Depositing large components (>10 kg) in titanium, aluminium, steel and other metals is possible using Wire + Arc Additive Manufacturing. This technology adopts arc welding tools and wire as feedstock ...for additive manufacturing purposes. High deposition rates, low material and equipment costs, and good structural integrity make Wire+Arc Additive Manufacturing a suitable candidate for replacing the current method of manufacturing from solid billets or large forgings, especially with regards to low and medium complexity parts. A variety of components have been successfully manufactured with this process, including Ti-6Al-4V spars and landing gear assemblies, aluminium wing ribs, steel wind tunnel models and cones. Strategies on how to manage residual stress, improve mechanical properties and eliminate defects such as porosity are suggested. Finally, the benefits of non-destructive testing, online monitoring and in situ machining are discussed.

The worm Caenorhabditis elegans is a model system for studying many aspects of biology, including host responses to bacterial pathogens, but it is not known to support replication of any virus. ...Plants and insects encode multiple Dicer enzymes that recognize distinct precursors of small RNAs and may act cooperatively. However, it is not known whether the single Dicer of worms and mammals is able to initiate the small RNA-guided RNA interference (RNAi) antiviral immunity as occurs in plants and insects. Here we show complete replication of the Flock house virus (FHV) bipartite, plus-strand RNA genome in C. elegans. We show that FHV replication in C. elegans triggers potent antiviral silencing that requires RDE-1, an Argonaute protein essential for RNAi mediated by small interfering RNAs (siRNAs) but not by microRNAs. This immunity system is capable of rapid virus clearance in the absence of FHV B2 protein, which acts as a broad-spectrum RNAi inhibitor upstream of rde-1 by targeting the siRNA precursor. This work establishes a C. elegans model for genetic studies of animal virus-host interactions and indicates that mammals might use a siRNA pathway as an antiviral response.

In antiviral RNA interference (RNAi), the DICER enzyme processes virus-derived double-stranded RNA (dsRNA) into small interfering RNAs (siRNAs) that guide ARGONAUTE proteins to silence complementary ...viral RNA. As a counterdefense, viruses deploy viral suppressors of RNAi (VSRs). Well-established in plants and invertebrates, the existence of antiviral RNAi remains unknown in mammals. Here, we show that undifferentiated mouse cells infected with encephalomyocarditis virus (EMCV) or Nodamura virus (NoV) accumulate ~22-nucleotide RNAs with all the signature features of siRNAs. These derive from viral dsRNA replication intermediates, incorporate into AGO2, are eliminated in Dicer knockout cells, and decrease in abundance upon cell differentiation. Furthermore, genetically ablating a NoV-encoded VSR that antagonizes DICER during authentic infections reduces NoV accumulation, which is rescued in RNAi-deficient mouse cells. We conclude that antiviral RNAi operates in mammalian cells.

A proof of concept integrating binary p–n heterojunctions into a semiconductor hybrid photocatalyst is demonstrated by non-covalent doping of graphite-like carbon nitride (g-C3N4) with ultrathin GO and MoS2 nanosheets using a facile sonochemical method. In this unique ternary hybrid, the layered MoS2 and GO nanosheets with a large surface area enhance light absorption to generate more photoelectrons. On account of the coupling between MoS2 and GO with g-C3N4, the ternary hybrid possesses binary p–n heterojunctions at the g-C3N4/MoS2 and g-C3N4/GO interfaces. The space charge layers created by the p–n heterojunctions not only enhance photogeneration, but also promote charge separation and transfer of electron–hole pairs. In addition, the ultrathin MoS2 and GO with high mobility act as electron mediators to facilitate separation of photogenerated electron–hole pairs at each p–n heterojunction. As a result, the ternary hybrid photocatalyst exhibits improved photoelectrochemical and photocatalytic activity under visible light irradiation compared to other reference materials. The results provide new insights into the large-scale production of semiconductor photocatalysts.
We clarify the physical origin of the dc voltage generation in a bilayer of a conducting polymer film and a micrometer-thick magnetic insulator Y_{3}Fe_{5}O_{12} (YIG) film under ferromagnetic ...resonance and/or spin wave excitation conditions. The previous attributed mechanism, the inverse spin Hall effect in the polymer Nat. Mater. 12, 622 (2013)NMAACR1476-112210.1038/nmat3634, is excluded by two control experiments. We find an in-plane temperature gradient in YIG which has the same angular dependence with the generated voltage. Both vanish when the YIG thickness is reduced to a few nanometers. Thus, we argue that the dc voltage is governed by the Seebeck effect in the polymer, where the temperature gradient is created by the nonreciprocal magnetostatic surface spin wave propagation in YIG.
This paper presents an experimental investigation of the fracture and fatigue crack growth properties of Ti‐6Al‐4V produced by the Wire + Arc Additive Manufacture (WAAM®) process. First, fracture ...toughness was measured for two different orientations with respect to the build direction; the effect of wire oxygen content and build strategy were also evaluated in the light of microstructure examination. Second, fatigue crack growth rates were measured for fully additive manufactured samples, as well as for samples containing an interface between WAAM® and wrought materials. The latter category covers five different scenarios of crack location and orientation with respect to the interface. Fatigue crack growth rates are compared with that of the wrought or WAAM® alone conditions. Crack growth trajectory of these tests is discussed in relation to the microstructure characteristics.
This study provides a new approach to estimate both anthropogenic non-point-source and point-source nitrogen (N) inputs to the landscape, and determines their impacts on riverine ammonia–nitrogen ...(AN) flux, providing a foundation for further exploration of anthropogenic effects on N pollution. Our study site is Huai River basin of China, a water–shed with one of the highest levels of N input in the world. Multi-year average (2003–2010) inputs of N to the watershed are 27 200 ± 1100 kg N km−2 yr−1. Non-point sources comprised about 98 % of total N input, and only 2 % of inputs are directly added to the aquatic ecosystem as point sources. Fertilizer application was the largest non-point source of new N to the Huai River basin (69 % of net anthropogenic N inputs), followed by atmospheric deposition (20 %), N fixation in croplands (7 %), and N content of imported food and feed (2 %). High N inputs showed impacts on riverine AN flux: fertilizer application, point-source N input, and atmospheric N deposition were proved as more direct sources to riverine AN flux. Modes of N delivery and losses associated with biological denitrification in rivers, water consumption, interception by dams may influence the extent of export of riverine AN flux from N sources. Our findings highlight the importance of anthropogenic N inputs from both point sources and non-point sources in heavily polluted watersheds, and provide some implications for AN prediction and management.
Cell-free DNA in plasma has been used for noninvasive prenatal testing and cancer liquid biopsy. The physical properties of cell-free DNA fragments in plasma, such as fragment sizes and ends, have ...attracted much recent interest, leading to the emerging field of cell-free DNA fragmentomics. However, one aspect of plasma DNA fragmentomics as to whether double-stranded plasma molecules might carry single-stranded ends, termed a jagged end in this study, remains underexplored. We have developed two approaches for investigating the presence of jagged ends in a plasma DNA pool. These approaches utilized DNA end repair to introduce differential methylation signals between the original sequence and the jagged ends, depending on whether unmethylated or methylated cytosines were used in the DNA end-repair procedure. The majority of plasma DNA molecules (87.8%) were found to bear jagged ends. The jaggedness varied according to plasma DNA fragment sizes and appeared to be in association with nucleosomal patterns. In the plasma of pregnant women, the jaggedness of fetal DNA molecules was higher than that of the maternal counterparts. The jaggedness of plasma DNA correlated with the fetal DNA fraction. Similarly, in the plasma of cancer patients, tumor-derived DNA molecules in patients with hepatocellular carcinoma showed an elevated jaggedness compared with nontumoral DNA. In mouse models, knocking out of the
gene reduced jaggedness, whereas knocking out of the
gene enhanced jaggedness. Hence, plasma DNA jagged ends represent an intrinsic property of plasma DNA and provide a link between nuclease activities and the fragmentation of plasma DNA.
We report on the highest energy photons from the Crab Nebula observed by the Tibet air shower array with the underground water-Cherenkov-type muon detector array. Based on the criterion of a muon ...number measured in an air shower, we successfully suppress 99.92% of the cosmic-ray background events with energies E>100 TeV. As a result, we observed 24 photonlike events with E>100 TeV against 5.5 background events, which corresponds to a 5.6σ statistical significance. This is the first detection of photons with E>100 TeV from an astrophysical source.
Long-distance quantum communication and networking require quantum memory nodes with efficient optical interfaces and long memory times. We report the realization of an integrated two-qubit network ...node based on silicon-vacancy centers (SiVs) in diamond nanophotonic cavities. Our qubit register consists of the SiV electron spin acting as a communication qubit and the strongly coupled silicon-29 nuclear spin acting as a memory qubit with a quantum memory time exceeding 2 seconds. By using a highly strained SiV, we realize electron-photon entangling gates at temperatures up to 1.5 kelvin and nucleus-photon entangling gates up to 4.3 kelvin. We also demonstrate efficient error detection in nuclear spin–photon gates by using the electron spin as a flag qubit, making this platform a promising candidate for scalable quantum repeaters.
A robust quantum network node
The ability to develop quantum networks and communicate quantum information over long distances requires quantum memory nodes with efficient optical interfaces and long memory times. Because of their long coherence times and efficient optical interface, color centers in diamond are promising candidates to achieve this goal. Stas
et al
. use silicon vacancies in diamond and integrated the properties into a single device (see the Perspective by Gangloff). The authors demonstrate a quantum memory with a lifetime exceeding 2 seconds and full optical control of the quantum states of the two-qubit register. With the capability of built-in error detection, such a platform is promising for the development of scalable quantum networks. —ISO
A quantum network node combining a long-lived memory, robust optical interface, and integrated error detection is realized.
