This review covers different theoretical and experimental aspects of quantum simulation-the emulation of the time evolution of one quantum system by another. The main concepts of digital and analog ...simulation are discussed, followed by an overview of the required resources and potential limitations. The strengths and weaknesses of some of the physical systems that can be used as quantum simulators are evaluated and the areas of application of quantum simulation are presented with numerous examples ranging from atomic and condensed matter physics to cosmology. Simulating quantum mechanics is known to be a difficult computational problem, especially when dealing with large systems. However, this difficulty may be overcome by using some controllable quantum system to study another less controllable or accessible quantum system, i.e., quantum simulation. Quantum simulation promises to have applications in the study of many problems in, e.g., condensed-matter physics, high-energy physics, atomic physics, quantum chemistry, and cosmology. Quantum simulation could be implemented using quantum computers, but also with simpler, analog devices that would require less control, and therefore, would be easier to construct. A number of quantum systems such as neutral atoms, ions, polar molecules, electrons in semiconductors, superconducting circuits, nuclear spins, and photons have been proposed as quantum simulators. This review outlines the main theoretical and experimental aspects of quantum simulation and emphasizes some of the challenges and promises of this fast-growing field.
The most populated state in the United States, California, is projected to add millions of new inhabitants through the end of the current century, requiring considerable landscape conversion to the ...built environment. A suite of continuous multiyear, medium-range resolution (20-km grid spacing), ensemble-based simulations is examined to assess urban expansion climate effects on California in 2100, and potential strategies to alleviate them. Summertime June–August (JJA) warming due to urban expansion of 1°–2°C is greater relative to any other season, and is completely offset by a range of adaptation strategies: green roofs (highly transpiring), cool roofs (highly reflective), and hybrid roofs (with combined biophysical properties of green and cool roofs). After offsetting of urban-induced warming, cool and hybrid roofs lead to a further 1°–2°C reduction in JJA 2-m temperature, highlighting enhanced efficacy of these adaptation strategies. Guided by medium-range-resolution results, additional high-resolution (2-km grid spacing) experiments are conducted for a subset of the JJA periods conducted on a coarser scale. Urban-induced 1°–2°C warming (local maximum warming exceeds 4°C) is simulated, and is offset by cool and green roof deployment. In agreement with coarser-resolution results, maximum near-surface cooling is greater for cool roofs relative to green roofs. Reduced daytime warming associated with both cool and green roofs also modifies the convective mixed layer, reducing turbulent kinetic energy and planetary boundary layer height, although this impact is less for green roofs than for cool roofs. The results presented here demonstrate the importance of future urban expansion in California and illustrate climatic consequences with implications for regional air quality.

This article investigates the effect of air conditioning (AC) systems on air temperature and examines their electricity consumption for a semiarid urban environment. We simulate a 10 day extreme heat ...period over the Phoenix metropolitan area (U.S.) with the Weather Research and Forecasting model coupled to a multilayer building energy scheme. The performance of the modeling system is evaluated against 10 Arizona Meteorological Network weather stations and one weather station maintained by the National Weather Service for air temperature, wind speed, and wind direction. We show that explicit representation of waste heat from air conditioning systems improved the 2 m air temperature correspondence to observations. Waste heat release from AC systems was maximum during the day, but the mean effect was negligible near the surface. However, during the night, heat emitted from AC systems increased the mean 2 m air temperature by more than 1°C for some urban locations. The AC systems modified the thermal stratification of the urban boundary layer, promoting vertical mixing during nighttime hours. The anthropogenic processes examined here (i.e., explicit representation of urban energy consumption processes due to AC systems) require incorporation in future meteorological and climate investigations to improve weather and climate predictability. Our results demonstrate that releasing waste heat into the ambient environment exacerbates the nocturnal urban heat island and increases cooling demands.

Abstract

MYCN
genes. Both previously reported and novel recurrent copy number variations and somatic pathogenic mutations in chromatin remodeling, DNA damage response and PI3K/MAPK growth pathways were variably detected, either in multiple or isolated foci. Proteomic analysis showed global upregulation of histone H3, lack of H3-K27 trimethylation, and further impairment of polycomb repressive complex 2 by ASXL1 downregulation. Activation of oncogenic pathways resulted from combined upregulation of N-MYC, SOX2, p65/p50 NF-κB and STAT3 transcription factors, EGFR, FGFR2, PDGFRα/β receptor tyrosine kinases, and downregulation of PHLPP1/2, PTEN and p16/INK4A tumor suppressors. Upregulation of SMAD4, PAI-1, CD44, and c-SRC in multiple foci most likely contributed to invasiveness. This integrated comprehensive analysis revealed a complex spatiotemporal evolution in diffuse intrisic pontine glioma, recommending pontine and cerebellar biopsies for accurate populational genetic characterization, and delineated common signaling pathways and potential therapeutic targets. It also revealed an unsuspected activation of a multitude of oncogenic pathways, including cancer cell reprogramming, explaining the resistance of DMG to current therapies.
Abstract
Appropriately characterizing future changes in regional-scale precipitation requires assessment of the interactive effect owing to greenhouse gas-induced climate change and the physical ...growth of the built environment. Here we use a suite of medium resolution (20 km grid spacing) decadal scale simulations conducted with the Weather Research and Forecasting model coupled to an urban canopy parameterization to examine the interplay between end-of-century long-lived greenhouse gas (LLGHG) forcing and urban expansion on continental US (CONUS) precipitation. Our results show that projected changes in extreme precipitation are at least one order of magnitude greater than projected changes in mean precipitation; this finding is geographically consistent over the seven CONUS National Climate Assessment (NCA) regions and between the pair of dynamically downscaled global climate model (GCM) forcings. We show that dynamical downscaling of the Geophysical Fluid Dynamics Laboratory GCM leads to projected end-of-century changes in extreme precipitation that are consistently greater compared to dynamical downscaling of the Community Earth System Model GCM for all regions except the Southeast NCA region. Our results demonstrate that the physical growth of the built environment can either enhance or suppress extreme precipitation across CONUS metropolitan regions. Incorporation of LLGHGs indicates compensating effects between urban environments and greenhouse gases, shifting the probability spectrum toward broad enhancement of extreme precipitation across future CONUS metropolitan areas. Our results emphasize the need for development of management policies that address flooding challenges exacerbated by the twin forcing agents of urban- and greenhouse gas-induced climate change.
The PI3K-Akt pathway is a major survival pathway activated in cancer. Efforts to develop targeted therapies have not been fully successful, mainly because of extensive internal intrapathway or ...external interpathway negative feedback loops or because of networking between pathway suppressors. The PTEN tumor suppressor is the major brake of the pathway and a common target for inactivation in somatic cancers. This review will highlight the networking of PTEN with other inhibitors of the pathway, relevant to cancer progression. PTEN constitutes the main node of the inhibitory network, and a series of convergences at different levels in the PI3K-Akt pathway, starting from those with growth factor receptors, will be described. As PTEN exerts enzymatic activity as a phosphatidylinositol-3,4,5-trisphosphate (PIP(3)) phosphatase, thus opposing the activity of PI3K, the concerted actions to increase the availability of PIP(3) in cancer cells, relying either on other phosphoinositide enzymes or on the intrinsic regulation of PTEN activity by other molecules, will be discussed. In particular, the synergy between PTEN and the circle of its direct interacting proteins will be brought forth in an attempt to understand both the activation of the PI3K-Akt pathway and the connections with other parallel oncogenic pathways. The understanding of the interplay between the modulators of the PI3K-Akt pathway in cancer should eventually lead to the design of therapeutic approaches with increased efficacy in the clinic.
Yersinia pestis, the causative agent of plague, is a highly uniform clone that diverged recently from the enteric pathogen Yersinia pseudotuberculosis. Despite their close genetic relationship, they ...differ radically in their pathogenicity and transmission. Here, we report the complete genomic sequence of Y. pseudotuberculosis IP32953 and its use for detailed genome comparisons with available Y. pestis sequences. Analyses of identified differences across a panel of Yersinia isolates from around the world reveal 32 Y. pestis chromosomal genes that, together with the two Y. pestis-specific plasmids, to our knowledge, represent the only new genetic material in Y. pestis acquired since the the divergence from Y. pseudotuberculosis. In contrast, 149 other pseudogenes (doubling the previous estimate) and 317 genes absent from Y. pestis were detected, indicating that as many as 13% of Y. pseudotuberculosis genes no longer function in Y. pestis. Extensive insertion sequence-mediated genome rearrangements and reductive evolution through massive gene loss, resulting in elimination and modification of preexisting gene expression pathways, appear to be more important than acquisition of genes in the evolution of Y. pestis. These results provide a sobering example of how a highly virulent epidemic clone can suddenly emerge from a less virulent, closely related progenitor.
The phosphatidylinositol-3-OH kinase (PI3K)-Akt pathway is activated in cancer by genetic or epigenetic events and efforts are under way to develop targeted therapies. phosphatase and tensin homolog ...deleted on chromosome 10 (PTEN) tumor suppressor is the major brake of the pathway and a common target for inactivation in glioblastoma, one of the most aggressive and therapy-resistant cancers. To achieve potent inhibition of the PI3K-Akt pathway in glioblastoma, we need to understand its mechanism of activation by investigating the interplay between its regulators. We show here that PTEN modulates the PI3K-Akt pathway in glioblastoma within a tumor suppressor network that includes Na(+)/H(+) exchanger regulatory factor 1 (NHERF1) and pleckstrin-homology domain leucine-rich repeat protein phosphatases 1 (PHLPP1). The NHERF1 adaptor, previously characterized by our group as a PTEN ligand and regulator, shows also PTEN-independent Akt-modulating effects that led us to identify the PHLPP1/PHLPP2 Akt phosphatases as NHERF1 ligands. NHERF1 interacts via its PDZ domains with PHLPP1/PHLPP2 and scaffolds heterotrimeric complexes with PTEN. Functionally, PHLPP1 requires NHERF1 for membrane localization and growth-suppressive effects. PHLPP1 loss boosts Akt phosphorylation only in PTEN-negative cells and cooperates with PTEN loss for tumor growth. In a panel of low-grade and high-grade glioma patient samples, we show for the first time a significant disruption of all three members of the PTEN-NHERF1-PHLPP1 tumor suppressor network in high-grade tumors, correlating with Akt activation and patient's abysmal survival. We thus propose a PTEN-NHERF1-PHLPP PI3K-Akt pathway inhibitory network that relies on molecular interactions and can undergo parallel synergistic hits in glioblastoma.
This research evaluates the climatic summertime representation of the diurnal cycle of near‐surface temperature using the Weather Research and Forecasting System (WRF) over the rapidly urbanizing and ...water‐vulnerable Phoenix metropolitan area. A suite of monthly, high‐resolution (2 km grid spacing) simulations are conducted during the month of July with both a contemporary landscape and a hypothetical presettlement scenario. WRF demonstrates excellent agreement in the representation of the daily to monthly diurnal cycle of near‐surface temperatures, including the accurate simulation of maximum daytime temperature timing. Thermal sensitivity to anthropogenic land use and land cover change (LULCC), assessed via replacement of the modern‐day landscape with natural shrubland, is small on the regional scale. The WRF‐simulated characterization of the diurnal cycle, supported by previous observational analyses, illustrates two distinct and opposing impacts on the urbanized diurnal cycle of the Phoenix metro area, with evening and nighttime warming partially offset by daytime cooling. The simulated nighttime urban heat island (UHI) over this semiarid urban complex is explained by well‐known mechanisms (slow release of heat from within the urban fabric stored during daytime and increased emission of longwave radiation from the urban canopy toward the surface). During daylight hours, the limited vegetation and dry semidesert region surrounding metro Phoenix warms at greater rates than the urban complex. Although prior work has suggested that daytime temperatures are lower within the urban complex owing to the addition of residential and agricultural irrigation (i.e., “oasis effect”) we show that modification of Phoenix's surrounding environment to a biome more representative of temperate regions eliminates the daytime urban cooling. Our results indicate that surrounding environmental conditions, including land cover and availability of soil moisture, play a principal role in establishing the nature and evolution of the diurnal cycle of near‐surface temperature for the greater Phoenix, Arizona, metropolitan area relative to its rural and undeveloped counterpart.
