Most asexual species of fungi have either lost sexuality recently, or they experience recombination by cryptic sexual reproduction. Verticillium dahliae is a plant-pathogenic, ascomycete fungus with ...no known sexual stage, even though related genera have well-described sexual reproduction. V. dahliae reproduces mitotically and its population structure is highly clonal. However, previously described discrepancies in phylogenetic relationships among clonal lineages may be explained more parsimoniously by recombination than mutation; therefore, we looked for evidence of recombination within and between clonal lineages. Genotyping by sequencing was performed on 141 V. dahliae isolates from diverse geographic and host origins, resulting in 26,748 single-nucleotide polymorphisms (SNPs). We found a strongly clonal population structure with the same lineages as described previously by vegetative compatibility groups (VCGs) and molecular markers. We detected 443 recombination events, evenly distributed throughout the genome. Most recombination events detected were between clonal lineages, with relatively few recombinant haplotypes detected within lineages. The only three isolates with mating type MAT1-1 had recombinant SNP haplotypes; all other isolates had mating type MAT1-2. We found homologs of eight meiosis-specific genes in the V. dahliae genome, all with conserved or partially conserved protein domains. The extent of recombination and molecular signs of sex in (mating-type and meiosis-specific genes) suggest that V. dahliae clonal lineages arose by recombination, even though the current population structure is markedly clonal. Moreover, the detection of new lineages may be evidence that sexual reproduction has occurred recently and may potentially occur under some circumstances. We speculate that the current clonal population structure, despite the sexual origin of lineages, has arisen, in part, as a consequence of agriculture and selection for adaptation to agricultural cropping systems.

The objective of this study was the valorization of the essential oils from Spanish Artemisia absinthium domesticated plants from Teruel and Sierra Nevada (Spain). These populations were experimentally cultivated in the field and under controlled conditions. The insect antifeedant properties of their essential oils collected yearly from two locations were tested against Spodoptera littoralis, Myzus persicae and Rhopalosiphum padi. Additionally we studied their phytotoxic, antifungal and antiparasitic effects. The oils from cultivated A. absinthium were characterized by the presence of cis-epoxyocimene, chrysanthenol, and chrysanthenyl acetate. The variations observed in oil composition were mostly quantitative but also qualitative. (Z)-2,6-Dimethyl-5,7-octadien-2,3-diol has been isolated and identified by NMR. Among the oil samples, these rich in cis-epoxyocimene and sesquiterpenes were the most active ones against S. littoralis. (Z)-2,6-Dimethyl-5,7-octadien-2,3-diol showed moderate activity against S. littoralis. The strongest antifeedant effects were found for commercial A. absinthium oil samples rich in thujones and sabinyl acetate. F. oxysporum and F. solani were affected by oils from cultivated A. absinthium and commercial oil samples. Oils from cultivated A. absinthium showed antiparasitic effects against Leishmania infantum and Trypanosoma cruzi with better results than the commercial samples.
Fusarium wilt caused by Fusarium oxysporum f. sp. ciceris, a main threat to global chickpea production, is managed mainly by resistant cultivars whose efficiency is curtailed by Fusarium oxysporum f. ...sp. ciceris races.
We characterized compatible and incompatible interactions by assessing the spatial-temporal pattern of infection and colonization of chickpea cvs. P-2245, JG-62 and WR-315 by Fusarium oxysporum f. sp. ciceris races 0 and 5 labeled with ZsGreen fluorescent protein using confocal laser scanning microscopy.
The two races colonized the host root surface in both interactions with preferential colonization of the root apex and subapical root zone. In compatible interactions, the pathogen grew intercellularly in the root cortex, reached the xylem, and progressed upwards in the stem xylem, being the rate and intensity of stem colonization directly related with the degree of compatibility among Fusarium oxysporum f. sp. ciceris races and chickpea cultivars. In incompatible interactions, race 0 invaded and colonized 'JG-62' xylem vessels of root and stem but in 'WR-315', it remained in the intercellular spaces of the root cortex failing to reach the xylem, whereas race 5 progressed up to the hypocotyl. However, all incompatible interactions were asymptomatic.
The differential patterns of colonization of chickpea cultivars by Fusarium oxysporum f. sp. ciceris races may be related to the operation of multiple resistance mechanisms.

Hypoglycaemia caused by glucose-lowering therapy has been linked to cardiovascular (CV) events. The ORIGIN trial provides an opportunity to further assess this relationship.
A total of 12 537 ...participants with dysglycaemia and high CV-risk were randomized to basal insulin glargine titrated to a fasting glucose of ≤ 5.3 mmol/L (95 mg/dL) or standard glycaemic care. Non-severe hypoglycaemia was defined as symptoms confirmed by glucose ≤ 54 mg/dL and severe hypoglycaemia as a requirement for assistance or glucose ≤ 36 mg/dL. Outcomes were: (i) the composite of CV death, non-fatal myocardial infarction or stroke; (ii) mortality; (iii) CV mortality; and (iv) arrhythmic death. Hazards were estimated before and after adjustment for a hypoglycaemia propensity score. During a median of 6.2 years (IQR: 5.8-6.7), non-severe hypoglycaemic episodes occurred in 41.7 and 14.4% glargine and standard group participants, respectively, while severe episodes occurred in 5.7 and 1.8%, respectively. Non-severe hypoglycaemia was not associated with any outcome following adjustment. Conversely, severe hypoglycaemia was associated with a greater risk for the primary outcome (HR: 1.58; 95% CI: 1.24-2.02, P < 0.001), mortality (HR: 1.74; 95% CI: 1.39-2.19, P < 0.001), CV death (HR: 1.71; 95% CI: 1.27-2.30, P < 0.001) and arrhythmic death (HR: 1.77; 95% CI: 1.17-2.67, P = 0.007). Similar findings were noted for severe nocturnal hypoglycaemia for the primary outcome and mortality. The severe hypoglycaemia hazard for all four outcomes was higher with standard care than with insulin glargine.
Severe hypoglycaemia is associated with an increased risk for CV outcomes in people at high CV risk and dysglycaemia. Although allocation to insulin glargine vs. standard care was associated with an increased risk of severe and non-severe hypoglycaemia, the relative risk of CV outcomes with hypoglycaemia was lower with insulin glargine-based glucose-lowering therapy than with the standard glycaemic control. Jamming is a phenomenon shared by a wide variety of systems, such as granular materials, foams, and glasses in their high density regime. This has motivated the development of a theoretical framework ...capable of explaining many of their static critical properties with a unified approach. However, the dynamics occurring in the vicinity of the jamming point has received little attention and the problem of finding a connection with the local structure of the configuration remains unexplored. Here we address this issue by constructing physically well defined structural variables using the information contained in the network of contacts of jammed configurations, and then showing that such variables yield a resilient statistical description of the particle-wise dynamics near this critical point. Our results are based on extensive numerical simulations of systems of spherical particles that allow us to statistically characterize the trajectories of individual particles in terms of their first two moments. We first demonstrate that, besides displaying a broad distribution of mobilities, particles may also have preferential directions of motion. Next, we associate each of these features with a structural variable computed uniquely in terms of the contact vectors at jamming, obtaining considerably high statistical correlations. The robustness of our approach is confirmed by testing two types of dynamical protocols, namely molecular dynamics and Monte Carlo, with different types of interaction. We also provide evidence that the dynamical regime we study here is dominated by anharmonic effects and therefore it cannot be described properly in terms of vibrational modes. Finally, we show that correlations decay slowly and in an interaction-independent fashion, suggesting a universal rate of information loss.

In the last two decades, the price of agave in states holding Designation of Origin of Tequila (DOT) status has been subject to uncertainty. This mixed sequential investigation identifies the ...variables that affect its value, verifying the cause of the problem. Using Minitab software, Pearson’s correlation analysis is applied to the most important variables and time-series predictive methods are applied to forecast them. The correlation analysis verified that the production of agave is not correlated with its price, in contrast to the production of tequila, its export and the income from its export. Since 2016, the price and production of agave systematically reduced its annual average growth until reaching negative levels in 2021, contrary to the production, export of tequila and its income (the five variables maintain this trend for the next 10 years). It is shown that the problem of the agave price at the end of the last decade is not due to imbalances in its supply, suggesting that the oligopoly of leading companies of the tequila agroindustry in Mexico controls its value.

Asteraceae plants from arid lands are a source of biomass, resin and latex rich in terpenoids with diverse biological effects. Thirty‐six previously isolated terpenes, comprising sesquiterpenes, ...diterpenes, triterpenes and quassinoids, isolated from arid‐land plants and a series of metabolites from the biotransformation of some lead compounds were evaluated against insect pests (Spodoptera littoralis, Leptinotarsa decemlineata, Myzus persicae and Rhopalosiphum padi), cells (insect, hamster, murine and human tumoral cells) and parasites (Trypanosoma cruzi and Leishmania infantum). Among the insecticidal sesquiterpenes, maalian‐1α,8α‐diol (12) and γ‐eudesmol (17) were antifeedant against L. decemlineata, M. persicae and cytotoxic to Sf9 insect cells, and (−)‐maali‐3‐en‐8α‐ol (10), (+)‐maaliane‐5α,8α,9α‐triol (11), chrysothame (31) and holacanthone (35) were antifeedant against S. littoralis. The parasite L. infantum was slightly more sensitive than T. cruzi to the test compounds (39 % vs. 33 % of active compounds) with compound 17 and the biotransformed diterpene 27 being antiparasitic to L. infantum, with no cytotoxic effects on mammalian cells. Moreover, sesquiterpenes 3 and 17, and grindelane diterpenes 22, 23 and 26 showed selective activity against chemoresistant human colon, cervical and melanoma cancer cells. Thus, considering our results, the best candidates for future studies are compounds 17 and 3, due to their activity on insect pests, parasites (17) and tumoral cells (3, 17, 22, 23 and 26).
Leishmania infantum is a protozoan causing leishmaniasis in humans and in dogs, among other animals, which is an endemic disease in the Mediterranean basin. In recent years, the role of wildlife as a ...possible reservoir of the disease was analyzed and several species of carnivores were reported to have the highest infection rates, with foxes and wolves being the more widely studied species; the role of rabbits and hares as reservoirs of leishmaniasis has also been described. In addition, several studies highlighted the role of bats as suitable hosts for Leishmania species (L. braziliensis, L. major, L. mexicana, and L. tropica) in South and Central America and Africa, but no Leishmania spp. infection in Chiroptera has been reported in Europe. In this study, samples from spleen, hair, and blood were analyzed to detect L. infantum DNA in bats from the Community of Madrid (Spain). Infection by L. infantum was detected in 59.2% of the bats studied (n = 16/27), with the spleen being selected as the site for detection, yielding 14/16 positive results (87.5% sensitivity), followed by hair (n = 7/16) and blood (n = 6/16). In two animals, samples from all three anatomical sites tested positive (7.4% of the total animals), while in four animals the spleen and hair samples tested positive (14.8%), in one animal the blood and hair samples tested positive (3.7%), and in another animal the blood sample only tested positive (3.7%). This is the first report of L. infantum detection in the common urban bat (Pipistrellus pipistrellus) in Europe.

Leishmaniasis are neglected diseases caused by several species of Leishmania that affect humans and many domestic and wild animals with a worldwide distribution. The objectives of this review are to ...identify wild animals naturally infected with zoonotic Leishmania species as well as the organs infected, methods employed for detection and percentage of infection. A literature search starting from 1990 was performed following the PRISMA methodology and 161 reports were included. One hundred and eighty-nine species from ten orders (i.e., Carnivora, Chiroptera, Cingulata, Didelphimorphia, Diprotodontia, Lagomorpha, Eulipotyphla, Pilosa, Primates and Rodentia) were reported to be infected, and a few animals were classified only at the genus level. An exhaustive list of species; diagnostic techniques, including PCR targets; infected organs; number of animals explored and percentage of positives are presented. L. infantum infection was described in 98 wild species and L. (Viania) spp. in 52 wild animals, while L. mexicana, L. amazonensis, L. major and L. tropica were described in fewer than 32 animals each. During the last decade, intense research revealed new hosts within Chiroptera and Lagomorpha. Carnivores and rodents were the most relevant hosts for L. infantum and L. (Viannia) spp., with some species showing lesions, although in most of the studies clinical signs were not reported.
