ID15A is a newly refurbished beamline at the ESRF devoted to operando and time‐resolved diffraction and imaging, total scattering and diffraction computed tomography. The beamline is optimized for ...rapid alternation between the different techniques during a single operando experiment in order to collect complementary data on working systems. The high available energy (up to 120 keV) means that even bulky and highly absorbing systems may be studied. The beamline is equipped with optimized focusing optics and a photon‐counting CdTe pixel detector, allowing for both unprecedented data quality at high energy and for very rapid triggered experiments. A large choice of imaging detectors and ancillary probes and sample environments is also available.

Single crystal CVD diamonds have been developed for use as XBPMs on the beamlines of the French synchrotron SOLEIL. These devices have been calibrated and characterized directly on the beamline ...following an elaborate protocol. An example is given for an XBPM installed on PROXIMA 2. This device has shown a charge collection efficiency of 100% for only 0.2V/μm with total signal-to-noise ratio better than 105. A complete 2D scan gives the homogeneity of the scale factors over the active surface with a very low error corresponding to 0.6μm (beam off-center and maximum calibration error). The current sensitivity measurement shows a fluctuation less than 0.02% and the responsivity is calculated to measure the incident flux. And finally, the spatial resolution for these XBPMs is better than 1μm rms at 1kHz and 100 nm rms at 5 Hz.

PROXIMA 2A is a new micro-focus and energy tunable beamline dedicated to biological macromolecular crystallography at Synchrotron SOLEIL. The beamline officially opened in March 2013, ...and its first year of user operation has yielded excellent results. The X-ray source is a powerful in-vacuum U24 undulator coupled to a cryo-cooled Si111 channel-cut monochromator and a pair of focussing bimorph mirrors in Kirpatrick-Baez configuration. This combination delivers a photon flux of over 10**12 ph/s into a focal spot of 10 μm × 5 μm (H×V FWHM), which is tunable over 6 – 15 keV. The supports of the optical elements have been designed to minimise the effects of vibrations and thermal dilations on the X-ray beam position, which is stable to within 5 microns over a day. The experimental station consists of a high performance micro-diffractometer, a cryostream, an area detector (ADSC Q315r), and an X-ray fluorescence detector. The X-ray energies for MAD experiments are directly calibrated on the sample. A robot equipped with a large 9 uni-puck dewar (CATS Irelec) is available to users for the automated transfer and screening of cryo-cooled samples. The users launch their experiments via an MXCuBE interface 1, which permits the centering of the sample, collecting of diffraction images, recording of X-ray spectra and the transfer of samples. The X-ray diffraction data are of an excellent quality, and the users readily exploit the micro-focused X-rays to select the best zones of their crystals. The first year of results from users has yielded a variety of success stories including novel protein structures resolved from crystals as small as 5 microns, as well as those solved by SAD & MAD methods. The future perspectives include automated helical and grid scans, in situ plate screening and multi-crystal merging techniques.
O artigo analisa a qualidade e a consistência de um roteiro de entrevista semiestruturada, adaptado para o estudo do suicídio de pessoas idosas e apresenta o método das autópsias psicossociais que ...resultou da aplicação desse instrumento. O objetivo é demonstrar como o uso da entrevista em profundidade e sua forma de organização e análise de dados foram testados e aperfeiçoados por uma rede de pesquisadores de vários centros de pesquisa do Brasil. O método envolveu a aplicação do instrumento em que se socializou um manual de instruções sobre a coleta, sistematização e análise de dados. A metodologia foi aplicada no estudo de 51 casos de idosos que faleceram por suicídio em dez municípios brasileiros, e permitiu a verificação da consistência do instrumento usado e a aplicabilidade do seu método, durante o processo e ao final, por meio de uma avaliação em rede. O roteiro aperfeiçoado e as instruções para replicá-lo e analisá-lo são aqui apresentados. Os resultados apontam o rigor e a credibilidade dessa abordagem metodológica testada e qualificada de um modo interdisciplinar e interinstitucional.
We are currently analyzing marine sediment cores collected aboard the Nathaniel B. Palmer in the Ross Sea during the 1994 and 1995 field seasons. These cores were collected to help fulfill the goal ...of understanding the past behavior of the West Antarctic Ice Sheet set forward by the West Antarctic Ice Sheet Initiative. Previous research on western Ross Sea cores (collected on Eltanin and Deep Freeze cruises) has been used to show that the ice did not advance to the continental shelf edge during marine isotope stage 2 and provided carbon-14 ( super(14)C) dates that constrain the timing of ice retreat across the continental shelf. In addition to these results, a bathymetric high has been recognized in the western Ross Sea based on seismic data and is interpreted as a grounding-line feature; this feature probably represents the last-glacial-maximum grounding-line position. Our objective is to determine the maximum grounded ice extent and ice retreat rate for the east-central Ross Sea by completing sedimentological analyses and by radiocarbon dating sediments that mark the transition from glacial to postglacial conditions.

. Sequential tissue sampling from these studies allowed for detailed on-treatment biomarker analysis. Here, we present a single-arm phase 2 study, investigating two cycles of atezolizumab before cystectomy in 95 patients with muscle-invasive urothelial cancer (ClinicalTrials.gov identifier: NCT02662309). Pathological complete response was the primary endpoint. Secondary endpoints focused on safety, relapse-free survival and biomarker analysis. The pathological complete response rate was 31% (95% confidence interval: 21-41%), achieving the primary efficacy endpoint. Baseline biomarkers showed that the presence of preexisting activated T cells was more prominent than expected and correlated with outcome. Other established biomarkers, such as tumor mutational burden, did not predict outcome, differentiating this from the metastatic setting. Dynamic changes to gene expression signatures and protein biomarkers occurred with therapy, whereas changes in DNA alterations with treatment were uncommon. Responding tumors showed predominant expression of genes related to tissue repair after treatment, making tumor biomarker interpretation challenging in this group. Stromal factors such as transforming growth factor-β and fibroblast activation protein were linked to resistance, as was high expression of cell cycle gene signatures after treatment.
The development of collagen hydrogels with tailored properties for improved applications in biomedicine represents an area of opportunity for materials science. The collagen can form ...semi‐interpenetrated networks (semi‐IPN) with various natural and/or synthetic polymers. This work aims the preparation of novel hydrogels generated from a collagen matrix cross‐linked with polyurethane (PU), and the subsequent inclusion of polysaccharide chains to form semi‐IPN systems with improved properties. The choice of polysaccharides for this purpose is related to their ability to modulate the biocompatibility and the antibacterial capacity in various biomedical strategies. The work contemplates to study the effect of the chemical structure of polysaccharide (hydroxyethylcellulose (HEC), hydroxypropylmethylcellulose (HPMC) or starch (Alm)) on the properties of these novel hydrogels. The results indicate that the semi‐IPN hydrogels that include Alm exhibit the formation of stronger intermolecular interactions promoted by hydrogen bonds than HEC and HPMC, significantly improving the mechanical properties and their degradation rate in acidic, alkaline, and proteolytic media; also showing high capacity to inhibit the growth of E. colli. The semi‐IPN hydrogels based on HEC and HPMC exhibit excellent improvement in both thermal and proteolytic degradation, compared with the collagen‐PU matrix. On the other hand, this semi‐IPN system does not present cytotoxic character for monocytes and fibroblasts growing for up to 48 h of culture. Therefore, these innovative 3D matrices will be excellent candidates with potential application in biomedical strategies such as wound healing dressings.

Every four years, the 10 national teams members of the South American Football Confederation (CONMEBOL) compete for one of the South American slots in the final phase of the FIFA World Cup. The qualifying competition consists of a double round robin tournament. The matches are scheduled in 9 closely spaced pairs known as double rounds. Every team plays twice in each double round. The tournament is spread over 2 years, so the double rounds are months apart. After using the same mirrored schedule for about twenty years, and persistent complaints from its members, CONMEBOL decided to change the schedule for the 2018 World Cup. Supported by one of CONMEBOL’s members, we used integer programmming to construct schedules that overcome the main drawbacks of the previous approach. After exploring many design criteria, we proposed a candidate schedule based on a French scheme. The main feature of the proposed schedule is that every team plays once at home and once away on each double round, a departure from traditional symmetric (mirrored) schemes. This proposal was unanimously approved by CONMEBOL members and is currently being used in the qualifier tournament for the 2018 FIFA World Cup in Russia.
