Interest in methods that routinely and accurately measure and predict animal characteristics is growing in importance, both for quality characterization of livestock products and for genetic ...purposes. Mid-infrared spectroscopy (MIRS) is a rapid and cost-effective tool for recording phenotypes at the population level. Mid-infrared spectroscopy is based on crossing matter by electromagnetic radiation and on the subsequent measure of energy absorption, and it is commonly used to determine traditional milk quality traits in official milk laboratories. The aim of this review was to focus on the use of MIRS to predict new milk phenotypes of economic relevance such as fatty acid and protein composition, coagulation properties, acidity, mineral composition, ketone bodies, body energy status, and methane emissions. Analysis of the literature demonstrated the feasibility of MIRS to predict these traits, with different accuracies and with margins of improvement of prediction equations. In general, the reviewed papers underlined the influence of data variability, reference method, and unit of measurement on the development of robust models. A crucial point in favor of the application of MIRS is to stimulate the exchange of data among countries to develop equations that take into account the biological variability of the studied traits under different conditions. Due to the large variability of reference methods used for MIRS calibration, it is essential to standardize the methods used within and across countries.
Hyperketonemia (HYK) is one of the most frequent and costly metabolic disorders in high-producing dairy cows and its diagnosis is based on β-hydroxybutyrate (BHB) concentration in blood. In the last ...10 years, the number of papers that have dealt with the impact of elevated BHB levels in dairy cattle has increased. Therefore, this paper reviewed the recent literature on BHB concentration in blood and milk, and its relationships with dairy cow health and performance, and farm profitability. Most studies applied the threshold of 1.2 mmol/l of BHB concentration in blood to indicate HYK; several authors considered BHB concentrations between 1.2 and 2.9 mmol/l as subclinical ketosis, and values ⩾3.0 mmol/l as clinical ketosis. Results on HYK frequency (prevalence and incidence) and cow performance varied according to parity and days in milk, being greater in multiparous than in primiparous cows, and in the first 2 weeks of lactation than in later stages. Hyperketonemia has been associated with greater milk fat content, fat-to-protein ratio and energy-corrected milk, and lower protein and urea nitrogen in milk. The relationships with milk yield and somatic cell count are still controversial. In general, HYK impairs health of dairy cows by increasing the risk of the onset of other early lactation diseases, and it negatively affects reproductive performance. The economic cost of HYK is mainly due to impaired reproductive performance and milk loss. From a genetic point of view, results from the literature suggested the feasibility of selecting cows with low susceptibility to HYK. The present review highlights that milk is the most promising matrix to identify HYK, because it is easy to sample and allows a complete screening of the herd through BHB concentration predicted using mid-IR spectroscopy during routine milk recording. Further research is needed to validate accurate and convenient methods to discriminate between cows in risk of HYK and healthy animals in field conditions and to support farmers to achieve an early detection and minimise the economic losses.
Lactose is the main carbohydrate in mammals' milk, and it is responsible for the osmotic equilibrium between blood and alveolar lumen in the mammary gland. It is the major bovine milk solid, and its ...synthesis and concentration in milk are affected mainly by udder health and the cow's energy balance and metabolism. Because this milk compound is related to several biological and physiological factors, information on milk lactose in the literature varies from chemical properties to heritability and genetic associations with health traits that may be exploited for breeding purposes. Moreover, lactose contributes to the energy value of milk and is an important ingredient for the food and pharmaceutical industries. Despite this, lactose has seldom been included in milk payment systems, and it has never been used as an indicator trait in selection indices. The interest in lactose has increased in recent years, and a summary of existing information about lactose in the dairy sector would be beneficial for the scientific community and the dairy industry. The present review collects and summarizes knowledge about lactose by covering and linking several aspects of this trait in bovine milk. Finally, perspectives on the use of milk lactose in dairy cattle, especially for selection purposes, are outlined.
Immunoglobulin G is the fundamental antibody for acquisition of passive transfer of immunity in ruminant newborns. Colostrum, in fact, must be administered as soon as possible after birth to ensure a ...successful transfer of IgG from the dam to the calf. Assessment of colostrum Ig concentration and gross composition via gold standards is expensive, time consuming, and hardly implementable for large-scale investigations. Therefore, in the present study we evaluated the predictive ability of mid-infrared spectroscopy (MIRS) as an indirect determination method. A total of 714 colostrum samples collected within 6 h from parturition from Italian Holstein cows, 30% primiparous and 70% pluriparous, were scanned using a benchtop spectrometer after dilution in pure water. The prediction models were developed by correlating spectral information with the reference measurements: IgG concentration (93.54 ± 33.87 g/L), total Ig concentrations (102.82 ± 35.04 g/L), and content of protein (14.71 ± 3.51%), fat (4.61 ± 3.04%), and lactose (2.36 ± 0.51 mg/100 mg). We found a good to excellent performance in prediction of colostrum IgG concentration and traditional composition traits in cross-validation (R2CV ≥ 0.92) and a promising and good predictive ability in external validation with R2V equal to 0.84, 0.89, and 0.74 for IgG, protein, and fat, respectively. In the case of IgG and protein content, for example, the coefficient of determination in external validation was greater than 0.84. The other Ig fractions, A and M, presented insufficient prediction accuracy likely due to their extremely low concentration compared with IgG (4.56 and 5.06 g/L vs. 93.54 g/L). The discriminant ability of MIRS-predicted IgG and protein content was outstanding when trying to classify samples according to the quality level (i.e., low vs. high concentration of IgG). In particular, the cut-off that better discriminate low- from high-quality colostrum was 75.40 g/L in the case of the MIRS-predicted IgG and 13.32% for the MIRS-predicted protein content. Therefore, MIRS is proposed as a rapid and cheap tool for large-scale punctual IgG, protein, and lactose quantification and for the screening of low-quality samples. From a practical perspective, there is the possibility to install colostrum models in the MIRS benchtop machineries already present in laboratories in charge of official milk testing. Colostrum phenotypes collected on an individual basis will be useful to breeders for the definition of specific selection strategies and to farmers for management scopes. Finally, our findings may be relevant for other stakeholders, given the fact that colostrum is an emerging ingredient for the animal and human food and pharmaceutical industry.
Milk preservative and freezing are used as strategies to prevent microbial growth and milk degradation, especially when immediate analytical processing is not feasible. The effects of the addition of ...preservative and freezing procedures have been investigated mainly in relation to milk gross chemical composition predicted through mid-infrared spectroscopy. This study aimed to determine whether different preservatives (i.e., no preservative, hydrogen peroxide, Bronopol, and Azidiol), freezing times (i.e., 0, 7, and 30 d), and temperatures of analysis (i.e., 5 and 21°C) influence the composition of milk protein fractions determined through reversed-phase HPLC. Bulk milk samples for the analysis of protein profile were collected from 5 commercial dairy farms. Data were analyzed with a linear mixed model, which included type of preservative, time of storage, temperature of analysis, and the interaction between type of preservative and time of storage as fixed effects, with the farm and the residual as random effects. Samples with no preservative had the greatest amount of all protein fractions, whereas Bronopol-preserved milk had the lowest amount. Increasing storage time under freezing conditions had a nonlinear detrimental effect on milk protein fractions. The temperature of analysis significantly contributed to the variation of κ-casein, β-casein, αS1-casein, β-lactoglobulin, and α-lactalbumin fractions. The z-scores were calculated to evaluate the similarity between detailed protein profile of fresh milk without preservative analyzed at 5°C and detailed protein profile of milk treated according to the tested conditions. Overall results suggested a good agreement between different analytical conditions. Still, short storage time under freezing conditions is recommended to avoid degradation of milk protein fractions and consequent analytical underestimation.
Colostrum quality and volume are fundamental for calves because it is the primary supplier of antibodies and the first source of energy, carbohydrates, lipids, proteins, minerals, and vitamins for ...the newborn. Assessing the detailed composition (i.e., AA and mineral content) of bovine colostrum (BC) on-line and at a reasonable cost would help dairy stakeholders such as farmers or veterinarians for precision feeding purposes and industries producing preparations containing BC such as foodstuff, supplements, and medicaments. In the present study we evaluated mid- (MIRS) and near-infrared spectroscopy (NIRS) prediction ability for AA and mineral composition of individual BC. Second, we the investigated the major factors affecting the phenotypic variability of such traits also evaluating the correlations with the Ig concentration. Results demonstrated that MIRS and NIRS were able to provide sufficiently accurate predictions for all the AA. The coefficient of determination in external validation (R
) fell, in fact, within the range of 0.70 to 0.86, with the exception of Ile, His, and Met. Only some minerals reached a sufficient accuracy (i.e., Ca, P, S, and Mg; R
≥ 0.66) using MIRS, and also S (R
= 0.87) using NIRS. Phenotypically, both parity and calving season affected the variability of these BC composition traits. Heifers' colostrum was the one with the greatest concentration of Ca and P, the 2 most abundant minerals. These minerals were however very low in cows calving in summer compared with the rest of the year. The pattern of AA across parities and calving season was not linear, likely because their variability was scarcely (or not) affected by these effects. Finally, samples characterized by high IgG concentration were those presenting on average greater concentration of AA. Findings suggest that infrared spectroscopy has the potential to be used to predict certain AA and minerals, outlining the possibility of implementing on-site analyses for the evaluation of the broad-sense BC quality.

Iodine represents a fundamental element for human health, with particular regard to thyroid function. Dietary intake of milk naturally rich in iodine becomes of primary importance in the prevention of syndromes related to iodine deficiency. The concentration of iodine in milk is characterized by wide variability, mainly related to animal feed and level of mineral supplementation. Therefore, there is interest in the development of fast analytical techniques which are able to predict milk iodine concentration. The aim of the present study was to investigate the effectiveness of energy-dispersive X-ray fluorescence (ED-XRF) for the prediction of iodine in cow milk. Results showed moderate accuracy of the ED-XRF technique, with a coefficient of determination in cross validation of 0.60. This study represents a first contribution towards the possibility to discriminate milk with high or low iodine concentration, as an essential preliminary step for the introduction into the market of naturally fortified milk.
An edited data set of 700 bulk and 46,338 test-day records collected between 2019 and 2021 in 42 Holstein-dominated farms in the Veneto Region (North of Italy) was available for the present study. ...Information on protein, fat and lactose content, somatic cell count, and somatic cell score was available in bulk milk as well as individual test-day records, whereas urea concentration (mg/dL), differential somatic cell count (%), and milk yield (kg/d) were available for test-day records only. Milk features were merged with meteorological data retrieved from 8 weather stations located maximum 10 km from the farms. The daily and weekly temperature-humidity index (THI; wTHI) and maximum daily (MTHI) and weekly temperature-humidity index were associated with each record to evaluate the effect of heat stress conditions on milk-related traits through linear mixed models. Least squares means were estimated to evaluate the effect of THI and, separately, of MTHI on milk characteristics correcting for conventional systematic factors. Overall, heat stress conditions lowered the quality of both bulk milk and test-day records, with fat and protein content being greatly reduced, and somatic cell score and differential somatic cell count augmented. Milk yield was not affected by either THI or MTHI in this data set, but the effect of elevated THI and MTHI was in general stronger on test-day records than on bulk milk. Farm-level economic losses of reduced milk quality rather than reduced yield as consequence of elevated THI or MTHI was estimated to be between $23.57 and $43.98 per farmer per day, which is of comparable magnitude to losses resulting from reduced production. Furthermore, MTHI was found to be a more accurate indicator of heat stress experienced by a cow, explaining more variability of traits compared with THI. The negative effect of heat stress conditions on quality traits commences at lower THI/MTHI values compared with milk yield. Thus, a progressive farmers' income loss due to climatic changes is already a reality and it is mainly due to deterioration of milk quality rather than quantity in the studied area.
In Southern Italy, buffalo (Bubalus bubalis) milk is mostly intended for the manufacture of Mozzarella di Bufala Campana Protected Denomination of Origin (PDO) cheese. Despite the economic boost of ...the last 2 decades, the buffalo farming system should be improved to maximize the efficiency of the dairy industry, improve yield and quality of milk and cheese, and work toward better animal welfare. Milk somatic cell count (SCC) is used worldwide as an indicator of udder health in individual milk and is useful for monitoring farm hygiene in bulk milk. Mastitis data are currently not available on a large scale in Italy; thus, SCC is essential for identifying animals with suspected udder infection and inflammation. Moreover, high milk SCC is associated with altered composition and acidity, and poor technological properties of milk. However, payment systems of the PDO area are based simply on the delivered volume of milk rather than on quality characteristics. Hence, currently there are no penalties for elevated SCC in bulk milk in the Italian buffalo dairy industry. In addition, SCC for buffalo milk is not mentioned by either the European Community regulations or the PDO protocol, evidencing a lack of rules for the maximum SCC limit. To provide a phenotypic characterization of SCC at the population level and to improve knowledge on buffalo milk quality, 876,299 test-day records of 70,156 buffaloes reared in the PDO area were analyzed. Data revealed that around 11% of herd-test-dates (≥5 animals sampled each) showed average milk SCC ≥400,000 cells/mL (i.e., above the threshold fixed by the European Community for bovine milk). This suggests that there is room to improve SCC at both the farm and individual level. Within first parity, more than 28 and 15% of lactations had average SCC ≥200,000 and ≥300,000 cells/mL, respectively. Both percentages increased with parity and were 39 and 25% in sixth parity, respectively. Supporting this, the proportion of lactations with average SCC ≥500,000 cells/mL increased from 6% in first parity to 12% in sixth parity. Milk yield and SCC were negatively correlated with each other, especially when SCC level was high. An ANOVA was carried out on test-day record milk yield and composition traits, with fixed effects of parity, lactation stage, class of somatic cell score (n = 6), month of calving, and their interactions; buffalo, herd-test-date, and residual were considered random effects. Significantly lower milk yield and lactose percentage were estimated in progressively higher classes of somatic cell score, whereas no significant differences were observed for fat and protein percentages. This is the first attempt to investigate milk SCC in a large data set of Italian dairy buffaloes. These findings may be helpful for defining reliable and effective SCC thresholds to be adopted whenever specific penalties for high SCC are included in milk payment systems. Finally, these results could be used in mastitis monitoring plans aiming to reduce SCC and udder issues at both the individual and farm levels in the Italian buffalo population.
High milk somatic cell count (SCC) influences milk production and quality; however, very little is known about the effect of low SCC on milk quality, especially in terms of mineral content and ...coagulation properties. Thus, the present study aimed to investigate the effects of somatic cell score (SCS), calculated as log2(SCC/100) + 3, on milk yield, composition (fat, crude protein, casein, lactose, milk urea nitrogen, protein fractions, and mineral contents), and coagulation properties of Brown Swiss, Holstein Friesian, and Simmental cows from multibreed herds. Milk composition and coagulation traits were predicted using mid-infrared spectroscopy. The data set comprised 95,591 observations of 6,940 cows in 313 multibreed herds, collected from January 2011 to December 2017. Observations were divided into 8 classes based on SCS. Statistical analysis was performed using a linear mixed model, which included breed, parity, stage of lactation, SCS class, and their interactions as fixed effects, and herd test day, cow, and residual as random effects. The probability that cows experienced SCS > 4.00 at 30 ± 5, 60 ± 5, or 90 ± 5 d after the observation test day was calculated for each SCS class, and odds ratios to the reference class (−1.00 < SCS ≤ 0.00) were reported. Results showed that the relationship between SCS and milk traits followed a third-order polynomial regression. The average loss of milk, fat, and crude protein yields were 0.43, 0.01, and 0.01 kg/d, respectively, for each SCS unit higher than 1.00. Very low SCS (<−1.00) had detrimental effects on milk yield and quality traits similar to or even stronger than high SCS (>4.00). Moreover, cows with SCS lower than −1.00 on a test day were about 7 times more likely to present high SCS within the following 90 ± 5 d than cows with SCS between −1.00 and 0.00. Breeds responded similarly to the increase of SCS, but the overall loss of fat and crude protein yields, and several minerals among Holstein Friesian were lower with increasing SCS. The best milk yield and quality were observed between SCS 0.00 and 1.00, but milk quality of Holstein Friesians started to decrease at lower SCS compared with milk quality of Brown Swiss and Simmental cows. Results suggest a breed-dependent optimum of SCS, and highlighted that very low SCS can be an indicator of udder health problems and, thus, may be used for early detection of mastitis.
