Dentin is an important structural component of the tooth. Odontoblast differentiation is an essential biological process that guarantees normal dentin formation, which is precisely regulated by ...various proteins. Murine double minute 2 (Mdm2) is an E3 ubiquitin ligase, and it plays a pivotal role in the differentiation of different cell types, such as osteoblasts and myoblasts. However, whether Mdm2 plays a role in odontoblast differentiation remains unknown. Here, we investigated the spatiotemporal expression of Mdm2 by immunostaining and found that Mdm2 was highly expressed in the odontoblasts and slightly in the dental papilla cells of mouse incisors and molars. Gene knockdown and overexpression experiments verified that Mdm2 promoted the odontoblast-like differentiation of mouse dental papilla cells (mDPCs). Intranuclear colocalization and physical interaction between Mdm2 and distal-less 3 (Dlx3), a transcription factor important for odontoblast differentiation, was found during the odontoblast-like differentiation of mDPCs by double immunofluorescence and immunoprecipitation. Mdm2 was proved to monoubiquitinate Dlx3, which enhanced the expression of Dlx3 target gene Dspp. In addition, p53, the canonical substrate of Mdm2, was validated to be also ubiquitinated but degraded by Mdm2 during the odontoblast-like differentiation of mDPCs. Gene knockdown experiments confirmed that p53 inhibited the odontoblast-like differentiation of mDPCs. p53 and Mdm2 double knockdown partially rescued the reduced odontoblast-like differentiation by knockdown of Mdm2 alone. Taken together, our study revealed that Mdm2 promoted the odontoblast-like differentiation of mDPCs by ubiquitinating both Dlx3 and p53. On one hand, the monoubiquitination of Dlx3 by Mdm2 led to upregulation of Dspp, which is a marker of the odontoblast differentiation. On the other hand, ubiquitination of p53 by Mdm2 resulted in its degradation, which eliminated the inhibitory effect of p53 on the odontoblast-like differentiation of mDPCs.
Spatial pattern information of carbon density in forest ecosystem including forest litter carbon (FLC) plays an important role in evaluating carbon sequestration potentials. The spatial variation of ...FLC density in the typical subtropical forests in southeastern China was investigated using Moran's I, geostatistics and a geographical information system (GIS). A total of 839 forest litter samples were collected based on a 12 km (south–north) × 6 km (east–west) grid system in Zhejiang province. Forest litter carbon density values were very variable, ranging from 10.2 kg ha−1 to 8841.3 kg ha−1, with an average of 1786.7 kg ha−1. The aboveground biomass had the strongest positive correlation with FLC density, followed by forest age and elevation. Global Moran's I revealed that FLC density had significant positive spatial autocorrelation. Clear spatial patterns were observed using local Moran's I. A spherical model was chosen to fit the experimental semivariogram. The moderate "nugget-to-sill" (0.536) value revealed that both natural and anthropogenic factors played a key role in spatial heterogeneity of FLC density. High FLC density values were mainly distributed in northwestern and western part of Zhejiang province, which were related to adopting long-term policy of forest conservation in these areas, while Hang-Jia-Hu (HJH) Plain, Jin-Qu (JQ) Basin and coastal areas had low FLC density due to low forest coverage and intensive management of economic forests. These spatial patterns were in line with the spatial-cluster map described by local Moran's I. Therefore, Moran's I, combined with geostatistics and GIS, could be used to study spatial patterns of environmental variables related to forest ecosystem.
The nearly continuous light curves with micromagnitude precision provided by the space mission Kepler are revolutionizing our view of pulsating stars. They have revealed a vast sea of low-amplitude ...pulsation modes that were undetectable from Earth. The long time base of Kepler light curves allows for the accurate determination of the frequencies and amplitudes of pulsation modes needed for in-depth asteroseismic modeling. However, for an asteroseismic study to be successful, the first estimates of stellar parameters need to be known and they cannot be derived from the Kepler photometry itself. The Kepler Input Catalog provides values for the effective temperature, surface gravity, and metallicity, but not always with sufficient accuracy. Moreover, information on the chemical composition and rotation rate is lacking. We are collecting low-resolution spectra for objects in the Kepler field of view with the Large Sky Area Multi-Object Fiber Spectroscopic Telescope (LAMOST, Xinglong observatory, China). All of the requested fields have now been observed at least once. In this paper, we describe those observations and provide a useful database for the whole astronomical community.
Body mass index (BMI) has been associated with the risk of oesophageal cancer. But the influence of BMI on postoperative complication and prognosis has always been controversial.
In total, 2031 ...consecutive patients who underwent oesophagectomy between 1998 and 2008 were classified according to Asian-specific BMI (kg m(-2)) cutoff values. The impact of BMI on overall survival (OS) was estimated using the Kaplan-Meier method and Cox proportional hazard models. We performed a meta-analysis to examine the association of BMI with OS and postoperative complication.
Patients with higher BMI had more postoperative complication (P=0.002), such as anastomotic leakage (P=0.016) and cardiovascular diseases (P<0.001), but less incidence of chylous leakage (P=0.010). Logistic regression analysis showed that BMI (P=0.005) was a confounding factor associated with postoperative complication. Multivariate analysis showed that overweight and obese patients had a more favourable survival than normal weight patients (HR (hazard ratio) = 0.80, 95% CI (confidence interval): 0.70-0.92, P=0.001). Subgroup analysis showed that the association with higher BMI and increased OS was observed in patients with oesophageal squamous cell carcinoma (ESCC) (P<0.001), oesophageal adenocarcinoma (EA) (P=0.034), never-smoking (P=0.035), ever-smoking (P=0.035), never alcohol consumption (P=0.005), weight loss (P=0.003) and advanced pathological stage (P<0.001). The meta-analysis further corroborated that higher BMI was associated with increased complication of anastomotic leakage (RR (risk ratio)=1.04, 95% CI: 1.02-1.06, P=0.001), wound infection (RR=1.03, 95% CI: 1.00-1.05, P=0.031) and cardiovascular diseases (RR=1.02, 95% CI: 1.00-1.05, P=0.039), but decreased incidence of chylous leakage (RR=0.98, 95% CI: 0.96-0.99, P<0.001). In addition, high BMI could significantly improved OS (HR=0.78, 95% CI: 0.71-0.85, P<0.001).
Preoperative BMI was an independent prognostic factor for survival, and strongly associated with postoperative complications in oesophageal cancer.
Check dams are commonly used for soil conservation. In the Loess Plateau of China, check dams have been widely constructed as the principal means to retain floodwater and intercept soil sediments ...since the 1970s. For instance, there are more than 6572 check dams in the Yanhe watershed with an area of 7725 km2 in the Loess Plateau. However, little research has been done to quantify the hydrological effects of the check dams. In this research, the SWAT model (Soil and Water Assessment Tool) was applied to simulate the runoff and sediment in the Yanhe watershed. We treated the 1950s to 1960s as the reference period since there were very few check dams during the period. The model was firstly calibrated and validated in the reference period. The calibrated model was then used in the later periods to simulate the hydrological effects of the check dams. The results showed that the check dams had a regulation effect on runoff and a retention effect on sediment. From 1984 to 1987, the runoff in rainy season (from May to October) decreased by 1.54 m3 s−1 (14.7%) to 3.13 m3 s−1 (25.9%) due to the check dams; while in dry season (from November to the following April), runoff increased by 1.46 m3 s−1 (60.5%) to 1.95 m3 s−1 (101.2%); the sediment in rainy season decreased by 2.49 × 106 ton (34.6%) to 4.35 × 106 ton (48.0%). From 2006 to 2008, the runoff in rainy season decreased by 0.79 m3 s−1 (15.5%) to 1.75 m3 s−1 (28.9%), and the runoff in dry season increased by 0.51 m3 s−1 (20.1%) to 0.97 m3 s−1 (46.4%); the sediment in rainy season decreased by 2.03 × 106 ton (79.4%) to 3.12 × 106 ton (85.5%). Construction of the large number of check dams in the Loess Plateau has enhanced the region's capacity to control the runoff and sediment. In the Yanhe watershed, the annual runoff was reduced by less than 14.3% due to the check dams; and the sediment in rainy season was blocked by up to 85.5%. Thus, check dams are effective measures for soil erosion control in the Loess Plateau.
The general relationships between vegetation and water yield under different climatic regimes are well established at a small watershed scale in the past century. However, applications of these basic ...theories to evaluate the regional effects of land cover change on water resources remain challenging due to the complex interactions of vegetation and climatic variability and hydrologic processes at the large scale. The objective of this study was to explore ways to examine the spatial and temporal effects of a large ecological restoration project on water yield across the Loess Plateau region in northern China. We estimated annual water yield as the difference between precipitation input and modelled actual evapotranspiration (ET) output. We constructed a monthly ET model using published ET data derived from eddy flux measurements and watershed streamflow data. We validated the ET models at a watershed and regional levels. The model was then applied to examine regional water yield under land cover change and climatic variability during the implementation of the Grain-for-Green (GFG) project during 1999-2007. We found that water yield in 38% of the Loess Plateau area might have decreased (1-48 mm per year) as a result of land cover change alone. However, combined with climatic variability, 37% of the study area might have seen a decrease in water yield with a range of 1-54 mm per year, and 35% of the study area might have seen an increase with a range of 1-10 mm per year. Across the study region, climate variability masked or strengthened the water yield response to vegetation restoration. The absolute annual water yield change due to vegetation restoration varied with precipitation regimes with the highest in wet years, but the relative water yield changes were most pronounced in dry years. We concluded that the effects of land cover change associated with ecological restoration varied greatly over time and space and were strongly influenced by climatic variability in the arid region. The current regional vegetation restoration projects have variable effects on local water resources across the region. Land management planning must consider the influences of spatial climate variability and long-term climate change on water yield to be more effective for achieving environmental sustainability.
An innovative extended response surface modeling technique (ERSM v1.0) is developed to characterize the nonlinear response of fine particles (PM2.5) to large and simultaneous changes of multiple ...precursor emissions from multiple regions and sectors. The ERSM technique is developed based on the conventional response surface modeling (RSM) technique; it first quantifies the relationship between PM2.5 concentrations and the emissions of gaseous precursors from each single region using the conventional RSM technique, and then assesses the effects of inter-regional transport of PM2.5 and its gaseous precursors on PM2.5 concentrations in the target region. We apply this novel technique with a widely used regional chemical transport model (CTM) over the Yangtze River delta (YRD) region of China, and evaluate the response of PM2.5 and its inorganic components to the emissions of 36 pollutant–region–sector combinations. The predicted PM2.5 concentrations agree well with independent CTM simulations; the correlation coefficients are larger than 0.98 and 0.99, and the mean normalized errors (MNEs) are less than 1 and 2% for January and August, respectively. It is also demonstrated that the ERSM technique could reproduce fairly well the response of PM2.5 to continuous changes of precursor emission levels between zero and 150%. Employing this new technique, we identify the major sources contributing to PM2.5 and its inorganic components in the YRD region. The nonlinearity in the response of PM2.5 to emission changes is characterized and the underlying chemical processes are illustrated.
To explore the differences in clinical characteristics and interocular interactions between patients with anisometropic amblyopia and ametropic amblyopia.
Cross-sectional study. The newly diagnosed ...anisometropic (the binocular difference in spherical equivalent≥1.00 D) amblyopia patients and ametropic amblyopia patients (aged 4 to 6 years) in Beijing Tongren Hospital from January 2020 to December 2022 were involved. Patients were further categorized by the refractive status after cycloplegia, including hyperopia, myopia, astigmatism, hyperopia with astigmatism, myopia with astigmatism, mild anisometropia and severe anisometropia. Quantitative measurements of best-corrected visual acuity (logMAR), stereoacuity (transformed to log units), perceptual eye position and interocular suppression were performed, and the differences between groups were analyzed. The rank sum test was used for statistical evaluation.

Dendritic cells (DCs) can mediate inflammation-related bone resorption that is crucial in the development of periodontitis. Butyrate is a critical by-product of microbes with antibacterial and ...anti-inflammatory properties. Here, we found that butyrate inhibited the activation of lipopolysaccharide (LPS)–induced DCs and generation of inflammatory cytokines by DCs. Moreover, butyrate regulated glycolysis in LPS-induced DCs via the G-protein-coupled receptor/hypoxia-inducible factor–1α pathway. In addition, butyrate inhibited the maturation of CD11c+MHC-II+ DCs in vivo, suppressing local inflammatory infiltration and ultimately alleviating bone resorption in a periodontitis model. Our results imply that butyrate suppresses the activation of LPS-induced DCs by modulating their metabolism, highlighting its potential as a therapeutic agent for inflammatory diseases.
