In racehorses, feeding a high-concentrate diet could cause abnormal fermentation in the hindgut. This feeding management regime is not suitable for the nutritional physiology of horses. However, ...studies on the hindgut environment have yet to be reported in Japanese draft horses, so feeding management needs to be investigated in these horses. Therefore, the objective of this study was to investigate the effects of a high-concentrate diet on hindgut fermentation in Japanese draft horses. Feces were collected from 20 male Japanese draft horses managed by two stables with different feeding designs (65% weight ratio of concentrate feed, HC; 50% weight ratio of concentrate, MC), and fecal metabolic characteristics and the microbiome were analyzed. Higher lactate concentrations and lower fecal pH levels were observed in the HC group (P=0.0011, P=0.0192, respectively). Fecal microbiome analysis revealed a decrease in microbial diversity (P=0.0360) and an increase in the relative abundance of Streptococcus lutetiensis/equinus/infantarius (P=0.0011) in the HC group. On the other hand, fibrolytic bacteria in the MC group had similarities with Clostridium sacchalolyticum and Ruminococcus albus. This study revealed that overfeeding of concentrates induced abnormal fermentation in the hindgut of Japanese draft horses. This suggests that the establishment of a feeding design based on not only the chemical compositions of feeds but also microbial dynamics is needed.

Because individual ruminants exhibit different methane conversion efficiencies, the microbial characteristics of low-methane-emitting animals can be essential for successful rumen manipulation and environment-friendly methane mitigation. Several bacterial species, including Sharpea, uncharacterized Succinivibrionaceae, and certain Prevotella phylotypes have been listed as key players in low-methane-emitting sheep and cows. The functional characteristics of the unclassified bacteria remain unclear, as they are yet to be cultured. Here, we review ruminal methane production and mitigation strategies, focusing on rumen fermentation and the functional role of rumen microbiota, and describe the phylogenetic and physiological characteristics of a novel Prevotella species recently isolated from low methane-emitting and high propionate-producing cows. This review may help to provide a better understanding of the ruminal digestion process and rumen function to identify holistic and environmentally friendly methane mitigation approaches for sustainable ruminant production.

Organic amendments are important sources of nitrous oxide (N2O) emissions from agricultural soils. In 2020, the total amount of N in organic amendments applied to Japanese agricultural soils (440 ...ktN) was larger than that of synthetic fertilizer (374 ktN). However, N2O emissions from organic amendments were estimated by using the country-specific N2O emission factor (EF) for synthetic fertilizer (0.31 % for rice paddy, 2.9 % for tea, and 0.62 % for other crops) in the National Greenhouse Gas Inventory Report of Japan. Thus, we conducted a N2O flux measurement campaign at 12 different experimental sites across Japan to estimate fertilizer-induced N2O EFs for major organic amendments in Japan, that is, poultry manure compost, swine manure compost, cattle manure compost, and organic fertilizer pellets. In addition, we conducted systematic review of N2O emissions and EFs for organic amendments, including data from our measurement campaign and published data from peer-reviewed papers in Japan. The final dataset, including the field measurement campaign and published data, resulted in 404 observations (including synthetic fertilizer and zero-N control) in 29 sites. Results showed that soil type affected EFs, that is, the mean EF of Andosols was lower than that of non-Andosols, which is similar to the case of EFs for synthetic fertilizer. Mean EFs for poultry manure compost, swine manure compost, cattle manure (compost and slurry), and non-animal manure organic fertilizers were 0.83 % (uncertainty range of 2.5th and 97.5th percentile: 0.09 % to 3.46 %), 0.70 % (0.02 % to 2.45 %), 0.39 % (0.00 % to 1.62 %), and 1.16 % (0.41 % to 3.03 %), respectively, when weighted by area of soil types. The mean EF of all organic amendments was 0.84 % (0.00 % to 2.91 %), when the area of soil type and amount of organic amendment used in Japan were considered. Our study provides country-specific EFs to estimate N2O emission from organic amendments in Japan.

Methane mitigation strategies have a two-sided benefit for both environment and efficient livestock production. This preliminary short-term
trial using Mootral (garlic and citrus extracts), a novel ...natural feed supplement, was conducted to evaluate its efficacy on rumen fermentation characteristics, methane production, and the bacterial and archaeal community. The experiment was performed as a batch culture using rumen fluid collected from sheep, and Mootral was supplemented in three concentrations: 0% (Control), 10%, and 20% of the substrate (50% Grass:50% Concentrate). The rumen fermentation data and alpha diversity of microbial community were analyzed by ordinary one-way analysis of variance. The relative abundance and statistical significance of families and operational taxonomic units (OTUs) among the groups were compared by Kruskal-Wallis H test using Calypso software. After 24-h incubation at 39°C, Mootral in a dose-dependent manner improved the production of total volatile fatty acids and propionate while it reduced the acetate proportion and acetate/propionate ratio. The total produced gas was two times higher in the Mootral-supplemented groups than control (
< 0.01), while the proportion of methane in the produced gas was reduced by 22% (
< 0.05) and 54% (
< 0.01) for 10 and 20% Mootral, respectively. Mootral did not change pH, digestibility, and ammonia-nitrogen. Microbial community analyses showed that Mootral effectively changed the ruminal microbiome. The bacterial community showed an increase of the relative abundance of the propionate-producing family such as
(
= 0.014) and
(
= 0.030), while there was a decrease in the relative abundance of some hydrogen-producing bacteria by Mootral supplementation. In the archaeal community,
was decreased by Mootral supplementation compared with control (
= 0.032), while the
family increased in a dose-dependent effect (
= 0.038). The results of the study showed the efficacy of the new mixture to alter the ruminal microbial community, produce more propionate, and reduce microbial groups associated with methane production, thus suggesting that Mootral is a promising natural mixture for methane reduction from ruminants.

AIM:To investigate the relationship between the ironmetabolism-related gene expression profiles and efficacy of antiviral therapy in chronic hepatitis C patients.METHODS:The hepatic expression ...profile of ironmetabolism-related genes was analyzed and its association with virological response to pegylated-interferon plus ribavirin combination therapy was evaluated.A hundred patients with chronic hepatitis C(genotype1b,n = 50; genotype 2,n = 50) were enrolled and retrospectively analyzed.Liver biopsy samples were subjected to quantitative polymerase chain reaction for iron-metabolismrelated genes and protein expression(Western blotting analysis) for ferroportin.As a control,normal liver tissue was obtained from 18 living donors of liver transplantation.Serum hepcidin level was measured by sensitive liquid chromatography/electrospray ionization tandem mass spectrometry.RESULTS:Iron overload is associated with liver damage by increasing oxidative stress and hepatitis C virus(HCV)is reported to induce iron accumulation in hepatocytes in vivo.Conversely,iron administration suppresses HCV replication in vitro.Therefore,the association between HCV infection and iron metabolism remains unclear.Compared with controls,patients had significantly higher gene expression for transferrin,iron-regulatoryproteins 1 and 2,divalent metal transporter 1,and ferroportin,but similar for transferrin receptors 1 and2,and hepcidin.When the expression profiles were compared between sustained virological response(SVR)and non-SVR patients,the former showed significantly lower transcription and protein expression of hepcidin and ferroportin.Expression of hepcidin-regulating genes,BMPR1,BMPR2,and hemojuvelin,was significantly increased,whereas BMP2 was decreased in HCV-infected liver.BMPR2 and hemojuvelin expression was significantly lower in the SVR than non-SVR group.HCV infection affects the expression of iron-metabolism-related genes,leading to iron accumulation in hepatocytes.CONCLUSION:Decreased expression of hepcidin and ferroportin in SVR patients indicates the importance of hepatocytic iron retention for viral response during pegylated-interferon plus ribavirin treatment.
Classically activated macrophages (CAMs) play a crucial protective role in the host by killing the invading pathogens. However, excessive activation of CAMs causes chronic inflammation leading to ...host tissue damage. Thus, control of macrophage activity is necessary to prevent chronic inflammation. To date, regulation of CAMs in the development of chronic inflammatory diseases has not been elucidated. In this study, we investigated the effect of a pan-caspase inhibitor, zVAD-fmk, in cell death in lipopolysaccharide (LPS)-activated macrophages, CAMs. Necrostatin-1, an inhibitor of necroptosis, inhibited zVAD-fmk-induced cell death in CAMs. The expression of mixed lineage kinase domain-like protein (MLKL) involved in the necroptosis pathway was up-regulated by LPS in CAMs. zVAD-fmk enhanced the phosphorylation of MLKL in CAMs. Moreover, inhibition of activation of mitogen activated protein kinase p38 and generation of reactive oxygen species (ROS) reduced zVAD-fmk-induced cell death in CAMs. Inhibition of ROS generation decreased the activation of MLKL and p38 in zVAD-fmk-treated CAMs. These results, taken together, indicate that zVAD-fmk-induced cell death occurred by necroptosis through ROS-mediated activation of MLKL and p38 in CAMs. Elucidation of the molecular mechanism underlying zVAD-fmk-induced necroptosis in CAMs might help in better understanding its significance in chronic inflammatory diseases.

Hypertensive disorders of pregnancy (HDP) lead to the death of approximately 30,000 women annually, and the identification of biomarkers to predict their onset before symptom occurrence is crucial. ...Here, we aimed to identify the first-trimester maternal serum biomarkers for predicting early-onset HDP via a comprehensive metabolomic analysis. This study was conducted by the Fukushima Regional Center as an adjunct study to the Japan Environment and Children's Study. The study comprised 12 patients with early-onset HDP and 12 control subjects with healthy pregnancy whose medical background information was matched with that of the patients by propensity-score matching. Capillary electrophoresis and mass spectrometry-based quantitative analysis of charged metabolites were performed with the first-trimester maternal serum samples. Welch's t-test was used to analyse metabolite peak areas in the two groups. A total of 166 charged metabolites were identified. The peak area of N-dimethylglycine and S-methylcysteine was significantly higher in the first-trimester serum of patients with early-onset HDP than in the controls. Conversely, the peak area of munic acid was significantly decreased in the serum of patients with early-onset HDP. Although we identified potential biomarkers for the prediction and diagnosis of early-onset HDP, no clear marker was identified because of a low statistical power.
