Digital information is fragile, yet access to digital information over time is a critical underpinning of core values and functions in our society, from open government to research and scholarship. ...Digital preservation research has focused on identifying technical, economic, and organizational sources of risk and has relied on an assumption that individuals will behave in rational and predictable ways in response to the same information. This article asserts that viewing digital preservation as a process that takes place in complex social contexts is just as important as thinking about digital preservation in terms of technological or economic issues. This is particularly important for understanding how individuals involved in digital preservation construct their understanding of risk because social factors influence how people construct their understanding of, and behave in response to, risk information. The author proposes an eight‐factor model for the social construction of risk, which includes: communication, complexity, expertise, organizations, political culture, trust, uncertainty, and vulnerability. The article demonstrates how these factors influence individuals as they construct their understanding of risk in the context of digital preservation and how this in turn affects digital preservation decisions.
Teleost fish represent the most diverse of the vertebrate groups and play important roles in food webs, as ecosystem engineers, and as vectors for microorganisms. However, the microbial ecology of ...fishes remains underexplored for most host taxa and for certain niches on the fish body. This is particularly true for the gills, the key sites of respiration and waste exchange in fishes. Here we provide a comprehensive analysis of the gill microbiome. We focus on ecologically diverse taxa from coral reefs around Moorea, sampling the gills and intestines of adults and juveniles representing 15 families. The gill microbiome composition differed significantly from that of the gut for both adults and juveniles, with fish-associated niches having lower alpha diversity values and higher beta diversity values than those for seawater, sediment, and alga-associated microbiomes. Of ∼45,000 operational taxonomic units (OTUs) detected across all samples, 11% and 13% were detected only in the gill and the intestine, respectively. In adult fish, both gill and intestinal microbiomes varied significantly among host species grouped by diet category. Gill and intestinal microbiomes from the same individual were more similar to one another than to gill and intestinal microbiomes from different individuals. These results demonstrate that distinct body sites are jointly influenced by host-specific organizing factors operating at the level of the host individual. The results also identify taxonomic signatures unique to the gill and the intestine, confirming fish-associated niches as distinct reservoirs of marine microbial diversity.
Fish breathe and excrete waste through their gills. The gills are also potential sites of pathogen invasion and colonization by other microbes. However, we know little about the microbial communities that live on the gill and the factors shaping their diversity. Focusing on ecologically distinct types of coral reef fish, we provide a comprehensive analysis of the fish gill microbiome. By comparison to microbiomes of the gut and the surrounding environment, we identify microbes unique to the gill niche. These microbes may be targets for further studies to determine the contribution of the microbiome to waste exchange or host immunity. We also show that despite exhibiting a unique taxonomic signature, the gill microbiome is influenced by factors that also influence the gut microbiome. These factors include the specific identity of the host individual. These results suggest basic principles describing how association with fishes structures the composition of microbial communities.
Risk is a foundational concept in digital preservation. While it has been examined from technical, economic, and organizational perspectives, I argue that it is also a social phenomenon. In this ...study I report on the results from 42 interviews with stakeholders in the Trustworthy Repositories Audit & Certification (TRAC) system, and analysis of documents relating to the ISO 16363 standard in order to examine how standard developers, auditors, and repository staff members understand the concept of risk for digital repositories. The results of this research demonstrate that members of these three stakeholder groups identified risk in the TRAC audit and certification process in terms of specific potential threats or sources of risk, which I have organized into five main categories: finance, legal, organizational governance, repository processes, and technical infrastructure. While standard developers, auditors, and repository staff generally shared an understanding of the major sources of potential risk that face digital repositories, they disagreed about whether and how these risks can be mitigated and how mitigation can be proven. Individuals who were more removed from the day-to-day work of the repositories undergoing an audit were more likely to accept well-documented risk identification and mitigation strategies as sufficient evidence of trustworthiness, while repository staff were skeptical that documentation was sufficient evidence of risk assessment and mitigation and thus questioned whether this would translate to actual trustworthiness for long-term digital preservation.

von Economo neurons (VENs) are bipolar, spindle-shaped neurons restricted to layer 5 of human frontoinsula and anterior cingulate cortex that appear to be selectively vulnerable to neuropsychiatric ...and neurodegenerative diseases, although little is known about other VEN cellular phenotypes. Single nucleus RNA-sequencing of frontoinsula layer 5 identifies a transcriptomically-defined cell cluster that contained VENs, but also fork cells and a subset of pyramidal neurons. Cross-species alignment of this cell cluster with a well-annotated mouse classification shows strong homology to extratelencephalic (ET) excitatory neurons that project to subcerebral targets. This cluster also shows strong homology to a putative ET cluster in human temporal cortex, but with a strikingly specific regional signature. Together these results suggest that VENs are a regionally distinctive type of ET neuron. Additionally, we describe the first patch clamp recordings of VENs from neurosurgically-resected tissue that show distinctive intrinsic membrane properties relative to neighboring pyramidal neurons.
While the apicomplexan parasites Plasmodium falciparum and Toxoplasma gondii are thought to primarily depend on glycolysis for ATP synthesis, recent studies have shown that they can fully catabolize ...glucose in a canonical TCA cycle. However, these parasites lack a mitochondrial isoform of pyruvate dehydrogenase and the identity of the enzyme that catalyses the conversion of pyruvate to acetyl-CoA remains enigmatic. Here we demonstrate that the mitochondrial branched chain ketoacid dehydrogenase (BCKDH) complex is the missing link, functionally replacing mitochondrial PDH in both T. gondii and P. berghei. Deletion of the E1a subunit of T. gondii and P. berghei BCKDH significantly impacted on intracellular growth and virulence of both parasites. Interestingly, disruption of the P. berghei E1a restricted parasite development to reticulocytes only and completely prevented maturation of oocysts during mosquito transmission. Overall this study highlights the importance of the molecular adaptation of BCKDH in this important class of pathogens.

Objective
Little is known about early knee osteoarthritis (OA). The significance of lesions on magnetic resonance imaging (MRI) in older persons without radiographic OA is unclear. Our objectives ...were to determine the extent of tissue pathology by MRI and evaluate its significance by testing the following hypotheses: cartilage damage, bone marrow lesions, and meniscal damage are associated with prevalent frequent knee symptoms and incident persistent symptoms; bone marrow lesions and meniscal damage are associated with incident tibiofemoral (TF) cartilage damage; and bone marrow lesions are associated with incident patellofemoral (PF) cartilage damage.
Methods
In a cohort study of 849 Osteoarthritis Initiative (OAI) participants who had a bilateral Kellgren/Lawrence (K/L) score of 0, we assessed cartilage damage, bone marrow lesions, and meniscal damage using the MRI OA Knee Score, as well as prevalent frequent knee symptoms, incident persistent symptoms, and incident cartilage damage. Multiple logistic regression (one knee per person) was used to evaluate associations between MRI lesions and each of these outcomes.
Results
Of the participants evaluated, 76% had cartilage damage, 61% had bone marrow lesions, 21% had meniscal tears, and 14% had meniscal extrusion. Cartilage damage (any; TF and PF), bone marrow lesions (any; TF and PF), meniscal extrusion, and body mass index (BMI) were associated with prevalent frequent symptoms. Cartilage damage (isolated PF; TF and PF), bone marrow lesions (any; isolated PF; TF and PF), meniscal tears, and BMI were associated with incident persistent symptoms. Hand OA, but no individual lesion type, was associated with incident TF cartilage damage, and bone marrow lesions (any; any PF) with incident PF damage. Having more lesion types was associated with a greater risk of outcomes.
Conclusion
MRI‐detected lesions are not incidental and may represent early disease in persons at increased risk of knee OA.
Research on rumination has demonstrated that compared with distraction, rumination intensifies and prolongs negative emotion. However, rumination and distraction differ both in
what
one thinks about ...and
how
one thinks about it. Do the negative outcomes of rumination result from how people think about negative events or simply that they think about them at all? To address this question, participants in 2 studies recalled a recent anger-provoking event and then thought about it in 1 of 2 ways: by ruminating or by reappraising. The authors examined the impact of these strategies on subsequent ratings of anger experience (Study 1) as well as on perseverative thinking and physiological responding over time (Study 2). Relative to reappraisal, rumination led to greater anger experience, more cognitive perseveration, and greater sympathetic nervous system activation. These findings provide compelling new evidence that how one thinks about an emotional event can shape the emotional response one has.
Purpose
Taking the researchers’ perspective, the purpose of this paper is to examine the types of context information needed to preserve data’s meaning in ways that support data reuse.
...Design/methodology/approach
This paper is based on a qualitative study of 105 researchers from three disciplinary communities: quantitative social science, archaeology and zoology. The study focused on researchers’ most recent data reuse experience, particularly what they needed when deciding whether to reuse data.
Findings
Findings show that researchers mentioned 12 types of context information across three broad categories: data production information (data collection, specimen and artifact, data producer, data analysis, missing data, and research objectives); repository information (provenance, reputation and history, curation and digitization); and data reuse information (prior reuse, advice on reuse and terms of use).
Originality/value
This paper extends digital curation conversations to include the preservation of context as well as content to facilitate data reuse. When compared to prior research, findings show that there is some generalizability with respect to the types of context needed across different disciplines and data sharing and reuse environments. It also introduces several new context types. Relying on the perspective of researchers offers a more nuanced view that shows the importance of the different context types for each discipline and the ways disciplinary members thought about them. Both data producers and curators can benefit from knowing what to capture and manage during data collection and deposit into a repository.
We describe convergent evidence from transcriptomics, morphology, and physiology for a specialized GABAergic neuron subtype in human cortex. ) matching a single transcriptomically defined cell type whose specific molecular marker signature is not seen in mouse cortex. Rosehip cells in layer 1 make homotypic gap junctions, predominantly target apical dendritic shafts of layer 3 pyramidal neurons, and inhibit backpropagating pyramidal action potentials in microdomains of the dendritic tuft. These cells are therefore positioned for potent local control of distal dendritic computation in cortical pyramidal neurons.
