There has been a large increase in basic science activity in cell therapy and a growing portfolio of cell therapy trials. However, the number of industry products available for widespread clinical ...use does not match this magnitude of activity. We hypothesize that the paucity of engagement with the clinical community is a key contributor to the lack of commercially successful cell therapy products. To investigate this, we launched a pilot study to survey clinicians from five specialities and to determine what they believe to be the most significant barriers to cellular therapy clinical development and adoption. Our study shows that the main concerns among this group are cost-effectiveness, efficacy, reimbursement, and regulation. Addressing these concerns can best be achieved by ensuring that future clinical trials are conducted to adequately answer the questions of both regulators and the broader clinical community.
On 15 May 2003, two ground-based, mobile, Doppler radars scanned a supercell that moved through the Texas Panhandle and cyclically produced mesocyclones. The two radars collected data from the storm ...during a rapid cyclic mesocyclogenesis stage and a more slowly evolving tornadic period. A 3-cm-wavelength radar scanned the supercell continuously for a short time after it was cyclic but close to the time of tornadogenesis. A 5-cm-wavelength radar scanned the supercell the entire time it exhibited cyclic behavior and for an additional 30 min after that. The volumetric data obtained with the 5-cm-wavelength radar allowed for the individual circulations to be analyzed at multiple levels in the supercell. Most of the circulations that eventually dissipated moved rearward with respect to storm motion and were located at distances progressively farther away from the region of rear-flank outflow. The circulations associated with a tornado did not move nearly as far rearward relative to the storm. The mean circulation diameters were approximately 1-4 km and had lifetimes of 10-30 min. Circulation dissipation often, but not always, occurred following decreases in circulation diameter, while changes in maximum radial wind shear were not reliable indicators of circulation dissipation. In one instance, a pair of circulations rotated cyclonically around, and moved toward, each other; the two circulations then combined to form one circulation. Single-Doppler radial velocities from both radars were used to assess the differences between the pretornadic circulations and the tornadic circulations. Storm outflow in the rear flank of the storm increased notably during the time cyclic mesocyclogenesis slowed and tornado formation commenced. Large storm-relative inflow likely advected the pretornadic circulations rearward in the absence of organized outflow. The development of strong outflow in the rear flank probably balanced the strong inflow, allowing the tornadic circulations to stay in areas rich in vertical vorticity generation. The present study describes how the development of a pair bond modifies social, sexual and aggressive behavior. Five heterosexual pairs of marmosets, previously unknown to each other, were formed at ...the beginning of the study. At the onset of pairing, social, sexual, exploratory and aggressive behaviors were recorded for 40 min. The animals were then observed for 20 min, both in the morning and afternoon for 21 days. The frequency and/or duration of behaviors recorded on Day 1 were compared to those recorded at later observations. The behavior displayed shortly after pairing should be completely unaffected by the pair bond, while such a bond should be present at later observations. Thus, it was possible to determine how the behavior between the pair was modified by the development of a pair bond. Social behaviors increased from Day 1 to Days 2–6 and all subsequent days observed. Conversely, other behaviors, such as open mouth displays (usually considered to be an invitation to sexual activity), had a high frequency during the early part of cohabitation but declined towards the end. Consequently, pair bonding manifests itself in an increased intensity of social behaviors. It is suggested that the intrinsically rewarding properties of grooming and perhaps other social behaviors turn the pair mate into a positive incentive, activating approach and further interactions when possible. Thus, the pair bond may be a motivational state activated by the conditioned incentive properties of the partner. This notion can explain all forms of pair bonds, including those occurring between individuals of the same sex and in promiscuous species.

The long-term reliability and durability of photovoltaic (PV) modules have been critical to the recent growth of solar power. The analysis of timeseries data and ultraviolet fluorescent (UVF) images ...provide insights into critical degradation modes that affect the long-term performance of PV modules. From the timeseries data, we automate the calculation of performance loss rates for 67 different systems located throughout the state of Florida. From UVF imaging, we utilize a pre-trained Mask-RCNN deep learning network for the segmentation of PV modules out of noisy images. In order to perform both of these tasks, a considerable amount of time has to be put into cleaning and reorganizing the input data. This is important both for human understanding of the data and machine action-ability to perform analysis. FAIR principles can be used to assist in this cleaning process to produce datasets that are of high quality for use in machine learning tasks. Here, we apply the FAIR principles to timeseries and image analysis as a showcase for FAIRified workflows for the automation and standardization of datasets for use in machine learning.
The surface bidirectional reflectance distribution function (BRDF) contains valuable information on canopy physiognomy for desert grassland and grass–shrub transition communities. This information ...may be accessed by inverting a BRDF model against sets of observations, which encompass important variations in viewing and illumination angles. This paper shows that structural canopy attributes can be derived through inversion of the Simple Geometric Model (SGM) of the BRDF developed in this paper. It is difficult to sample BRDF features from the ground because of the discontinuous nature of the canopies and long intrinsic length scales in remotely sensed spectral measures (>10 m). A multispectral digital camera was therefore used to derive spatial multiangular reflectance data sets from the air and the SGM was validated against and inverted with these. It was also validated using 3-D radiosity simulations driven with maps of field-measured plant dimensions. The interpretation of the retrieved parameter maps (shrub density, shrub width and canopy height) reveals variations in canopy structure within desert grassland and grassland–shrubland transition communities, which are clearly related to structural and optical features in high resolution panchromatic and vegetation index images. To our knowledge, this paper reports on the first attempts to acquire structural canopy attributes of desert landscapes using multiple view angle data at scales less than 1 km. The results point to further opportunities to exploit multiangular data from spaceborne sensors such as the Multiangle Imaging SpectroRadiometer (MISR) and the Compact High Resolution Imaging Spectrometer (CHRIS) on the NASA Terra and European Space Agency's PROBA satellites, respectively.

A committee assembled by the American Academy of Neurology (AAN) reassessed the evidence related to the care of women with epilepsy (WWE) during pregnancy, including preconceptional folic ...acid and prenatal vitamin K use and the clinical implications of placental and breast‐milk transfer of antiepileptic drugs (AEDs). The committee evaluated the available evidence based on a structured literature review and classification of relevant articles. Preconceptional folic acid supplementation is possibly effective in preventing major congenital malformations in the newborns of WWE taking AEDs. There is inadequate evidence to determine if the newborns of WWE taking AEDs have a substantially increased risk of hemorrhagic complications. Primidone and levetiracetam probably transfer into breast milk in clinically important amounts. Valproate, phenobarbital, phenytoin, and carbamazepine probably are not transferred into breast milk in clinically important amounts. Pregnancy probably causes an increase in the clearance and a decrease in the concentrations of lamotrigine, phenytoin, and, to a lesser extent carbamazepine, and possibly decreases the level of levetiracetam and the active oxcarbazepine metabolite, the monohydroxy derivative (MHD). Supplementing WWE with at least 0.4 mg of folic acid before pregnancy may be considered. Monitoring of lamotrigine, carbamazepine, and phenytoin levels during pregnancy should be considered, and monitoring of levetiracetam and oxcarbazepine (as MHD) levels may be considered. A paucity of evidence limited the strength of many recommendations.
BRCA1 is a breast cancer susceptibility gene that is down-regulated in a significant proportion of sporadic breast cancers. BRCA1 is posttranscriptionally regulated by RNA-binding proteins, the ...identities of which are unknown. HuR is an RNA binding protein implicated in posttranscriptional regulation of many genes and is overexpressed in sporadic breast cancer. To investigate the possibility that these two molecules are functionally linked in breast cancer, we performed bioinformatic analysis of the BRCA1 3' untranslated region (UTR), RNA-protein assays with the HuR protein and the BRCA1 3'UTR, and immunohistochemical analysis of a cohort of breast tumors using antibodies against BRCA1 and HuR. Here, we describe the identification of two predicted HuR-binding sites in the BRCA1 3'UTR, one of which binds specifically to HuR. We also show that this interaction is disrupted by single nucleotide substitutions in the BRCA1 3'UTR and that endogenous HuR protein associates with BRCA1 transcripts in T47D and MCF7 breast cancer cells. Expression of ectopic HuR results in a significant decrease in BRCA1 protein expression and also BRCA1 3'UTR activity. Immunohistochemical analysis revealed that although BRCA1 and HuR expression were associated with some clinicopathologic features of the tumors, there was no statistically significant correlation between BRCA1 and HuR protein expression. These results identify the first posttranscriptional protein regulator of BRCA1 and have implications for understanding BRCA1 regulation in human breast cancer.
