Context.
With more than 1000 h of observation from Feb. 2016 to Oct. 2019, the
Spitzer
Exploration Program Red Worlds (ID: 13067, 13175 and 14223) exclusively targeted TRAPPIST-1, a nearby (12 pc) ...ultracool dwarf star, finding that it is orbited by seven transiting Earth-sized planets. At least three of these planets orbit within the classical habitable zone of the star, and all of them are well-suited for a detailed atmospheric characterization with the upcoming JWST.
Aims.
The main goals of the
Spitzer
Red Worlds program were (1) to explore the system for new transiting planets, (2) to intensively monitor the planets’ transits to yield the strongest possible constraints on their masses, sizes, compositions, and dynamics, and (3) to assess the infrared variability of the host star. In this paper, we present the global results of the project.
Methods.
We analyzed 88 new transits and combined them with 100 previously analyzed transits, for a total of 188 transits observed at 3.6 or 4.5
μ
m. For a comprehensive study, we analyzed all light curves both individually and globally. We also analyzed 29 occultations (secondary eclipses) of planet b and eight occultations of planet c observed at 4.5
μ
m to constrain the brightness temperatures of their daysides.
Results.
We identify several orphan transit-like structures in our
Spitzer
photometry, but all of them are of low significance. We do not confirm any new transiting planets. We do not detect any significant variation of the transit depths of the planets throughout the different campaigns. Comparing our individual and global analyses of the transits, we estimate for TRAPPIST-1 transit depth measurements mean noise floors of ~35 and 25 ppm in channels 1 and 2 of
Spitzer
/IRAC, respectively. We estimate that most of this noise floor is of instrumental origins and due to the large inter-pixel inhomogeneity of IRAC InSb arrays, and that the much better interpixel homogeneity of JWST instruments should result in noise floors as low as 10 ppm, which is low enough to enable the atmospheric characterization of the planets by transit transmission spectroscopy. Our analysis reveals a few outlier transits, but we cannot conclude whether or not they correspond to spot or faculae crossing events. We construct updated broadband transmission spectra for all seven planets which show consistent transit depths between the two
Spitzer
channels. Although we are limited by instrumental precision, the combined transmission spectrum of planet b to g tells us that their atmospheres seem unlikely to be CH
4
-dominated. We identify and model five distinct high energy flares in the whole dataset, and discuss our results in the context of habitability. Finally, we fail to detect occultation signals of planets b and c at 4.5
μ
m, and can only set 3-
σ
upper limits on their dayside brightness temperatures (611 K for b 586 K for c).
Ultra-intense MeV photon and neutron beams are indispensable tools in many research fields such as nuclear, atomic and material science as well as in medical and biophysical applications. For ...applications in laboratory nuclear astrophysics, neutron fluxes in excess of 10
n/(cm
s) are required. Such ultra-high fluxes are unattainable with existing conventional reactor- and accelerator-based facilities. Currently discussed concepts for generating high-flux neutron beams are based on ultra-high power multi-petawatt lasers operating around 10
W/cm
intensities. Here, we present an efficient concept for generating γ and neutron beams based on enhanced production of direct laser-accelerated electrons in relativistic laser interactions with a long-scale near critical density plasma at 10
W/cm
intensity. Experimental insights in the laser-driven generation of ultra-intense, well-directed multi-MeV beams of photons more than 10
ph/sr and an ultra-high intense neutron source with greater than 6 × 10
neutrons per shot are presented. More than 1.4% laser-to-gamma conversion efficiency above 10 MeV and 0.05% laser-to-neutron conversion efficiency were recorded, already at moderate relativistic laser intensities and ps pulse duration. This approach promises a strong boost of the diagnostic potential of existing kJ PW laser systems used for Inertial Confinement Fusion (ICF) research.
Magnetic skyrmions are stabilized by a combination of external magnetic fields, stray field energies, higher-order exchange interactions and the Dzyaloshinskii-Moriya interaction (DMI). The last ...favours homochiral skyrmions, whose motion is driven by spin-orbit torques and is deterministic, which makes systems with a large DMI relevant for applications. Asymmetric multilayers of non-magnetic heavy metals with strong spin-orbit interactions and transition-metal ferromagnetic layers provide a large and tunable DMI. Also, the non-magnetic heavy metal layer can inject a vertical spin current with transverse spin polarization into the ferromagnetic layer via the spin Hall effect. This leads to torques that can be used to switch the magnetization completely in out-of-plane magnetized ferromagnetic elements, but the switching is deterministic only in the presence of a symmetry-breaking in-plane field. Although spin-orbit torques led to domain nucleation in continuous films and to stochastic nucleation of skyrmions in magnetic tracks, no practical means to create individual skyrmions controllably in an integrated device design at a selected position has been reported yet. Here we demonstrate that sub-nanosecond spin-orbit torque pulses can generate single skyrmions at custom-defined positions in a magnetic racetrack deterministically using the same current path as used for the shifting operation. The effect of the DMI implies that no external in-plane magnetic fields are needed for this aim. This implementation exploits a defect, such as a constriction in the magnetic track, that can serve as a skyrmion generator. The concept is applicable to any track geometry, including three-dimensional designs.

African swine fever is a devastating viral disease of domestic and wild pigs against which no vaccine or therapy is available. Therefore, we applied the CRISPR (clustered regularly interspaced short ...palindromic repeats) - Cas9 nuclease system to target the double-stranded DNA genome of African swine fever virus (ASFV). To this end, a permissive wild boar lung (WSL) cell line was modified by stable transfection with a plasmid encoding Cas9 and a guide RNA targeting codons 71 to 78 of the phosphoprotein p30 gene (CP204L) of ASFV. Due to targeted Cas9 cleavage of the virus genome, plaque formation of ASFV was completely abrogated and virus yields were reduced by four orders of magnitude. The specificity of these effects could be demonstrated by using a natural ASFV isolate and escape mutants possessing nucleotide exchanges within the target sequence, which were not inhibited in the Cas9-expressing cell line. Growth of the cell line was not affected by transgene expression which, as well as virus inhibition, proved to be stable over at least 50 passages. Thus, CRISPR-Cas9 mediated targeting of the ASFV p30 gene is a valid strategy to convey resistance against ASF infection, which may also be applied in its natural animal host.
Abstract
The development of high intensity petawatt lasers has created new possibilities for ion acceleration and nuclear fusion using solid targets. In such laser-matter interaction, multiple ion ...species are accelerated with broad spectra up to hundreds of MeV. To measure ion yields and for species identification, CR-39 solid-state nuclear track detectors are frequently used. However, these detectors are limited in their applicability for multi-ion spectra differentiation as standard image recognition algorithms can lead to a misinterpretation of data, there is no unique relation between track diameter and particle energy, and there are overlapping pit diameter relationships for multiple particle species. In this report, we address these issues by first developing an algorithm to overcome user bias during image processing. Second, we use calibration of the detector response for protons, carbon and helium ions (alpha particles) from 0.1 to above 10 MeV and measurements of statistical energy loss fluctuations in a forward-fitting procedure utilizing multiple, differently filtered CR-39, altogether enabling high-sensitivity, multi-species particle spectroscopy. To validate this capability, we show that inferred CR-39 spectra match Thomson parabola ion spectrometer data from the same experiment. Among all photosynthetic organisms, green bacteria have evolved one of the most efficient light-harvesting antenna, the chlorosome, that contains hundreds of thousands of bacteriochlorophyll ...molecules, allowing these bacteria to grow photosynthetically by absorbing only a few photons per bacteriochlorophyll molecule per day. In contrast to other photosynthetic light-harvesting antenna systems, for which a protein scaffold imposes the proper positioning of the chromophores with respect to each other, in chlorosomes, this is accomplished solely by self-assembly. This has aroused enormous interest in the structure–function relations of these assemblies, as they can serve as blueprints for artificial light harvesting systems. In spite of these efforts, conclusive structural information is not available yet, reflecting the sample heterogeneity inherent to the natural system. Here we combine mutagenesis, polarization-resolved single-particle fluorescence–excitation spectroscopy, cryo-electron microscopy, and theoretical modeling to study the chlorosomes of the green sulfur bacterium Chlorobaculum tepidum. We demonstrate that only the combination of these techniques yields unambiguous information on the structure of the bacteriochlorophyll aggregates within the chlorosomes. Moreover, we provide a quantitative estimate of the curvature variation of these aggregates that explains ongoing debates concerning the chlorosome structure.

In pharmacology, particularly ...receptor theory, the drug dose-effect relation of bio-active substances is frequently described by a sigmoidal function formulated by A.V. Hill. In biomechanics and muscle physiology then again, H. Hatze had elaborated a mathematical model for the stimulation- and length-dependent dynamics of the calcium-induced activation of mammalian skeletal muscle. Here, we prove that muscular activity-pCa curves described by the Hill equation and the equilibrium state predicted by Hatze’s activation dynamics are equivalent. Thus, the exponent introduced by Hatze can be directly identified with its counterpart in the Hill equation, by which the former model gains further physiological interpretability. Conversely, the Hill constant can now be interpreted as a function of the fibre length, generally allowing for advanced Hill plots based on model ideas. We derive and examine the complementary relation of both model approaches, highlight the benefits of mutually viewing one approach from the perspective of the other, and address the physiology behind sigmoidal curves.

ABSTRACT
SPECULOOS-South, an observatory composed of four independent 1-m robotic telescopes, located at ESO Paranal, Chile, started scientific operation in 2019 January. This Southern hemisphere ...facility operates as part of the Search for Habitable Planets EClipsing ULtra-cOOl Stars (SPECULOOS), an international network of 1-m-class telescopes surveying for transiting terrestrial planets around the nearest and brightest ultracool dwarfs (UCDs). To automatically and efficiently process the observations of SPECULOOS-South, and to deal with the specialized photometric requirements of UCD targets, we present our automatic pipeline. This pipeline includes an algorithm for automated differential photometry and an extensive correction technique for the effects of telluric water vapour, using ground measurements of the precipitable water vapour. Observing very red targets in the near-infrared can result in photometric systematics in the differential light curves, related to the temporally-varying, wavelength-dependent opacity of the Earth’s atmosphere. These systematics are sufficient to affect the daily quality of the light curves, the longer time-scale variability study of our targets and even mimic transit-like signals. Here we present the implementation and impact of our water vapour correction method. Using the 179 nights and 98 targets observed in the I + z′ filter by SPECULOOS-South since 2019 January, we show the impressive photometric performance of the facility (with a median precision of ∼1.5 mmag for 30-min binning of the raw, non-detrended light curves) and assess its detection potential. We compare simultaneous observations with SPECULOOS-South and TESS, to show that we readily achieve high-precision, space-level photometry for bright, UCDs, highlighting SPECULOOS-South as the first facility of its kind.
We report on enhanced laser driven electron beam generation in the multi MeV energy range that promises a tremendous increase of the diagnostic potential of high energy sub-PW and PW-class laser ...systems. In the experiment, an intense sub-picosecond laser pulse of ∼1019 Wcm−2 intensity propagates through a plasma of near critical electron density (NCD) and drives the direct laser acceleration (DLA) of plasma electrons. Low-density polymer foams were used for the production of hydrodynamically stable long-scale NCD-plasmas. Measurements show that relativistic electrons generated in the DLA-process propagate within a half angle of 12 ± 1° to the laser axis. Inside this divergence cone, an effective electron temperature of 10-13 MeV and a maximum of the electron energy of 100 MeV were reached. The high laser energy conversion efficiency into electrons with energies above 2 MeV achieved 23% with a total charge approaching 1 μC. For application purposes, we used the nuclear activation method to characterize the MeV bremsstrahlung spectrum produced in the interaction of the high-current relativistic electrons with high-Z samples and measured top yields of gamma-driven nuclear reactions. The optimization of the high-Z target geometry predicts an ultra-high MeV photon number of ∼1012 per shot at moderate relativistic laser intensity of 1019 Wcm−2. A good agreement between the experimental data and the results of the 3D-PIC and GEANT4-simulations was demonstrated.
