Breastfeeding or gestation in schistosomotic mothers can cause long-term alterations in the immune response of offspring.
Evaluate the expression of histone deacetylases (HDACs) (all classes), the ...production of cytokines by T and B lymphocytes and macrophages, and the frequency of CD4+CD25+FoxP3+-cells in adult offspring born and/or suckled by schistosomotic mothers.
We harvested splenocytes from offspring born to (BIM), suckled by (SIM), or born to/suckled by (BSIM) schistosomotic mothers and animals from noninfected mothers (Control) at seven-weeks old and cultured them with/without Concanavalin A. HDAC expression was evaluated by real-time quantitative polymerase chain reaction (qPCR), and cytokines and membrane markers were evaluated by fluorescence-activated cell sorting (FACS).
Compared to Control, BIM mice showed increased expression of HDAC9 and frequency of CD4+IL-10+-cells. The SIM group had increased expression of HDAC1, HDAC2, HDAC6, HDAC7, HDAC10, Sirt2, Sirt5, Sirt6, and Sirt7. The BSIM group only had increased HDAC10 expression. The SIM and BSIM groups exhibited decreased frequencies of CD4+IL-4+-cells and CD4+CD25+FoxP3+-cells, along with a higher frequency of CD14+IL-10+-cells and an increase in CD45R/B220+IL-10+-cells. The BSIM group also showed a high frequency of CD4+IL10+-cells.
Breastfeeding induced the expression of HDACs from various classes involved in reducing inflammatory responses. However, gestation enhanced the expression of a single HDAC and breastfeeding or gestation appears to favour multiple IL-10-dependent pathways, but not cells with a regulatory phenotype.
Medicinal plant (MP) use supports comprehensiveness of care in Primary Health Care (PHC), enabling appreciation of popular knowledge and self-care. This integrative literature review aims to analyze ...researches that approach the insertion of using MP in PHC. PICO strategy was used as a guideline in search of evidence, reuniting 18 articles published between January 2015 and August 2020, in the Virtual Health Library and PubMed databases. The variables of analysis were knowledge of PHC healthcare professionals about MP and associated policies, MP use by its users, highlighting their profile, the reasons that lead to the use and lack of security in MP use. The results show insufficient knowledge of healthcare professionals about Integrative and Complementary Practices policies and the medicinal uses of plants. The main users are women, elderly, with low income and education, either in Brazil or other countries. Regarding safety in MP use, frequently there is no correct identification of species, its origin, its preparation and the appropriate dose for each case. Finally, failure to approach these contents during training of healthcare professionals generates less knowledge, less research and more prejudice due to lack of information, impairing incentive and dissemination to the community.
To evaluate the effect of plasma-enhanced chemical vapor deposition (PECVD) with silicon hydride (SiH4) at different times on HT-zirconia surface characteristics and bonding of composite cement ...before and after thermocycling.
Blocks of HT zirconia were obtained, polished, sintered and divided into five groups, according to PECVD time (n = 31): Zr-30 (30 s), Zr-60 (60 s), Zr-120 (120 s) and Zr-300 (300 s). The control group (Zr-0) did not receive PECVD. X-ray diffraction (XRD), Fourier-transform infrared spectroscopy (FTIR), energy dispersive spectroscopy (EDS) in conjunction with field-emission scanning electron microscopy (FE-SEM), x-ray photoelectron spectroscopy (XPS), goniometry, and profilometry tests were used for chemical and topographic characterization. Monobond N silane (Ivoclar Vivadent) was applied to the surface, and a cylinder of composite cement (Variolink N) was made (3 x 3 mm). Half of the specimens of each group were stored for 24 h or subjected to thermocycling (6 x 103 cycles). A shear bond strength (SBS) test was performed. Results were subjected to one-way ANOVA and Tukey's tests (α = 0.05).
For experimental groups, XPS showed that formation of Si-O bonds contributed to increased surface free energy (SFE). FE-SEM and EDS showed that the longer the deposition time, the greater the amount of silicon on the surface. Zr-60 and Zr-300 presented higher and lower surface roughnesses, respectively. The silicon penetrated the microstructure, causing higher stress concentrations. The bond strength to composite cement was improved after all PECVD deposition times.
The PECVD technique with SiH4, associated with chemical treatment with primer based on silane methacrylate, is a solely chemical surface treatment capable of maintaining bonding between composite cement and HT zirconia.
