Here we report a pilot-sized study to compare the taxonomic composition of sputum microbiome in 17 newly-diagnosed lung cancer (LC) patients and 17 controls. Another object was to compare the ...representation of individual bacterial genera and species in sputum with the frequency of chromosomal aberrations in the blood lymphocytes of LC patients and in controls. Both groups were male; average age 56.1 ± 11.5 in patients and 55.7 ± 4.1 in controls. Differences in the species composition of bacterial communities in LC patients and controls were significant (pseudo-F = 1.94; p = 0.005). Increased prevalence in LC patients was detected for the genera Haemophilus and Bergeyella; whereas a decrease was observed for the genera Atopobium, Stomatobaculum, Treponema and Porphyromonas. Donors with high frequencies of chromosomal aberrations had a significant reduction in the microbiome of representatives of the genus Atopobium in the microbiome and a simultaneous increase in representatives of the species Alloprevotella compared to donors with a low level of chromosomal aberrations in lymphocytes. Thus, a comparison of the bacterial composition in the sputum of donors with cytogenetic damages in theirs lymphocytes, warrants further investigations on the potential role of microorganisms in the process of mutagenesis in somatic cells of the host body.
Introduction. As is currently known, the epidemic process in the Kaliningrad Region was mainly associated with the spread of the recombinant form of HIV-1 (CRF03_AB); however, regular HIV ...importations from other countries and continents has created favorable conditions for emergence and spread of various recombinant forms of the virus.
The most complete information on the diversity of recombinant forms in the region is also necessary to understand the structure of drug resistance (DR).
The aim of the study was to explore the HIV-1 genetic diversity in the Kaliningrad Region.
Materials and methods. We studied 162 blood plasma samples obtained from patients from the Kaliningrad Region, both with confirmed virological failure of antiretroviral therapy (ART) and with newly diagnosed HIV infection. For reverse transcription and amplification of HIV genome fragments, diagnostic AmpliSense HIVResist-Seq.
Results and discussion. The various recombinants between subtypes A and B (74%) were predominant in study group: recombinant was between CRF03_AB and subtype A (33.95%) and CRF03_AB-like (13.58%) were the most common. Among the pure subtypes of the virus, subtype A6 (16.67%). The circulation of subtypes B (3.70%) and G (1.23%) was also noted.
Ninety-six patients (59.26%) were identified with at least one mutation associated with antiretroviral (ARV) drug resistance.
Conclusion. The observed diversity of subtypes and recombinant forms of the virus implies that the new recombinants are actively emerging in the studied region, both between existing recombinant forms and pure subtypes, as well as between pure subtypes.
Studies into photochemical oxidation can enhance the current understanding of degradation processes within aqueous solutions of active pharmaceutical ingredients (APIs). APIs are complex decomposable ...compounds that, once in reservoirs, cause irreversible consequences in living organisms. The results obtained on the photodestruction of various drugs in water in the presence of hydrogen peroxide and peroxydisulfate contribute to gaining new practical and theoretical knowledge in the field of water treatment, post-treatment, and purification. The paper presents laboratory results on the oxidation of model aqueous solutions of nitrofural, tetracycline, and paracetamol under the combined effect of UV radiation with micro-additives of oxidants (hydrogen peroxide and peroxydisulfate). The reaction order of APIs destruction is determined by the least square method. The results show that the combined effect of UV radiation and microadditives of hydrogen peroxide and peroxydisulfate contributes to both a high degree of purification (up to 98%) and a high rate of oxidative degradation of APIs (nitrofural, tetracycline, and paracetamol) compared to the use of UV radiation separately. The studied drugs can be arranged in the following order in terms of their oxidative degradation transformation ability: nitrofural>tetracycline>paraceta mol. The paper theoretically proves that photochemical destruction in the presence of peroxydisulfate leads to the formation of more highly reactive oxygen-containing radicals, which are involved in the decomposition of nitrofural, tetracycline, and paracetamol.
Aim. Study cases virologic failure ART HIV-1 in children.Materials and methods. The blood plasma samples of 6 patients with HIV infection were used, identified under 1 year of life, from the ...North-West Federal District of the Russian Federation, received in 2014–2016. Presented in a group of patients were aimed at identifying drug resistance in the virus and correction of ART due to virological failure. In the present study we used genotyping by direct sequencing of the site of the polymerase gene (pol) length of 1285 nt., The gene encoding the protease (PR) length of 465 nt. and a portion of the reverse transcriptase (RT) gene length of 820 nt.Results. In all cases, I was diagnosed with HIV-1 subtype A1, the so-called IDU-A, which is the most common form of HIV-1 in the Russian Federation. Among the obtained isolates of HIV have been identified typical drug resistance mutations to NRTI and NNRTI. Detected multiple natural polymorphic variants from the land of the nucleotide sequences. Thus, all samples show protease mutation M36I, R41K, H69K, and L89M, five samples E35D, four samples I13V. Only one among the surveyed our children with combined variant protease gene mutations L63T + V77I + I93L and reverse transcriptase gene V35I + K166R, despite ongoing therapy and absence of significant drug resistance mutations, according to Stanford University-based data showed high viral load – 2.03×107 copies/ml. The importance of the influence of natural polymorphisms in the drug resistance of the virus is discussed.Conclusion. The high frequency of natural polymorphic variants of the protease and reverse transcriptase genes of the virus in clinical samples submitted. The number of such mutations, apparently does not depend on the number of treatment regimens, but found some correlation with adherence to treatment.
The fungi Trichophyton mentagrophytes and T. interdigitale are closely related species, causing superficial infections in humans and other mammals. The status of these taxa is a field of long-lasting ...debates. To clarify their phylogenetic relationships within the genus Trichophyton and sharpen the species boundaries, we performed sequencing of four T. mentagrophytes genomes and also evaluated three previously published multilocus data sets. We performed computational species delimitation analysis on all available in GenBank internal transcribed spacer region (ITS) sequences of Trichophyton spp. Phylogenomic data, phylogenetic network, and species delimitation analyses implied that T. mentagrophytes and T. interdigitale belong to the same phylogenetic species. However, we argue that taxonomic status quo should be retained, from the perspective of epidemiological data and the principle of taxonomic stability. Since there is a correlation between ITS genotype and epidemiological source of an isolate, restriction of T. interdigitale to purely anthropophilic ITS genotypes seems to be reasonable.
To identify an imbalance of cardiac remodeling mediators and monocytes subpopulation in blood, distribution of myocardium macrophages in patients with ischemic cardiomyopathy (ICMP).
The study ...engaged 30 patients with ICMP, 26 patients with coronary heart disease (CHD) without ICMP, 15 healthy donors. Concentrations of TGFβ, MMP-9, MCP-1, galectin-3 were measured in plasma of blood from the coronary sinus and peripheral blood in CHD patients, as well as in peripheral blood in healthy donors, by enzyme immunoassay method. The ration of classical, intermediate, non-classical, transitional monocytes in peripheral blood of patients and healthy donors was assessed by flow cytometry (expression CD14, CD16); the content of CD68+ macrophages in myocardium – by immunohistochemistry method.
In both samples of blood, the content of galectin-3 in patients with ICMP was higher than in CHD patients without ICMP and the level of TGFβ was comparable between the groups. At ICMP, the concentration of MMP-9 in sinus blood was higher than that in CHD patients without ICMP in whom an excess of MCP-1 in the general blood flow was determined. The density of distribution of CD68+ cells in the myocardium in patients with ICMP was higher in the perianeurysmal zone than in the right atrium appendage. ICMP was characterized by a deficiency of non-classical monocytes, and CHD without ICMP – by an excess of intermediate cells in peripheral blood.
Myocardium remodeling at ICMP is mediated by not so much TGFβ but intracardiac galectin-3, which determines the subpopulation composition of blood monocytes.
Aim. Epidemiologic examination of the case of group HIV-1 infection based on data obtained by general diagnostics methods for confirmation of the investigation hypothesis of deliberate HIV infection ...during heterosexual intercourse. Materials and methods. Sera samples from 8 HIV infected patients (5 female and 3 male) from Veliky Novgorod sent for epidemiologic investigation were used. Determination was carried out based on 1285 nt sequence analysis of polymerase gene segment (pol). Results. The study allowed to identify HIV in clinical samples sent to the expertise and establish phylogenetic connections between virus isolates obtained from both target and control group patients. Analysis of the results allowed to isolate samples grouped in a separate cluster that indicates tight cordial connections between the vims isolates from clinical material of these patients. Patients of the target group were infected with HIV-1 isolate ofthe circulating recombinant form CRF03_AB from the same origin that is confirmed by high homology ofthe nucleotide sequences. Conclusion. Epidemiologic investigation of a group case of HIV-1 infection has determined that the infection of the women of the target group occurred from the same source. Phylogenetic analysis results indicate the presence of an epidemiologic connection within the examined group that confirms the HIV infection transmission and conclusions ofthe investigation. Use of molecular-phylogenetic analysis of data obtained by laboratory diagnostics methods of HIV resistance to antiretroviral preparations allows with anamnestic and investigation information (in the context of available evidence) to investigate cases of HIV infection.
Abstract
Candida auris is an emergent yeast pathogen, easily transmissible between patients and with high percent of multidrug resistant strains. Here we present a draft genome sequence of the first ...known Russian strain of C. auris, isolated from a case of candidemia. The strain clustered within South Asian C. auris clade and seemingly represented an independent event of dissemination from the original species range. Observed fluconazole resistance was probably due to F105L and K143R mutations in ERG11.
