The diversity of identified phenolic compounds was different among ecotypes, allowing them to be distinguished by chemical diversity, free radical-scavenging capacities and heat Scoville units. The total capsaicinoid content was higher in Population I (23.5 mg/g DW) than in Populations II and III, which had contents of 15.3 and 10.7 mg/g DW, respectively. This variability could lead to phytochemical exploitation and the conservation of the natural populations of wild chili.
Systemic lupus erythematosus (SLE) is a clinically heterogeneous disease with limited reliable diagnostic biomarkers. We investigated whether gene methylation could meet sensitivity and specificity ...criteria for a robust biomarker.
IFI44L promoter methylation was examined using DNA samples from a discovery set including 377 patients with SLE, 358 healthy controls (HCs) and 353 patients with rheumatoid arthritis (RA). Two independent sets including 1144 patients with SLE, 1350 HCs, 429 patients with RA and 199 patients with primary Sjögren's syndrome (pSS) were used for validation.
Significant hypomethylation of two CpG sites within IFI44L promoter, Site1 (Chr1: 79 085 222) and Site2 (Chr1: 79 085 250; cg06872964), was identified in patients with SLE compared with HCs, patients with RA and patients with pSS. In a comparison between patients with SLE and HCs included in the first validation cohort, Site1 methylation had a sensitivity of 93.6% and a specificity of 96.8% at a cut-off methylation level of 75.5% and Site2 methylation had a sensitivity of 94.1% and a specificity of 98.2% at a cut-off methylation level of 25.5%. The IFI44L promoter methylation marker was also validated in an European-derived cohort. In addition, the methylation levels of Site1 and Site2 within IFI44L promoter were significantly lower in patients with SLE with renal damage than those without renal damage. Patients with SLE showed significantly increased methylation levels of Site1 and Site2 during remission compared with active stage.
The methylation level of IFI44L promoter can distinguish patients with SLE from healthy persons and other autoimmune diseases, and is a highly sensitive and specific diagnostic marker for SLE.
Lowest-energy structures, the distribution of isomers, and their molecular properties depend significantly on geometry and temperature. Total energy computations using DFT methodology are typically ...carried out at a temperature of zero K; thereby, entropic contributions to the total energy are neglected, even though functional materials work at finite temperatures. In the present study, the probability of the occurrence of one particular Be4B8 isomer at temperature T is estimated by employing Gibbs free energy computed within the framework of quantum statistical mechanics and nanothermodynamics. To identify a list of all possible low-energy chiral and achiral structures, an exhaustive and efficient exploration of the potential/free energy surfaces is carried out using a multi-level multistep global genetic algorithm search coupled with DFT. In addition, we discuss the energetic ordering of structures computed at the DFT level against single-point energy calculations at the CCSD(T) level of theory. The total VCD/IR spectra as a function of temperature are computed using each isomer’s probability of occurrence in a Boltzmann-weighted superposition of each isomer’s spectrum. Additionally, we present chemical bonding analysis using the adaptive natural density partitioning method in the chiral putative global minimum. The transition state structures and the enantiomer–enantiomer and enantiomer–achiral activation energies as a function of temperature evidence that a change from an endergonic to an exergonic type of reaction occurs at a temperature of 739 K.
Dogs with spontaneous tumors treated in veterinary hospitals offer an excellent opportunity for studying immunotherapies, including oncolytic viruses. Oncolytic viruses have advanced into the clinic ...as an intratumorally administered therapeutic; however, intravenous delivery has been hindered by neutralization in the blood. To circumvent this hurdle, mesenchymal stem cells have been used as a "Trojan horse." Here, we present the treatment of 27 canine patients with cancer with canine mesenchymal stem cells infected with ICOCAV17, a canine oncolytic adenovirus. No significant adverse effects were found. The response rate was 74%, with 14.8% showing complete responses, including total remissions of lung metastasis. We detected virus infection, stromal degeneration, and immune cell infiltration in tumor biopsies after 4 weeks of treatment. The increased presence of antiadenoviral antibodies in the peripheral blood of treated dogs did not appear to prevent the clinical benefit of this therapy. These data indicate that oncolytic viruses loaded in mesenchymal stem cells represent an effective cancer immunotherapy.
The classical clinical limitations of antitumoral viroimmunotherapy can be overcome by use of mesenchymal stem cells.

Abstract
Objective
To compare the clinical effectiveness of V‐bend bonded retainers (BR) versus vacuum‐formed retainers (VFR) regarding their capacity to maintain treatment stability and survival ...rates after 12 months.
Materials and Methods
Patients finishing orthodontic treatment were randomly allocated into two groups. The BR group received maxillary and mandibular BRs in the lingual surfaces of the anterior teeth. The VFR group received VFRs right after fixed appliances removal. The patients were evaluated at four time‐points: at fixed appliances removal (T0), after 3 (T1), 6 (T2) and 12 months (T3). In each time‐point digital models were obtained and analysed with the OrthoAnalyzer™ software. Treatment stability based on occlusal outcomes and retainers' survival rates were evaluated. Intergroup comparisons were performed using Mann–Whitney
U
‐tests. The Kaplan–Meier survival plot and the log‐rank test were employed to assess the retainers' survival.
Results
Both BR and VFR groups included 25 patients. The groups were comparable regarding their baseline characteristics. Up to 6 months, both retainers were equally effective; however, after 12 months, BRs were more effective in maintaining the incisors' alignment in the maxilla and the mandible compared to the VFRs. No differences were noticed in the intercanine and intermolar widths, overjet and overbite. There were no differences regarding the retainers' survivability in both arches.
Conclusions
BRs were more effective in maintaining the alignment of the incisors in the maxilla and mandible compared to VFRs after 12 months. Both retainers presented the same survival rates after the same period.
The use of mixtures of synthetic and natural polymers is a potential option to reduce the pollution by plastic waste. In this work, the method for the chemical modification of chitosan with ...poly(lactic acid) was developed; then, the preparation of films of blends of polyethylene and chitosan-poly(lactic acid) produced by an extrusion method using polyethylene-graft maleic anhydride as a compatibilizer. It was possible to obtain films with a maximum content of 20 wt% and 30 wt%, chitosan, with and without compatibilizer, respectively. Scanning electron microscope (SEM) analysis showed a homogeneous surface on all films. The addition of the compatibilizer had a significant effect on the mechanical properties of the films, such as an increase in Young's modulus and a decrease in the elongation at break; additionally, the compatibilizer promotes thermal degradation in a single step and gives the film a slight increase in thermal resistance. These results are attributed to an improved interaction in the interface of polyethylene and chitosan-poly(lactic acid), promoted by the compatibilizer.
Adequate testing is critically important for control of the SARS-CoV-2 pandemic. Antibody testing is an option for case management and epidemiologic studies, with high specificity and variable ...sensitivity. However, characteristics of local populations may affect performance of these tests. For this reason, the National Institute of Health (INS) and regulatory agencies in Colombia require verification of diagnostic accuracy of tests introduced to the Colombian market. We conducted a validation study of the Abbott SARS-CoV-2 test for qualitative detection of IgG using the Abbott Architect i2000SR. Participants and retrospective samples were included from patients with suspected SARS-CoV-2 infection, age greater than or equal to18 years, and greater than or equal to8 days elapsed since initiation of symptoms. Pre-pandemic plasma samples (taken before October 2019) were used as controls. We estimated the sensitivity, specificity and agreement (kappa) of the Abbott IgG test compared to the gold standard (RT-PCR). The overall sensitivity was 83.1% (95% CI: 75.4-100). Sensitivity among patients with greater than or equal to14 days since the start of symptoms was 85.7%, reaching 88% in samples collected from patients with COVID-19 symptoms onset >60 days. Specificity was 100% and the kappa index of agreement was 0.804 (95% CI: 0.642-0.965). Our findings show high sensitivity and specificity of the Abbott IgG test in a Colombian population, which meet the criteria set by the Colombian INS to aid in the diagnosis of COVID-19. Data from our patient groups also suggest that IgG response is detectable in a high proportion of individuals (88.1%) during the first two months following onset of symptoms.

Photothermal nanocomposite hydrogels are promising materials for remotely triggering drug delivery by near-infrared (NIR) radiation stimuli. In this work, a novel hydrogel based on poly(vinyl ...alcohol), poly(vinyl methyl ether-alt-maleic acid), poly(vinyl methyl ether), and functionalized multiwalled carbon nanotubes (MWCNT-f) was prepared by the freeze/thaw method. A comparative characterization of materials (with and without MWCNT-f) was carried out by infrared spectroscopy, differential scanning calorimetry, scanning electron microscopy, mechanical assays, swelling kinetics measurements, and photothermal analysis under NIR irradiation. Hydrophilic chemotherapeutic 5-fluorouracil (5-FU) and hydrophobic ibuprofen drugs were independently loaded into hydrogels, and the drug release profiles were obtained under passive and NIR-irradiation conditions. The concentration-dependent cytotoxicity of materials was studied in vitro using noncancerous cells and cancer cells. Notable changes in the microstructure and physicochemical properties of hydrogels were observed by adding a low content (0.2 wt %) of MWCNT-f. The cumulative release amounts of 5-FU and ibuprofen from the hydrogel containing MWCNT-f were significantly increased by 21 and 39%, respectively, through the application of short-term NIR irradiation pulses. Appropriate concentrations of the nanocomposite hydrogel loaded with 5-FU produced cytotoxicity in cancer cells without affecting noncancerous cells. The overall properties of the MWCNT-f-containing hydrogel and its photothermal behavior make it an attractive material to promote the release of hydrophilic and hydrophobic drugs, depending on the treatment requirements.
