Copaifera is a genus of large trees found in Brazil, mainly in Amazon forest, but also in Atlantic forest and cerrado biomes. It has also been found in other countries in South America. In Africa, it ...is found mainly in Congo, Cameroon, Guinea and Angola. Its oleoresin has been used in folk medicine in the treatment of numerous healthy disorders, such as urinary, respiratory, skin and inflammatory diseases, for which there are several studies corroborating its ethnopharmacological uses. It is also extensively employed in the pharmaceutical and cosmetic industries in the development of ointments, pills, soaps, perfumes, among others. Copaifera oleoresin contains mainly diterpenes, such as: kaurenoic acid, kaurenol, copalic acid, agathic acid, hardwiickic acid, polyalthic acid, and sesquiterpenes, comprising β-caryophyllene, caryophyllene oxide, α-copaene, α–humulene, γ-muurolene and β-bisabolol, among other compounds. On the other hand, Copaifera leaves contain mainly phenolic compounds, such as flavonoids and methylated galloylquinic acid derivatives. Therefore, considering the economic importance of Copaifera oleoresin, its ethnopharmacological uses, the need to develop new pharmaceuticals for the treatment of many diseases, as well as the pharmacological potential of the compounds found in Copaifera spp., it was undertaken a review covering mostly the last two decades on the distribution, chemistry, pharmacology, quality control and safety of Copaifera species.
Invasion of periodontal tissues by Porphyromonas gingivalis and Aggregatibacter actinomycetemcomitans can be associated with aggressive forms of periodontitis. Oleoresins from different copaifera ...species and their compounds display various pharmacological properties. The present study evaluates the antibacterial and antivirulence activity of oleoresins obtained from different copaifera species and of ten isolated compounds against two causative agents of periodontitis. The following assays were performed: determination of the minimum inhibitory concentration (MIC), determination of the minimum bactericidal concentration (MBC), and determination of the antibiofilm activity by inhibition of biofilm formation and biofilm eradication tests. The antivirulence activity was assessed by hemagglutination, P. gingivalis Arg-X and Lis-X cysteine protease inhibition assay, and A. actinomycetemcomitans leukotoxin inhibition assay. The MIC and MBC of the oleoresins and isolated compounds 1, 2, and 3 ranged from 1.59 to 50 μg/mL against P. gingivalis (ATCC 33277) and clinical isolates and from 6.25 to 400 μg/mL against A. actinomycetemcomitans (ATCC 43717) and clinical isolates. About the antibiofilm activity, the oleoresins and isolated compounds 1, 2, and 3 inhibited biofilm formation by at least 50% and eradicated pre-formed P. gingivalis and A. actinomycetemcomitans biofilms in the monospecies and multispecies modes. A promising activity concerning cysteine protease and leucotoxin inhibition was also evident. In addition, molecular docking analysis was performed. The investigated oleoresins and their compounds may play an important role in the search for novel sources of agents that can act against periodontal pathogens.
Bacterial and viral infections are serious public health issue. The toxicity of the BRP was evaluated in the Caenorhabditis elegans animal model. The MIC values for the crude hydroalcoholic extract sample ranged from 3.12 to 100 μg/mL, while fractions and isolated compounds the MIC values ranged from 1.56 to 400 μg/mL.The BRP crude hydroalcoholic extract, oblongifolin B, and gutiferone E presented MICB
values ranging from 1.56 to 100 μg/mL against monospecies and multispecies biofilms. Neovestitol and vestitol inhibited CHIKV infection by 93.5 and 96.7%, respectively. The tests to evaluate toxicity in C. elegans demonstrated that the BRP was not toxic below the concentrations 750 μg/mL. The results constitute an alternative approach for treating various infectious diseases.
Candida species have been responsible for a high number of invasive infections worldwide. In this sense, Rottlerin has demonstrated a wide range of pharmacological activities. Therefore, this study ...aimed to evaluate the antifungal, antibiofilm and antivirulence activity of Rottlerin in vitro against Candida spp. and its toxicity and antifungal activity in vivo. Rottlerin showed antifungal activity against all yeasts evaluated, presenting Minimum Inhibitory and Fungicidal Concentration (MIC and MFC) values of 7.81 to > 1000 µg/mL. Futhermore, it was able to significantly inhibit biofilm production, presenting Biofilm Inhibitory Concentration (MICB
) values that ranged from 15.62 to 250 µg/mL and inhibition of the cell viability of the biofilm by 50% (IC
) from 2.24 to 12.76 µg/mL. There was a considerable reduction in all hydrolytic enzymes evaluated, with emphasis on hemolysin where Rottlerin showed a reduction of up to 20%. In the scanning electron microscopy (SEM) analysis, Rottlerin was able to completely inhibit filamentation by C. albicans. Regarding in vivo tests, Rottlerin did not demonstrate toxicity at the therapeutic concentrations demonstrated here and was able to increase the survival of C. elegans larvae infected. The results herein presented are innovative and pioneering in terms of Rottlerin's multipotentiality against these fungal infections.
The use of models capable of forecasting the production of photovoltaic (PV) energy is essential to guarantee the best possible integration of this energy source into traditional distribution grids. ...Long Short-Term Memory networks (LSTMs) are commonly used for this purpose, but their use may not be the better option due to their great computational complexity and slower inference and training time. Thus, in this work, we seek to evaluate the use of neural networks MLPs (Multilayer Perceptron), Recurrent Neural Networks (RNNs), and LSTMs, for the forecast of 5 min of photovoltaic energy production. Each iteration of the predictions uses the last 120 min of data collected from the PV system (power, irradiation, and PV cell temperature), measured from 2019 to mid-2022 in Maceió (Brazil). In addition, Bayesian hyperparameters optimization was used to obtain the best of each model and compare them on an equal footing. Results showed that the MLP performs satisfactorily, requiring much less time to train and forecast, indicating that they can be a better option when dealing with a very short-term forecast in specific contexts, for example, in systems with little computational resources.

Two novel silver(I) complexes with the antibacterial sulfa drugs mafenide (Maf) and ethyl-mafenide (eMaf) are presented in this article. Elemental analysis indicated the 1:1 metal:ligand compositions AgC7H9N2O2S for Ag-Maf and AgC8H11N2O2S for Ag-eMaf. Infrared (IR), and 1H and 13C nuclear magnetic resonance (NMR) spectroscopic analyses indicated coordination of the sulfa drugs to silver(I) by the nitrogen and oxygen atoms of the deprotonated sulfonamide (SO2NH) group in a bidentate chelate mode. The proposition of the coordination mode of the sulfa drugs to silver(I) was also supported by a combination of experimental and theoretical (DFT) data. The Ag-Maf and Ag-eMaf complexes exhibited antibacterial activity over Staphylococcus aureus, Burkholderia cepacia, Staphylococcus epidermidis, and Pseudomonas aeruginosa aerobic bacteria, with MIC (minimum inhibitory concentration) in the range 21.3–170 µmol L−1, being comparable to the commercial drug silver-sulfadiazine (SSD). Antiviral studies against Chikungunya virus (CHIKV) showed that the complexes exhibit anti-CHIKV activity at non-cytotoxic concentrations to the host cells. Biophysical studies based on gel electrophoresis indicated that the complexes do not interact with albumin and lysozyme, which suggests that proteins are not the main targets of the compounds.

Since the novel coronavirus disease outbreak, over 179.7 million people have been infected by SARS-CoV-2 worldwide, including the population living in dengue-endemic regions, particularly Latin ...America and Southeast Asia, raising concern about the impact of possible co-infections. Thirteen SARS-CoV-2/DENV co-infection cases reported in Midwestern Brazil between April and September of 2020 are described. Information was gathered from hospital medical records regarding the most relevant clinical and laboratory findings, diagnostic process, therapeutic interventions, together with clinician-assessed outcomes and follow-up. Of the 13 cases, seven patients presented Acute Undifferentiated Febrile Syndrome and six had pre-existing co-morbidities, such as diabetes, hypertension and hypopituitarism. Two patients were pregnant. The most common symptoms and clinical signs reported at first evaluation were myalgia, fever and dyspnea. In six cases, the initial diagnosis was dengue fever, which delayed the diagnosis of concomitant infections. The most frequently applied therapeutic interventions were antibiotics and analgesics. In total, four patients were hospitalized. None of them were transferred to the intensive care unit or died. Clinical improvement was verified in all patients after a maximum of 21 days. The cases reported here highlight the challenges in differential diagnosis and the importance of considering concomitant infections, especially to improve clinical management and possible prevention measures. Failure to consider a SARS-CoV-2/DENV co-infection may impact both individual and community levels, especially in endemic areas.

The combination of pyrimethamine and sulfadiazine is the standard care in cases of congenital toxoplasmosis. However, therapy with these drugs is associated with severe and sometimes life-threatening ...side effects. The investigation of phytotherapeutic alternatives to treat parasitic diseases without acute toxicity is essential for the advancement of current therapeutic practices. The present study investigates the antiparasitic effects of oleoresins from different species of Copaifera genus against T. gondii. Oleoresins from C. reticulata, C. duckei, C. paupera, and C. pubiflora were used to treat human trophoblastic cells (BeWo cells) and human villous explants infected with T. gondii. Our results demonstrated that oleoresins were able to reduce T. gondii intracellular proliferation, adhesion, and invasion. We observed an irreversible concentration-dependent antiparasitic action in infected BeWo cells, as well as parasite cell cycle arrest in the S/M phase. The oleoresins altered the host cell environment by modulation of ROS, IL-6, and MIF production in BeWo cells. Also, Copaifera oleoresins reduced parasite replication and TNF-α release in villous explants. Anti-T. gondii effects triggered by the oleoresins are associated with immunomodulation of the host cells, as well as, direct action on parasites.
Undoubtedly, the advent of antibiotics in the 19th century had a substantial impact, increasing human life expectancy. However, a multitude of scientific investigations now indicate that we are ...currently experiencing a phase known as the post-antibiotic era. There is a genuine concern that we might regress to a time before antibiotics and confront widespread outbreaks of severe epidemic diseases, particularly those caused by bacterial infections. These investigations have demonstrated that epidemics thrive under environmental stressors such as climate change, the depletion of natural resources, and detrimental human activities such as wars, conflicts, antibiotic overuse, and pollution. Moreover, bacteria possess a remarkable ability to adapt and mutate. Unfortunately, the current development of antibiotics is insufficient, and the future appears grim unless we abandon our current approach of generating synthetic antibiotics that rapidly lose their effectiveness against multidrug-resistant bacteria. Despite their vital role in modern medicine, medicinal plants have served as the primary source of curative drugs since ancient times. Numerous scientific reports published over the past three decades suggest that medicinal plants could serve as a promising alternative to ineffective antibiotics in combating infectious diseases. Over the past few years, phenolic compounds, alkaloids, saponins, and terpenoids have exhibited noteworthy antibacterial potential, primarily through membrane-disruption mechanisms, protein binding, interference with intermediary metabolism, anti-quorum sensing, and anti-biofilm activity. However, to optimize their utilization as effective antibacterial drugs, further advancements in omics technologies and network pharmacology will be required in order to identify optimal combinations among these compounds or in conjunction with antibiotics.
Diphylla ecaudata is a hematophagous bat endemic of South America, with food preference for bird blood. Given the lack of information about the reproductive activity of this species, this study aimed ...to describe the testicular morphology and histomorphometry of D. ecaudata in order to understand its reproductive biology, specially spermatogenesis. The animals were collected in Lajes city, Rio Grande do Norte, Brazil. Following euthanasia, the testes were histologically processed for morphological, morphometric, ultrastructural and immunohistochemical analyses. Their average body weight was 24.64g, with a gonadosomatic index of 0.49%, tubulesomatic index of 0.47%, and a total of 32.20m of seminiferous tubules per gram of testis. The pre-meiotic, meiotic, and post-meiotic phases accounted for 56.20%, 9.30%, and 34.50% of the seminiferous epithelium cycle, respectively. The ultrastructure of spermiogenesis was similar to that described in other mammals and the perforatorium was not observed in the sperm. Androgen receptors were detected in Sertoli cell nuclei and Leydig cell cytoplasm, while aromatase enzyme was detected only in Sertoli cell nuclei. FGF2 and BCL-2 activities were detected in the cytoplasm of zygotene and pachytene primary spermatocytes, as well as round and elongated spermatids. D. ecaudata showed testicular pattern similar to other mammals and characteristics common to other bat species. This species stood out for its high efficiency of Sertoli cells, which presented high capacity to support germ cells, besides the highest sperm production rates among those already recorded. This study is the first step towards the knowledge of D. ecaudata reproduction and the first description of its spermatogenesis.
