Studies on activated cell-signaling pathways responsible for neoplastic transformation are numerous in solid tumors and in adult leukemias. Despite of positive results in the evolution of pediatric ...hematopoietic neoplasias, there are some high-risk subtypes at worse prognosis. The aim of this study was to asses the expression and activation status of crucial proteins involved in cell-signaling pathways in order to identify molecular alterations responsible for the proliferation and/or escape from apoptosis of leukemic blasts. The quantitative and qualitative expression and activation of Erk-1, c-Jun, Caspase8, and Gadd45a was analyzed, by immunocytochemical (ICC) and western blotting methods, in bone marrow blasts of 72 patients affected by acute myeloid leukemia (AML), T-cell acute lymphoblastic leukemia (ALL) and stage IV non-Hodgkin Lymphoma (NHL). We found an upregulation of Erk-1, Caspase8, c-Jun, and Gadd45a proteins with a constitutive activation in 95.8%, 91.7%, 86.2%, 83.4% of analyzed specimens, respectively. It is worth noting that all AML patients showed an upregulation of all proteins studied and the high expression of GADD45a was associated to the lowest DFS median (p = 0.04). On univariate analysis, only Erk-1 phosphorylation status was found to be correlated with a significantly shorter 5-years DFS in all disease subgroups (p = 0.033) and the lowest DFS median in ALL/NHL subgroup (p = 0.04). Moreover, the simultaneous activation of multiple kinases, as we found for c-Jun and Erk-1 (r = 0.26; p = 0.025), might synergistically enhance survival and proliferation potential of leukemic cells. These results demonstrate an involvement of these proteins in survival of blast cells and, consequently, on relapse percentages of the different subgroups of patients.
This study evaluated the effect of ultrasonic vibration on the tensile strength required to remove intraradicular post cemented with different materials.
Bovine teeth were selected, and 7 mm of the ...cervical root canals were prepared to size 5 Largo drill, the posts were cemented with zinc phosphate, Enforce (resin) or Rely X (glass ionomer). The specimens were divided into six groups (n = 10), according to the following procedures: GI-cementation with zinc phosphate associated with traction force; GII-cementation with zinc phosphate associated with ultrasonic activation and traction force; G111-cementation with Enforce associated with traction force; GIV-cementation with Enforce associated with ultrasonic activation and traction force; GV-cementation with Rely X associated with traction force; and GVI-cementation with Rely X associated with ultrasonic activation and traction force. The tensile test was conducted using the electromechanical testing machine, the force was determined by a specialized computer program and ultrasonic activation using the Jet Sonic Four Plus (Gnatus) device in 10P.
Concerning to average ranking, GI showed statistically significant difference in comparison with GII and GVI (p < 0.05); there was no statistical difference in GIII and GIV when compared to other groups (p > 0.05).
The ultrasound favored the intraradicular post traction regardless of the employed cement in greater or lesser extent.
The post removal is a routine practice in the dental office, therefore, new solutions and better alternatives are need to the practitioner. We did not find in the literature many articles referring to this practice. Thus, the results from this study are relevant in the case planning and to promote more treatment options.
Abstract The aim of this study was to evaluate the diagnostic value of a grid of molecular genetic markers detectable in sputum and plasma samples of individuals enrolled in a lung cancer screening ...program with low-dose CT. Subjects enrolled in the baseline screening round of the ITALUNG (randomised) screening trial were invited to provide biological specimens for molecular analysis (1356 subjects out of 1406). We included 98 subjects in this analysis. There was a highly statistically significant difference between proportion of subjects with a negative baseline CT screening test who were positive to allelic imbalance, and those with a non-calcified nodule (NCN greater than or equal to 5 mm), the reason of recall for all suspects at CT Scan ( χ2 : 22.9; P < 0.0001). Allelic imbalance showed good performance for screening of NCN ≥5 mm. In subjects recalled for NCN ≥5 mm, LOH, K-ras mutations and high levels of free plasma DNA (>5 ng/ml plasma) might be important to support clinical decision making for further follow-up and repeated screening. This study, embedded in an early diagnosis randomised trial, suggests that a multi-screening approach integrating imaging technique and a biomolecular marker panel is worth of further investigation.
O policloreto de vinila (PVC) é um plástico muito utilizado por ser compatível com vários plastificantes, como o DEHP ( di (2-etil) ftalato) que o torna mais flexível. Utilizou-se 50 ratos com 180g e ...200g. No grupo 1: 10 animais não receberam nenhum tipo de substância. Nos grupos 2 e 3 com 20 animais em cada grupo receberam doses de óleo de milho (1 ml) e 2,85 mg / kg de peso corporal / dia DEPH dissolvido em 1 ml de óleo de milho por gavagem uma vez ao dia. Após 4 semanas os animais foram sacrificados e o fígado e pulmão, foram retirados para análise histopatológica. No fígado dos animais dos grupo 1e 2 observou-se um parênquima hepático normal, hepatócitos dispostos em arranjos cordonais com capilares sinusóides normais, presença de células de Kupffer, hepatócitos com núcleos normais, entretanto no grupo 2 observou-se núcleos binucleados. No grupo 3 encontrou-se no espaço porta uma proliferação de ductos biliares e presença de hepatócitos binucleares, megalócitos e capilares congestos. No pulmão do grupo 1 observou-se aspectos normais nas áreas da região alveolar e bronquiolar, septos interalveolares de aspecto normal com hemácias. No grupo 2 encontrou-se um discreto espessamento alveolar, mas com preservação da luz alveolar evidente congestão pulmonar. No grupo 3 observou-se uma hiperplasia de nódulos linfóides associados aos brônquios, grande área de infarto hemorrágico e acentuado espessamento de septos interalveolares. Conclui-se que foram constatadas alterações histopatológicas do grupo tratado e a preservação das características normais do grupo controle.
A obesidade é fator de risco para diversas outras doenças tornando a pessoa obesa mais propensa a desenvolver problemas como hipertensão arterial, doenças cardiovasculares, diabetes tipo 2, entre ...outras. Dos fármacos antiobesidade, destaca-se a Sibutramina, utilizada como antidepressivo e após algum tempo constatou-se o seu potencial efeito emagrecedor. Esta investigação teve como objetivo avaliar os efeitos da sibutramina sobre o sistema reprodutor feminino de ratas Wistar. Os animais foram distribuídos em 2 grupos com 10 animais cada grupo: o grupo 1 (controle) recebeu 0,3mL de soro fisiológico ( SF) a 0,9%, o grupo 2 recebeu 15mg/Kg de peso diluído em 0,3mL de água destilada de sibutramina, por gavagem, diariamente, durante 24 dias. Após esse período os animais foram previamente anestesiados para coleta de sangue por punção cardíaca em seguida os animais foram eutanasiados de acordo com as normas do Comitê de ética e pesquisa animal (CEPan). Os ovários e o útero foram fixados em formol 10% e corados com Hematoxilina /Eosina (H.E.) O peso corpóreo das ratas foi influenciado significativamente (p<0,001) pelo tratamento com sibutramina, em comparação ao grupo controle, assim como o consumo alimentar e hídrico, o grupo que recebeu tratamento com sibutramina ingeriu menos ração e água comparados ao grupo controle. Na histopatologia encontrou-se no ovário folículos atrésicos e intensa vascularização no estroma ovariano e no útero acentuadas projeções papilares e intensa proliferação epitelial. Conclui-se que a administração da sibutramina, na dosagem utilizada nesta pesquisa, reduziu o ganho de peso nas ratas e alterou o parênquima normal dos tecidos reprodutivos de ratas Wistar.
