The purpose of this study was to determine the efficacy of stent-based delivery of sirolimus (SRL) alone or in combination with dexamethasone (DEX) to reduce in-stent neointimal hyperplasia. SRL is a ...potent immunosuppressive agent that inhibits SMC proliferation by blocking cell cycle progression.
Stents were coated with a nonerodable polymer containing 185 microgram SRL, 350 microgram DEX, or 185 microgram SRL and 350 microgram DEX. Polymer biocompatibility studies in the porcine and canine models showed acceptable tissue response at 60 days. Forty-seven stents (metal, n=13; SRL, n=13; DEX, n=13; SRL and DEX, n=8) were implanted in the coronary arteries of 16 pigs. The tissue level of SRL was 97+/-13 ng/artery, with a stent content of 71+/-10 microgram at 3 days. At 7 days, proliferating cell nuclear antigen and retinoblastoma protein expression were reduced 60% and 50%, respectively, by the SRL stents. After 28 days, the mean neointimal area was 2.47+/-1.04 mm(2) for the SRL alone and 2.42+/-1.04 mm(2) for the combination of SRL and DEX compared with the metal (5.06+/-1.88 mm(2), P<0.0001) or DEX-coated stents (4.31+/-3.21 mm(2), P<0.001), resulting in a 50% reduction of percent in-stent stenosis.
Stent-based delivery of SRL via a nonerodable polymer matrix is feasible and effectively reduces in-stent neointimal hyperplasia by inhibiting cellular proliferation.
Spectroscopic and spectropolarimetric observations of the pre-main sequence early-G star HD 141943 were obtained at four observing epochs (in 2006, 2007, 2009 and 2010). The observations were ...undertaken at the 3.9-m Anglo-Australian Telescope using the UCLES echelle spectrograph and the SEMPOL spectropolarimeter visitor instrument. Brightness and surface magnetic field topologies were reconstructed for the star using the technique of least-squares deconvolution to increase the signal-to-noise ratio of the data.
The reconstructed brightness maps show that HD 141943 had a weak polar spot and a significant amount of low-latitude features, with little change in the latitude distribution of the spots over the 4 yr of observations. The surface magnetic field was reconstructed at three of the epochs from a high-order (l≤ 30) spherical harmonic expansion of the spectropolarimetric observations. The reconstructed magnetic topologies show that in 2007 and 2010 the surface magnetic field was reasonably balanced between poloidal and toroidal components. However, we find tentative evidence of a change in the poloidal/toroidal ratio in 2009 with the poloidal component becoming more dominant. At all epochs the radial magnetic field is predominantly non-axisymmetric while the azimuthal field is predominantly axisymmetric with a ring of positive azimuthal field around the pole similar to that seen on other active stars.
Hyperacute rejection of porcine organs by old world primate recipients is mediated through preformed antibodies against galactosyl-α-1,3-galactose (Galα-1,3-Gal) epitopes expressed on the pig cell ...surface. Previously, we generated inbred miniature swine with a null allele of the α-1,3-galactosyltransferase locus (GGTA1) by nuclear transfer (NT) with gene-targeted fibroblasts. To expedite the generation of GGTA1 null pigs, we selected spontaneous null mutant cells from fibroblast cultures of heterozygous animals for use in another round of NT. An unexpectedly high rate of spontaneous loss of GGTA1 function was observed, with the vast majority of null cells resulting from loss of the WT allele. Healthy piglets, hemizygous and homozygous for the gene-targeted allele, were produced by NT by using fibroblasts that had undergone deletional and crossover/gene conversion events, respectively. Aside from loss of Galα-1,3-Gal epitopes, there were no obvious phenotypic differences between these null piglets and WT piglets from the same inbred lines. In fact, congenital abnormalities observed in the heterozygous NT animals did not reappear in the serially produced null animals.

ABSTRACT
We present a spectropolarimetric magnetic snapshot survey of 55 stars which includes 53 F-type stars ranging from spectral types F0 to F9 plus 2 chemically peculiar stars β CrB, and δ Cap. ...We look for magnetic fields in stars spanning a range of effective temperatures where the transition from fossil to dynamo magnetic fields is believed to occur. High-resolution spectropolarimetry using circularly polarized spectra is used to look for a magnetic detection in the Stokes V profile, determine the mean longitudinal magnetic field (Bl), and to look for correlations with stellar parameters. Surface magnetic fields are detected on 14 F-stars, and present in every spectral class from F3V-F9V ranging in strength from 0.3 ± 0.1 G (36 UMa, F8V) to 8.3 ± 0.9 G (h Dra, F8V). Thus we find photospheric magnetic fields are present in stars as early as spectral type F3V with an outer convection zone thickness less than a few per cent of the stellar radius.

We report the detection of sixteen binary systems from the Anglo-Australian Planet Search. Solutions to the radial velocity data indicate that the stars have companions orbiting with a wide range of ...masses, eccentricities and periods. Three of the systems potentially contain brown-dwarf companions while another two have eccentricities that place them in the extreme upper tail of the eccentricity distribution for binaries with periods less than 1000 d. For periods up to 12 years, the distribution of our stellar companion masses is fairly flat, mirroring that seen in other radial velocity surveys, and contrasts sharply with the current distribution of candidate planetary masses, which rises strongly below 10 M
J. When looking at a larger sample of binaries that have FGK star primaries as a function of the primary star metallicity, we find that the distribution maintains a binary fraction of ∼43 ± 4 per cent between −1.0 and +0.6 dex in metallicity. This is in stark contrast to the giant exoplanet distribution. This result is in good agreement with binary formation models that invoke fragmentation of a collapsing giant molecular cloud, suggesting that this is the dominant formation mechanism for close binaries and not fragmentation of the primary star's remnant protoplanetary disc.
We report the discovery of two long-period giant planets from the Anglo-Australian Planet Search. HD 154857c is in a multiple-planet system, while HD 114613b appears to be solitary. HD 114613b has an ...orbital period P = 10.5 yr, and a minimum mass m sin i of 0.48 M sub(Jup); HD 154857c has P = 9.5 yr and m sin i = 2.6 M sub(Jup). These new data confirm the planetary nature of the previously unconstrained long-period object in the HD 154857 system.We have performed detailed dynamical stability simulations which show that the HD 154857 two-planet system is stable on timescales of at least 10 super(8) yr. These results highlight the continued importance of "legacy" surveys with long observational baselines; these ongoing campaigns are critical for determining the population of Jupiter analogs, and hence of those planetary systems with architectures most like our own solar system.
We present a detailed analysis of high-resolution spectropolarimetric observations of the weak-line T Tauri stars (wTTSs) TWA 9A and V1095 Sco as part of a wider survey of magnetic properties and ...activity in weak-line T Tauri stars, called MaTYSSE (Magnetic Topologies of Young Stars and the Survival of close-in giant Exoplanets). Our targets have similar masses but differing ages which span the stage of radiative core formation in solar-mass stars. We use the intensity line profiles to reconstruct surface brightness maps for both stars. The reconstructed brightness maps for TWA 9A and V1095 Sco were used to model and subtract the activity-induced jitter, reducing the RMS in the radial velocity measurements of TWA 9A by a factor of ˜7, and for V1095 Sco by a factor of ˜3. We detect significant circular polarization for both stars, but only acquired a high-quality circular polarisation time-series for TWA 9A. Our reconstructed large-scale magnetic field distribution of TWA 9A indicates a strong, non-axisymmetric field. We also analyse the chromospheric activity of both stars by investigating their H α emission, finding excess blue-ward emission for most observations of V1095 Sco, and symmetric, double-peaked emission for TWA 9A, with enhanced emission at one epoch likely indicating a flaring event.
In this paper we present chromospheric emission levels of the solar-type stars in the young open clusters IC 2391 and IC 2602. High-resolution spectroscopic data were obtained for over 50 F, G and K ...stars from these clusters over several observing campaigns using the University College London Echelle Spectrograph on the 3.9-m Anglo-Australian Telescope. Unlike older clusters, the majority (28/52) of the solar-type stars in the two clusters are rapid rotators (v sin i > 20 km s−1) with five of the stars being classified as ultra-rapid rotators (v sin i > 100 km s−1). The emission levels in the calcium infrared triplet lines were then used as a measure of the chromospheric activity of the stars. When plotted against the Rossby number (NR), the star's chromospheric emission levels show a plateau in the emission for log(NR) ≲−1.1 indicating chromospheric saturation similar to the coronal saturation seen in previously observed X-ray emission from the same stars. However, unlike the coronal emission, the chromospheric emission of the stars shows little evidence of a reduction in emission (i.e. supersaturation) for the ultra-rapid rotators in the clusters. Thus we believe that coronal supersaturation is not the result of an overall decrease in magnetic dynamo efficiency for ultra-rapid rotators.
