The average human genome contains a small cohort of active L1 retrotransposons that encode two proteins (ORF1p and ORF2p) required for their mobility (i.e., retrotransposition). Prior studies ...demonstrated that human ORF1p, L1 RNA, and an ORF2p-encoded reverse transcriptase activity are present in ribonucleoprotein (RNP) complexes. However, the inability to physically detect ORF2p from engineered human L1 constructs has remained a technical challenge in the field. Here, we have employed an epitope/RNA tagging strategy with engineered human L1 retrotransposons to identify ORF1p, ORF2p, and L1 RNA in a RNP complex. We next used this system to assess how mutations in ORF1p and/or ORF2p impact RNP formation. Importantly, we demonstrate that mutations in the coiled-coil domain and RNA recognition motif of ORF1p, as well as the cysteine-rich domain of ORF2p, reduce the levels of ORF1p and/or ORF2p in L1 RNPs. Finally, we used this tagging strategy to localize the L1-encoded proteins and L1 RNA to cytoplasmic foci that often were associated with stress granules. Thus, we conclude that a precise interplay among ORF1p, ORF2p, and L1 RNA is critical for L1 RNP assembly, function, and L1 retrotransposition.

L1 retrotransposons express proteins (ORF1p and ORF2p) that preferentially mobilize their encoding RNA in cis, but they also can mobilize Alu RNA and, more rarely, cellular mRNAs in trans. Although ...these RNAs differ in sequence, each ends in a 3′ polyadenosine (poly(A)) tract. Here, we replace the L1 polyadenylation signal with sequences derived from a non-polyadenylated long non-coding RNA (MALAT1), which can form a stabilizing triple helix at the 3′ end of an RNA. L1/MALAT RNAs accumulate in cells, lack poly(A) tails, and are translated; however, they cannot retrotranspose in cis. Remarkably, the addition of a 16 or 40 base poly(A) tract downstream of the L1/MALAT triple helix restores retrotransposition in cis. The presence of a poly(A) tract also allows ORF2p to bind and mobilize RNAs in trans. Thus, a 3′ poly(A) tract is critical for the retrotransposition of sequences that comprise approximately one billion base pairs of human DNA.

Doucet et al. demonstrate that a 3′ poly(A) tract is required for human LINE-1-mediated retrotransposition. The ORF2p LINE-1-encoded protein preferentially associates with 3′ poly(A) tracts in LINE-1 and Alu RNAs, which helps explain their abundance in the human genome.
Rationnel L’étude prospective GERODIAB recherche un lien entre l’équilibre glycémique et la morbi-mortalité à 5 ans de diabétiques de type 2 de 70 ans et plus. Patients et méthodes En 2009-2010, 56 ...centres métropolitains ont recruté 987 diabétiques autonomes (femmes 52 %), âgés de 77 ± 5 ans (65 % ≥ 75 ans) et ayant un IMC de 30 ± 5 kg/m². L’ancienneté du diabète était de 18 ± 11 ans et l’HbA1c de 7,6 ± 1,3 % (60 ± 14mmol/mol) ; 90 % avaient une HTA et 85 % une anomalie lipidique. Les résultats (m ± SD, %) ont été comparés par analyse de variance et test du chi² ; les analyses multivariées ont été effectuées par régression linéaire ou modèle logistique. Résultats Moins de 1 % des patients avaient présenté une hyperosmolarité ou une cétose, 13 % une infection et 34 % une hypoglycémie dans les 6 mois précédents. La rétinopathie (26 %) était associée à l’ancienneté du diabète, à l’IMC, au score de dépression, aux hypoglycémies et à l’HbA1c ; la néphropathie (MDRD < 60 ml/min: 35 %), à l’âge, à l’ancienneté du diabète, à l’hypercholestérolémie, aux hypoglycémies, à l’HTA et à l’IMC. La coronaropathie (31 %) était associée à l’hypercholestérolémie, au score d’autonomie, au sexe masculin et à l’HTA ; l’insuffisance cardiaque (10 %) au tour de taille, à l’âge, et au HDL-cholestérol. L’artériopathie des membres inférieurs (26 %), à l’ancienneté du diabète, aux scores d’autonomie, à l’HTA et au sexe masculin ; l’ischémie cérébrale ; aux scores d’autonomie, au sexe et à l’HTA ; l’hypotension orthostatique (31 %), à l’HTA sévère. La neuropathie périphérique était liée à la PAD, à l’ancienneté du diabète et aux scores gériatriques. Les troubles cognitifs (29 %) étaient associés à l’âge, au sexe féminin, à l’HbA1c, à la neuropathie, à l’ischémie cérébrale et à l’insuffisance cardiaque. De grandes différences régionales étaient observées au détriment du Nord-Ouest et du Nord-Est. Conclusion GERODIAB a permis d’ores et déjà la description d’une population souvent ignorée des études cliniques. L’étude souligne l’intérêt des tests gériatriques systématiques. Objectif Décrire la fréquence et les facteurs associés aux hypoglycémies dans la cohorte observationnelle Gerodiab. Patients et méthodes Les hypoglycémies étaient définies par des événements ...symptomatiques et l’auto-surveillance (< 0,60 g/l = 3,3 mmol/l) dans les 6 mois précédant l’inclusion de 987 diabétiques français âgés de 70 ans et plus de la cohorte Gerodiab. Les résultats (moyenne ± SD, %) ont été analysés par les tests t de Student et du chi2 ; l’analyse multivariée a été réalisée par régression logistique. Résultats Une hypoglycémie au moins était observée chez 33,6 % des patients. Ils présentaient une plus grande ancienneté du diabète (21 ± 11 vs. 16 ± 10 ans ; P < 0,001) mais un âge semblable (77 ± 5 vs. 77 ± 5 ans ; NS). Ils avaient des valeurs plus faibles de l’IMC (29 ± 5 vs. 30 ± 5 kg/m2 ; P < 0,05), de la PAD (73 ± 11 vs. 75 ± 10 mmHg ; P < 0,05), du LDL-cholestérol (2,3 ± 0,8 vs. 2,6 ± 1,0 mmol/L ; P < 0,001), du MDRD (65 ± 22 vs. 69 ± 23 ml/min ; P < 0,01) mais non significativement inférieures de l’HbA1c (58 ± 11 vs. 60 ± 15 mmol/mol 7,5 ± 1,0 vs. 7,6 ± 1,4 % ; NS). Ils présentaient plus fréquemment une rétinopathie (36 vs. 21 % ; P < 0,001), une néphropathie (54 vs. 44 % ; P < 0,05) et une neuropathie périphérique (36 vs 24 % ; P < 0,01), mais une fréquence similaire des accidents coronariens ou vasculaires cérébraux et de l’insuffisance cardiaque. Les scores des échelles gériatriques MMSE, MNA, IADL et ADL étaient similaires dans les 2 groupes, mais le mini-GDS était plus souvent altéré (42 vs. 31 % ; P < 0,01). L’insuline était le seul traitement associé à une fréquence accrue des hypoglycémies (75 vs. 48 % ; P < 0,001). Avec le modèle logistique, l’hypoglycémie était associée successivement avec la rétinopathie (P < 0,01), le LDL-cholestérol et le mini-GDS (P < 0,05) pour une concordance globale de 44 % (P < 0,001). Conclusion Ces résultats soulignent la fréquence de l’association entre les hypoglycémies et la rétinopathie chez des diabétiques de type 2 âgés. Bien que son interprétation soit complexe, cette association confirme l’importance d’objectifs individualisés pour prévenir les hypoglycémies.
The idea that tropical forest and savanna are alternative states is crucial to how we manage these biomes and predict their future under global change. Large-scale empirical evidence for alternative ...stable states is limited, however, and comes mostly from the multimodal distribution of structural aspects of vegetation. These approaches have been criticized, as structure alone cannot separate out wetter savannas from drier forests for example, and there are also technical challenges to mapping vegetation structure in unbiased ways. Here, we develop an alternative approach to delimit the climatic envelope of the two biomes in Africa using tree species lists gathered for a large number of forest and savanna sites distributed across the continent. Our analyses confirm extensive climatic overlap of forest and savanna, supporting the alternative stable states hypothesis for Africa, and this result is corroborated by paleoecological evidence. Further, we find the two biomes to have highly divergent tree species compositions and to represent alternative compositional states. This allowed us to classify tree species as forest vs. savanna specialists, with some generalist species that span both biomes. In conjunction with georeferenced herbarium records, we mapped the forest and savanna distributions across Africa and quantified their environmental limits, which are primarily related to precipitation and seasonality, with a secondary contribution of fire. These results are important for the ongoing efforts to restore African ecosystems, which depend on accurate biome maps to set appropriate targets for the restored states but also provide empirical evidence for broad-scale bistability.
Mosquitoes develop long-lasting viral infections without substantial deleterious effects, despite high viral loads. This makes mosquitoes efficient vectors for emerging viral diseases with enormous ...burden on public health. How mosquitoes resist and/or tolerate these viruses is poorly understood. Here we show that two species of Aedes mosquitoes infected with two arboviruses from distinct families (dengue or chikungunya) generate a viral-derived DNA (vDNA) that is essential for mosquito survival and viral tolerance. Inhibition of vDNA formation leads to extreme susceptibility to viral infections, reduction of viral small RNAs due to an impaired immune response, and loss of viral tolerance. Our results highlight an essential role of vDNA in viral tolerance that allows mosquito survival and thus may be important for arbovirus dissemination and transmission. Elucidating the mechanisms of mosquito tolerance to arbovirus infection paves the way to conceptualize new antivectorial strategies to selectively eliminate arbovirus-infected mosquitoes.
LINE-1 (L1) retrotransposons represent approximately one sixth of the human genome, but only the human-specific L1HS-Ta subfamily acts as an endogenous mutagen in modern humans, reshaping both ...somatic and germline genomes. Due to their high levels of sequence identity and the existence of many polymorphic insertions absent from the reference genome, the transcriptional activation of individual genomic L1HS-Ta copies remains poorly understood. Here we comprehensively mapped fixed and polymorphic L1HS-Ta copies in 12 commonly-used somatic cell lines, and identified transcriptional and epigenetic signatures allowing the unambiguous identification of active L1HS-Ta copies in their genomic context. Strikingly, only a very restricted subset of L1HS-Ta loci - some being polymorphic among individuals - significantly contributes to the bulk of L1 expression, and these loci are differentially regulated among distinct cell lines. Thus, our data support a local model of L1 transcriptional activation in somatic cells, governed by individual-, locus-, and cell-type-specific determinants.
Large tropical trees and a few dominant species were recently identified as the main structuring elements of tropical forests. However, such result did not translate yet into quantitative approaches ...which are essential to understand, predict and monitor forest functions and composition over large, often poorly accessible territories. Here we show that the above-ground biomass (AGB) of the whole forest can be predicted from a few large trees and that the relationship is proved strikingly stable in 175 1-ha plots investigated across 8 sites spanning Central Africa. We designed a generic model predicting AGB with an error of 14% when based on only 5% of the stems, which points to universality in forest structural properties. For the first time in Africa, we identified some dominant species that disproportionally contribute to forest AGB with 1.5% of recorded species accounting for over 50% of the stock of AGB. Consequently, focusing on large trees and dominant species provides precise information on the whole forest stand. This offers new perspectives for understanding the functioning of tropical forests and opens new doors for the development of innovative monitoring strategies.
Introduction Les diabétiques âgés sont surexposés à l’hypotension orthostatique (HO), elle-même néfaste sur le pronostic cardiovasculaire. L’objectif était d’évaluer la fréquence de l’HO et ses ...facteurs associés dans la cohorte GERODIAB à l’inclusion. Patients et méthodes GERODIAB a recruté 987 diabétiques de type 2 autonomes, de 70 ans ou plus. La pression artérielle était mesurée, après 10 min de repos, et à 1, 3 et 5 minutes après passage en orthostatisme. L’HO était définie par une baisse de PAS d’au moins 20 mmHg et/ou de PAD d’au moins 10 mmHg à 1,3 ou 5 minutes. Les données (m ± SD ; %) ont été comparées par test t ou du chi2 ; l’analyse multivariée était réalisée par modèle logistique. Résultats 301 (30,5 %) patients avaient une OH ; La PAS et la PAD étaient plus élevées au repos chez les patients présentant une HO (146 ± 21 et 78 ± 11mmHg vs 138 ± 17 et 72 ± 10mmHg ; p < 0,001). Ils avaient également une HTA plus sévère (p < 0,01). Les patients avec HO présentaient une pression pulsée plus élevée (68 ± 18 vs 65 ± 15mmHg ; p < 0,05) et une fréquence cardiaque accélérée (78 ± 11 vs 72 ± 10 min−1 ; p < 0,001). Leur ratio taillehanche était augmenté (p < 0,01). Aucune différence significative n’était constatée pour les traitements antihypertenseurs, notamment bêta-bloquants (p = 0,19) et diurétiques (p = 0,84), l’âge, l’ancienneté du diabète ou les scores gériatriques. Les patients avec HO avaient plus souvent une artériopathie oblitérante des membres inférieurs (31 % vs 24 % ; p < 0,05) et des amputations (3,3 % vs 1,5 % ; p = 0,05). Ils n’avaient pas plus fréquemment de neuropathie périphérique, d’AVC, d’insuffisance cardiaque, de cardiopathie ischémique. En analyse multivariée, l’HO était associée successivement à l’HTA sévère (p < 0,01), au ratio taille-hanche (p < 0,05) et aux amputations (p < 0,05). Conclusion Un tiers environ des patients diabétiques âgés autonomes présente une HO. Ils ont plus souvent une HTA plus sévère. L’HO pourrait constituer une limitation à l’intensification du traitement anti-HTA dans cette population.
