Chitinase 3-Like-1 (CHI3L1) is a secreted 40 kDa glycoprotein that is upregulated in a number of human cancers and in non-neoplastic disease states characterized by chronic inflammation and tissue ...remodeling. Increased serum levels of CHI3L1 parallel disease severity, poorer prognosis, and shorter survival in many human neoplasias, including cancers of the breast, colon, prostate, ovaries, brain, thyroid, lung, and liver. Increased serum CHI3L1 also correlates with disease severity in rheumatoid arthritis, osteoarthritis, liver fibrosis, inflammatory bowel disease, and bacterial septicemia. CHI3L1 is a rheumatoid arthritis (RA) autoantigen, and MHC complexes containing specific CHI3L1 peptides have been found in RA patients; however, intranasal introduction of these same CHI3L1 peptides can induce tolerance towards them. CHI3L1 is a nonhydrolytic member of the human chitinase family that binds chitin tightly and heparin at lower affinity. Interactions with type I collagen, CHI3L1's only known protein-binding partner, helps regulate collagen fibril formation. The principal sources of CHI3L1 are activated macrophages and chondrocytes, neutrophils, and some tissue and tumor cells. CHI3L1 can act as a fibroblast mitogen and can activate several signaling pathways, however, no cell surface-binding partner for CHI3L1 has been identified. The ability of CHI3L1 to bind both proteins and carbohydrates allows potential interactions with a variety of cell-surface and extracellular-matrix proteins, proteoglycans, and polysaccharides, and thus CHI3L1 can interface between proteomics and glycomics.

Sea spray aerosol (SSA) consists of both sea salt and organic components. These aerosols affect Earth's climate by scattering solar radiation and by altering cloud properties. Here we present ...observations of SSA particles generated at sea using an over‐the‐side bubbling system (Sea Sweep) and an onboard plunging wave mesocosm (Marine Aerosol Reference Tank—MART) during five cruises in the North Atlantic. The cruises were timed to sample different stages of the North Atlantic plankton bloom and included transects from the oligotrophic Sargasso Sea to the biologically productive western subarctic. The results show that the North Atlantic plankton bloom has little effect on the emission flux, organic fraction, or cloud condensation nuclei (CCN) activity of SSA, and therefore, plankton ecosystems do not need to be included in modeling aerosol indirect effects of primary SSA in global climate models or in chemical transport models.

These particles affect Earth's climate by scattering solar radiation and altering cloud properties (e.g., cloud brightness, extent, and rain). SSA consists of both sea salt and organic components. A key question in chemical transport and climate models is whether marine plankton ecosystems affect the organic component of SSA. The results presented here show that marine plankton ecosystems have little effect on SSA and that the ocean source of organic SSA comes from the large pool of dissolved organic carbon in the ocean. Chemical transport and climate models, therefore, can treat the ocean as a uniform organic SSA source, affected only by sea surface temperature.

Synaptotagmin (Syt) family proteins contain tandem C2 domains, C2A and C2B, which insert into anionic membranes in response to increased cytosolic Ca2+ concentration and facilitate exocytosis in ...neuronal and endocrine cells. The C2A domain from Syt7 binds lipid membranes much more tightly than the corresponding domain from Syt1, but the implications of this difference for protein function are not yet clear. In particular, the ability of the isolated Syt7 C2A domain to initiate membrane apposition and/or aggregation has been previously unexplored. Here, we demonstrate that Syt7 C2A induces apposition and aggregation of liposomes using Förster resonance energy transfer (FRET) assays, dynamic light scattering, and spectroscopic techniques involving lipid-coated gold nanoparticles (LCAuNPs). Protein–membrane binding, membrane apposition, and macroscopic aggregation are three separate phenomena with distinct Ca2+ requirements: the threshold Ca2+ concentration for membrane binding is lowest, followed by apposition and aggregation. However, aggregation is highly sensitive to protein concentration and can occur even at submicromolar Syt7 C2A; thus, highly sensitive assays are needed for measuring apposition without complications arising from aggregation. Notably, the localized surface plasmon resonance of the LCAuNP is sensitive to ≤10 nM Syt7 C2A concentrations. Furthermore, when the LCAuNPs were added into a FRET-based liposome apposition assay, the resultant energy transfer increased; possible explanations are discussed. Overall, LCAuNP-based methods allow for highly sensitive detection of protein-induced membrane apposition under conditions that miminize large-scale aggregation.

The UN Sustainable Development Goals (SDGs) are the blueprint to achieve a better and more sustainable future. To achieve the goal, tracking progress — not just on a national level, but locally — is crucial to guide future policy development. While sustainability assessment at the national evel is quite advanced in China, similar assessments focusing at the regional or even at the city-level are currently lacking. Here, we advanced the Inclusive Wealth Index (IWI) framework, which is firstly proposed by the United Nations Development Programme, through taking water wealth into account and adjusting the variable based on data availability. Then we investigate the sustainability performance of 210 cities in China in 2016 via the advanced version of the IWI framework. The analysis makes a holistic assessment based on produced, human, and natural capital, as well as considering heterogeneities in economy, social, and environmental conditions across these cities. We find that cities clustered in the eastern parts of China are characterized by high levels of sustainability performance and increasing capacities for sustainability, largely driven by their high quality and quantity of human capital. In comparison, the western cities have a large amount of low-skilled human capital and low levels of produced capital, which determines their low sustainability performance. Cities clustered in the north are heavily dependent on low value-added products and resource-intensive industries. Furthermore, we make projections of the IWI and its three components for different cities from 2020 to 2030, referring to the index systems presented in city planning which describe the development speed of income, education, fixed asset investment, forests etc. In the future, cities in central and western clusters show considerable potential for increasing IWI per capita, whereas cities with a dominant energy sector in the north would face declining capacity for sustainability due to the exhaustion of fossil fuels and raw materials. By fully taking account of and adapting to local circumstances, we tailor-design pathways for different types of cities to grow their sustainability potentials. Those resources-dependent cities in the north could avoid the impending decline by gradually developing their human and produced capital while abandoning their resource dependency. Our study contributes to city-level sustainable development in China through the lens of per capita IWI and the potential future dynamics of changing compositions in their capital.
Responsive feeding, where parents are guided by children's hunger and satiation cues and provide appropriate structure and support for eating, is believed to promote healthier weight status. However, ...few studies have assessed prospective associations between observed parental feeding and toddler growth. We characterized toddler growth from 18 to 36 months and, in a subset of families, examined whether observed maternal responsiveness to toddler satiation cues and encouraging prompts to eat at 18 and 24 months were associated with toddler body mass index z-score (BMIz) from 18 to 36 months. Participants included 163 toddlers and their mothers with overweight/obesity who had participated in a lifestyle intervention during pregnancy. Anthropometrics were measured at 18, 24, and 36 months. In a subsample, mealtime interactions were recorded in families' homes at 18 (n = 77) and 24 (n = 75) months. On average, toddler BMIz remained stable from 18 to 36 months with 31.3% (n = 51) categorized with a healthy weight, 56.4% (n = 92) with at risk for overweight and 12.3% (n = 20) with overweight. Fewer maternal prompts to eat at 18 months was associated with both higher probability of having at risk for overweight/overweight (p < .05), and higher child 36-month BMIz (p = .002). Higher child weight status at 12 months was also associated with both higher probability of having at risk for overweight/overweight (p < .05), and higher child 36-month BMIz (p < .001). Neither 24-month maternal prompts nor 18 or 24 month responsiveness to satiation cues were associated with toddler BMIz. In this diverse sample, weight status was relatively stable from 18 to 36 months. Maternal prompts to eat measured earlier in toddlerhood and prior child weight status were associated with toddler BMIz.
Emissions factors (EFs) for gas and sub-micron particle-phase species were measured in intercepted plumes as a function of vessel speed from an underway research vessel, the NOAA ship Miller Freeman, ...operating a medium-speed diesel engine on low-sulfur marine gas oil (fuel sulfur content ~0.1% by weight). The low-sulfur fuel in use conforms to the MARPOL fuel sulfur limit within emission control areas set to take effect in 2015 and to California-specific limits set to take effect in 2014. For many of the particle-phase species, EFs were determined using multiple measurement methodologies, allowing for an assessment of how well EFs from different techniques agree. The total sub-micron PM (PM1) was dominated by particulate black carbon (BC) and particulate organic matter (POM), with an average POM / BC ratio of 1.3. Consideration of the POM / BC ratios observed here with literature studies suggests that laboratory and in-stack measurement methods may overestimate primary POM EFs relative to those observed in emitted plumes. Comparison of four different methods for black carbon measurement indicates that careful attention must be paid to instrument limitations and biases when assessing EFBC. Particulate sulfate (SO42-) EFs were extremely small and the particles emitted by Miller Freeman were inefficient as cloud condensation nuclei (CCN), even at high super saturations, consistent with the use of very low-sulfur fuel and the overall small emitted particle sizes. All measurement methodologies consistently demonstrate that the measured EFs (fuel mass basis) for PM1 mass, BC and POM decreased as the ship slowed. Particle number EFs were approximately constant across the speed change, with a shift towards smaller particles being emitted at slower speeds. Emissions factors for gas-phase CO and formaldehyde (HCHO) both increased as the vessel slowed, while EFs for NOx decreased and SO2 EFs were approximately constant.
The sea breeze/land breeze diurnal circulation within the Los Angeles Basin and adjacent waters transports marine air into the basin during the day and urban air to Santa Monica Bay during the night. ...Nitryl chloride, ClNO2 is a nocturnal trace gas formed from the heterogeneous reaction of dinitrogen pentaoxide (N2O5) with chloride containing aerosol. Its photolysis after sunrise produces atomic chlorine radicals and regenerates NO2, both of which may increase ozone production. Mixing of the chloride source from marine sea salt with the urban NOx source in Los Angeles provides conditions ideal for the production of ClNO2. This paper presents an analysis using a wind profiler on the coast and measurements of ClNO2 and its precursors made from both ship and aircraft to assess the prevailing meteorological conditions important for ClNO2 production in this region, with a particular focus on the production over water within the land breeze phase of the circulation. A box model is used to calculate an upper limit to the amount of ClNO2 capable of being produced strictly over Santa Monica Bay during the land breeze. On three out of the four nights of ClNO2 measurements in Santa Monica Bay, the ClNO2 exceeds the upper limit calculated using the box model and shows that the majority of the ClNO2 is produced over the city and transported to Santa Monica Bay by the land breeze. This ClNO2 transport suggests the sea breeze more efficiently transports aerosol chloride inland than land breeze transports NOx offshore.
