Summary
A rice node is a hub for distribution of mineral elements; however, most genes highly expressed in the node have not been functionally characterized.
Transcriptomic analysis of a rice node ...revealed that two metallothionein genes, OsMT2b and OsMT2c, were highly expressed in the node I. We functionally characterized these genes in terms of gene expression pattern, cellular and subcellular localization, phenotypic analysis of the single and double knockout mutants and metal‐binding ability.
Both OsMT2b and OsMT2c were mainly and constitutively expressed in the phloem region of enlarged and diffuse vascular bundles in the nodes and of the anther. Knockout of either OsMT2b or OsMT2c increased zinc (Zn) accumulation in the nodes, but decreased Zn distribution to the panicle, resulting in decreased grain yield. A double mutant, osmt2bmt2c, showed further negative effects on the Zn distribution and grain yield. By contrast, knockout of OsMT2b had a small effect on copper (Cu) accumulation. Both OsMT2b and OsMT2c showed binding ability with Zn, whereas only OsMT2b showed binding ability with Cu in yeast.
Our results suggest that both OsMT2b and OsMT2c play an important role mainly in the distribution of Zn to grain through chelation and subsequent transport of Zn in the phloem in rice.
Macrophages are heterogeneous cell populations that are present in all tissues. Macrophages can be divided into classically activated inflammatory macrophages (M1) and alternatively activated ...anti-inflammatory macrophages (M2). It has been generally accepted that M1 macrophages are polarised in an inflammatory environment to produce pro-inflammatory cytokines, whilst M2 macrophages are involved in anti-inflammation and aid tissue repair in wound healing. Bacterial endotoxin (lipopolysaccharide; LPS) is a potent factor in infection, which induces M1 macrophages resulting in higher levels of iNOS, TNFα and IL-12p70 which dictate inflammatory T cell responses. M2 macrophages can be transformed into M1 macrophages following LPS stimulation to promote inflammation. Candida albicans is a commensal fungal microorganism, which has been suggested to induce immune tolerance; however, the mechanism of C. albicans-induced immune tolerance has not been investigated in detail. IL-35 is a recently identified anti-inflammatory cytokine which is a heterodimeric protein consisting of the Epstein-Barr virus-induced gene 3 (EBI3) and IL-12p35. IL-35 shares the protein subunit p35, with IL-12p70. IL-12p70 is the most potent cytokine to induce Th1 responses during inflammation. In this study, we demonstrate that heat-killed C. albicans (HKC) strongly suppressed LPS-induced IL-12p70 production in M2 macrophages. Candida albicans induced a high level of EBI3 expression in M2 macrophages, which served as a mechanism for IL-12p70 suppression by competitive binding of the common protein subunit (p35) of IL-35 and IL-12p70. To demonstrate that EBI3 expression had the ability to block IL-12p70 production intracellularly, a Chinese Hamster Ovary (CHO) cell line with biscistronic expression of IL-12p40 and p35 was constructed, followed by ectopic over-expression of EBI3. The over-expression of EBI3 in the IL-12p70 producing cell line effectively suppressed IL-12p70 production. IL-35 secretion was also detected in the cell line, with suppressed IL-12p70 production by immune-precipitation Western blotting. However, this secretion was not evident in M2 macrophages following stimulation by HKC. This can be explained by the constitutive expression of IL-35 receptors (gp130 and IL-12Rβ2) in M2 macrophages for cytokine consumption. Our results have indicated that C. albicans can suppress host inflammatory responses in mucosal skin by suppressing LPS-induced IL-12p70 production. Lower IL-12p70 production may avoid an unnecessary Th1 response in order to retain immune tolerance, which may be one of the mechanisms by which C. albicans achieves a successful commensal lifestyle without having a detrimental effect on the host's health.

We performed proteomic analysis with the leaf tonoplast-rich fraction and identified two half-size ABC transporters; FeASL1.1 and FeALS1.2. We investigated the gene expression patterns and subcellular localization. To demonstrate their physiological role, we expressed FeALS1.1 or FeALS1.2 in the Arabidopsis atals1 mutant under the control of AtALS1 promoter.
FeALS1.1 expression was upregulated by Al in both the leaves and the roots, and its expression level in the roots was six times higher than its homologous gene (AtALS1) of Arabidopsis. FeALS1.2 expression, however, was not affected by Al but showed a 39 times higher expression level than AtALS1 in the leaves. When FeALS1.1 or FeALS1.2 was expressed in atals1, both of them recovered their Al tolerance through altering the subcellular localization of Al in root cells.
Taken together, our results indicate that FeALS1.1 and FeALS1.2 are involved in the internal detoxification of Al in the roots and leaves, respectively, by sequestering Al into the vacuoles. Their high expression is probably required for high Al tolerance in buckwheat.
With a huge amount of protein sequence data, the computational method for protein-protein interaction (PPI) prediction using only the protein sequences information have drawn increasing interest. In ...this article, we propose a sequence-based method based on a novel representation of local protein sequence descriptors. Local descriptors account for the interactions between residues in both continuous and discontinuous regions of a protein sequence, so this method enables us to extract more PPI information from the sequence. A series of elaborate experiments are performed to optimize the prediction model by varying the parameter k and the distance measuring function of the k-nearest neighbors learning system and the ways of coding a protein pair. When performed on the PPI data of Saccharomyces cerevisiae, the method achieved 86.15% prediction accuracy with 81.03% sensitivity at the precision of 90.24%. An independent data set of 986 Escherichia coli PPIs was used to evaluate this prediction model and the prediction accuracy is 73.02%. Given the complex nature of PPIs, the performance of our method is promising, and it can be a helpful supplement for PPIs prediction.
The wide Mg alloy sheets produced by hot extrusion usually can easily form an inhomogeneous texture, resulting in anisotropic mechanical properties and poor formability. However, few studies have ...been carried out on the bulk texture investigation at different areas of as-extruded Mg alloy sheets, especially the Mg alloys with different alloying elements. In this work, the effect of Al on the bulk texture and mechanical properties at different areas for three wide Mg-Al-Zn alloy sheets with different Al contents (Mg-3Al-0.5Zn, Mg-8Al-0.5Zn and Mg-9Al-0.5Zn) are mainly investigated by neutron diffraction. The results showed that a strong and uneven basal texture was formed in the Mg-3Al-0.5Zn sheet. Meanwhile, the intensity of the basal texture was significantly weakened due to the numerous fine precipitates of Mg17Al12 particles, with the Al content increasing, which hinder the grain growth during extrusion, while fine recrystallized grains have a more random orientation. The enhanced tensile properties in Mg-8Al-0.5Zn and Mg-9Al-0.5Zn alloy sheets are ascribed to the cooperation effect of a refined microstructure, precipitates and weakened basal texture. Among the three Mg alloy sheets, the Mg-8Al-0.5Zn alloy sheet has a yield strength of about 270 MPa, an ultimate tensile strength of about 330 MPa and ultimate elongation of about 16% in the extrusion direction, which possesses the most excellent comprehensive mechanical properties.
Soybean mosaic virus （SMV） disease is one of the most destructive viral diseases in soybean （Glycine max （L.） Merr.）. SMV strain SC3 is the major prevalent strain in huang-huai and Yangtze valleys, ...China. The soybean cultivar Qihuang 1 is of a rich resistance spectrum and has a wide range of application in breeding programs in China. In this study, F1, F2 and F2：3 from Qihuang 1×nannong 1138-2 were used to study inheritance and linkage mapping of the SC3 resistance gene in Qihuang 1. The secondary F2 population and near isogenic lines （nILs） derived from residual heterozygous lines （RhLs） of Qihuang 1×nannong 1138-2 were separatively used in the ifne mapping and candidate gene analysis of the resistance gene. Results indicated that a single dominant gene （designated RSC3Q） controls resistance, which was located on chromosome 13. Two genomic-simple sequence repeat （SSR） markers BARCSOYSSR_13_1114 and BARCSOYSSR_13_1136 were found lfanking the two sides of the RSC3Q. The interval between the two markers was 651 kb. Quantitative real-time PCR analysis of the candidate genes showed that ifve genes （Glyma13g25730, 25750, 25950, 25970 and 26000） were likely involved in soybean SMV resistance. These results would have utility in cloning of RSC3Q resistance candidate gene and marker-assisted selection （MaS） in resistance breeding to SMV.
Five previously undescribed monoterpenoid indole alkaloids, mappianines A−E, along with twelve known analogues, were isolated from the stems of Mappianthus iodoides Hand.-Mazz. Their structures and ...absolute configurations were determined by spectroscopic analysis, single-crystal X-ray diffraction, and ECD calculations. The plausible biogenetic pathway of mappianine A was proposed. All the isolated compounds were evaluated for their cytotoxic effects on MGC-803, Bel-7404, A549, NCI-H460, and HepG2 cancer cell lines. Mappianine B, tetrahydroalstonine, β-carbolin-1-one, and 1,2,3,4-tetrahydronorharman-1-one displayed moderate cytotoxicity against all cell lines tested, with IC50 values ranging from 5.19 to 42.86 μM.

Mermithid nematodes, such as Ovomermis sinensis, are used as biological control agents against many insect pests, including cotton bollworm (Helicoverpa armigera). However, given the host's robust ...immune system, the infection rate of O. sinensis is low, thus restricting its widespread use. To understand the host defense mechanisms against mermithid nematodes, we identified and characterized a protein involved in the recognition of O. sinensis, the potential O. sinensis‐binding protein C‐type lectin 1 (HaCTL1a and/or HaCTL1b), which was eluted from the surface of O. sinensis after incubation with H. armigera plasma. HaCTL1b is homologous to the previously reported HaCTL1a protein. HaCTL1 was predominantly expressed in hemocytes and was induced by the steroid hormone 20‐hydroxyecdysone through ecdysone receptor (HaEcR) or ultraspiracle (HaUSP), or both. Binding assays confirmed the interactions of the HaCTL1 proteins with O. sinensis but not with Romanomermis wuchangensis, a parasitic nematode of mosquito. Moreover, the HaCTL1 proteins were secreted into the hemocoel and promoted hemocyte‐mediated encapsulation and phagocytosis. A knockdown of HaEcR and/or HaUSP resulted in compromised encapsulation and phagocytosis. Thus, HaCTL1 appears to modulate cellular immunity in the defense against parasitic nematodes, and the 20‐hydroxyecdysone–HaEcR–HaUSP complex is involved in regulating the process.
A proposed model for the induction of HaCTL1 upon 20E signaling, thereby promoting cellular immunity. 20E‐HaEcR‐HaUSP complex induces the expression of HaCTL1 in hemocytes, followed by the secretion of HaCTL1 into the hemocoel. HaCTL1 then recognizes pathogens (such as parasitic nematodes and bacteria) and interacts with the unknown hemocytic receptor, which then activates cellular immune reactions such as encapsulation and phagocytosis.
