Context. Be stars are fast-rotating early-type emission line stars. It is generally assumed that observed emission is generated in a rotating disk-like envelope, as supported by the observed ...correlation between the stellar projected rotational velocity vsini and the width of the emission lines. Then, high-resolution spectroscopic observations of Balmer lines profiles play an important role in putting constraints on Be stars modeling. Aims. We present Balmer lines spectroscopy for a sample of 48 Be stars. For most of them, Hα and Hβ have been observed more than two times, in a total period spanning almost two years between 2008 and 2009. Methods. Spectral synthesis of the Hα profile was performed following two steps: photospheric contribution was computed by using Kurucz’s code ATLAS9 and SYNTHE, and disk emission was derived by the approach of Hummel & Vrancken (2000, A&A, 302, 751). Results. For 26 out of 48 stars, a modeling of the total Hα emission, i.e. photospheric absorption plus disk net emission, has been attempted. By this modeling we derived an estimation of the disk radius, as well as the inclination angle between the rotational axis with line of sight and the base density at the stellar equator. For the stars observed more than once, we also discuss the variability of Hα and Hβ for what concerns both the equivalent width and the spectral profile. We found 16 stars with variable equivalent width and 7 stars with clear signs of profile variations. Conclusions. For all the stars in our sample, we derive all the fundamental astrophysical quantities, such as, effective temperature, gravity, and projected rotational velocity. We found 13 stars whose equivalent width is variable with a confidence level greater than 80% and 7 object for which spectral profiles show change with time. According to the commonly used classification scheme, we classified 16 stars as belonging to class 1, 13 to class 2, 11 are shell stars, 6 objects do not show net emission, and 2 stars display transitions from class 1 and 2. For the class 1 stars, we confirm the correlation between vsini and peak separation. Concerning the geometry of the disk, we derived the base density at the stellar equator, the radius, and the inclination angle between rotational axis and line of sight. The maximum concentration of stars occurs for disk dimensions ranging in the interval of 6 to 8 stellar radii and for inclination angles going from 23° to 35°.
Rapid molecular diagnostics for detecting multidrug-resistant and extensively drug-resistant tuberculosis (M/XDR-TB) primarily identify mutations in Mycobacterium tuberculosis (Mtb) genes associated ...with drug resistance. Their accuracy, however, is dependent largely on the strength of the association between a specific mutation and the phenotypic resistance of the isolate with that mutation, which is not always 100%. While this relationship is well established and reliable for first-line anti-TB drugs, rifampin and isoniazid, it is less well-studied and understood for second-line, injectable drugs, amikacin (AMK), kanamycin (KAN) and capreomycin (CAP).
We conducted a systematic review of all published studies evaluating Mtb mutations associated with resistance to AMK, KAN, CAP in order to characterize the diversity and frequency of mutations as well as describe the strength of the association between specific mutations and phenotypic resistance in global populations. Our objective was to determine the potential utility and reliability of these mutations as diagnostic markers for detecting AMK, KAN and CAP resistance. Mutation data was reviewed for 1,585 unique clinical isolates from four continents and over 18 countries. Mutations in the rrs, tlyA, eis promoter and gidB genes were associated with AMK, KAN and/or CAP resistance.
The rrs A1401G mutation was present in the majority of AMK, KAN and CAP resistant Mtb strains reviewed, but was also found in 7% of CAP susceptible strains. The 1401 mutation alone, however, was not found with sufficient frequency to detect more than 70-80% of global Mtb strains resistant to AMK and CAP, and 60% of strains resistant to KAN. Additional mutations in the rrs, eis promoter, tlyA and gidB genes appear to be associated with resistance and could improve sensitivity and specificity of future diagnostics.

Accurate, comprehensive, and timely detection of drug-resistant tuberculosis (TB) is essential to inform patient treatment and enable public health surveillance. This is crucial for effective control ...of TB globally. Whole-genome sequencing (WGS) and targeted next-generation sequencing (NGS) approaches have potential as rapid in vitro diagnostics (IVDs), but the complexity of workflows, interpretation of results, high costs, and vulnerability of instrumentation have been barriers to broad uptake outside of reference laboratories, especially in low- and middle-income countries. A new, solid-state, tabletop sequencing instrument, Illumina iSeq100, has the potential to decentralize NGS for individual patient care.
In this study, we evaluated WGS and targeted NGS for TB on both the new iSeq100 and the widely used MiSeq (both manufactured by Illumina) and compared sequencing performance, costs, and usability. We utilized DNA libraries produced from Mycobacterium tuberculosis clinical isolates for the evaluation. We conducted WGS on three strains and observed equivalent uniform genome coverage with both platforms and found the depth of coverage obtained was consistent with the expected data output. Utilizing the standardized, cloud-based ReSeqTB bioinformatics pipeline for variant analysis, we found the two platforms to have 94.0% (CI 93.1%-94.8%) agreement, in comparison to 97.6% (CI 97%-98.1%) agreement for the same libraries on two MiSeq instruments. For the targeted NGS approach, 46 M. tuberculosis-specific amplicon libraries had 99.6% (CI 98.0%-99.9%) agreement between the iSeq100 and MiSeq data sets in drug resistance-associated SNPs. The upfront capital costs are almost 5-fold lower for the iSeq100 ($19,900 USD) platform in comparison to the MiSeq ($99,000 USD); however, because of difference in the batching capabilities, the price per sample for WGS was higher on the iSeq100. For WGS of M. tuberculosis at the minimum depth of coverage of 30x, the cost per sample on the iSeq100 was $69.44 USD versus $28.21 USD on the MiSeq, assuming a 2 × 150 bp run on a v3 kit. In terms of ease of use, the sequencing workflow of iSeq100 has been optimized to only require 27 minutes total of hands-on time pre- and post-run, and the maintenance is simplified by a single-use cartridge-based fluidic system. As these are the first sequencing attempts on the iSeq100 for M. tuberculosis, the sequencing pool loading concentration still needs optimization, which will affect sequencing error and depth of coverage. Additionally, the costs are based on current equipment and reagent costs, which are subject to change.
The iSeq100 instrument is capable of running existing TB WGS and targeted NGS library preparations with comparable accuracy to the MiSeq. The iSeq100 has reduced sequencing workflow hands-on time and is able to deliver sequencing results in <24 hours. Reduced capital and maintenance costs and lower-throughput capabilities also give the iSeq100 an advantage over MiSeq in settings of individualized care but not in high-throughput settings such as reference laboratories, where sample batching can be optimized to minimize cost at the expense of workflow complexity and time.

ABSTRACT
In this paper, we present the results of a spectroscopic campaign devoted to ascertaining the actual nature of a sample of five objects reported as chemically peculiar stars in the Catalogue ...of Ap, HgMn and Am stars (2009, A&A, 498, 961). Spectroscopic observations were obtained with Catania Astrophysical Observatory Spectropolarimeter. For each of the objects, we derive the effective temperature, gravity, rotational and radial velocities, and chemical abundances using the spectral synthesis method. All the targets were found to be SB1 systems. Our abundance analysis led us to classify HD 40788 and HD 187959 as marginal Am stars and HD 202431 as an Am star, and to confirm HD 134793 as an SrEuCr Ap star and, finally, HD 189652 as a normal star. Finally, we confirm that Am stars can possibly be used as an accurate tool to date stellar populations in galaxies.
In this paper, we present the results of a comprehensive study of six eclipsing binaries whose components are confirmed or suspected Am stars. By combining long-term, high-resolution Catania ...Astrophysical Observatory Spectropolarimeter (CAOS) spectroscopy and TESS photometry, we have been able to accurately obtain the orbital parameters of each system as well as the atmospheric parameters of its components. We performed an in-depth chemical analysis and provided chemical abundances of C, O, Na, Mg, Si, Ca, Sc, Ti, Cr, Mn, Fe, Ni, Zn, Sr, Y, Zr, and Ba. From the solution of the light and radial curves, we have determined the masses, radii, and temperatures with a good degree of accuracy. We observe apsidal motion in the eccentric system HD 216429, in which the Rossiter-McLaughlin effect is also noted. We inferred the age of our targets by fitting isochrones on the HR diagram and find that both components in each system are properly described with the same isochrone, which reinforces our results. Furthermore, dynamical and evolutionary masses, independently obtained, show an excellent agreement. According to the out-of-eclipse variability shown in their TESS light curves and their position on the Hertzsprung-Russell diagram, we claim that the stars HD 42954 (as δ Sct type) and HD 151604 ( γ Dor) have a pulsating nature. Based on the chemical analysis, we corroborate that four of the systems studied here are formed by Am stars, while in the remaining ones (HD 126031 and HD 216429) only the primary component exhibits a peculiar composition. Additionally, the age distribution found in Am stars supports their suitability as age tracers in stellar populations.
ABSTRACT
ASCC 123 is a little-studied young and dispersed open cluster. Recently, we conducted the first research devoted to it. In this paper, we complement our previous work with Transiting ...Exoplanet Survey Satellite (TESS) photometry for the 55 likely members of the cluster. We pay special attention to seven of these high-probability members, all with FGK spectral types, for which we have high-resolution spectra from our preceding work. By studying the TESS light curves of the cluster members, we determine the rotational period and the amplitude of the rotational modulation for 29 objects. The analysis of the distribution of the periods allows us to estimate a gyrochronological age for ASCC 123 similar to that of the Pleiades, confirming the value obtained in our previous investigation. A young cluster age is also suggested by the distribution of variation amplitudes. In addition, for those stars with spectroscopic data, we calculate the inclination of their rotation axis. These values appear to follow a random distribution, as already observed in young clusters, with no indication of spin alignment. However, our sample is too small to confirm this on more solid statistical grounds. Finally, for these seven stars, we study the level of magnetic activity from the Hα and Ca ii H&K lines. Despite the small number of data points, we find a correlation of the Hα and Ca ii flux with Rossby number. The position of these stars in flux–flux diagrams follows the general trends observed in other active late-type stars.
Undetected and untreated, low-levels of drug resistant (DR) subpopulations in clinical Mycobacterium tuberculosis (Mtb) infections may lead to development of DR-tuberculosis, potentially resulting in ...treatment failure. Current phenotypic DR susceptibility testing has a theoretical potential for 1% sensitivity, is not quantitative, and requires several weeks to complete. The use of "single molecule-overlapping reads" (SMOR) analysis with next generation DNA sequencing for determination of ultra-rare target alleles in complex mixtures provides increased sensitivity over standard DNA sequencing. Ligation free amplicon sequencing with SMOR analysis enables the detection of resistant allele subpopulations at ≥0.1% of the total Mtb population in near real-time analysis. We describe the method using standardized mixtures of DNA from resistant and susceptible Mtb isolates and the assay's performance for detecting ultra-rare DR subpopulations in DNA extracted directly from clinical sputum samples. SMOR analysis enables rapid near real-time detection and tracking of previously undetectable DR sub-populations in clinical samples allowing for the evaluation of the clinical relevance of low-level DR subpopulations. This will provide insights into interventions aimed at suppressing minor DR subpopulations before they become clinically significant.

ABSTRACT
In this paper, we present a detailed spectroscopic study of the SB1 system HD 161660. New spectroscopic observations have been obtained by us with Catania Astrophysical Observatory ...Spectropolarimeter (CAOS@OAC). Combining these observations with archive data from HARPS@3.6ESO, we derived atmospheric parameters as temperature and gravity (from the fit of Balmer lines), microturbulence and rotational velocity (from metal lines), and chemical composition. We found underabundances of helium, carbon, magnesium, sulphur and chromium, overabundances of neon, phosphorus, argon, manganese, xenon, and mercury. All other elements have solar composition. In particular, mercury abundance is derived taking into account an isotopic mixture different from the terrestrial one (essentially pure 202Hg). Considering this chemical pattern, we definitively confirm HD 161660 is an HgMn star. Further, variability of equivalent widths points out a non-homogeneous distribution of helium and magnesium over stellar surface. As to iron and phosphorus, we found a non-constant abundance with the optical depth, a result currently considered an evidence of vertical stratification. Finally, we improved the fundamental parameters characterizing the HD 161660 orbit.
There is still incomplete knowledge of which Mycobacterium tuberculosis (Mtb) antigens can trigger distinct T cell responses at different stages of infection. Here, a proteome-wide screen of 20,610 ...Mtb-derived peptides in 21 patients mid-treatment for active tuberculosis (ATB) reveals IFNγ-specific T cell responses against 137 unique epitopes. Of these, 16% are recognized by two or more participants and predominantly derived from cell wall and cell processes antigens. There is differential recognition of antigens, including TB vaccine candidate antigens, between ATB participants and interferon-gamma release assay (IGRA + /-) individuals. We developed an ATB-specific peptide pool (ATB116) consisting of epitopes exclusively recognized by ATB participants. This pool can distinguish patients with pulmonary ATB from IGRA + /- individuals from various geographical locations, with a sensitivity of over 60% and a specificity exceeding 80%. This proteome-wide screen of T cell reactivity identified infection stage-specific epitopes and antigens for potential use in diagnostics and measuring Mtb-specific immune responses.
ABSTRACT
Classical Cepheids (DCEPs) are the first fundamental step in the calibration of the cosmological distance ladder. Furthermore, they represent powerful tracers in the context of Galactic ...studies. We have collected high-resolution spectroscopy with UVES@VLT for a sample of 65 DCEPs. The majority of them are the faintest DCEPs ever observed in the Milky Way. For each target, we derived accurate atmospheric parameters, radial velocities, and abundances for 24 different species. The resulting iron abundances range between +0.3 and −1.1 dex with the bulk of stars at Fe/H ∼ −0.5 dex. Our sample includes the most metal-poor DCEPs observed so far with high-resolution spectroscopy. We complement our sample with literature data obtaining a complete sample of 637 DCEPs and use Gaia Early Data Release 3 (EDR3) photometry to determine the distance of the DCEPs from the period–Wesenheit–Metallicity relation. Our more external data trace the Outer arm at Galactocentric radius (RGC) ∼ 16–18 kpc which appears significantly warped. We investigate the metallicity gradient of the Galactic disc using this large sample, finding a slope of −0.060 ± 0.002 dex kpc−1, in very good agreement with previous results based both on DCEPs and open clusters. We also report a possible break in the gradient at RGC = 9.25 kpc with slopes of −0.063 ± 0.007 and −0.079 ± 0.003 dex kpc−1 for the inner and outer sample, respectively. The two slopes differ by more than 1σ. A more homogeneous and extended DCEPs sample is needed to further test the plausibility of such a break.
