Lung macrophage subpopulations have been identified based on size. We investigated characteristics of small and large macrophages in the alveolar spaces and lung interstitium of COPD patients and ...controls. Alveolar and interstitial cells were isolated from lung resection tissue from 88 patients. Macrophage subpopulation cell-surface expression of immunological markers and phagocytic ability were assessed by flow cytometry. Inflammatory related gene expression was measured. Alveolar and interstitial macrophages had subpopulations of small and large macrophages based on size and granularity. Alveolar macrophages had similar numbers of small and large cells; interstitial macrophages were mainly small. Small macrophages expressed significantly higher cell surface HLA-DR, CD14, CD38 and CD36 and lower CD206 compared to large macrophages. Large alveolar macrophages showed lower marker expression in COPD current compared to ex-smokers. Small interstitial macrophages had the highest pro-inflammatory gene expression levels, while large alveolar macrophages had the lowest. Small alveolar macrophages had the highest phagocytic ability. Small alveolar macrophage CD206 expression was lower in COPD patients compared to smokers. COPD lung macrophages include distinct subpopulations; Small interstitial and small alveolar macrophages with more pro-inflammatory and phagocytic function respectively, and large alveolar macrophages with low pro-inflammatory and phagocytic ability.
Previous research demonstrating that positive episodic simulation enhances future expectancies has relied on explicit expectancy measures. The current study investigated the effects of episodic ...simulation on implicit expectancies. Using the Future Thinking Implicit Relational Assessment Procedure (FT-IRAP), participants made true/false decisions to indicate whether or not they expected positive/negative outcomes after adopting orientations consistent or inconsistent with an optimistic disposition. The outcome measure, DIRAP, was based on response time differences between consistent and inconsistent blocks. Participants then engaged in either positive simulation training, in which they imagined positive future events, or a neutral visualisation task before repeating the FT-IRAP twice following 10-minute intervals. Positive simulation training increased DIRAP scores for don't-expect-negative trials-boosting participants' readiness to affirm that negative events were unlikely to happen to them. Although findings did not generalise across all trial types, they show potential for positive simulation training to enhance implicit future expectancies.

Abstract
Dietary niche is fundamental for determining species ecology; thus, a detailed understanding of what drives variation in dietary niche is vital for predicting ecological shifts and could ...have implications for species management. Gut microbiota can be important for determining an organism’s dietary preference, and therefore which food resources they are likely to exploit. Evidence for whether the composition of the gut microbiota is plastic in response to changes in diet is mixed. Also, the extent to which dietary preference can be changed following colonisation by new gut microbiota from different species is unknown. Here, we use
Drosophila spp.
to show that: (1) the composition of an individual’s gut microbiota can change in response to dietary changes, and (2) ingestion of foreign gut microbes can cause individuals to be attracted to food types they previously had a strong aversion to. Thus, we expose a mechanism for facilitating rapid shifts in dietary niche over short evolutionary timescales.
Laser wakefield accelerators (LWFAs) can produce high-energy electron bunches in short distances. Successfully coupling these sources with undulators has the potential to form an LWFA-driven ...free-electron laser (FEL), providing high-intensity short-wavelength radiation. Electron bunches produced from LWFAs have a correlated distribution in longitudinal phase space: a chirp. However, both LWFAs and FELs have strict parameter requirements. The bunch chirp created using ideal LWFA parameters may not suit the FEL; for example, a chirp can reduce the high peak current required for free-electron lasing. We, therefore, design a flexible beamline that can accept either positively or negatively chirped LWFA bunches and adjust the chirp during transport to an undulator. We have used the accelerator design program MAD8 to design a beamline in stages, and to track particle bunches. The final beamline design can produce ambidirectional values of longitudinal dispersion (Formula: see text): we demonstrate values of + 0.20 mm, 0.00 mm and - 0.22 mm. Positive or negative values of Formula: see text apply a shear forward or backward in the longitudinal phase space of the electron bunch, which provides control of the bunch chirp. This chirp control during the bunch transport gives an additional free parameter and marks a new approach to matching future LWFA-driven FELs.
Data storage at the UK Tier-1 facility at RAL is provided through its ECHO storage, serving the requirements for the WLGC and increasing numbers of other HEP and astronomy related communities. ECHO ...is a Cephbacked erasure-coded object store, currently providing in excess of 40PB of usable space, with frontend access to data provided via XRootD or GridFTP, using the libradosstriper library of Ceph. The storage must service the needs of: high-throughput compute, with staged and direct file access passing through an XCache on each workernode; data access to compute running at storageless satellite sites; and, managed inter-site data transfers using the recently adopted HTTPS protocol (via WebDav), which includes multi-hop data transfers to and from RAL’s newly commissioned CTA tape endpoint. A review of the experiences of providing data access via an object store within these data workflows is presented, including the details of the improvements necessary for the transition to WebDav, used for most inter-site data movements, and enhancements for direct-IO file access, where the development and optimisation of buffering and vector read strategies is explored.

Two experiments used a dual task methodology to investigate the role of visual imagery and executive resources in the retrieval of specific autobiographical memories. In Experiment 1, dynamic visual noise led to a reduction in the number of specific memories retrieved in response to both high and low imageability cues, but did not affect retrieval times. In Experiment 2, irrelevant pictures reduced the number of specific memories but only in response to low imageability cues. Irrelevant pictures also increased response times to both high and low imageability cues. The findings are in line with previous work suggesting that disrupting executive resources may impair generative, but not direct, retrieval of autobiographical memories. In contrast, visual distractor tasks appear to impair access to specific autobiographical memories via both the direct and generative retrieval routes, thereby highlighting the potential role of visual imagery in both pathways.

More than one thousand physicists analyse data collected by the ATLAS experiment at the Large Hadron Collider (LHC) at CERN through 150 computing facilities around the world. Efficient distributed ...analysis requires optimal resource usage and the interplay of several factors: robust grid and software infrastructures, and system capability to adapt to different workloads. The continuous automatic validation of grid sites and the user support provided by a dedicated team of expert shifters have been proven to provide a solid distributed analysis system for ATLAS users. Typical user workflows on the grid, and their associated metrics, are discussed. Measurements of user job performance and typical requirements are also shown.

The Science and Technology Facilities Council (STFC) datacentre provides large- scale High Performance Computing facilities for the scientific community. It currently consumes approximately 1.5MW and ...this has risen by 25% in the past two years. STFC has been investigating leveraging preemption in the Tier 1 batch farm to save power. HEP experiments are increasing using jobs that can be killed to take advantage of opportunistic CPU resources or novel cost models such as Amazon's spot pricing. Additionally, schemes from energy providers are available that offer financial incentives to reduce power consumption at peak times. Under normal operating conditions, 3% of the batch farm capacity is wasted due to draining machines. By using preempt-able jobs, nodes can be rapidly made available to run multicore jobs without this wasted resource. The use of preempt-able jobs has been extended so that at peak times machines can be hibernated quickly to save energy. This paper describes the implementation of the above and demonstrates that STFC could in future take advantage of such energy saving schemes.
Depression is associated with difficulties in goal pursuit which could be related to deficits in emotional anticipation regarding goal success. Therefore, the reported study investigated emotional ...anticipation for personal goals and whether this differs as a function of depressive symptoms. After listing approach and avoidance goals, 263 participants made predictions about these goals (e.g. likelihood of achievement and controllability) and rated the vividness and perspective with which they envisaged goal achievement. They also provided ratings of either anticipated (predicted emotions that would accompany goal success) or anticipatory (in-the-moment emotions when imagining goal success) positive emotions. Higher levels of depressive symptomatology were associated with pessimistic predictions about goal achievement, coupled with reduced vividness and greater adoption of observer perspective when envisaging achievement. Furthermore, those experiencing higher levels of depressive symptoms evidenced biases in both anticipated and anticipatory positive emotions associated with goal success. They believed that goal achievement would bring them lower levels of positive emotion and also reported less in-the-moment happiness, satisfaction, and pleasure when thinking about achieving their goals. Irrespective of depressive symptom level, anticipated emotions were generally stronger than anticipatory emotions. These findings have implications both for research on future-oriented emotions (anticipated and anticipatory) and for the development of therapeutic techniques to aid depression.
