Recent research has revealed potential advantages of feeding an elevated plane of nutrition to calves during the preweaning period. However, calves fed more nutrients preweaning may be more ...susceptible to depressed growth and weaning stress during the transition from liquid to solid feed. The objective of this study was to investigate the relationship between the age of weaning and feed intake, and its influence on growth, gastrointestinal development, and behavioral indicators in dairy calves fed an elevated plane of nutrition during the preweaning period. To meet this objective, 20 female Holstein calves were randomly assigned at birth to be weaned at 6 or 8 wk. Milk replacer (mixed at 150 g/L) was offered at 1.2 kg/calf per day in 2 meals until a 1-wk step-down, when meals were reduced by 50% 1 wk before weaning. Daily starter, chopped oat straw, water intake, and weekly body weights were measured until d 70 of life. To assess digestive tract development, rumen fluid, fecal, and blood samples were taken before and after weaning (d 35, 49, and 63) and analyzed for ruminal short-chain fatty acids, blood β-hydroxybutyrate, and fecal starch, respectively. Behavioral indicators of weaning stress, including vocalizing and non-nutritive oral behavior, were measured by visual observation for 1 h, 3 times per week, before the second feeding of the day during the period from 2 wk before weaning to 2 wk after weaning. The calves weaned at 8 wk compared with 6 wk had higher average daily gain for the week preweaning (0.79±0.09 vs. 0.34±0.10 kg/d) and postweaning (1.05±0.09 vs. 0.35±0.11 kg/d), and were heavier at d 70 (99.9±1.81 vs. 91.0±2.26 kg). From 5 to 8 wk of age, starter and water intakes were lower in calves weaned at 8 wk of age. However, overall starter intake did not differ during the last week of the experiment. Furthermore, calves weaned at 8 wk compared with 6 wk had higher starter intake for 1 wk preweaning (1.36±0.13 vs. 0.40±0.08 kg/d) and postweaning (2.51±0.20 vs. 1.16±0.15 kg/d). In both treatments, weaning increased ruminal short-chain fatty acids, blood β-hydroxybutyrate, and fecal starch, yet the differences between the week before and after weaning were greater for calves weaned at 6 wk compared with those weaned at 8 wk. Treatment × week relative to weaning interactions indicated that several behaviors varied between early- and later-weaned calves during the week before weaning; calves weaned at 6 wk tended to exhibit 75% more non-nutritive oral behavior and spent 55% less time ruminating, and 36% less time lying compared with calves weaned at 8 wk. Under the conditions of this study, the results suggest that calves fed an elevated plane of nutrition preweaning have higher starter intakes and average daily gain during the weaning period when weaning is extended from 6 to 8 wk of age.
Disturbances alter ecosystem carbon dynamics, often by reducing carbon uptake and stocks. We compared the impact of two types of disturbances that represent the most likely future conditions of ...currently dense ponderosa pine forests of the southwestern United States: (1) high-intensity fire and (2) thinning, designed to reduce fire intensity. High-severity fire had a larger impact on ecosystem carbon uptake and storage than thinning. Total ecosystem carbon was 42% lower at the intensely burned site, 10 years after burning, than at the undisturbed site. Eddy covariance measurements over two years showed that the burned site was a net annual source of carbon to the atmosphere whereas the undisturbed site was a sink. Net primary production (NPP), evapotranspiration (ET), and water use efficiency were lower at the burned site than at the undisturbed site. In contrast, thinning decreased total ecosystem carbon by 18%, and changed the site from a carbon sink to a source in the first post-treatment year. Thinning also decreased ET, reduced the limitation of drought on carbon uptake during summer, and did not change water use efficiency. Both disturbances reduced ecosystem carbon uptake by decreasing gross primary production (55% by burning, 30% by thinning) more than total ecosystem respiration (TER; 33-47% by burning, 18% by thinning), and increased the contribution of soil carbon dioxide efflux to TER. The relationship between TER and temperature was not affected by either disturbance. Efforts to accurately estimate regional carbon budgets should consider impacts on carbon dynamics of both large disturbances, such as high-intensity fire, and the partial disturbance of thinning that is often used to prevent intense burning. Our results show that thinned forests of ponderosa pine in the southwestern United States are a desirable alternative to intensively burned forests to maintain carbon stocks and primary production.
Continuity of coastal Indigenous cultures relies on healthy ecosystems and opportunity to fulfill cultural practices. Owing to resource stewardship practice over millennia, Indigenous nations possess ...Indigenous knowledge that positions them as leaders in contemporary resource management. However, Indigenous peoples possibly face social-ecological traps, situations in which feedbacks between social and ecological systems result in an undesirable state, that are challenging to overcome. Centuries of compounding colonization and environmental degradation have negatively impacted Indigenous knowledge and culturally mediated stewardship practices. Our partnership, comprising academics and four First Nations on the Central Coast of British Columbia, Canada, mobilized information from semistructured interviews with knowledge holders to explore Indigenous knowledge of a culturally important but vulnerable species, yelloweye rockfish (Sebastes ruberrimus). We analyzed interviews and discovered evidence of an extant but transcendable social-ecological trap. The emergent themes represent an exploration beyond our original project goals and research questions. Our study revealed that external forces of colonization, i.e., via forced assimilation, and species declines created a social-ecological trap. However, participants ubiquitously described stewardship principles, and noted ongoing cultural revitalization efforts, active recovery of depleted species, and reassertion of Indigenous management rights as ways they are rebelling against, and overcoming, the trap. Although the framing of a social-ecological trap may be perceived as diminishing the authority of Indigenous governance systems, we found the opposite to be true. Despite external pressures, coastal First Nations are reasserting cultural and management rights and shaping their futures. We suggest that ongoing Indigenous cultural renewal and ecosystem recovery in the face of the historically entrenched trap be supported through recognizing and implementing inherent Indigenous marine management rights. The social-ecological trap described here differs from others in the literature in that the creation of the trap was external; moving beyond it is happening through internal, i.e., led by the First Nations, efforts.
A new and improved setup of the SF6 retrieval together with a newly calibrated version of MIPAS-ENVISAT level 1b spectra (version 5, ESA data version 5.02/5.06) was used to obtain a new global SF6 ...data set, covering the total observational period of MIPAS from July 2002 to April 2012 for the first time. Monthly and zonally averaged SF6 profiles were converted into mean age of air using a tropospheric SF6-reference curve. The obtained data set of age of air was compared to airborne age of air measurements. The temporal evolution of the mean age of air was then investigated in 10° latitude and 1–2 km altitude bins. A regression model consisting of a constant and a linear trend term, two proxies for the quasi-biennial oscillation variation, sinusoidal terms for the seasonal and semiannual variation and overtones was fitted to the age of air time series. The annual cycle for particular regions in the stratosphere was investigated and compared to other studies. The age of air trend over the total MIPAS period consisting of the linear term was assessed and compared to previous findings of Stiller et al. (2012). While the linear increase of mean age is confirmed to be positive for the northern midlatitudes and southern polar middle stratosphere, differences are found in the northern polar upper stratosphere, where the mean age is now found to increase as well. The magnitude of trends in the northern midlatitude middle stratosphere is slightly lower compared to the previous version and the trends fit remarkably well to the trend derived by Engel et al. (2009). Negative age of air trends found by Stiller et al. (2012) are confirmed for the lowermost tropical stratosphere and lowermost southern midlatitudinal stratosphere. Differences to the previous data versions occur in the middle tropical stratosphere around 25 km, where the trends are now negative. Overall, the new latitude–altitude distribution of trends appears to be less patchy and more coherent than the previous one. The new data provide evidence of an accelerating shallow branch of the Brewer–Dobson circulation, at least in the Southern Hemisphere. Finally the age of air decadal trends are compared to trends calculated with simulated SF6 values by the Karlsruhe Simulation Model of the Middle Atmosphere (KASIMA) and good agreement is found. The hemispheric asymmetry in the trends found in the MIPAS data is also indicated in the trends calculated with simulated SF6 values by the KASIMA model.

Diarrhea, resulting from gastrointestinal infection by pathogens, is a common cause of the high mortality and morbidity of neonatal calves. The objective of this study was to evaluate the effects of supplementing a yeast product in milk replacer (MR) on growth and health of calves, and on fecal populations of some targeted microorganisms related to calf health and growth (i.e., total bacteria, Escherichia coli, Clostridium cluster XIVa, Faecalibacterium prausnitzii and Bifidobacterium spp.). We hypothesized that feeding a Saccharomyces cerevisiae var boulardii (SCB) product would improve gastrointestinal health and growth performance of calves. Forty-two Holstein bull calves (42.6±0.77kg at birth) were randomly assigned on day 2 of age to either a control or SCB treatment. The SCB was supplemented in MR and fed at 5g/d per head to supply 10 billion colony-forming units per day. All calves received high quality colostrum (>50mg/mL of immunoglobulin G) during the first 24h of life, and were fed with 8L MR (150g/L mixed with 40°C water) daily from day 2–35, and 4L daily from day 35–42. Calves were also fed calf starter ad libitum from day 7–56. Daily MR and starter offered and refused, daily fecal scores, nasal scores, ear scores, and weekly body weight of calves were recorded. Fecal samples were collected on day 7, 35 and 56 after the first feeding of that day for microbial targets analysis. Overall, there is no serious disease challenge for all the calves during the entire experimental period. No differences were observed in MR intake, starter intake, metabolizable energy (ME) intake, average daily gain, ME intake to gain ratio, fecal score, nasal score, eye score or any targeted microorganisms between treatments throughout the experiment. These results suggest that supplementing SCB in MR has no additive effects on animal growth or fecal biomarkers of gut health when calves do not show deteriorated health status.
The impact of altered amino acid metabolism on cancer progression is not fully understood. We hypothesized that a metabolic transcriptome shift during metastatic evolution is crucial for brain ...metastasis. Here, we report a powerful impact in this setting caused by epigenetic upregulation of glutamate decarboxylase 1 (GAD1), a regulator of the GABA neurotransmitter metabolic pathway. In cell-based culture and brain metastasis models, we found that downregulation of the DNA methyltransferase DNMT1 induced by the brain microenvironment-derived clusterin resulted in decreased GAD1 promoter methylation and subsequent upregulation of GAD1 expression in brain metastatic tumor cells. In a system to dynamically visualize cellular metabolic responses mediated by GAD1, we monitored the cytosolic NADH:NAD
equilibrium in tumor cells. Reducing GAD1 in metastatic cells by primary glia cell coculture abolished the capacity of metastatic cells to utilize extracellular glutamine, leading to cytosolic accumulation of NADH and increased oxidative status. Similarly, genetic or pharmacologic disruption of the GABA metabolic pathway decreased the incidence of brain metastasis
Taken together, our results show how epigenetic changes in GAD1 expression alter local glutamate metabolism in the brain metastatic microenvironment, contributing to a metabolic adaption that facilitates metastasis outgrowth in that setting.

We analyze temporal trends of peroxyacetyl nitrate (PAN) retrievals from the Michelson Interferometer for Passive Atmospheric Sounding (MIPAS) during 2002–2011 in the altitude range 8–23 km over the ...Asian summer monsoon (ASM) region. The greatest enhancements of PAN mixing ratios in the upper troposphere and lower stratosphere (UTLS) are seen during the summer monsoon season from June to September. During the monsoon season, the mole fractions of PAN show statistically significant (at 2σ) positive trends from 0.2 ± 0.05 to 4.6 ± 3.1 ppt yr−1 (except between 12 and 14 km) which is higher than the annual mean trends of 0.1 ± 0.05 to 2.7 ± 0.8 ppt yr−1. These rising concentrations point to increasing NOx (= NO + NO2) and volatile organic compound (VOC) emissions from developing nations in Asia, notably India and China. We analyze the influence of monsoon convection on the distribution of PAN in UTLS with simulations using the global chemistry–climate model ECHAM5-HAMMOZ. During the monsoon, transport into the UTLS over the Asian region primarily occurs from two convective zones, one the South China Sea and the other over the southern flank of the Himalayas. India and China host NOx-limited regimes for ozone photochemical production, and thus we use the model to evaluate the contributions from enhanced NOx emissions to the changes in PAN, HNO3 and O3 concentrations in the UTLS. From a set of sensitivity experiments with emission changes in particular regions, it can be concluded that Chinese emissions have a greater impact on the concentrations of these species than Indian emissions. According to SCanning Imaging Absorption SpectroMeter for Atmospheric CHartographY (SCIAMACHY) NO2 retrievals NOx emissions increases over India have been about half of those over China between 2002 and 2011.
The ozone radiometer GROMOS (GROund-based Millimeter-wave Ozone Spectrometer) has been performing continuous observations of stratospheric ozone profiles since 1994 above Bern, Switzerland (46.95 ...degree N, 7.44 degree E, 577 m). GROMOS is part of the Network for the Detection of Atmospheric Composition Change (NDACC). From November 1994 to October 2011, the ozone line spectra were measured by a filter bench (FB). In July 2009, a fast Fourier transform spectrometer (FFTS) was added as a back end to GROMOS. The new FFTS and the original FB measured in parallel for over 2 years. The ozone profiles retrieved separately from the ozone line spectra of FB and FFTS agree within 5 % at pressure levels from 30 to 0.5 hPa, from October 2009 to August 2011. A careful harmonisation of both time series has been carried out by taking the FFTS as the reference instrument for the FB. This enables us to assess the long-term trend derived from stratospheric ozone observations at Bern. The trend analysis was performed by using a robust multilinear parametric trend model which includes a linear term, the solar variability, the El Nino-Southern Oscillation (ENSO) index, the quasi-biennial oscillation (QBO), the annual and semi-annual oscillation and several harmonics with period lengths between 3 and 24 months. Over the last years, some experimental and modelling trend studies have shown that the stratospheric ozone trend is levelling off or even turning positive. With our observed ozone profiles, we are able to support this statement by reporting a statistically significant trend of +3.14 % decade-1 at 4.36 hPa (37.76 km), covering the period from January 1997 to January 2015, above Bern. Additionally, we have estimated a negative trend over this period of -3.94 % decade-1 at 0.2 hPa (59 km).
Recent research suggests that circulating β-hydroxybutyrate (BHB) levels may be a meaningful indicator of grain intake and rumen development in pre-ruminant calves. As such, BHB levels may be a ...surrogate measure of rumen function to contribute to minimal weaning stress during the transition from liquid to solid feed. The primary objective of this study was to determine the optimal cut-point of circulating BHB levels that would be predictive of sufficient grain intake and rumen development for a successful transition from liquid to solid feed at the time of weaning. An additional objective was to validate the Precision Xtra (Abbott Diabetes Care, Abingdon, UK) calf-side test for determination of BHB in whole blood in calves around weaning, as compared with the gold standard laboratory method. A total of 20 Holstein female calves were randomly assigned at birth to be weaned at 6 wk (n=10) or 8 wk (n=10) of age. Milk replacer (mixed at 150 g/L) was offered at 1.2kg/calf per d in 2 meals until a 1-wk step down, when milk meals were reduced by 50% 1 wk before weaning. Daily measurements included the intakes of starter grain, chopped straw, and water. Weekly measurements included body weight and blood BHB, until 70 d of life. To assess digestive tract development, rumen fluid samples were taken before and after weaning (d 35, 49, and 63) and analyzed for ruminal short-chain fatty acids. Whole blood was collected by jugular venipuncture, and BHB was determined by the Precision Xtra test at calf-side. In addition, serum was separated from a clotted sample, frozen, and stored until laboratory analysis was performed. Laboratory BHB results were correlated with both the Precision Xtra test (r=0.95) and starter intake over 1 d (r=0.89), a 3-d average (r=0.90), and a 7-d average (r=0.90). Additionally, laboratory BHB results were associated with total ruminal volatile fatty acids (r=0.82), ruminal butyrate (r=0.77), and body weight (r=0.69). A receiver operating characteristic curve was created to plot the true positive rate against the false positive rate at 10μmol/L BHB intervals to determine the optimal cut-point of circulating BHB that is predictive of an average starter intake of 1,000 g/d over a 3-d period. The optimal combination of sensitivity (95.7%) and specificity (96.1%) was at 100µmol of BHB/L of blood. A value of 0.2 mmol/L on the Precision Xtra test yielded a sensitivity and specificity of 84.0 and 97.2%, respectively, over the 3-d average period of starter intake. These results show considerable promise for use of the Precision Xtra whole blood BHB test in the decision-making process of determining sufficient starter grain intake and rumen development for a successful transition from liquid to solid feed, and indicate that this test conducted at calf-side is highly accurate.
