Recent progress in plasmon-based sensors has greatly overcome the limitations of conventional optical sensors, in terms of sensitivity, tunability, photostability, and in vivo applicability, by ...employing nanostructured materials. In this review, we summarize three types of plasmon-based nanosensors categorized by the geometries of sensing platforms as plasmonic transducers, which are planar nanofilm, periodic nanoarrays, and individual nanoparticles, respectively. We then introduce the notable accomplishments to enhance the sensing performances for each sensing platform. Additionally, we present recent works for signal enhancement strategy based on the targeted nanoassemblies of particles, which can be applied to all plasmon-based sensing platforms. The focus of this review will be on how plasmonic nanostructures can be applied to detect biological and chemical analytes, and notable approaches to improve the sensitivity.

To gain insight into the human health implications of microplastics, in this study, we investigated the possible mechanisms affecting the toxicity of high-density polyethylene (HDPE) in the nematode Caenorhabditis elegans using RNAi screening and a bioinformatics-based unbiased approach. The candidate pathways identified from C. elegans study were also confirmed using vertebrate model, zebrafish, Danio rerio and human relevance was then inferred using Comparative Toxicogenomics Database (CTD) analysis. Prior to evaluating the toxicity, label-free Raman mapping was conducted to investigate whether or not the organisms could uptake HDPE. C. elegans transcription factor RNAi screening results showed that the nucleotide excision repair (NER) and transforming growth factor-beta (TGF-β) signaling pathways were significantly associated with HDPE exposure, which was also confirmed in zebrafish model. Gene-disease interaction analysis using the CTD revealed the possible human health implications of microplastics. Finally, based on this finding, related AOPs were identified from AOP Wiki (http://aopwiki.org), which are “Peroxisome proliferator-activated receptors γ inactivation leading to lung fibrosis” and “AFB1: Mutagenic Mode-of-Action leading to Hepatocellular Carcinoma”. Further studies are needed for the validation of these AOPs with various microplastics.
Controlled assembly of colloidal nanoparticles onto solid substrates generally needs to overcome their thermal diffusion in water. For this purpose, several techniques that are based on chemical ...bonding, capillary interactions with substrate patterning, optical force, and optofluidic heating of light‐absorbing substrates are proposed. However, the direct assembly of colloidal nanoparticles on generic substrates without chemical linkers and substrate patterning still remains challenging. Here, photothermal convection lithography is proposed, which allows the rapid placement of colloidal nanoparticles onto the surface of diverse solid substrates. It is based on local photothermal heating of colloidal nanoparticles by resonant light focusing without substrate heating, which induces convective flow. The convective flow, then, forces the colloidal nanoparticles to assemble at the illumination point of light. The size of the assembly is increased by either increasing the light intensity or illumination time. It is shown that three types of colloidal gold nanoparticles with different shapes (rod, star, and sphere) can be uniformly assembled by the proposed method. Each assembly with a diameter of tens of micrometers can be completed within a minute and its patterned arrays can also be achieved rapidly.

Under the resonant light illumination, colloidal nanoparticles are spontaneously moved toward the focal point and densely assembled together. Each assembly can be completed within a minute and its patterned arrays can also be achieved rapidly.

Because HCCs have strong resistance to conventional chemotherapeutic ...agents, novel therapeutic strategies are needed to improve survival in patients with HCC. The multicellular tumor spheroid (MCTS) model is a powerful method for anticancer research because of its ability to mimic the complexity and heterogeneity of tumor tissue, the three-dimensional cellular context of tumor tissue, and the pathophysiological gradients of in vivo tumors. However, it is difficult to obtain meaningful results from the MCTS model without considering the conditions of clinical tumors. We, therefore, provided a proof of concept to determine whether spheroid models simulate in vivo tumor microenvironments. Histones are closely related to the state of chromatin, and epigenetic modification of their tail results in regulation in cells. Therefore, developing various analytical tools to map the changes in ...position and distribution of histone modifications is helpful in studying underlying mechanisms. Herein, we propose a high-spatial and colourimetric imaging method using plasmonic nanoparticles as probes to visualize heterochromatin histone markers in a single nucleus. We visualized the reorganization between repressive histone markers, H3K9me3 and H3K27me3, caused by oncogene-induced senescence based on the scattering colours and spectral shift of plasmonic nanoprobes to longer wavelengths using their distance-dependent coupling effect. The measured scattering profiles were correlated with the computation results simulating the scattering spectra according to the arrangements and distances among the plasmonic nanoprobes. The plasmonic nanoprobe-based high-spatial hyperspectral imaging provides an advanced way to study the dynamics of histone modifications for predicting the progression of diseases or senescence.
We report graphene nanopores with integrated optical antennae. We demonstrate that a nanometer-sized heated spot created by photon-to-heat conversion of a gold nanorod resting on a graphene membrane ...forms a nanoscale pore with a self-integrated optical antenna in a single step. The distinct plasmonic traits of metal nanoparticles, which have a unique capability to concentrate light into nanoscale regions, yield the significant advantage of parallel nanopore fabrication compared to the conventional sequential process using an electron beam. Tunability of both the nanopore dimensions and the optical characteristics of plasmonic nanoantennae are further achieved. Finally, the key optical function of our self-integrated optical antenna on the vicinity of graphene nanopore is manifested by multifold fluorescent signal enhancement during DNA translocation.
In recent nanobiotechnology developments, a wide variety of functional nanomaterials and engineered biomolecules have been created, and these have numerous applications in cell biology. For these ...nanomaterials to fulfill their promises completely, they must be able to reach their biological targets at the subcellular level and with a high level of specificity. Traditionally, either nanocarrier- or membrane disruption-based method has been used to deliver nanomaterials inside cells; however, these methods are suboptimal due to their toxicity, inconsistent delivery, and low throughput, and they are also labor intensive and time-consuming, highlighting the need for development of a next-generation, intracellular delivery system. This study reports on the development of an intracellular nanomaterial delivery platform, based on unexpected cell-deformation phenomena via spiral vortex and vortex breakdown exerted in the cross- and T-junctions at moderate Reynolds numbers. These vortex-induced cell deformation and sequential restoration processes open cell membranes transiently, allowing effective and robust intracellular delivery of nanomaterials in a single step without the aid of carriers or external apparatus. By using the platform described here (termed spiral hydroporator), we demonstrate the delivery of different nanomaterials, including gold nanoparticles (200 nm diameter), functional mesoporous silica nanoparticles (150 nm diameter), dextran (hydrodynamic diameters between 2–55 nm), and mRNA, into different cell types. We demonstrate here that the system is highly efficient (up to 96.5%) with high throughput (up to 1 × 106 cells/min) and rapid delivery (∼1 min) while maintaining high levels of cell viability (up to 94%).
