It has been suggested that cross-frequency phase-amplitude coupling (PAC), particularly in temporal brain structures, serves as a neural mechanism for coordinated working memory storage. In this ...magnetoencephalography study, we show that during visual working memory maintenance, temporal cortex regions, which exhibit enhanced PAC, interact with prefrontal cortex via enhanced low-frequency phase synchronization. Healthy human participants were engaged in a visual delayed match-to-sample task with pictures of natural objects. During the delay period, we observed increased spectral power of beta (20-28 Hz) and gamma (40-94 Hz) bands as well as decreased power of theta/alpha band (7-9 Hz) oscillations in visual sensory areas. Enhanced PAC between the phases of theta/alpha and the amplitudes of beta oscillations was found in the left inferior temporal cortex (IT), an area known to be involved in visual object memory. Furthermore, the IT was functionally connected to the prefrontal cortex by increased low-frequency phase synchronization within the theta/alpha band. Together, these results point to a mechanism in which the combination of PAC and long-range phase synchronization subserves enhanced large-scale brain communication. They suggest that distant brain regions might coordinate their activity in the low-frequency range to engage local stimulus-related processing in higher frequencies via the combination of long-range, within-frequency phase synchronization and local cross-frequency PAC.
Working memory maintenance, like other cognitive functions, requires the coordinated engagement of brain areas in local and large-scale networks. However, the mechanisms by which spatially distributed brain regions share and combine information remain primarily unknown. We show that the combination of long-range, low-frequency phase synchronization and local cross-frequency phase-amplitude coupling might serve as a mechanism to coordinate memory processes across distant brain areas. In this study, low-frequency phase synchronization between prefrontal and temporal cortex co-occurred with local cross-frequency phase-amplitude coupling to higher frequencies in the latter. By such means, ongoing working memory storage taking place in higher frequencies in temporal regions might be effectively coordinated by distant frontal brain regions through synchronized activity in the low-frequency range.
Intrinsic cortical dynamics modulates the processing of sensory information and therefore may be critical for conscious perception 1–3. We tested this hypothesis by electroencephalographic recording ...of ongoing and stimulus-related brain activity during stepwise drug-induced loss of consciousness in healthy human volunteers. We found that progressive loss of consciousness was tightly linked to the emergence of a hypersynchronous cortical state in the alpha frequency range (8–14 Hz). This drug-induced ongoing alpha activity was widely distributed across the frontal cortex. Stimulus-related responses to median nerve stimulation consisted of early and midlatency response components in primary somatosensory cortex (S1) and a late component also involving temporal and parietal regions. During progressive sedation, the early response was maintained, whereas the midlatency and late responses were reduced and eventually vanished. The antagonistic relation between the late sensory response and ongoing alpha activity held for constant drug levels on the single-trial level. Specifically, the late response component was negatively correlated with the power and long-range coherence of ongoing frontal alpha activity. Our results suggest blocking of intracortical communication by hypersynchronous ongoing activity as a key mechanism for the loss of consciousness.

The ascending modulatory systems of the brain stem are powerful regulators of global brain state. Disturbances of these systems are implicated in several major neuropsychiatric disorders. Yet, how ...these systems interact with specific neural computations in the cerebral cortex to shape perception, cognition, and behavior remains poorly understood. Here, we probed into the effect of two such systems, the catecholaminergic (dopaminergic and noradrenergic) and cholinergic systems, on an important aspect of cortical computation: its intrinsic variability. To this end, we combined placebo-controlled pharmacological intervention in humans, recordings of cortical population activity using magnetoencephalography (MEG), and psychophysical measurements of the perception of ambiguous visual input. A low-dose catecholaminergic, but not cholinergic, manipulation altered the rate of spontaneous perceptual fluctuations as well as the temporal structure of "scale-free" population activity of large swaths of the visual and parietal cortices. Computational analyses indicate that both effects were consistent with an increase in excitatory relative to inhibitory activity in the cortical areas underlying visual perceptual inference. We propose that catecholamines regulate the variability of perception and cognition through dynamically changing the cortical excitation-inhibition ratio. The combined readout of fluctuations in perception and cortical activity we established here may prove useful as an efficient and easily accessible marker of altered cortical computation in neuropsychiatric disorders.

3. Here, we used magnetoencephalography (MEG) to link the dynamics of neural population activity in human motor cortex to perceptual choices in a “yes/no” visual motion detection task. We found that (1) motor response-selective MEG activity in the “gamma” (64–100 Hz) and “beta” (12–36 Hz) frequency ranges predicted subjects' choices several seconds before their overt manual response; (2) this choice-predictive activity built up gradually during stimulus viewing toward both “yes” and “no” choices; and (3) the choice-predictive activity in motor cortex reflected the temporal integral of gamma-band activity in motion-sensitive area MT during stimulus viewing. Because gamma-band activity in MT reflects visual motion strength
4, these findings suggest that, during motion detection, motor plans for both “yes” and “no” choices result from continuously accumulating sensory evidence. We conclude that frequency-specific neural population activity at the cortical output stage of sensorimotor pathways provides a window into the mechanisms underlying perceptual decisions.
The extent to which deep brain stimulation (DBS) can improve quality of life may be perceived as a permanent trade-off between neurological improvements and complications of therapy, comorbidities, ...and disease progression.
We retrospectively investigated 123 consecutive and non-preselected patients. Indications for DBS surgery were Parkinson's disease (82), dystonia (18), tremor of different etiology (21), Huntington's disease (1) and Gilles de la Tourette syndrome (1). AEs were defined as any untoward clinical occurrence, sign or patient complaint or unintended disease if related or unrelated to the surgical procedures, implanted devices or ongoing DBS therapy.
Over a mean/median follow-up period of 4.7 years (578 patient-years) 433 AEs were recorded in 106 of 123 patients (86.2%). There was no mortality or persistent morbidity from the surgical procedure. All serious adverse events (SAEs) that occurred within 4 weeks of surgery were reversible. Neurological AEs (193 in 85 patients) and psychiatric AEs (78 in 48 patients) were documented most frequently. AEs in 4 patients (suicide under GPI stimulation, weight gain >20 kg, impairment of gait and speech, cognitive decline >2 years following surgery) were severe or worse, at least possibly related to DBS and non reversible. In PD 23.1% of the STN-stimulated patients experienced non-reversible (or unknown reversibility) AEs that were at least possibly related to DBS in the form of impaired speech or gait, depression, weight gain, cognitive disturbances or urinary incontinence (severity was mild or moderate in 15 of 18 patients). Age and Hoehn&Yahr stage of STN-simulated PD patients, but not preoperative motor impairment or response to levodopa, showed a weak correlation (r = 0.24 and 0.22, respectively) with the number of AEs.
DBS-related AEs that were severe or worse and non-reversible were only observed in PD (4 of 82 patients; 4.9%), but not in other diseases. PD patients exhibited a significant risk for non-severe AEs most of which also represented preexisting and progressive axial and non-motor symptoms of PD. Mild gait and/or speech disturbances were rather frequent complaints under VIM stimulation. GPI stimulation for dystonia could be applied with negligible DBS-related side effects.

Transcranial alternating current stimulation (tACS), applied to two brain sites with different phase lags, has been shown to modulate stimulation-outlasting functional EEG connectivity between the targeted regions. Given the lack of knowledge on mechanisms of tACS aftereffects, it is difficult to further enhance effect sizes and reduce variability in experiments. In this computational study, we tested if spike-timing-dependent plasticity (STDP) can explain stimulation-outlasting connectivity modulation by dual-site tACS and explored the effects of tACS parameter choices. Two populations of spiking neurons were coupled with synapses subject to STDP, and results were validated via a re-analysis of EEG data. Our simulations showed stimulation-outlasting connectivity changes between in- and anti-phase tACS, dependent on both tACS frequency and synaptic conduction delays. Importantly, both a simple network entraining to a wide range of tACS frequencies as well as a more realistic network that spontaneously oscillated at alpha frequency predicted that the largest effects would occur for short conduction delays between the stimulated regions. This finding agreed with experimental EEG connectivity modulation by 10Hz tACS, showing a clear negative correlation of tACS effects with estimated conduction delays between regions. In conclusion, STDP can explain connectivity aftereffects of dual-site tACS. However, not all combinations of tACS frequency and application sites are expected to effectively modulate connectivity via STDP. We therefore suggest using appropriate computational models and/or EEG analysis for planning and interpretation of dual-site tACS studies relying on aftereffects.

A promising approach for brain-computer interfaces (BCIs) employs the steady-state visual evoked potential (SSVEP) for extracting control information. Main advantages of these SSVEP BCIs are a simple ...and low-cost setup, little effort to adjust the system parameters to the user and comparatively high information transfer rates (ITR). However, traditional frequency-coded SSVEP BCIs require the user to gaze directly at the selected flicker stimulus, which is liable to cause fatigue or even photic epileptic seizures. The spatially coded SSVEP BCI we present in this article addresses this issue. It uses a single flicker stimulus that appears always in the extrafoveal field of view, yet it allows the user to control four control channels. We demonstrate the embedding of this novel SSVEP stimulation paradigm in the user interface of an online BCI for navigating a 2-dimensional computer game. Offline analysis of the training data reveals an average classification accuracy of 96.9±1.64%, corresponding to an information transfer rate of 30.1±1.8 bits/min. In online mode, the average classification accuracy reached 87.9±11.4%, which resulted in an ITR of 23.8±6.75 bits/min. We did not observe a strong relation between a subject's offline and online performance. Analysis of the online performance over time shows that users can reliably control the new BCI paradigm with stable performance over at least 30 minutes of continuous operation.

The spatially-coded SSVEP BCI exploits changes in the topography of the steady-state visual evoked response to visual flicker stimulation in the extrafoveal field of view. In contrast to ...frequency-coded SSVEP BCIs, the operator does not gaze into any flickering lights; therefore, this paradigm can reduce visual fatigue. Other advantages include high classification accuracies and a simplified stimulation setup. Previous studies of the paradigm used stimulation intervals of a fixed duration. For frequency-coded SSVEP BCIs, it has been shown that dynamically adjusting the trial duration can increase the system's information transfer rate (ITR). We therefore investigated whether a similar increase could be achieved for spatially-coded BCIs by applying dynamic stopping methods. To this end we introduced a new stopping criterion which combines the likelihood of the classification result and its stability across larger data windows. Whereas the BCI achieved an average ITR of 28.4±6.4 bits/min with fixed intervals, dynamic intervals increased the performance to 81.1±44.4 bits/min. Users were able to maintain performance up to 60 minutes of continuous operation. We suggest that the dynamic response time might have worked as a kind of temporal feedback which allowed operators to optimize their brain signals and compensate fatigue.

Brain processing depends on the interactions between neuronal groups. Those interactions are governed by the pattern of anatomical connections and by yet unknown mechanisms that modulate the ...effective strength of a given connection. We found that the mutual influence among neuronal groups depends on the phase relation between rhythmic activities within the groups. Phase relations supporting interactions between the groups preceded those interactions by a few milliseconds, consistent with a mechanistic role. These effects were specific in time, frequency, and space, and we therefore propose that the pattern of synchronization flexibly determines the pattern of neuronal interactions.
