination of the flux conversion factors (FCF) for the 850 and 450 μm arrays. Descriptions of the instrument beam shape and photometry methods are presented. The calibration factors are well determined, with relative calibration accuracy better than 5 per cent at 850 μm and 10 per cent at 450 μm, reflecting the success of the derived opacity relations as well as the stability of the performance of the instrument over several months. The sample size of the calibration observations and accurate FCFs have allowed the determination of the 850 and 450 μm fluxes of several well-known submillimetre sources, and these results are compared with previous measurements from SCUBA.

Abstract
We present the results of the first, deep Atacama Large Millimeter Array (ALMA) imaging covering the full ≃4.5 arcmin2 of the Hubble Ultra Deep Field (HUDF) imaged with Wide Field Camera ...3/IR on HST. Using a 45-pointing mosaic, we have obtained a homogeneous 1.3-mm image reaching σ1.3 ≃ 35 μJy, at a resolution of ≃0.7 arcsec. From an initial list of ≃50 > 3.5σ peaks, a rigorous analysis confirms 16 sources with S
1.3 > 120 μJy. All of these have secure galaxy counterparts with robust redshifts (〈z〉 = 2.15). Due to the unparalleled supporting data, the physical properties of the ALMA sources are well constrained, including their stellar masses (M
*) and UV+FIR star formation rates (SFR). Our results show that stellar mass is the best predictor of SFR in the high-redshift Universe; indeed at z ≥ 2 our ALMA sample contains seven of the nine galaxies in the HUDF with M
* ≥ 2 × 1010 M⊙, and we detect only one galaxy at z > 3.5, reflecting the rapid drop-off of high-mass galaxies with increasing redshift. The detections, coupled with stacking, allow us to probe the redshift/mass distribution of the 1.3-mm background down to S
1.3 ≃ 10 μJy. We find strong evidence for a steep star-forming ‘main sequence’ at z ≃ 2, with SFR ∝M
* and a mean specific SFR ≃ 2.2 Gyr−1. Moreover, we find that ≃85 per cent of total star formation at z ≃ 2 is enshrouded in dust, with ≃65 per cent of all star formation at this epoch occurring in high-mass galaxies (M
* > 2 × 1010 M⊙), for which the average obscured:unobscured SF ratio is ≃200. Finally, we revisit the cosmic evolution of SFR density; we find this peaks at z ≃ 2.5, and that the star-forming Universe transits from primarily unobscured to primarily obscured at z ≃ 4.
Riboswitches are structured non-coding RNAs often located upstream of essential genes in bacterial messenger RNAs. Such RNAs regulate expression of downstream genes by recognizing a specific cellular ...effector. Although nearly 50 riboswitch classes are known, only a handful recognize multiple effectors. Here, we report the 2.60-Å resolution co-crystal structure of a class I type I preQ
-sensing riboswitch that reveals two effectors stacked atop one another in a single binding pocket. These effectors bind with positive cooperativity in vitro and both molecules are necessary for gene regulation in bacterial cells. Stacked effector recognition appears to be a hallmark of the largest subgroup of preQ
riboswitches, including those from pathogens such as Neisseria gonorrhoeae. We postulate that binding to stacked effectors arose in the RNA World to closely position two substrates for RNA-mediated catalysis. These findings expand known effector recognition capabilities of riboswitches and have implications for antimicrobial development.
The essential pre-mRNA splicing factor U2AF2 (also called U2AF65) identifies polypyrimidine (Py) tract signals of nascent transcripts, despite length and sequence variations. Previous studies have ...shown that the U2AF2 RNA recognition motifs (RRM1 and RRM2) preferentially bind uridine-rich RNAs. Nonetheless, the specificity of the RRM1/RRM2 interface for the central Py tract nucleotide has yet to be investigated. We addressed this question by determining crystal structures of U2AF2 bound to a cytidine, guanosine, or adenosine at the central position of the Py tract, and compared U2AF2-bound uridine structures. Local movements of the RNA site accommodated the different nucleotides, whereas the polypeptide backbone remained similar among the structures. Accordingly, molecular dynamics simulations revealed flexible conformations of the central, U2AF2-bound nucleotide. The RNA binding affinities and splicing efficiencies of structure-guided mutants demonstrated that U2AF2 tolerates nucleotide substitutions at the central position of the Py tract. Moreover, enhanced UV-crosslinking and immunoprecipitation of endogenous U2AF2 in human erythroleukemia cells showed uridine-sensitive binding sites, with lower sequence conservation at the central nucleotide positions of otherwise uridine-rich, U2AF2-bound splice sites. Altogether, these results highlight the importance of RNA flexibility for protein recognition and take a step towards relating splice site motifs to pre-mRNA splicing efficiencies.
RNA recognition by proteins is central to biology. Here we demonstrate the existence of a recurrent structural motif, the “arginine fork”, that codifies arginine readout of cognate backbone and ...guanine nucleobase interactions in a variety of protein–RNA complexes derived from viruses, metabolic enzymes, and ribosomes. Nearly 30 years ago, a theoretical arginine fork model was posited to account for the specificity between the HIV-1 Tat protein and TAR RNA. This model predicted that a single arginine should form four complementary contacts with nearby phosphates, yielding a two-pronged backbone readout. Recent high-resolution structures of TAR–protein complexes have unveiled new details, including (i) arginine interactions with the phosphate backbone and the major-groove edge of guanine and (ii) simultaneous cation−π contacts between the guanidinium group and flanking nucleobases. These findings prompted us to search for arginine forks within experimental protein–RNA structures retrieved from the Protein Data Bank. The results revealed four distinct classes of arginine forks that we have defined using a rigorous but flexible nomenclature. Examples are presented in the context of ribosomal and nonribosomal interfaces with analysis of arginine dihedral angles and structural (suite) classification of RNA targets. When arginine fork chemical recognition principles were applied to existing structures with unusual arginine–guanine recognition, we found that the arginine fork geometry was more consistent with the experimental data, suggesting the utility of fork classifications to improve structural models. Software to analyze arginine–RNA interactions has been made available to the community.
The discovery in the mid-1970s that occupational exposures to pesticides could diminish or destroy the fertility of workers sparked concern about the effects of hazardous substances on male ...reproductive health. More recently, there is evidence that sperm quantity and quality may have declined worldwide, that the incidence of testicular cancer has progressively increased in many countries, and that other disorders of the male reproductive tract such as hypospadias and cryptorchidism may have also increased. There is growing concern that occupational factors and environmental chemical exposures, including in utero and childhood exposures to compounds with estrogenic activity, may be correlated with these observed changes in male reproductive health and fertility. We review the evidence and methodologies that have contributed to our current understanding of environmental effects on male reproductive health and fertility and discuss the methodologic issues which confront investigators in this area. One of the greatest challenges confronting researchers in this area is assessing and comparing results from existing studies. We elaborate recommendations for future research. Researchers in the field of male reproductive health should continue working to prioritize hazardous substances; elucidate the magnitude of male reproductive health effects, particularly in the areas of testicular cancer, hypospadias, and cryptorchidism; develop biomarkers of exposure to reproductive toxins and of reproductive health effects for research and clinical use; foster collaborative interdisciplinary research; and recognize the importance of standardized laboratory methods and sample archiving.

The Balloon-borne Large Aperture Submillimeter Telescope (BLAST) is a suborbital surveying experiment designed to study the evolutionary history and processes of star formation in local galaxies ...(including the Milky Way) and galaxies at cosmological distances. The BLAST continuum camera, which consists of 270 detectors distributed between three arrays, observes simultaneously in broadband (30%) spectral windows at 250, 350, and 500 mum. The optical design is based on a 2 m diameter telescope, providing a diffraction-limited resolution of 30 super(image ) at 250 mum. The gondola pointing system enables raster mapping of arbitrary geometry, with a repeatable positional accuracy of image30 super(image ); postflight pointing reconstruction to image5 super(image ) rms is achieved. The onboard telescope control software permits autonomous execution of a preselected set of maps, with the option of manual override. In this paper we describe the primary characteristics and measured in-flight performance of BLAST. BLAST performed a test flight in 2003 and has since made two scientifically productive long- duration balloon flights: a 100 hr flight from ESRANGE (Kiruna), Sweden to Victoria Island, northern Canada in 2005 June; and a 250 hr, circumpolar flight from McMurdo Station, Antarctica, in 2006 December.
We present a catalogue of ~3000 submillimetre sources detected ( greater than or equal to 3.5...) at 850 ...m over ~5 deg super( 2) surveyed as part of the James Clerk Maxwell Telescope (JCMT) ...SCUBA-2 Cosmology Legacy Survey (S2CLS). This is the largest survey of its kind at 850 ...m, increasing the sample size of 850 ...m selected submillimetre galaxies by an order of magnitude. The wide 850 ...m survey component of S2CLS covers the extragalactic fields: UKIDSS-UDS, COSMOS, Akari-NEP, Extended Groth Strip, Lockman Hole North, SSA22 and GOODS-North. The average 1... depth of S2CLS is 1.2 mJy beam super( -1), approaching the SCUBA-2 850 ...m confusion limit, which we determine to be ... 0.8 mJy beam super( -1). We measure the 850 ...m number counts, reducing the Poisson errors on the differential counts to approximately 4 per cent at S sub( 850) ... 3 mJy. With several independent fields, we investigate field-to-field variance, finding that the number counts on 0.5...-1... scales are generally within 50 per cent of the S2CLS mean for S sub( 850) > 3 mJy, with scatter consistent with the Poisson and estimated cosmic variance uncertainties, although there is a marginal (2...) density enhancement in GOODS-North. The observed counts are in reasonable agreement with recent phenomenological and semi-analytic models, although determining the shape of the faint-end slope (S sub( 850) < 3 mJy) remains a key test. The large solid angle of S2CLS allows us to measure the bright-end counts: at S sub( 850) > 10 mJy there are approximately 10 sources per square degree, and we detect the distinctive up-turn in the number counts indicative of the detection of local sources of 850 ...m emission, and strongly lensed high-redshift galaxies.
