Cortical iron has been shown to be elevated in Alzheimer's disease (AD), but the impact of the directly measured iron on the clinical syndrome has not been assessed. We investigated the association ...between post-mortem iron levels with the clinical and pathological diagnosis of AD, its severity, and the rate of cognitive decline in the 12 years prior to death in subjects from the Memory and Aging Project (n = 209). Iron was elevated (β SE = 9.7 2.6; P = 3.0 × 10
) in the inferior temporal cortex only in subjects who were diagnosed with clinical AD during life and had a diagnosis of AD confirmed post-mortem by standardized criteria. Although iron was weakly associated with the extent of proteinopathy in tissue with AD neuropathology, it was strongly associated with the rate of cognitive decline (e.g., global cognition: β SE = -0.040 0.005, P = 1.6 × 10
). Thus, cortical iron might act to propel cognitive deterioration upon the underlying proteinopathy of AD, possibly by inducing oxidative stress or ferroptotic cell death, or may be related to an inflammatory response.
The mechanisms leading to neurodegeneration in Alzheimer's disease (AD) may involve oxidative stress and neuroinflammation. Ceruloplasmin (Cp) is a circulating protein that intersects both these ...pathways, since its expression is increased during the acute phase response, and the protein acts to lower pro-oxidant iron in cells. Since the role of Cp in AD, and its potential for use as a biomarker is not established, we investigated CSF Cp and its association with longitudinal outcome measures related to AD.
This was an observational study of 268 people from the Alzheimer's Disease Neuroimaging (ADNI) cohort. Subjects were classified clinically as having AD, mild cognitive impairment (MCI) or were cognitively normal (CN), and were also classified as being positive for β-amyloid using established thresholds in the CSF t-tau/Aβ42 ratio. Subjects underwent cognitive tests and MRI studies every 6 months for 2 years, then yearly thereafter for up to 6 years.
At baseline, CSF Cp was not associated with clinical or pathological diagnosis, but we found an unexpected association between CSF Cp and levels of CSF apolipoprotein E. In longitudinal analysis, high level of CSF Cp was associated with accelerated cognitive decline (as assessed by ADAS-Cog, CDR-SB, and MMSE) and ventricular volume enlargement in people classified as MCI and who had underlying β-amyloid pathology.
These results raise new questions into the role of Cp in neuroinflammation, oxidative stress, and APOE pathways involved in AD, and reveal the potential for this protein to be used as a biomarker in disease prognostication.
•CSF ceruloplasmin is correlated with CSF apolipoprotein E levels.•In people with Aβ pathology, CSF ceruloplasmin predicts future cognitive decline.•CSF ceruloplasmin predicts future brain atrophy upon high Aβ pathology.
The aim of this study is to find the most suitable heat wave definition among 15 different ones and to evaluate its impact on total, age-, and gender-specific mortality for Bandafassi, Senegal.
Daily ...weather station data were obtained from Kedougou situated at 17 km from Bandafassi from 1973 to 2012. Poisson generalized additive model (GAM) and distributed lag non-linear model (DLNM) are used to investigate the effect of heat wave on mortality and to evaluate the nonlinear association of heat wave definitions at different lag days, respectively.
Heat wave definitions, based on three or more consecutive days with both daily minimum and maximum temperatures greater than the 90th percentile, provided the best model fit. A statistically significant increase in the relative risk (RRs 1.4 (95% Confidence Interval (CI): 1.2-1.6), 1.7 (95% CI: 1.5-1.9), 1.21 (95% CI: 1.08-1.3), 1.2 (95% CI: 1.04-1.5), 1.5 (95% CI: 1.3-1.8), 1.4 (95% CI: 1.2-1.5), 1.5 (95% CI: 1.07-1.6), and 1.5 (95% CI: 1.3-1.8)) of total mortality was observed for eight definitions. By using the definition based on the 90th percentile of minimum and maximum temperature with a 3-day duration, we also found that females and people aged ≥ 55 years old were at higher risks than males and other different age groups to heat wave related mortality.
The impact of heat waves was associated with total-, age-, gender-mortality. These results are expected to be useful for decision makers who conceive of public health policies in Senegal and elsewhere. Climate parameters, including temperatures and humidity, could be used to forecast heat wave risks as an early warning system in the area where we conduct this research. More broadly, our findings should be highly beneficial to climate services, researchers, clinicians, end-users and decision-makers.

β-Amyloid pathology is elevated in ~30% of cognitively normal people over 65, and is associated with accelerated neurodegeneration in the pre-clinical stages of Alzheimer's disease. Recent findings ...reveal that brain iron might also act to propel neurodegeneration in people with underlying amyloid pathology. Here, repeated PET scans of fluorodeoxyglucose (FDG) were used as a biomarker for brain hypometabolism and a downstream biomarker of neurodegeneration to investigate whether levels of ferritin in the cerebrospinal fluid (CSF; a reporter of brain iron load) are associated with prodromal disease progression of people with high β-amyloid pathology determined by established cut-off values in CSF t-tau/Aβ42 ratio. Nineteen cognitively normal participants with low t-tau/Aβ42, and 71 participants with high t-tau/Aβ42 who were cognitively normal or had mild cognitive impairment were included as participants from the Alzheimer's Disease Neuroimaging Initiative (ADNI) study. These subjects had repeated FDG-PET scans at 6-month intervals for 2 years, and yearly intervals for up to a further 3 years. In mixed-effects linear models of FDG signal, baseline CSF ferritin was associated with an accelerated decline in FDG PET in high t-tau/Aβ42 participants (βSE = −0.066 0.017; P = .0002), but not in people with low t-tau/Aβ42 (−0.029 0.049; P = .554). These data implicate iron as a contributing factor to neurodegeneration associated with β-amyloid pathology, and highlight CSF ferritin as a complementary prognostic biomarker to the t-tau/Aβ42 ratio that predicts near-term risk for disease progression.

This study examines the relationship between climate change and public health in Haiti, a country already facing socioeconomic challenges. The well-being of Haiti’s vulnerable population is expected ...to be further affected by climate change, leading to an increase in vector-borne, water-borne, and heat-related diseases. As one of the most vulnerable countries to climate change effects, Haiti is currently experiencing an increase in vector-borne diseases such as malaria, dengue, and chikungunya, as well as water-borne diseases and emerging zoonotic outbreaks. This study aims to improve planning, decision-making, and responses to public health challenges by utilizing health data, climatic information, and impact models. The methodology involves the creation of a comprehensive climate and health database to uncover detailed spatial-temporal relationships on a national scale. By evaluating disease indicators from historical periods (1950-2014) and future projections (2015-2100) using the Shared Socio-Economic Pathways (SSPs) from the multi-model ensemble mean of the CMIP6 models, target diseases, including malaria, meningitis, dengue, and heat-sensitive chronic diseases are assessed. Our results highlight a decrease in rainfall and a strong increase in temperatures, especially within western Haiti under the extreme SSP585 scenario. The ability of the impact models to simulate the seasonality and spatial distribution of malaria incidence, dengue and heatwaves was performed. The analysis of risks related to climate-sensitive diseases’ climatic parameters shows that Haiti’s west and central regions are mostly exposed to vector-borne and water-borne diseases. Models predict a decrease in malaria cases due to climate change with hot temperatures and a decline in rainfall, while dengue transmission patterns may undergo changes. These findings will inform the implementation of context-specific early-warning systems and adaptation strategies for climate-sensitive diseases while acknowledging the challenges of integrating climate-altered data into health policies.
Several vector-borne diseases, such as malaria, are sensitive to climate and weather conditions. When unusual conditions prevail, for example, during periods of heavy rainfall, mosquito populations ...can multiply and trigger epidemics. This study, which consists of better understanding the link between malaria transmission and climate factors at a national level, aims to validate the VECTRI model (VECtor borne disease community model of ICTP, TRIeste) in Senegal. The VECTRI model is a grid-distributed dynamical model that couples a biological model for the vector and parasite life cycles to a simple compartmental Susceptible-Exposed-Infectious-Recovered (SEIR) representation of the disease progression in the human host. In this study, a VECTRI model driven by reanalysis data (ERA-5) was used to simulate malaria parameters, such as the entomological inoculation rate (EIR) in Senegal. In addition to the ERA5-Land daily reanalysis rainfall, other daily rainfall data come from different meteorological products, including the CPC Global Unified Gauge-Based Analysis of Daily Precipitation (CPC for Climate Prediction Center), satellite data from the African Rainfall Climatology 2.0 (ARC2), and the Climate Hazards InfraRed Precipitation with Station data (CHIRPS). Observed malaria data from the National Malaria Control Program in Senegal (PNLP/Programme National de Lutte contre le Paludisme au Senegal) and outputs from the climate data used in this study were compared. The findings highlight the unimodal shape of temporal malaria occurrence, and the seasonal malaria transmission contrast is closely linked to the latitudinal variation of the rainfall, showing a south–north gradient over Senegal. This study showed that the peak of malaria takes place from September to October, with a lag of about one month from the peak of rainfall in Senegal. There is an agreement between observations and simulations about decreasing malaria cases on time. These results indicate that the southern area of Senegal is at the highest risk of malaria spread outbreaks. The findings in the paper are expected to guide community-based early-warning systems and adaptation strategies in Senegal, which will feed into the national malaria prevention, response, and care strategies adapted to the needs of local communities.
The saliva of haematophagous arthropods contains an array of anti-haemostatic, anti-inflammatory and immunomodulatory molecules that contribute to the success of the blood meal. The saliva of ...haematophagous arthropods is also involved in the transmission and the establishment of pathogens in the host and in allergic responses. This survey provides a comprehensive overview of the pharmacological activity and immunogenic properties of the main salivary proteins characterised in various haematophagous arthropod species. The potential biological and epidemiological applications of these immunogenic salivary molecules will be discussed with an emphasis on their use as biomarkers of exposure to haematophagous arthropod bites or vaccine candidates that are liable to improve host protection against vector-borne diseases.
Understanding the regional impact of future climate change is one of the major global challenges of this century. This study investigated possible effects of climate change on malaria in West Africa ...in the near future (2006–2035) and the far future (2036–2065) under two representative concentration pathway (RCP) scenarios (RCP4.5 and RCP8.5), compared to an observed evaluation period (1981–2010). Projected rainfall and temperature were obtained from the coordinated regional downscaling experiment (CORDEX) simulations of the Rossby Centre Regional Atmospheric regional climate model (RCA4). The malaria model used is the Liverpool malaria model (LMM), a dynamical malaria model driven by daily time series of rainfall and temperature obtained from the CORDEX data. Our results highlight the unimodal shape of the malaria prevalence distribution, and the seasonal malaria transmission contrast is closely linked to the latitudinal variation of the rainfall. Projections showed that the mean annual malaria prevalence would decrease in both climatological periods under both RCPs but with a larger magnitude of decreasing under the RCP8.5. We found that the mean malaria prevalence for the reference period is greater than the projected prevalence for 6 of the 8 downscaled GCMs. The study enhances understanding of how malaria is impacted under RCP4.5 and RCP8.5 emission scenarios. These results indicate that the southern area of West Africa is at most risk of epidemics, and the malaria control programs need extra effort and help to make the best use of available resources by stakeholders.
