The role for insufficient iron has been examined in detail, but manganese also has an essential function in photosynthesis and dissolved concentrations in the Southern Ocean can be strongly depleted. However, clear evidence for or against manganese limitation in this system is lacking. Here we present results from ten experiments distributed across Drake Passage. We found manganese (co-)limited phytoplankton growth and macronutrient consumption in central Drake Passage, whilst iron limitation was widespread nearer the South American and Antarctic continental shelves. Spatial patterns were reconciled with the different rates and timescales for removal of each element from seawater. Our results suggest an important role for manganese in modelling Southern Ocean productivity and understanding major nutrient drawdown in glacial periods.
The sea-surface microlayer (SML) is at the uppermost surface of the ocean, linking the hydrosphere with the atmosphere. The presence and enrichment of organic compounds in the SML have been suggested ...to influence air–sea gas exchange processes as well as the emission of primary organic aerosols. Here, we report on organic matter components collected from an approximately 50 µm thick SML and from the underlying water (ULW), ∼ 20 cm below the SML, in December 2012 during the SOPRAN METEOR 91 cruise to the highly productive, coastal upwelling regime off the coast of Peru. Samples were collected at 37 stations including coastal upwelling sites and off-shore stations with less organic matter and were analyzed for total and dissolved high molecular weight (> 1 kDa) combined carbohydrates (TCCHO, DCCHO), free amino acids (FAA), total and dissolved hydrolyzable amino acids (THAA, DHAA), transparent exopolymer particles (TEP), Coomassie stainable particles (CSPs), total and dissolved organic carbon (TOC, DOC), total and dissolved nitrogen (TN, TDN), as well as bacterial and phytoplankton abundance. Our results showed a close coupling between organic matter concentrations in the water column and in the SML for almost all components except for FAA and DHAA that showed highest enrichment in the SML on average. Accumulation of gel particles (i.e., TEP and CSP) in the SML differed spatially. While CSP abundance in the SML was not related to wind speed, TEP abundance decreased with wind speed, leading to a depletion of TEP in the SML at about 5 m s−1. Our study provides insight to the physical and biological control of organic matter enrichment in the SML, and discusses the potential role of organic matter in the SML for air–sea exchange processes.

kDa) dissolved organic matter and in ...particles from seawater samples. Monomeric sugars are determined after acid hydrolysis and neutralization through acid evaporation using high performance anion exchange chromatography (HPAEC) coupled with pulsed amperometric detection (PAD). The separation of sugars during chromatography is achieved in two steps, an isocratic elution (18
mM NaOH) followed by a gradient course of two mobile eluent phases (NaOH and CH
3COONa). HPAEC-PAD has previously been applied to measure neutral and amino sugars in marine samples. Since salt anions interfere with the measurement, some of the earlier studies used ion exchange resins for seawater desalting. Thereby, variable losses of neutral and amino sugars, and the complete removal of acidic sugars have been reported. Here, we show that desalting by membrane dialysis (1
kDa) is an efficient alternative to ion exchange resins and yields recoveries of >
90% for HMW carbohydrates. We conducted several tests to determine the accuracy and reproducibility of the method. Sugar concentrations determined with our protocol were compared to results obtained with the colorimetric TPTZ-method, and with earlier HPAEC-PAD protocols using cation/anion exchange resins. Applications of our protocol to field samples indicated that acidic sugars can comprise a substantial fraction (30–50%) of HMW dissolved carbohydrates in seawater. The simultaneous analysis of the three classes of sugars appears promising to detect a larger fraction of marine combined carbohydrates, and thus to improve our understanding of organic matter cycling in the ocean.

Nutrient limitation of oceanic primary production exerts a fundamental control on marine food webs and the flux of carbon into the deep ocean. The extensive boundaries of the oligotrophic ...sub-tropical gyres collectively define the most extreme transition in ocean productivity, but little is known about nutrient limitation in these zones. Here we present the results of full-factorial nutrient amendment experiments conducted at the eastern boundary of the South Atlantic gyre. We find extensive regions in which the addition of nitrogen or iron individually resulted in no significant phytoplankton growth over 48 hours. However, the addition of both nitrogen and iron increased concentrations of chlorophyll a by up to approximately 40-fold, led to diatom proliferation, and reduced community diversity. Once nitrogen-iron co-limitation had been alleviated, the addition of cobalt or cobalt-containing vitamin B
could further enhance chlorophyll a yields by up to threefold. Our results suggest that nitrogen-iron co-limitation is pervasive in the ocean, with other micronutrients also approaching co-deficiency. Such multi-nutrient limitations potentially increase phytoplankton community diversity.
About 5 trillion plastic particles are present in our oceans, from the macro to the micro size. Like any other aquatic particulate, plastics and microplastics can create a micro-environment, within ...which microbial and chemical conditions differ significantly from the surrounding water. Despite the high and increasing abundance of microplastics in the ocean, their influence on the transformation and composition of marine organic matter is largely unknown. Chromophoric dissolved organic matter (CDOM) is the photo-reactive fraction of the marine dissolved organic matter (DOM) pool. Changes in CDOM quality and quantity have impacts on marine microbial dynamics and the underwater light environment. One major source of CDOM is produced by marine bacteria through their alteration of pre-existing DOM substrates. In a series of microcosm experiments in controlled marine conditions, we explored the impact of microplastics on the quality and quantity of microbial CDOM. In the presence of microplastics we observed an increased production of CDOM with changes in its molecular weight, which resulted from either an increased microbial CDOM production or an enhanced transformation of DOM from lower to higher molecular weight CDOM. Our results point to the possibility that marine microplastics act as localized hot spots for microbial activity, with the potential to influence marine carbon dynamics.
The chemical speciation of iron (Fe) in oceans is influenced by ambient pH, dissolved oxygen, and the concentrations and strengths of the binding sites of dissolved organic matter (DOM). Here, we ...derived new nonideal competitive adsorption (NICA) constants for Fe(III) binding to marine DOM via pH-Fe titrations. We used the constants to calculate Fe(III) speciation and derive the apparent Fe(III) solubility (SFe(III)app) in the ambient water column across the Peruvian shelf and slope region. We define SFe(III)app as the sum of aqueous inorganic Fe(III) species and Fe(III) bound to DOM at a free Fe (Fe3+) concentration equal to the limiting solubility of Fe hydroxide (Fe(OH)3(s)). A ca. twofold increase in SFe(III)app in the oxygen minimum zone (OMZ) compared to surface waters is predicted. The increase results from a one order of magnitude decrease in H+ concentration which impacts both Fe(III) hydroxide solubility and organic complexation. A correlation matrix suggests that changes in pH have a larger impact on SFe(III)app and Fe(III) speciation than DOM in this region. Using Fe(II) measurements, we calculated ambient DFe(III) and compared the value with the predicted SFe(III)app. The underlying distribution of ambient DFe(III) largely reflected the predicted SFe(III)app, indicating that decreased pH as a result of OMZ intensification and ocean acidification may increase SFe(III)app with potential impacts on surface DFe inventories.
The sea surface microlayer (SML) covers more than 70% of the Earth's surface and is the boundary layer interface between the ocean and the atmosphere. This important biogeochemical and ecological ...system is critical to a diverse range of Earth system processes, including the synthesis, transformation and cycling of organic material, and the air-sea exchange of gases, particles and aerosols. In this review we discuss the SML paradigm, taking into account physicochemical and biological characteristics that define SML structure and function. These include enrichments in biogenic molecules such as carbohydrates, lipids and proteinaceous material that contribute to organic carbon cycling, distinct microbial assemblages that participate in air-sea gas exchange, the generation of climate-active aerosols and the accumulation of anthropogenic pollutants with potentially serious implications for the health of the ocean. Characteristically large physical, chemical and biological gradients thus separate the SML from the underlying water and the available evidence implies that the SML retains its integrity over wide ranging environmental conditions. In support of this we present previously unpublished time series data on bacterioneuston composition and SML surfactant activity immediately following physical SML disruption; these imply timescales of the order of minutes for the reestablishment of the SML following disruption. A progressive approach to understanding the SML and hence its role in global biogeochemistry can only be achieved by considering as an integrated whole, all the key components of this complex environment.
The sea surface microlayer (SML) represents the boundary layer at the air–sea
interface. Microbial eukaryotes in the SML potentially influence air–sea gas
exchange directly by taking up and producing ...gases and indirectly by
excreting and degrading organic matter, which may modify the viscoelastic
properties of the SML. However, little is known about the distribution of
microbial eukaryotes in the SML. We studied the composition of the microbial
community, transparent exopolymer particles and polysaccharides in the SML
during the PEACETIME cruise along a west–east transect in the Mediterranean
Sea, covering the western basin, Tyrrhenian Sea and Ionian Sea. At the
stations located in the Ionian Sea, fungi – likely of continental origin and
delivered by atmospheric deposition – were found in high relative abundances,
making up a significant proportion of the sequences recovered. Concomitantly,
bacterial and picophytoplankton counts decreased from west to east, while
transparent exopolymer particle (TEP) abundance and total carbohydrate (TCHO)
concentrations remained constant in all basins. Our results suggest that the
presence of substrates for fungi, such as Cladosporium, known to take up phytoplankton-derived polysaccharides, in combination with decreased
substrate competition by bacteria, might favor fungal dominance in the neuston
of the Ionian Sea and other low-nutrient, low-chlorophyll (LNLC) regions.
When the K
channel-like protein Kesv from Ectocarpus siliculosus virus 1 is heterologously expressed in mammalian cells, it is sorted to the mitochondria. This targeting can be redirected to the ...endoplasmic reticulum (ER) by altering the codon usage in distinct regions of the gene or by inserting a triplet of hydrophobic amino acids (AAs) into the protein's C-terminal transmembrane domain (ct-TMD). Systematic variations in the flavor of the inserted AAs and/or its codon usage show that a positive charge in the inserted AA triplet alone serves as strong signal for mitochondria sorting. In cases of neutral AA triplets, mitochondria sorting are favored by a combination of hydrophilic AAs and rarely used codons; sorting to the ER exhibits the inverse dependency. This propensity for ER sorting is particularly high when a common codon follows a rarer one in the AA triplet; mitochondria sorting in contrast is supported by codon uniformity. Since parameters like positive charge, hydrophobic AAs, and common codons are known to facilitate elongation of nascent proteins in the ribosome the data suggest a mechanism in which local changes in elongation velocity and co-translational folding in the ct-TMD influence intracellular protein sorting.
The interface layer between ocean and atmosphere is only a couple of micrometers thick but plays a critical role in climate relevant processes, including the air-sea exchange of gas and heat and the ...emission of primary organic aerosols (POA). Recent findings suggest that low-level cloud formation above the Arctic Ocean may be linked to organic polymers produced by marine microorganisms. Sea ice harbors high amounts of polymeric substances that are produced by cells growing within the sea-ice brine. Here, we report from a research cruise to the central Arctic Ocean in 2012. Our study shows that microbial polymers accumulate at the air-sea interface when the sea ice melts. Proteinaceous compounds represented the major fraction of polymers supporting the formation of a gelatinous interface microlayer and providing a hitherto unrecognized potential source of marine POA. Our study indicates a novel link between sea ice-ocean and atmosphere that may be sensitive to climate change.
