genotype in Chilean pediatric kidney recipients using extended-release TAC, to evaluate different LSS-AUC
formulas and dose requirements.
We analyzed pediatric kidney recipients using different extended-release TAC brands to determine their trapezoidal AUC
and
genotypes (SNP rs776746). Daily TAC dose (TAC-D mg/kg) and AUC
normalized by dose were compared between CYP3A5 expressors (*1/*1 and *1/*3) and non-expressors (*3/*3). We evaluated the single and combined time-points to identify the best LSS-AUC
model. We compared the performance of this model with two pediatric LSS-AUC
equations for clinical validation.
Fifty-one pharmacokinetic profiles were obtained from kidney recipients (age 13.1 ± 2.9 years). When normalizing AUC
by TAC-D significant differences were found between CYP3A5 expressors and non-expressors (1701.9 vs. 2718.1 ng*h/mL/mg/kg,
< 0.05). C
had a poor fit with AUC

, showed the best performance to predict LSS-AUC

= 0.8765) and yielded the lowest precision error (7.1% ± 6.4%) with the lowest fraction (9.8%) of deviated AUC
, in comparison to other LSS equations.
Estimation of LSS-AUC
with 3 time-points is an advisable and clinically useful option for pediatric kidney recipients using extended-release TAC to provide better guidance of decisions if toxicity or drug inefficacy is suspected. The different
genotypes associated with variable dose requirements reinforce considering genotyping before KTx. Further multi-centric studies with admixed cohorts are needed to determine the short- and long-term clinical benefits.
Desde la antigüedad la relación del hombre con los recursos naturales ha sido estrecha, en especial con el uso de las plantas, como alimento, vestido, material de construcción para curar ...enfermedades. El objetivo del presente estudio fue registrar los usos de las plantas medicinales más comunes en la zona metropolitana de Saltillo, Coahuila. De octubre 2021 a mayo del 2022 se entrevistaron 113 informantes (17 hombres y 96 mujeres, de 20 a 70 años) a través de un cuestionario etnobotánico semiestructurado dividido en dos partes: aspectos socioeconómicos y conocimiento tradicional de plantas medicinales. El reporte de usos obtenido se dividió en 13 categorías basadas en las Clasificación Estadística Internacional de Enfermedades y Problemas de Salud de la Organización Mundial de la Salud. Se registro un uso promedio de 4.6 plantas y cinco usos por planta. Las mujeres registraron 4.9 plantas en promedio, a comparación de los hombres con un 3.1. El análisis de regresión, mostro correlación positiva entre edad y número de planta. Se utilizan 82 plantas para tratamiento de enfermedades distribuidas en 37 familias botánicas. Las enfermedades que más afectan son del sistema respiratorio, gastrointestinal y nervioso. Las especies medicinales con mayor Índice de Valor de Uso fueron: Matricaria chamomilla (IVU=0.65) y Litsea parvifolia (IVU=0.35). Las familias más representativas fueron Asteraceae (10 especies, 125 reportes de uso), Lamiaceae (12 especies, 119 RU) y Lauraceae (cuatro especies, 64 RU). La forma de preparación más comunes fueron la infusión (88.6 %) y emplasto (3.68 %).
Introducción: la fuerza muscular puede disminuir en los pacientes con insuficiencia renal crónica por diversas causas. Objetivos: determinar las variables antropométricas, clínicas y la fuerza ...muscular en pacientes adultos con insuficiencia renal crónica. Metodología: se aplicó un diseño observacional, descriptivo, transversal. Se incluyeron varones y mujeres, mayores de 17 años, portadores de insuficiencia renal crónica que asistían al Hospital Nacional (Itauguá) y Hospital Militar (Asunción), Paraguay, entre abril y noviembre 2021. Se determinaron variables antropométricas, clínicas y laboratoriales. La fuerza muscular se midió con un dinamómetro de mano. Se utilizó un grupo de sujetos jóvenes sanos como grupo control para la comparación de la fuerza muscular. Se aplicó estadística descriptiva con el programa estadístico Epi Info 7™. La investigación fue aprobada por el Comité de Ética de la Universidad Privada del Este, Paraguay. Resultados: ingresaron al estudio 119 sujetos, siendo 62 (52%) del sexo masculino con edad media 56±15 años y 57 (48%) del sexo femenino, con edad media 51±16 años. La media de la depuración de creatinina fue 16,4±17,9 mL/min. La etiología más común fue la asociación de hipertensión arterial y diabetes mellitus (45%). Actuaron de grupo control 99 mujeres con edad media 25±5 años y 51 varones con edad media 26±5 años. Al comparar la fuerza entre los pacientes y los sujetos sanos se halló 66,4% de fuerza muscular disminuida entre los portadores de insuficiencia renal crónica. Conclusión: la fuerza muscular se halla disminuida en 66,4% de los pacientes insuficiencia renal crónica. Se recomienda la detección oportuna y el tratamiento precoz del déficit de la fuerza muscular en este grupo de pacientes.

The protist parasite Tritrichomonas foetus displays a pear-shaped (PS) and a pseudocystic or endoflagellar form (EFF). Here, we characterised the ecto-phosphatase activity on the surface of EFF and compare its biochemical properties to that of the PS regarding rate of substrate hydrolysis, pH activation profile and sensitivity to well-known phosphatases inhibitors. Two strains exhibiting low- and high-cytotoxicity were used. The enzyme activities of PS and EFF exhibited similar characteristics of protein tyrosine phosphatases (PTP). However, the ecto-phosphatase activities for both forms presented distinct kinetic parameters and different inhibition patterns by PTP inhibitors, suggesting the presence of distinct ecto-enzyme activities between PS and EFF, as well, between both strains. Ultrastructural cytochemistry confirmed the differential distribution of the ecto-phosphatase activity during the EFF transformation. An increase in the percentage of the EFF resulted in a proportional increase in the ecto-phosphatase activity. During EFF reversion, ecto-phosphatase activity decreased and was restored to the level found in the parasites before EFF induction. PS and EFF from the high-cytotoxic strain exhibited higher ecto-phosphatase activities than PS and EFF from the low-cytotoxic strain, respectively. In both strains, the EFF was more cytotoxic and exhibited higher ecto-phosphatase activity when compared to the PS. A large part of the ecto-phosphatase activities of EFF from both strains and PS from the high-cytotoxic strain was irreversibly inhibited when the parasites were pre-treated with a specific antibody against amoebic PTP (anti-EhPRL). Immunoreaction assays revealed that the anti-EhPRL antibody cross-reacted with a 24-kDa protein differentially expressed on the cell surface of PS and EFF T. foetus. A positive correlation was observed between the surface expression of 24-kDa protein and ecto-phosphatase activity. Irreversible inhibition of a part of the ecto-phosphatase activities partially blocked the EFF induction and the cytotoxic effects exerted by both forms. These results suggest that the ecto-phosphatase activities could play a role on the EFF transformation and cytotoxicity of T. foetus.
