The Snail family of zinc-finger transcription factors are evolutionarily conserved proteins that control processes requiring cell movement. Specifically, they regulate epithelial-to-mesenchymal ...transitions (EMT) where an epithelial cell severs intercellular junctions, degrades basement membrane and becomes a migratory, mesenchymal-like cell. Interestingly, Slug expression has been observed in angiogenic endothelial cells (EC) in vivo, suggesting that angiogenic sprouting may share common attributes with EMT. Here, we demonstrate that sprouting EC in vitro express both Slug and Snail, and that siRNA-mediated knockdown of either inhibits sprouting and migration in multiple in vitro angiogenesis assays. We find that expression of MT1-MMP, but not of VE-Cadherin, is regulated by Slug and that loss of sprouting as a consequence of reduced Slug expression can be reversed by lentiviral-mediated re-expression of MT1-MMP. Activity of MMP2 and MMP9 are also affected by Slug expression, likely through MT1-MMP. Importantly, we find enhanced expression of Slug in EC in human colorectal cancer samples compared with normal colon tissue, suggesting a role for Slug in pathological angiogenesis. In summary, these data implicate Slug as an important regulator of sprouting angiogenesis, particularly in pathological settings.
The anatomically simple plants that first colonized land must have acquired molecular and biochemical adaptations to drought stress. Abscisic acid (ABA) coordinates responses leading to desiccation ...tolerance in all land plants. We identified ABA nonresponsive mutants in the model bryophyte Physcomitrella patens and genotyped a segregating population to map and identify the ABA NON-RESPONSIVE (ANR) gene encoding a modular protein kinase comprising an N-terminal PAS domain, a central EDR domain, and a C-terminal MAPKKK-like domain. anr mutants fail to accumulate dehydration tolerance-associated gene products in response to drought, ABA, or osmotic stress and do not acquire ABA-dependent desiccation tolerance. The crystal structure of the PAS domain, determined to 1.7-Å resolution, shows a conserved PAS-fold that dimerizes through a weak dimerization interface. Targeted mutagenesis of a conserved tryptophan residue within the PAS domain generates plants with ABA nonresponsive growth and strongly attenuated ABA-responsive gene expression, whereas deleting this domain retains a fully ABA-responsive phenotype. ANR orthologs are found in early-diverging land plant lineages and aquatic algae but are absent from more recently diverged vascular plants. We propose that ANR genes represent an ancestral adaptation that enabled drought stress survival of the first terrestrial colonizers but were lost during land plant evolution.
We previously published a method for measuring several physiological traits causing obstructive sleep apnea (OSA). The method, however, had a relatively low success rate (76%) and required ...mathematical modeling, potentially limiting its application. This paper presents a substantial revision of that technique. To make the measurements, continuous positive airway pressure (CPAP) was manipulated during sleep to quantify 1) eupneic ventilatory demand, 2) the level of ventilation at which arousals begin to occur, 3) ventilation off CPAP (nasal pressure = 0 cmH(2)O) when the pharyngeal muscles are activated during sleep, and 4) ventilation off CPAP when the pharyngeal muscles are relatively passive. These traits could be determined in all 13 participants (100% success rate). There was substantial intersubject variability in the reduction in ventilation that individuals could tolerate before having arousals (difference between ventilations #1 and #2 ranged from 0.7 to 2.9 liters/min) and in the amount of ventilatory compensation that individuals could generate (difference between ventilations #3 and #4 ranged from -0.5 to 5.5 liters/min). Importantly, the measurements accurately reflected clinical metrics; the difference between ventilations #2 and #3, a measure of the gap that must be overcome to achieve stable breathing during sleep, correlated with the apnea-hypopnea index (r = 0.9, P < 0.001). An additional procedure was added to the technique to measure loop gain (sensitivity of the ventilatory control system), which allowed arousal threshold and upper airway gain (response of the upper airway to increasing ventilatory drive) to be quantified as well. Of note, the traits were generally repeatable when measured on a second night in 5 individuals. This technique is a relatively simple way of defining mechanisms underlying OSA and could potentially be used in a clinical setting to individualize therapy.
Objective: To demonstrate the diagnostic ability of label-free, point-scanning, fiber-based Fluorescence Lifetime Imaging (FLIm) as a means of intraoperative guidance during oral and oropharyngeal ...cancer removal surgery. Methods: FLIm point-measurements acquired from 53 patients (n = 67893 pre-resection in vivo , n = 89695 post-resection ex vivo ) undergoing oral or oropharyngeal cancer removal surgery were used for analysis. Discrimination of healthy tissue and cancer was investigated using various FLIm-derived parameter sets and classifiers (Support Vector Machine, Random Forests, CNN). Classifier output for the acquired set of point-measurements was visualized through an interpolation-based approach to generate a probabilistic heatmap of cancer within the surgical field. Classifier output for dysplasia at the resection margins was also investigated. Results: Statistically significant change (P <inline-formula><tex-math notation="LaTeX">< </tex-math></inline-formula> 0.01) between healthy and cancer was observed in vivo for the acquired FLIm signal parameters (e.g., average lifetime) linked with metabolic activity. Superior classification was achieved at the tissue region level using the Random Forests method (ROC-AUC: 0.88). Classifier output for dysplasia (% probability of cancer) was observed to lie between that of cancer and healthy tissue, highlighting FLIm's ability to distinguish various conditions. Conclusion: The developed approach demonstrates the potential of FLIm for fast, reliable intraoperative margin assessment without the need for contrast agents. Significance: Fiber-based FLIm has the potential to be used as a diagnostic tool during cancer resection surgery, including Transoral Robotic Surgery (TORS), helping ensure complete resections and improve the survival rate of oral and oropharyngeal cancer patients.
CAM photosynthesis is hypothesized to have evolved in atmospheres of low CO2 concentration in recent geological time because of its ability to concentrate CO2 around Rubisco and boost water use ...efficiency relative to C3 photosynthesis. We assess this hypothesis by compiling estimates of when CAM clades arose using phylogenetic chronograms for 73 CAM clades. We further consider evidence of how atmospheric CO2 affects CAM relative to C3 photosynthesis.
Where CAM origins can be inferred, strong CAM is estimated to have appeared in the past 30 million years in 46 of 48 examined clades, after atmospheric CO2 had declined from high (near 800 ppm) to lower (<450 ppm) values. In turn, 21 of 25 clades containing CAM species (but where CAM origins are less certain) also arose in the past 30 million years. In these clades, CAM is probably younger than the clade origin. We found evidence for repeated weak CAM evolution during the higher CO2 conditions before 30 million years ago, and possible strong CAM origins in the Crassulaceae during the Cretaceous period prior to atmospheric CO2 decline. Most CAM-specific clades arose in the past 15 million years, in a similar pattern observed for origins of C4 clades.
The evidence indicates strong CAM repeatedly evolved in reduced CO2 conditions of the past 30 million years. Weaker CAM can pre-date low CO2 and, in the Crassulaceae, strong CAM may also have arisen in water-limited microsites under relatively high CO2. Experimental evidence from extant CAM species demonstrates that elevated CO2 reduces the importance of nocturnal CO2 fixation by increasing the contribution of C3 photosynthesis to daily carbon gain. Thus, the advantage of strong CAM would be reduced in high CO2, such that its evolution appears less likely and restricted to more extreme environments than possible in low CO2.
Declines in invertebrate biodiversity1,2 pose a significant threat to key ecosystem services.3–5 Current analyses of biodiversity often focus on taxonomic diversity (e.g., species richness),6,7 which ...does not account for the functional role of a species. Functional diversity of species’ morphological or behavioral traits is likely more relevant to ecosystem service delivery than taxonomic diversity, as functional diversity has been found to be a key driver of a number of ecosystem services including decomposition and pollination.8–12 At present, we lack a good understanding of long-term and large-scale changes in functional diversity, which limits our capacity to determine the vulnerability of key ecosystem services with ongoing biodiversity change. Here we derive trends in functional diversity and taxonomic diversity over a 45-year period across Great Britain for species supporting freshwater aquatic functions, pollination, natural pest control, and agricultural pests (a disservice). Species supporting aquatic functions showed a synchronous collapse and recovery in functional and taxonomic diversity. In contrast, pollinators showed an increase in taxonomic diversity, but a decline and recovery in functional diversity. Pest control agents and pests showed greater stability in functional diversity over the assessment period. We also found that functional diversity could appear stable or show patterns of recovery, despite ongoing changes in the composition of traits among species. Our results suggest that invertebrate assemblages can show considerable variability in their functional structure over time at a national scale, which provides an important step in determining the long-term vulnerability of key ecosystem services with ongoing biodiversity change.

Natural Hazard-induced Disasters (NHD) cause a wide range of losses to built and natural environments, the latter often beyond standard measures. Precise accounting and characterisation of the losses ...can assist in developing effective management policies that help to build resilient communities. This study applies trans-disciplinary approaches to assess total, monetary and non-monetary, NHD-related losses, estimated at AUD 156 million per year (2010–2019 average), for Australia’s Northern Territory where bushfires, cyclones, storms and floods are destructive and frequent events. Non-monetary losses, often overlooked or omitted, were estimated at AUD103 million per year, accounting for two-thirds of total disaster-related losses. Marketable losses, estimated at AUD 53 million per year, were inferred, using standard and non-standard datasets, from the Australian Government’s Natural Disaster Relief and Recovery Arrangements, insurance costs (Insurance Council of Australia database), and other relevant sources. Non-monetary losses were accounted for by the loss of ecosystem services from natural systems caused by cyclones and bushfires only, applying ecological economics approaches, but without considering long-term losses over the duration of recovery. This study informs disaster management policies to invest in collective emergency and environmental management planning for reducing NHD risk and building resilience of local communities to manage and prepare for rapidly changing climates. Such an accounting approach is essential in contexts where NHDs disproportionately affect the lives and well-being of disadvantaged remote communities.
The interaction between environmental stressors may be a greater threat to biota than any individual ecological threat on its own. Land‐use change and inappropriate fire regimes are known to pose ...great challenges to biodiversity conservation worldwide. Despite much research being conducted into their singular impacts on ecosystems, very few have investigated how their interaction may be affecting the biota of a region. We used data from surveys in 1998/2000 and 2019/2020 to compare the feeding guild assemblages of bird communities in different habitats within the greater Darwin region. By compiling two sets of spatial data, land‐use change, and fire history mapping, we were able to investigate their interaction and impact on the avian assemblages in the Darwin urban area. Using Generalized Linear Mixed Models (GLMM) we found that an increase in urbanization significantly affected fire occurrence across study sites. Furthermore, we found that the interaction between land‐use change and fire regimes had a significant effect on species that primarily feed on fruit. We conclude that while an increase in urbanization did not directly affect the avian assemblages, the impact of land‐use change on the fire regimes indirectly impacted urban bird community structures.
Using spatial data and historical and recent bird surveys, we investigated the impact of land‐use change and fire on the Darwin bird assemblages. We determined that the interaction between land‐use change and fire regimes directly affected frugivores. Furthermore, we found that the effect of land‐use change on fire regimes indirectly impacted avian assemblages.

The growth of experimental studies of crassulacean acid metabolism (CAM) in diverse plant clades, coupled with recent advances in molecular systematics, presents an opportunity to re-assess the ...phylogenetic distribution and diversity of species capable of CAM. It has been more than two decades since the last comprehensive lists of CAM taxa were published, and an updated survey of the occurrence and distribution of CAM taxa is needed to facilitate and guide future CAM research. We aimed to survey the phylogenetic distribution of these taxa, their diverse morphology, physiology and ecology, and the likely number of evolutionary origins of CAM based on currently known lineages.
We found direct evidence (in the form of experimental or field observations of gas exchange, day-night fluctuations in organic acids, carbon isotope ratios and enzymatic activity) for CAM in 370 genera of vascular plants, representing 38 families. Further assumptions about the frequency of CAM species in CAM clades and the distribution of CAM in the Cactaceae and Crassulaceae bring the currently estimated number of CAM-capable species to nearly 7 % of all vascular plants. The phylogenetic distribution of these taxa suggests a minimum of 66 independent origins of CAM in vascular plants, possibly with dozens more. To achieve further insight into CAM origins, there is a need for more extensive and systematic surveys of previously unstudied lineages, particularly in living material to identify low-level CAM activity, and for denser sampling to increase phylogenetic resolution in CAM-evolving clades. This should allow further progress in understanding the functional significance of this pathway by integration with studies on the evolution and genomics of CAM in its many forms.
