Land use systems that integrate woody vegetation with livestock and/or crops and are recognised for their biodiversity and cultural importance can be termed high nature and cultural value (HNCV) ...agroforestry. In this review, based on the literature and stakeholder knowledge, we describe the structure, components and management practices of ten contrasting HNCV agroforestry systems distributed across five European bioclimatic regions. We also compile and categorize the ecosystem services provided by these agroforestry systems, following the Common International Classification of Ecosystem Services. HNCV agroforestry in Europe generally enhances biodiversity and regulating ecosystem services relative to conventional agriculture and forestry. These systems can reduce fire risk, compared to conventional forestry, and can increase carbon sequestration, moderate the microclimate, and reduce soil erosion and nutrient leaching compared to conventional agriculture. However, some of the evidence is location specific and a better geographical coverage is needed to generalize patterns at broader scales. Although some traditional practices and products have been abandoned, many of the studied systems continue to provide multiple woody and non-woody plant products and high-quality food from livestock and game. Some of the cultural value of these systems can also be captured through tourism and local events. However there remains a continual challenge for farmers, landowners and society to fully translate the positive social and environmental impacts of HNCV agroforestry into market prices for the products and services.
Histone deacetylases (HDACs) have been identified as therapeutic targets due to their regulatory function in chromatin structure and organization. Here, we analyzed the therapeutic effect of LBH589, ...a class I-II HDAC inhibitor, in acute lymphoblastic leukemia (ALL). In vitro, LBH589 induced dose-dependent antiproliferative and apoptotic effects, which were associated with increased H3 and H4 histone acetylation. Intravenous administration of LBH589 in immunodeficient BALB/c-RAG2(-/-)γc(-/-) mice in which human-derived T and B-ALL cell lines were injected induced a significant reduction in tumor growth. Using primary ALL cells, a xenograft model of human leukemia in BALB/c-RAG2(-/-)γc(-/-) mice was established, allowing continuous passages of transplanted cells to several mouse generations. Treatment of mice engrafted with T or B-ALL cells with LBH589 induced an in vivo increase in the acetylation of H3 and H4, which was accompanied with prolonged survival of LBH589-treated mice in comparison with those receiving vincristine and dexamethasone. Notably, the therapeutic efficacy of LBH589 was significantly enhanced in combination with vincristine and dexamethasone. Our results show the therapeutic activity of LBH589 in combination with standard chemotherapy in pre-clinical models of ALL and suggest that this combination may be of clinical value in the treatment of patients with ALL.
This paper introduces this special issue of Aquatic Sciences. It outlines a multi-scale, hierarchical framework for developing process-based understanding of catchment to reach hydromorphology that ...can aid design and delivery of sustainable river management solutions. The framework was developed within the REFORM (REstoring rivers FOR effective catchment Management) project, funded by the European Union’s FP7 Programme. Specific aspects of this ‘REFORM framework’ and some applications are presented in other papers in this special issue. The REFORM framework is founded on previous hierarchical frameworks, sixteen examples of which are reviewed. However, the REFORM framework has some particular properties that reflect the European context for which it was developed. The framework delineates regional landscapes into nested spatial units at catchment, landscape unit, segment, reach, geomorphic unit and finer scales. Reaches, regardless of their ‘naturalness’, are assigned to a river type based on valley confinement, planform and bed material. Indicators are quantified at each spatial scale to feed three groups of assessments. First, contemporary indicators at reach and geomorphic unit scales investigate present processes, forms and human pressures within each reach. These feed assessments of present reach hydromorphological function/alteration, including whether the reach is functioning appropriately for its type; riparian corridor function and alteration; and hydromorphological adjustment. Second, indicators at catchment to segment scales investigate water and sediment production and delivery to reaches and how these are affected by human pressures now and in the past. These are used to construct an inventory of changes over space and time. Third, historical reach and geomorphic unit scale indicators are used to construct the trajectory of reach-scale changes. Contemporary reach-scale assessments, space–time inventory, and trajectory of changes are then combined to establish how river reaches of different type, subject to different human pressures, and located in different environmental contexts behave in response to changes at all considered spatial scales. These support forecasts of the likely responses of reaches to future scenarios (e.g., changes in climate, land cover, channel interventions).
Aims Although transplantation of skeletal myoblast (SkM) in models of chronic myocardial infarction (MI) induces an improvement in cardiac function, the limited engraftment remains a major ...limitation. We analyse in a pre-clinical model whether the sequential transplantation of autologous SkM by percutaneous delivery was associated with increased cell engraftment and functional benefit. Methods and results Chronically infarcted Goettingen minipigs (n = 20) were divided in four groups that received either media control or one, two, or three doses of SkM (mean of 329.6 × 106 cells per dose) at intervals of 6 weeks and were followed for a total of 7 months. At the time of sacrifice, cardiac function was significantly better in animals treated with SkM in comparison with the control group. A significantly greater increase in the ΔLVEF was detected in animals that received three doses vs. a single dose of SkM. A correlation between the total number of transplanted cells and the improvement in LVEF and ΔLVEF was found (P < 0.05). Skeletal myoblast transplant was associated with an increase in tissue vasculogenesis and decreased fibrosis (collagen vascular fraction) and these effects were greater in animals receiving three doses of cells. Conclusion Repeated injection of SkM in a model of chronic MI is feasible and safe and induces a significant improvement in cardiac function.
Ochratoxin A (OTA) is a mycotoxin produced by species of the genus
Aspergillus and
Penicillium. Human exposure has been demonstrated worldwide and its origin seems to be the intake of contaminated ...foods. The kidneys are the target organ of this mycotoxin. Immunotoxic and genotoxic effects of OTA were investigated in Wistar male rats (aged 12 weeks), treated by gavage with 50, 150 or 450 μg OTA/kg body weight for 28 days, in the context of a general toxicity study, which was designed following the recommendations of OECD guideline 407. At the end of the study, the mean plasma concentration of the mycotoxin was determined, several immune function assays were performed and bone marrow smears were obtained and stained in order to analyse micronuclei in polychromatic erytrocytes. Mean plasma concentration was found to be 187, 600 and 807 μg/L, respectively. At the highest dose, a decrease in body weight gain was observed. Histopathological investigations revealed tubulonephrosis and acute tubular necrosis in the kidneys of the animals treated with OTA. The frequency and severity of the lesions increased with the dose. The response of splenocytes to sheep red blood cells was decreased in a dose-dependent manner; however, nonstatistically significant differences were obtained. The natural killer cell activity was strongly affected by OTA treatment. Cytotoxic T lymphocyte activity was lower in the animals exposed to 50 μg OTA/kg b.w. but was not modified in the groups exposed to 150 and 450 μg OTA/ kg b.w. The bacteriolytic capability of macrophages was significantly reduced in groups exposed to 50 and 450 μg OTA/ kg b.w. The number of micronuclei in bone marrow polychromatic erytrocytes did not vary significantly with respect to the control at any dose, but a false negative result can not be ruled out because the exposure doses were much lower than those recommended in OECD guideline 474.
Food security, climate change mitigation, and land use challenges are interlinked and need to be considered simultaneously. One possible solution is sustainable intensification, which is the practice ...of increasing food production per area of land whilst also reducing the environmental impacts associated with this. Agroforestry has been stated to be a practice that meets this definition. In this study, a new methodology is presented to assess the potential of different management options as sustainable intensification practices. The methodology is based on comparing the carbon emissions associated with the production of food and the carbon sequestered for that same activity for a particular quantity of food produced over a specific area and over a specific time. The resulting indicator, the “carbon balance” is the difference between the greenhouse gasses emitted (considered as negative values) and carbon sequestered (positive values) estimated in Mg CO2eq per Mg of food produced on one hectare of land for one year. The carbon balance quantifies the global warming potential associated with sustainable intensification by integrating a process-based model with life cycle analysis and is able to estimate above- and below-ground biomass and soil carbon content. This methodology is tested in Portugal for wheat production under crop monoculture and agroforestry systems. The results show agroforestry to be a suitable practice for sustainable intensification compared to a crop monoculture as it just slightly decreased wheat yields whilst providing a positive carbon balance from year 50 onwards of approximately 1 Mg of CO2eq sequestered per Mg of wheat produced.
Climate changes affect aquatic ecosystems by altering temperatures and precipitation patterns, and the rear edges of the distributions of cold-water species are especially sensitive to these effects. ...The main goal of this study was to predict in detail how changes in air temperature and precipitation will affect streamflow, the thermal habitat of a cold-water fish (the brown trout, Salmo trutta), and the synergistic relationships among these variables at the rear edge of the natural distribution of brown trout. Thirty-one sites in 14 mountain rivers and streams were studied in central Spain. Models of streamflow were built for several of these sites using M5 model trees, and a non-linear regression method was used to estimate stream temperatures. Nine global climate models simulations for Representative Concentration Pathways RCP4.5 and RCP8.5 scenarios were downscaled to the local level. Significant reductions in streamflow were predicted to occur in all of the basins (max. −49 %) by the year 2099, and seasonal differences were noted between the basins. The stream temperature models showed relationships between the model parameters, geology and hydrologic responses. Temperature was sensitive to streamflow in one set of streams, and summer reductions in streamflow contributed to additional stream temperature increases (max. 3.6 °C), although the sites that are most dependent on deep aquifers will likely resist warming to a greater degree. The predicted increases in water temperatures were as high as 4.0 °C. Temperature and streamflow changes will cause a shift in the rear edge of the distribution of this species. However, geology will affect the extent of this shift. Approaches like the one used herein have proven to be useful in planning the prevention and mitigation of the negative effects of climate change by differentiating areas based on the risk level and viability of fish populations.
Solar thermal technology for the production on electricity is one of the current technological challenges. In concentrating solar power (CSP) plants, in order to achieve high power it is required to ...use a high operating temperature to reach high conversion efficiencies. The majority of today's commercial solar thermal power plants are based on the parabolic trough collector technology with operating temperature around 400°C. However, the technology of solar tower is used in order to maximize the efficiency of the CSP plants. This technology reaches an operating temperature higher than 1000°C and the development of high temperature receivers that work in this temperature ranges is still in its early stages. The fundamental problems observed are related to materials durability and reliability.
The main objective of this paper has been to develop testing methods for solar receivers which guarantee their reliability and durability under demanding working conditions of high solar concentrating technology. Based on a revision of published or draft Standards, a qualification test methodology for durability tests has been developed.
INTRODUCTION AND OBJECTIVESThe incidence of upper urinary tract tumors is currently unknown. The aim of this study is to determine the real incidence of upper tract urothelial carcinoma (UTUC) in ...Spain. MATERIAL AND METHODSA descriptive, prospective and multicenter epidemiological study was conducted in 31 Spanish facilities by means of the Platform for Multicenter Studies of the Spanish Association of Urology. Recruitment was opened from May 1st, 2017 to April 30th, 2018. The original database was exported directly from the electronic Data Collection Logbook on December 15th, 2018, with a total of 404 cases registered (402 valid cases after depuration). Statistical analysis was performed using IBM SPSS software v 23 and EPIDAT v 3.4. RESULTSThe incidence adjusted to Spanish population from raw data was 3.27 cases per 100.000 inhabitants per year (2.93 - 3.61 95% CI) and 3,3 cases per 100.000 inhabitants per year (2.96-3.66 95%CI) when adjusted to European population by age. The mean age at diagnosis was 70 years, and 77% of patients were male. Thirty-four percent of patients had an incidental diagnosis. Tumors were most commonly located in the pyelocalyceal system (54%), followed by the distal ureter (22%). Prior ureteroscopy was performed in 114 patients: this technique modified the subsequent treatment indication in 58% of cases. Radical nephroureterectomy was performed in 311 patients. Kidney-sparing surgery was the elected treatment in 76 patients (20%). Complications were found in 69% of cases, most of them classified as Clavien 1 and 2 (86% of all complications). Postoperative mortality rate was 1.7%. CONCLUSIONSUTUC adjusted incidence rate in Spain is 3.27 and 3.3 in Europe. Prior URS modified the treatment indication in 18% of patients. We found a 69% complication rate and a 1.7% mortality rate.
