Silvoarable agroforestry could promote use of trees on farms in Europe, but its likely effect on production, farm profitability, and environmental services is poorly understood. Hence, from 2001 to ...2005, the Silvoarable Agroforestry for Europe project developed a systematic process to evaluate the biophysical and economic performance of arable, forestry, and silvoarable systems in Spain, France, and The Netherlands. A biophysical model called “Yield-SAFE” was developed to predict long-term yields for the different systems and local statistics and expert opinion were used to derive their revenue, costs, and pre- and post-2005 grant regimes. These data were then used in an economic model called “Farm-SAFE” to predict plot- and farm-scale profitability. Land equivalent ratios were greater than one, showing Yield-SAFE predicted that growing trees and crops in silvoarable systems was more productive than growing them separately. Pre-2005 grants in Spain and The Netherlands penalised silvoarable systems, but post-2005 grants were more equitable. In France, walnut and poplar silvoarable systems were consistently the most profitable system under both grant regimes. In Spain, holm oak and stone pine silvoarable systems were the least profitable system under pre-2005 grants, but only marginally less profitable than arable systems under post-2005 grants. In The Netherlands, low timber values and the opportunity cost of losing arable land for slurry manure application made silvoarable and forestry systems uncompetitive with arable systems under both grant regimes.
Therapeutic proteins, including monoclonal antibodies, are typically manufactured using clonally derived, stable host cell lines, since consistent and predictable cell culture performance is highly ...desirable. However, selecting and preparing banks of stable clones takes considerable time, which inevitably extends overall development timelines for new therapeutics by delaying the start of subsequent activities, such as the scale‐up of manufacturing processes. In the context of the coronavirus disease 2019 (COVID‐19) pandemic, with its intense pressure for accelerated development strategies, we used a novel transposon‐based Leap‐In Transposase® system to rapidly generate high‐titer stable pools and then used them directly for large scale‐manufacturing of an anti‐severe acute respiratory syndrome coronavirus 2 monoclonal antibody under cGMP. We performed the safety testing of our non‐clonal cell bank, then used it to produce material at a 200L‐scale for preclinical safety studies and formulation development work, and thereafter at 2000L scale for supply of material for a Phase 1 clinical trial. Testing demonstrated the comparability of critical product qualities between the two scales and, more importantly, that our final clinical trial product met all pre‐set product quality specifications. The above expediated approach provided clinical trial material within 4.5 months, in comparison to 12–14 months for production of clinical trial material via the conventional approach.
Use of well‐characterized nonclonal stable CHO cell pools and platform processes can expediate cGMP manufacturing of monoclonal antibodies for early clinical development during pandemic outbreaks of emerging infectious diseases.

Living marine stromatolites at Highborne Cay, Bahamas, are formed by microbial mat communities that facilitate precipitation of calcium carbonate and bind and trap small carbonate sand ...grains. This process results in a laminated structure similar to the layering observed in ancient stromatolites. In the modern marine system at Highborne Cay, lamination, lithification and stromatolite formation are associated with cycling between three types of microbial communities at the stromatolite surface (Types 1, 2 and 3, which range from a leathery microbial mat to microbially fused sediment). Examination of 923 universal small‐subunit rRNA gene sequences from these communities reveals that taxonomic richness increases during transition from Type 1 to Type 3 communities, supporting a previous model that proposed that the three communities represent different stages of mat development. The phylogenetic composition also changes significantly between these community types and these community changes occur in concert with variation in biogeochemical rates. The dominant bacterial groups detected in the stromatolites include Alphaproteobacteria, Planctomycetes, Cyanobacteria and Bacteroidetes. In addition, the stromatolite communities were found to contain novel cyanobacteria that may be uniquely associated with modern marine stromatolites. The implications of these findings are discussed in the context of current models for stromatolite formation.
Exocytotic incorporation of plasmalemmal precursor vesicles (PPVs) into the cell surface is necessary for axonal outgrowth and is known to occur mainly at the nerve growth cone. We have demonstrated ...recently that plasmalemmal expansion is regulated at the growth cone by IGF-1, but not by BDNF, in a manner that is quasi independent of the neuron's perikaryon. To begin elucidating the signaling pathway by which exocytosis of the plasmalemmal precursor is regulated, we studied activation of the IRS/PI3K/Akt pathway in isolated growth cones and hippocampal neurons in culture stimulated with IGF-1 or BDNF. Our results show that IGF-1, but not BDNF, significantly and rapidly stimulates IRS/PI3K/Akt and membrane expansion. Inhibition of PI3K with Wortmannin or LY294002 blocked IGF-1-stimulated plasmalemmal expansion at the growth cones of cultured neurons. Finally, our results show that, upon stimulation with IGF-1, most active PI3K becomes associated with distal microtubules in the proximal or central domain of the growth cone. Taken together, our results suggest a critical role for IGF-1 and the IRS/PI3K/Akt pathway in the process of membrane assembly at the axonal growth cone.
Mixed systems of agriculture incorporating combinations of trees and crops have formed key elements of the landscape of Europe throughout historical times, and many such systems continue to function ...in the present day. In many cases they represent formerly widespread traditional systems in decline and a number have already become extinct or exist only in a threatened state. The causes are both practical and economic. The agricultural subsidy regime within the European Union is presently unfavourable towards silvoarable practices, which has been a major factor in their recent decline. The silvoarable systems of Europe can be split into two classes according to location - northern Europe and the Mediterranean. The latter contains not only a greater area of silvoarable cultivation, but also a greater diversity of systems due to the broader range of commercial tree and crop species grown. In general, the systems of northern Europe are limited by light, whilst those of the Mediterranean are limited by the availability of water. Mixed systems of agriculture present an opportunity for future European rural development and have the potential to contribute towards the increased sustainability of agriculture and enhancement of biodiversity, whilst preserving landscapes that are both culturally important and aesthetically pleasing. A better understanding of the legacy of traditional silvoarable systems, combined with the formulation of a consistent definition and specific European policy towards them will be invaluable in ensuring that the benefits of mixed agriculture are fully exploited in the future.PUBLICATION ABSTRACT

Silvoarable agroforestry (SAF) is the cultivation of trees and arable crops on the same parcel of land. SAF may contribute to modern diversified land use objectives in Europe, such as enhanced ...biodiversity and productivity, reduced leaching of nitrogen, protection against flooding and erosion, and attractiveness of the landscape. Long-term yield predictions are needed to assess long-term economic profitability of SAF.

A model for growth, resource sharing and productivity in agroforestry systems was developed to act as a tool in forecasts of yield, economic optimization of farming enterprises and exploration of policy options for land use in Europe. The model is called Yield-SAFE; from “YIeld Estimator for Long term Design of Silvoarable AgroForestry in Europe”. The model was developed with as few equations and parameters as possible to allow model parameterization under constrained availability of data from long-term experiments.

The model consists of seven state equations expressing the temporal dynamics of: (1) tree biomass; (2) tree leaf area; (3) number of shoots per tree; (4) crop biomass; (5) crop leaf area index; (6) heat sum; and (7) soil water content. The main outputs of the model are the growth dynamics and final yields of trees and crops. Daily inputs are temperature, radiation and precipitation. Planting densities, initial biomasses of tree and crop species, and soil parameters must be specified.

A parameterization of Yield-SAFE is generated, using published yield tables for tree growth and output from the comprehensive crop simulation model STICS. Analysis of tree and crop growth data from two poplar agroforestry stands in the United Kingdom demonstrates the validity of the modelling concept and calibration philosophy of Yield-SAFE. A sensitivity analysis is presented to elucidate which biological parameters most influence short and long-term productivity and land equivalent ratio.

The conceptual model, elaborated in Yield-SAFE, in combination with the outlined procedure for model calibration, offers a valid tool for exploratory land use studies.
This paper explores transactions costs in the context of agri‐environmental policy schemes based on management agreements. While transactions costs encompass a wide range of organisational costs, the ...focus here is on the public‐sector administrative costs of policy implementation. Empirical administrative cost functions were estimated to investigate the factors affecting the magnitude of such costs, using panel data spanning five years for the 22 English Environmentally Sensitive Areas. The extent of participation appears to be important in explaining administrative cost variability across areas. The data suggested the existence of size economics with regard to the numbers of agreements made in any one ESA, and a significant effect of scheme experience in exerting downwards‐pressure on administrative costs. Policy budgeting and evaluation should take into account the non‐trivial costs of organisation, particularly if agri‐environmental schemes based on the procurement of conservation goods through management agreements are to be extended in future.
Microbial mats in the hypersaline lake of Salt Pan, Eleuthera, Bahamas, display a gradient of lithification along a transect from the center to the shore of the lake. These mats exist under similar ...geochemical conditions, with light quantity and quality as the sole major environmental difference. Therefore, we hypothesized that the microbial community may be driving the differences in lithification and, by extension, mineral biosignature formation. The lithifying and non-lithifying mat communities were compared (via 16S rRNA gene sequencing, 485 and 464 sequences, respectively) over both temporal and spatial scales. Seven bacterial groups dominated in all the microbial mat libraries: bacteriodetes, alphaproteobacteria, deltaproetobacteria, chloroflexi, spirochaetes, cyanobacteria, and planctomycetes. The mat communities were all significantly different over space, time, and lithification state. Species richness is significantly higher in the non-lithifying mats, potentially due to differences in mat structure and activity. This increased richness may impact lithification and, hence, biosignature production.
