Three years of multi-axis differential optical absorption spectroscopy (MAXDOAS) measurements (2011-2013) have been used for estimating the NO2 mixing ratio along a horizontal line of sight from the ...high mountain subtropical observatory of Izana, at 2370 m a.s.l. (NDACC station, 28.3 degree N, 16.5 degree W). The method is based on horizontal path calculation from the O2-O2 collisional complex at the 477 nm absorption band which is measured simultaneously to the NO2 column density, and is applicable under low aerosol-loading conditions. The MAXDOAS technique, applied in horizontal mode in the free troposphere, minimizes the impact of the NO2 contamination resulting from the arrival of marine boundary layer (MBL) air masses from thermally forced upwelling breeze during middle hours of the day. Comparisons with in situ observations show that during most of the measuring period, the MAXDOAS is insensitive or very slightly sensitive to the upwelling breeze. Exceptions are found for pollution events during southern wind conditions. On these occasions, evidence of fast, efficient and irreversible transport from the surface to the free troposphere is found. Background NO2 volume mixing ratio (vmr), representative of the remote free troposphere, is in the range of 20-45 pptv. The observed seasonal evolution shows an annual wave where the peak is in phase with the solar radiation. Model simulations with the chemistry-climate CAM-Chem model are in good agreement with the NO2 measurements, and are used to further investigate the possible drivers of the NO2 seasonality observed at Izana.

Rheumatoid arthritis (RA) patients can be classified based on presence or absence of anticitrullinated peptide antibodies (ACPA) in their serum. This heterogeneity among patients may reflect ...important biological differences underlying the disease process. To date, the majority of genetic studies have focused on the ACPA-positive group. Therefore, our goal was to analyse the genetic risk factors that contribute to ACPA-negative RA.
We performed a large-scale genome-wide association study (GWAS) in three Caucasian European cohorts comprising 1148 ACPA-negative RA patients and 6008 controls. All patients were screened using the Illumina Human Cyto-12 chip, and controls were genotyped using different genome-wide platforms. Population-independent analyses were carried out by means of logistic regression. Meta-analysis with previously published data was performed as follow-up for selected signals (reaching a total of 1922 ACPA-negative RA patients and 7087 controls). Imputation of classical HLA alleles, amino acid residues and single nucleotide polymorphisms was undertaken.
The combined analysis of the studied cohorts resulted in identification of a peak of association in the HLA-region and several suggestive non-HLA associations. Meta-analysis with previous reports confirmed the association of the HLA region with this subset and an observed association in the CLYBL locus remained suggestive. The imputation and deep interrogation of the HLA region led to identification of a two amino acid model (HLA-B at position 9 and HLA-DRB1 at position 11) that accounted for the observed genome-wide associations in this region.
Our study shed light on the influence of the HLA region in ACPA-negative RA and identified a suggestive risk locus for this condition.
Abstract
PCR is a molecular technique that multiplies DNA fragments in a logarithmical way. qPCR uses fluoroscopic dyes or probes to quantify amplicons but it is a complex and expensive technique ...that should be performed by highly trained personnel. PCR has been used in a wide variety of disciplines such as in food sciences, organ transplant, odontology, oncology and lately, as the standard diagnostic technique for COVID-19. Even when qPCR is a reliable and robust technique, it is hardly accessible for developing countries for its complex labelling procedures and expensive instrumentation, for that, it is of big relevance to search for simpler and cheaper alternative technologies for the detection and analysis of DNA. In this work, we explore the feasibility of using multifrequency bioimpedance measurements to detect label-free PCR products as a proof of principle for the future development of a gene biosensor on the basis of PCR and bioimpedance measurements.
We assessed the possible association between variants of the genes encoding for the angiotensin-converting enzyme ( ACE) and alpha-actinin-3 ( ACTN3) (both individually and combined) and several ...endurance phenotypic traits, e.g., peak power output (PPO), ventilatory (VT) and respiratory compensation threshold (RCT), among others, in professional road cyclists and sedentary controls (n = 46 each). We applied an ANCOVA test using the aforementioned phenotype traits as dependent variables, ACE and/or ACTN3 genotype as the fixed (independent) factor and age and body mass as covariates. We only found a significant genotype effect with no concomitant covariate effect for ACTN3, with cyclists who were not alpha-actinin-3 deficient (RR + RX genotypes) having higher PPO and VT values than their XX counterparts (mean SEM: 7.4 (0.1) vs. 7.1 (0.1) W/kg, p = 0.035; and 4.5 (0.1) vs. 4.3 (0.1) W/kg, p = 0.029, respectively). Cyclists with an "extreme" ACTN3 and ACE genotype combination, i.e., most strength/power oriented (DD + RR/RX), had higher RCT values than those with the "intermediate" combinations (II + RX/RR, p = 0.036; and DD + XX, p = .0004) but similar to those with the most endurance oriented genotype (II + XX). No significant differences (p > 0.05) were found in controls. In summary, in world-class cyclists, we only found an association between ACTN3 genotypes and VT and PPO, and between ACTN3/ACE genotype combinations and RCT.
The hypothesis being advanced in this paper is that there is a new medical paradigm emerging from the biomedical research carried out in this century, mainly due to the explosion of the so called ...“omics” and associated techniques. The main idea is that there is a common pathway from wellbeing and health to chronic disease (“chronopathy”) and even to death, which comprises following steps: 1) unhealthy diet, sedentary life style and permanent exposition to xenobiotics and all kinds of noxious stimuli;→2) intestinal dysbiosis;→3) alteration of the intestinal mucus layer (especially that of the colon);→4) disruption of the endothelial tight junctions;→5) metabolic endotoxemia+bacterial translocation;→6) inflammation;→7) exacerbation of the enteric nervous system (ENS) and consequent maladaptation and malfunctioning of the colon;→8) epigenetic manifestations;→9) “chronopathy” and premature death. Therefore, in order to maintain a good health or to improve or even reverse chronic diseases in a person, the main outcome to look for is a homeostatic balance of the intestinal microbiota (eubiosis), most of which is located in the colon. Lynn Margulis was one of the main scientists to highlight the importance of the role played by bacteria not only in the origin of all biological species now present on earth, but also on their role in global homeostasis. Bacteria do not rely on other living beings for their existence, while the latter depend completely on the former. Humans are no exemption, and new evidence emerges each day about the pivotal role of intestinal microbiota in human health, disease and, in general, in its wellbeing. The following facts about intestinal microbiota are nowadays generally accepted: there are about 10 times more bacteria in the gut than human cells in every human being; the microbioma is about 100–150 times bigger that the human genome, and there is a clear link between intestinal microbiota and many of the most common chronic diseases, from obesity and diabetes to depression and Parkinson disease and different kinds of cancer. The main implication of this theory is that we should become a sort of microbiota farmers, that is, we ought to be more conscious of our intestinal microbiota, take care of it and monitor it permanently. Thus, as part of our good life habits (healthy eating, physical exercise), we should probably undergo periodic seasons of fasting and colon cleansing, as it was common in older times.
The characterisation of digestive proteases in native freshwater fish such as the Mayan cichlid Cichlasoma urophthalmus provides scientific elements that may be used to design balanced feed that ...matches with the digestive capacity of the fish. The purpose of this study was to characterise the digestive proteases, including the effect of the pH and the temperature on enzyme activity and stability, as well as the effect of inhibitors using multienzymatic extracts of the stomach and intestine of C. urophthalmus juveniles. Results showed that the optimum activities of the acid and alkaline proteases occurred at pH values of 3 and 9, respectively, whereas their optimum temperatures were 55 and 65 °C, respectively. The acid proteases were most stable at pH values of 2–3 and at temperatures of 35–45 °C, whereas the alkaline proteases were most stable at pH values of 6–9 and at 25–55 °C. The inhibition assays recorded a residual activity of 4 % with pepstatin A for the acid proteases. The inhibition of the alkaline proteases was greater than 80 % with TPCK, TLCK, EDTA and ovalbumin, and of 60 and 43.8 % with PMSF and SBT1, respectively. The results obtained in this study make it possible to state that C. urophthalmus has a sufficiently complete digestive enzyme machinery to degrade food items characteristic of an omnivorous fish species, although specimens showed a tendency to carnivory.
Context. The circumstellar disk of the Herbig Fe star HD 142527 is host to several remarkable features including a warped inner disk, a 120 au-wide annular gap, a prominent dust trap and several ...spiral arms. A low-mass companion, HD 142527 B, was also found orbiting the primary star at ~14 au. Aims. This study aims to better characterize this companion, which could help explain its impact on the peculiar geometry of the disk. Method. We observed the source with VLT/SINFONI in H + K band in pupil-tracking mode. Data were post-processed with several algorithms based on angular differential imaging (ADI). Results. HD 142527 B is conspicuously re-detected in most spectral channels, which enables us to extract the first medium-resolution spectrum of a low-mass companion within 0.″1 from its central star. Fitting our spectrum with both template and synthetic spectra suggests that the companion is a young M2.5 ± 1.0 star with an effective temperature of 3500 ± 100 K, possibly surrounded with a hot (1700 K) circum-secondary environment. Pre-main sequence evolutionary tracks provide a mass estimate of 0.34 ± 0.06 M⊙, independent of the presence of a hot environment. However, the estimated stellar radius and age do depend on that assumption; we find a radius of 1.37 ± 0.05 R⊙ (resp. 1.96 ± 0.10 R⊙) and an age of 1.8-0.5+1.2 1.8 − 0.5 + 1.2 $ 1.8_{-0.5}^{+1.2} $ Myr (resp. 0.75 ± 0.25 Myr) in the case of the presence (resp. absence) of a hot environment contributing in H + K. Our new values for the mass and radius of the companion yield a mass accretion rate of 4.1–5.8 × 10−9 M⊙ yr−1 (2–3% that of the primary). Conclusions. We have constrained the physical properties of HD 142527 B, thereby illustrating the potential for SINFONI+ADI to characterize faint close-in companions. The new spectral type makes HD 142527 B a twin of the well-known TW Hya T Tauri star, and the revision of its mass to higher values further supports its role in shaping the disk.
Context. HR8799 is orbited by at least four giant planets, making it a prime target for the recently commissioned Spectro-Polarimetric High-contrast Exoplanet REsearch (VLT/SPHERE). As such, it was ...observed on five consecutive nights during the SPHERE science verification in December 2014. Aims. We aim to take full advantage of the SPHERE capabilities to derive accurate astrometric measurements based on H-band images acquired with the Infra-Red Dual-band Imaging and Spectroscopy (IRDIS) subsystem, and to explore the ultimate astrometric performance of SPHERE in this observing mode. We also aim to present a detailed analysis of the orbital parameters for the four planets. Methods. We performed thorough post-processing of the IRDIS images with the Vortex Imaging Processing (VIP) package to derive a robust astrometric measurement for the four planets. This includes the identification and careful evaluation of the different contributions to the error budget, including systematic errors. Combining our astrometric measurements with the ones previously published in the literature, we constrain the orbital parameters of the four planets using PyAstrOFit, our new open-source python package dedicated to orbital fitting using Bayesian inference with Monte-Carlo Markov Chain sampling. Results. We report the astrometric positions for epoch 2014.93 with an accuracy down to 2.0 mas, mainly limited by the astrometric calibration of IRDIS. For each planet, we derive the posterior probability density functions for the six Keplerian elements and identify sets of highly probable orbits. For planet d, there is clear evidence for nonzero eccentricity (e ~ 0.35), without completely excluding solutions with smaller eccentricities. The three other planets are consistent with circular orbits, although their probability distributions spread beyond e = 0.2, and show a peak at e ≃ 0.1 for planet e. The four planets have consistent inclinations of approximately 30° with respect to the sky plane, but the confidence intervals for the longitude of the ascending node are disjointed for planets b and c, and we find tentative evidence for non-coplanarity between planets b and c at the 2σ level.
Plant recruitment interactions (i.e., what recruits under what) shape the composition, diversity, and structure of plant communities. Despite the huge body of knowledge on the mechanisms underlying ...recruitment interactions among species, we still know little about the structure of the recruitment networks emerging in ecological communities. Modeling and analyzing the community‐level structure of plant recruitment interactions as a complex network can provide relevant information on ecological and evolutionary processes acting both at the species and ecosystem levels. We report a data set containing 143 plant recruitment networks in 23 countries across five continents, including temperate and tropical ecosystems. Each network identifies the species under which another species recruits. All networks report the number of recruits (i.e., individuals) per species. The data set includes >850,000 recruiting individuals involved in 118,411 paired interactions among 3318 vascular plant species across the globe. The cover of canopy species and open ground is also provided. Three sampling protocols were used: (1) The Recruitment Network (RN) protocol (106 networks) focuses on interactions among established plants (“canopy species”) and plants in their early stages of recruitment (“recruit species”). A series of plots was delimited within a locality, and all the individuals recruiting and their canopy species were identified; (2) The paired Canopy‐Open (pCO) protocol (26 networks) consists in locating a potential canopy plant and identifying recruiting individuals under the canopy and in a nearby open space of the same area; (3) The Georeferenced plot (GP) protocol (11 networks) consists in using information from georeferenced individual plants in large plots to infer canopy‐recruit interactions. Some networks incorporate data for both herbs and woody species, whereas others focus exclusively on woody species. The location of each study site, geographical coordinates, country, locality, responsible author, sampling dates, sampling method, and life habits of both canopy and recruit species are provided. This database will allow researchers to test ecological, biogeographical, and evolutionary hypotheses related to plant recruitment interactions. There are no copyright restrictions on the data set; please cite this data paper when using these data in publications.
