BACKGROUND AND AIMS: A major problem in all restoration and rehabilitation projects is to restore the function of the ecosystem. Nitrogen, next to water, is the most limiting factor for productivity ...of arid terrestrial ecosystems. METHODS: We used a successful restored area, completed 10 years earlier, in comparison with an undisturbed scrubland area and a remaining, disturbed area in the southern Sonoran Desert in Baja California, Mexico. We compared the abundance of the nifH gene, estimated by qPCR, potential N₂ fixation activity by acetylene reduction assay, and diversity of diazotrophs by denaturing gradient gel electrophoresis in the rhizosphere of the most representative plant species, the cardon cactus and the mesquite used for restoration. RESULTS: The abundance of N₂-fixing bacteria in the rhizosphere of cardon growing with mesquite had significantly higher abundance of nifH gene than the rhizosphere of cardon that grew separately. Across all samples, the potential N₂ fixation was significantly higher in soil samples from the restored site than samples from the undisturbed and disturbed sites. CONCLUSIONS: Successful long-term restoration improved the potential N₂ fixation to a level similar to undisturbed lands. Beneficial interactions between cardon and mesquite are a promising venue for desert reforestation by their contribution to improve N₂ fixing potential in degraded arid lands and increasing the population of diazotrophs.
The RNA subunit of telomerase is an essential component whose primary sequence and length are poorly conserved among eukaryotic organisms. The phytopathogen Ustilago maydis is a dimorphic fungus of ...the order Ustilaginales. We analyzed several species of Ustilaginales to computationally identify the TElomere RNA (TER) gene ter1. To confirm the identity of the TER gene, we disrupted the gene and characterized telomerase-negative mutants. Similar to catalytic TERT mutants, ter1Δ mutants exhibit phenotypes of growth delay, telomere shortening and low replicative potential. ter1-disrupted mutants were unable to infect maize seedlings in heterozygous crosses and showed defects such as cell cycle arrest and segregation failure. We concluded that ter1, which encodes the TER subunit of the telomerase of U. maydis, have similar and perhaps more extensive functions than trt1.

Development of highly sensitive molecular sensors is of immense current interest for biomedical diagnosis. Although the possibility of single molecule detection through plasmonic sensors based on ...surface enhanced Raman scattering (SERS) has been demonstrated, the practical application of such devices is hindered due their complex fabrication strategies and inhomogeneous Raman signal response. Here we present a simple biological method for large scale fabrication of plasmonic microtubes using selective nonpathogenic fungi as living templates. The biologically fabricated microstructures can be used as SERS substrates for the detection of organic molecules with signal enhancement factors as high as 1 × 1010, exploiting their rugged, highly porous surfaces with intense hot spots. Although they are structurally not as perfect as those of chemically or physically fabricated plasmonic SERS substrates, their morphological/geometrical inhomogeneity introduces only about a one order fluctuation of SERS enhancement factor along their lengths. The plasmonic microstructures have great potential for the fabrication of cheap, efficient, and highly sensitive gold-based biosensors for the detection and monitoring of organic molecules.
Zea mays L. ssp. mexicana (teosinte) is a naturally occurring grass related to maize. The two plants have developed foliar fungal diseases that can be controlled with beneficial bacterial ...antagonists. While the beneficial effects on stem and root development of the application of bacteria of the genus Azospirillum is widely known, the effects of the bacteria on the control of the phytopathogenic fungi associated with teosinte leaves and seeds are unknown. Bacterium of the species A. brasilense that present acetylene reducing activity, siderophore production and the ability to antagonise pathogenic fungi in vitro and in vivo in teosinte plants were selected for this study, in which the incidence of fungal diseases caused by Alternaria, Bipolaris and Fusarium were reduced in plants. Furthermore, biomass (root and stem) production increased, improving teosinte plant health in greenhouse and field conditions.
Se evaluó el desarrollo y producción de la col (Brassica oleracea var. Royal vantage) en invernadero, bajo diferentes presiones osmóticas de la solución nutritiva Steiner y biofertlizada con ...consorcios bacterianos. Se estableció un diseño factorial 4 × 3, donde los factores fueron cuatro niveles de presión osmótica (0.18, 0.36, 0.54 atm y un testigo sin solución nutritiva), y dos consorcios bacterianos: Azospirillum brasiliense + Acinetobacter calcoaceticus (AA) y Raoultella terrigena + Chromobacterium violaceum (RC), y un testigo sin inocular. Se establecieron seis repeticiones por cada tratamiento. Las semillas inoculadas con el consorcio AA presentaron el mayor porcentaje de emergencia (100%), así como la tasa de emergencia más alta (20.5), en comparación con consorcio RC y las plantas no inoculadas. La inoculación con el consorcio AA y la presión osmótica de 0.54 atm produjeron plantas más altas (60-65%), y un tallo con mayor diámetro (35-46%). El peso de la biomasa fresca y la acumulación de materia seca se duplicaron en las plantas inoculadas con respecto a las no inoculadas. Se observó mayor concentración de K en plantas inoculadas con AA y RC (20-35% y 25-45%, respectivamente) respecto a las plantas no inoculadas. La concentración de nitratos en las plantas se incrementó entre 200-350% y 60-230% con la inoculación de los consorcios AA y RC, respectivamente. La concentración de P se incrementó entre 200-500% con el consorcio AA y entre 100-400% con el consorcio RC. La inoculación con los consorcios AA y RC, a presión osmótica de 0.54 atm, indujo el mayor desarrollo de las cabezas de la col (clasif icadas como 5 y 4, respectivamente). Se concluye que la col, bajo fertilización mineral moderada en invernadero, responde favorablemente a la inoculación de bacterias promotoras de crecimiento vegetal.

Si se transfiere el modelo a los milperos menos eficientes, se puede mejorar la autosuficiencia alimentaria.
The diseases found in Pinus patula, a species present in the forest area of Puebla, Mexico cause foliage loss and damage to trees and we consider that fungi could be involved. The objective of the ...current investigation was to study the nutrients of the soil and the fungi associated with the lesions on the needles of Pinus patula in Tetela de Ocampo, Puebla, Mexico. It was found that the nitrogen, phosphorus and potassium content of the soils associated with pine, as well as the organic matter, were poor. In addition, we performed chemical, physical and biological analyses on soils from the study site. We isolated forty-five fungi from the lesions on the needles. The identification of twelve of them was both morphological and molecular by sequence analysis of the ITS1-ITS2 gene. The identified fungi included Alternaria alternata, Annulohypoxylon stygium, Botryosphaeria dothidea, Curvularia lunata, Curvularia pseudobrachyspora, Curvularia spicifera, Curvularia trifolii, Daldinia eschscholtzii, Daldinia sp. Diplodia sapinea, Lophodermium indianum and Myrmaecium rubricosum. We concluded that P. patula is affected by nutritional deficiency as well as by the presence of phytopathogenic fungi.
The chitinase enzyme was identified in isolated bacteria of maize rhizosphere as well as its potential for the biological control of fungi associated at seeds of the same plant. The production of ...chitinase enzyme was found in the genera identified as Acinetobacter, Bacterium, Burkholderia, Paenibacillus, Pseudomonas, Rhizobium, Shewanella, Sphingomonas and Stenotrophomonas. Bacterial isolates with ability to degrade fungal mycelium from maize fungi as Fusarium and Alternaria among others, were detected. Bacterial chitinase activity and the presence of the chiA gene were determined. The inoculation of chitinolytic bacteria showed a positive effect in the control of fungi in maize seeds. The results support the potential use of chitinase enzyme producing bacteria on the control of phytopathogenic fungi.
El trabajo se realizó con el objetivo de evaluar el efecto de dos biofertilizantes en la producción de maíz, solos y combinados con dos fórmulas de fertilización química. El experimento se realizó ...bajo un diseño experimental de bloques al azar, con nueve tratamientos y cuatro repeticiones, conformando un total de 36 unidades experimentales. Los biofertilizantes evaluados fueron las especies Azospirillum brasilense y Chromobactherium violaceum, y las fórmulas de fertilización química 80-23-15 y 160-46-30 de N, P y K. El mejor rendimiento de grano de maíz se logró con el tratamiento Azospirillum brasilense + 160-46-30 de N-P-K, con 5.97 t ha-1, en segundo lugar Chromobacterium violaceum + 80-23-15 de N-P-K con 5.87 t ha-1. El menor rendimiento de 4.30 t ha-¹ se tuvo con el Testigo absoluto. Los resultados obtenidos muestran que cuando se utiliza biofertilizantes existe un incremento en el rendimiento de maíz por efecto de la aplicación de microorganismos benéficos. El análisis económico mostró que los mayores beneficios netos se tuvieron con los tratamientos Azospirillum brasilense + Chromobacterium violaceum + fertilización química 160-46-30 y Azospirillum brasilense + fertilización quimica 160-46-30. Se concluye que el uso del biofertilizante Azospirillum brasilense representó un incremento en el rendimiento de grano de 27.98% (1.67 t ha-1) con respecto al testigo absoluto, lográndose un mayor beneficio neto.
Burkholderia es un género ambivalente debido a que algunas de sus especies establecen relaciones simbiótico-mutualistas con las plantas, y simbiótico-patogénicas con plantas, animales y humanos. ...Desde que la bacteria fitopatógena B. cepacia fue reportada como oportunista nosocomial, asociada a la fibrosis quística, surgió la preocupación de posibles infecciones en humanos. El objetivo de esta contribución fue hacer un análisis de la versatilidad funcional de Burkholderia y de su efecto en la salud humana. Burkholderia albergó cerca de 100 especies y al complejo B. cepacia (CBC) constituido por 22 especies. Inicialmente, se determinó la existencia de dos linajes dentro del género: el A que incluía varias especies que se asociaban con plantas, así como las saprofitas; y el B que contenía las especies del CBC (oportunistas patógenas de humanos), el subgrupo de B. pseudomallei que incluía patógenos de humanos y animales, y un grupo de especies fitopatógenas. Finalmente, se renombraron algunos individuos como Paraburkholderia y Caballeronia. Los análisis recientes de burkholderias provenientes de humanos y del ambiente, indican que no existe una subdivisión filogenética que distinga entre benéficas y patogénicas. De ahí la importancia de considerar los riesgos para la salud humana, cuando algún miembro de este grupo sea empleado en actividades agrícolas.
