Once limited to the military domain, unmanned aerial vehicles are now poised to gain widespread adoption in the commercial sector. One such application is to deploy these aircraft, also known as drones, for last-mile delivery in logistics operations. While significant research efforts are underway to improve the technology required to enable delivery by drone, less attention has been focused on the operational challenges associated with leveraging this technology. This paper provides two mathematical programming models aimed at optimal routing and scheduling of unmanned aircraft, and delivery trucks, in this new paradigm of parcel delivery. In particular, a unique variant of the classical vehicle routing problem is introduced, motivated by a scenario in which an unmanned aerial vehicle works in collaboration with a traditional delivery truck to distribute parcels. We present mixed integer linear programming formulations for two delivery-by-drone problems, along with two simple, yet effective, heuristic solution approaches to solve problems of practical size. Solutions to these problems will facilitate the adoption of unmanned aircraft for last-mile delivery. Such a delivery system is expected to provide faster receipt of customer orders at less cost to the distributor and with reduced environmental impacts. A numerical analysis demonstrates the effectiveness of the heuristics and investigates the tradeoffs between using drones with faster flight speeds versus longer endurance.
Prevalence of certain forms of psychopathology, such as autism and depression, differs between genders and understanding gender differences of the neurotypical brain may provide insights into risk ...and protective factors. In recent research, resting state functional magnetic resonance imaging (rfMRI) is widely used to map the inherent functional networks of the brain. Although previous studies have reported gender differences in rfMRI, the robustness of gender differences is not well characterized. In this study, we use a large data set to test whether rfMRI functional connectivity (FC) can be used to predict gender and identify FC features that are most predictive of gender. We utilized rfMRI data from 820 healthy controls from the Human Connectome Project. By applying a predefined functional template and partial least squares regression modeling, we achieved a gender prediction accuracy of 87% when multi‐run rfMRI was used. Permutation tests confirmed that gender prediction was reliable (
p<.001). Effects of motion, age, handedness, blood pressure, weight, and brain volume on gender prediction are discussed. Further, we found that FC features within the default mode (DMN), fronto‐parietal and sensorimotor networks contributed most to gender prediction. In the DMN, right fusiform gyrus and right ventromedial prefrontal cortex were important contributors. The above regions have been previously implicated in aspects of social functioning and this suggests potential gender differences in social cognition mediated by the DMN. Our findings demonstrate that gender can be reliably predicted using rfMRI data and highlight the importance of controlling for gender in brain imaging studies.
Colloidally suspended PbS quantum dots stabilized with 3-mercaptopropionic acid (3-MPA) were prepared via solution ligand exchange. The optical properties of the as-prepared colloidal QDs were ...investigated by absorption and emission spectroscopy. The 3-MPA exchanged QDs luminesced with intensities comparable to oleate capped PbS QDs. The nanocrystal ligand chemistry was investigated by solution 1H Nuclear Magnetic Resonance (NMR) spectroscopy, which indicated a dynamic ligand shell undergoing rapid exchange. Optical properties of the QD colloid evolved over time and were correlated to the oxidative conversion of 3-MPA into dithiodipropionic acid (DTDPA) in the presence of dimethyl sulfoxide (DMSO) observed by NMR.
Current methods to delete genomic sequences are based on clustered regularly interspaced short palindromic repeats (CRISPR)-Cas9 and pairs of single-guide RNAs (sgRNAs), but can be inefficient and ...imprecise, with errors including small indels as well as unintended large deletions and more complex rearrangements. In the present study, we describe a prime editing-based method, PRIME-Del, which induces a deletion using a pair of prime editing sgRNAs (pegRNAs) that target opposite DNA strands, programming not only the sites that are nicked but also the outcome of the repair. PRIME-Del achieves markedly higher precision than CRISPR-Cas9 and sgRNA pairs in programming deletions up to 10 kb, with 1-30% editing efficiency. PRIME-Del can also be used to couple genomic deletions with short insertions, enabling deletions with junctions that do not fall at protospacer-adjacent motif sites. Finally, extended expression of prime editing components can substantially enhance efficiency without compromising precision. We anticipate that PRIME-Del will be broadly useful for precise, flexible programming of genomic deletions, epitope tagging and, potentially, programming genomic rearrangements.
Although the different age groups had differences in sensitivity of asthma exacerbations (AEs) to environmental factors, no comprehensive study has examined the age-stratified effects of ...environmental factors on AEs.
We sought to examine the short-term effects in age-stratified groups (infants, preschool children, school-aged children, adults, and the elderly) of outdoor environmental factors (air pollutants, weather conditions, aeroallergens, and respiratory viral epidemics) on AEs.
We performed an age-stratified analysis of the short-term effects of 4 groups of outdoor environmental factors on AEs in Seoul Metropolitan City (Korea) from 2008 and 2012. The statistical analysis used a Poisson generalized linear regression model, with a distributed lag nonlinear model for identification of lagged and nonlinear effects and convergent cross-mapping for identification of causal associations.
Analysis of the total population (n = 10,233,519) indicated there were 28,824 AE events requiring admission to an emergency department during the study period. Diurnal temperature range had significant effects in pediatric (infants, preschool children, and school-aged children) and elderly (relative risk RR, 1.056-1.078 and 1.016, respectively) subjects. Tree and weed pollen, human rhinovirus, and influenza virus had significant effects in school-aged children (RR, 1.014, 1.040, 1.042, and 1.038, respectively). Tree pollen and influenza virus had significant effects in adults (RR, 1.026 and 1.044, respectively). Outdoor air pollutants (particulate matter of ≤10 μm in diameter, nitrogen dioxide, ozone, carbon monoxide, and sulfur dioxide) had significant short-term effects in all age groups (except for carbon monoxide and sulfur dioxide in infants).
These findings provide a need for the development of tailored strategies to prevent AE events in different age groups.

The ecological and oceanographic processes that drive the response of pelagic ocean microbiomes to environmental changes remain poorly understood, particularly in coastal upwelling ecosystems. Here ...we show that seasonal and interannual variability in coastal upwelling predicts pelagic ocean microbiome diversity and community structure in the Southern California Current region. Ribosomal RNA gene sequencing, targeting prokaryotic and eukaryotic microbes, from samples collected seasonally during 2014-2020 indicate that nitracline depth is the most robust predictor of spatial microbial community structure and biodiversity in this region. Striking ecological changes occurred due to the transition from a warm anomaly during 2014-2016, characterized by intense stratification, to cooler conditions in 2017-2018, representative of more typical upwelling conditions, with photosynthetic eukaryotes, especially diatoms, changing most strongly. The regional slope of nitracline depth exerts strong control on the relative proportion of highly diverse offshore communities and low biodiversity, but highly productive nearshore communities.
Motivated by recent technological advances in mobile robotics, this paper explores a novel approach for warehouse order picking. In particular, this work considers two types of commercially available ...mobile robots - one that can grasp items from a shelf (a picker) and another (a transporter) that can quickly deliver all items from the pick list to the packing station. A new vehicle routing problem is defined which seeks to minimise the time to deliver all items from a pick list to the packing station, a problem termed the pick, place, and transport vehicle routing problem. A mixed integer linear programming formulation is developed to answer three related research questions. First, what combination of picker and transport robots is required to obtain performance exceeding traditional human-based picking operations? Second, how should the composition of the robot fleet be altered to affect the greatest performance improvements? Finally, what are the impacts of warehouse layout designs when coordinated mobile robots are deployed? An extensive numerical analysis reveals that, (1) increasing the number of cross aisles decreases system performance; (2) centrally located packing stations improve system performance; and (3) the average distance from each pick location to the packing station and the average distance between pick locations are effective metrics for identifying specific fleet modifications that are likely to yield system improvements.
Chronic allograft vasculopathy (CAV) in murine heart allografts can be elicited by adoptive transfer of donor specific antibody (DSA) to class I MHC antigens and is independent of complement. Here we ...address the mechanism by which DSA causes CAV. B6.RAG1−/− or B6.RAG1−/−C3−/− (H‐2b) mice received B10.BR (H‐2k) heart allografts and repeated doses of IgG2a, IgG1 or F(ab’)2 fragments of IgG2a DSA (anti‐H‐2k). Intact DSA regularly elicited markedly stenotic CAV in recipients over 28 days. In contrast, depletion of NK cells with anti‐NK1.1 reduced significantly DSA‐induced CAV, as judged morphometrically. Recipients genetically deficient in mature NK cells (γ‐chain knock out) also showed decreased severity of DSA‐induced CAV. Direct NK reactivity to the graft was not necessary. F(ab’)2 DSA fragments, even at doses twofold higher than intact DSA, were inactive. Graft microvascular endothelial cells responded to DSA in vivo by increased expression of phospho‐extracellular signal‐regulated kinase (pERK), a response not elicited by F(ab’)2 DSA. We conclude that antibody mediates CAV through NK cells, by an Fc dependent manner. This new pathway adds to the possible mechanisms of chronic rejection and may relate to the recently described C4d‐negative chronic antibody‐mediated rejection in humans.
Chronic cardiac allograft vasculopathy in the mouse mediated by donor‐specific antibody is shown to require NK cells and the Fc portion of antibody but is independent of complement fixation. See editorial by Li and Baldwin on page 275.
