9. Among these, a dichloromethane extract of
was capable of causing a loss of viability of over 50%, exceeding the effect of the commercial insecticide chlorpyrifos. This extract elicited chromatin condensation and the fragmentation in treated cells. Nanoencapsulation assays of the cytotoxic plant extracts in soybean liposomes and chitosan nanostructures were carried out. The nanosystems exhibited sizes lower or around 200 nm, low polydispersity, and generally high encapsulation efficiencies. Release assays showed that chitosan nanoemulsions provide a fast and total extract release, while liposome-based systems are suitable for a more delayed release. These results represent a proof-of-concept for the future development of bioinsecticide nanoformulations based on the cytotoxic plant extracts.

The biodistribution profile of a new dextrin nanomagnetogel, which consists of γ-Fe2O3 superparamagnetic nanoparticles loaded within a polymeric matrix of modified dextrin, was studied in mice. The ...nanomagnetogel bear a monomodal size distribution profile (average diameter 110 nm) close to neutral surface charge and higher relaxivity (r 2 = 215–248 mM–1 s–1 and r 2/r 1 = 13–11) than those of commercial formulations (r 2 = 160–177 mM–1 s–1 and r 2/r 1 = 4–7). Also, the observed blood half-lifeapproximately 4 his superior to that of similar commercially available formulations, which remain for a few minutes in circulation. PEGylation resulted in 1.7- and 1.2-fold lower accumulation in the liver and spleen, respectively, within the first 24 h. Noteworthy, a good correlation was obtained between the amount of polymer (quantified by scintigraphy) in the spleen, 48 h after administration, and the amount of iron physically loaded through hydrophobic interactions (quantified by ICP) indicating the absence of iron leakage from the polymeric matrix. This study provides evidence of the in vivo stability of a self-assembled nanomagnetogel, a relevant feature which is seldom reported in the literature.

Rhipicephalus microplus, commonly known as southern cattle tick, causes huge economic losses in the cattle industry. Its infestation affects the production of meat and milk and causes discomfort to hosts. In addition, it is the vector of Babesia spp. and Anaplasma spp. The most frequent method used to control these parasites is through synthetic acaricides. However, their indiscriminate use can be toxic for hosts and environment as well as cause selection of resistant ticks. Plant extracts and essential oils emerge as promising alternatives to manage tick infestation. Carvacrol, an aromatic monoterpene extracted from plants, has recognized antimicrobial, antioxidant, insecticidal, repellent and acaricidal activities. Acetylation of carvacrol is believed to enhance its nematicidal and acaricidal activities and to decrease its toxicity to hosts. Thus, the aim of this study was to evaluate the effect of different concentrations of acetylcarvacrol in the morphology of ovaries of engorged R. microplus ticks. The most remarkable morphological alterations found in the female germ cells were irregular and thicker chorion, decreasing in size and irregular shape of female germ cells (oocytes), cytoplasmatic vacuolization as well as ring-shaped nucleoli. These alterations were analyzed through a semi-quantitative method proposed in this study for ixodid ticks. Treatment group IV, which was exposed to 4.5 μL/mL of carvacrol acetate, showed the most significant alterations, and it was also statistically different when compared to control groups. Therefore, sublethal concentrations of acetylcarvacrol demonstrated to impact the reproductive system of R. microplus by causing several damages in the female germ cells. This would hinder the generation of new individuals, probably contributing for a long-term control of tick infestation.
Type 1 diabetes (T1D) is a complex disease with metabolic and functional changes that can alter an individual's proteome. An LC-MS/MS analytical method, in an HDMSE system, was used to identify ...differentially expressed proteins in the high abundance protein-depleted serum of T1D patients and healthy controls. Samples were processed in Progenesis QI for Proteomics software. A functional enrichment of the proteins was performed with Gene Ontology and ToppGene, and the interactions were visualized by STRING 11.5. As a result, 139 proteins were identified, 14 of which were downregulated in the serum of patients with T1D compared to controls. Most of the differentially expressed proteins were shown to be involved with the immune system, inflammation, and growth hormone stimulus response, and were associated with the progression of T1D. Differential protein expression data showed for the first-time changes in CPN2 expression levels in the serum of patients with T1D. Our findings indicate that these proteins are targets of interest for future investigations and for validation of protein biomarkers in T1D.

The novel influenza A pandemic virus (H1N1pdm) caused considerable morbidity and mortality worldwide in 2009. The aim of the present study was to evaluate the clinical course, duration of viral ...shedding, H1N1pdm evolution and emergence of antiviral resistance in hospitalized cancer patients with severe H1N1pdm infections during the winter of 2009 in Brazil.
We performed a prospective single-center cohort study in a cancer center in Rio de Janeiro, Brazil. Hospitalized patients with cancer and a confirmed diagnosis of influenza A H1N1pdm were evaluated. The main outcome measures in this study were in-hospital mortality, duration of viral shedding, viral persistence and both functional and molecular analyses of H1N1pdm susceptibility to oseltamivir.
A total of 44 hospitalized patients with suspected influenza-like illness were screened. A total of 24 had diagnosed H1N1pdm infections. The overall hospital mortality in our cohort was 21%. Thirteen (54%) patients required intensive care. The median age of the studied cohort was 14.5 years (3-69 years). Eighteen (75%) patients had received chemotherapy in the previous month, and 14 were neutropenic at the onset of influenza. A total of 10 patients were evaluated for their duration of viral shedding, and 5 (50%) displayed prolonged viral shedding (median 23, range=11-63 days); however, this was not associated with the emergence of a resistant H1N1pdm virus. Viral evolution was observed in sequentially collected samples.
Prolonged influenza A H1N1pdm shedding was observed in cancer patients. However, oseltamivir resistance was not detected. Taken together, our data suggest that severely ill cancer patients may constitute a pandemic virus reservoir with major implications for viral propagation.

Subsyndromal delirium (SSD) in the Intensive Care Unit (ICU) is associated with an increased morbidity with unknown post-discharge functional and cognitive outcomes. We performed a prospective ...multicenter study to analyze the mental status of patients during their first 72 h after ICU admission and its trajectory, with follow-ups at 3 and 6 months after hospital discharge. Amongst the 106 included patients, SSD occurred in 24.5% (n = 26) and was associated with the duration of mechanical ventilation (p = 0.003) and the length of the ICU stay (p = 0.002). After the initial 72 h, most of the SSD patients (30.8%) improved and no longer had SSD; 19.2% continued to experience SSD and one patient (3.8%) progressed to delirium. The post-hospital discharge survival rate for the SSD patients was 100% at 3 months and 87.5% at 6 months. At admission, 96.2% of the SSD patients were fully independent in daily living activities, 66.7% at 3-month follow-up, and 100% at 6-month follow-up. Most SSD patients demonstrated a cognitive decline from admission to 3-month follow-up and improved at 6 months (IQCODE-SF: admission 3.13, p < 0.001; 3 months 3.41, p = 0.019; 6 months 3.19, p = 0.194). We concluded that early SSD is associated with worse outcomes, mainly a transitory cognitive decline after hospital discharge at 3 months, with an improvement at 6 months. This highlights the need to prevent and identify this condition during ICU stays.
This work compares the results of four different unfolding codes, MSANDB, MAXED, FRUIT and BONMA, which are based on different unfolding techniques. Additionally, Bayesian parameter estimation is ...also considered. All unfolding codes were supplied with the same set of input data acquired at the Environmental Research Station 'Schneefernerhaus' on the Zugspitze mountain, corresponding to continuous measurements of secondary neutrons from cosmic radiation. The HMGU high-energy extended Bonner sphere spectrometer (BSS), consisting of 16 measuring channels with (3)He proportional counters, was used as a reference BSS. The differences in the neutron spectra obtained with the different unfolding codes are discussed, and the uncertainties of integral quantities, like neutron fluence and ambient dose equivalent, are quantified.
Goat whey is normally discarded in the milk processing industry. However, several studies have addressed its biological properties and possible use in human or animal diet. The present study aimed to ...analysis the protein profile of goat whey to evaluate its possible oxidant, antioxidant, antibacterial, antitumour, and cytotoxic activities in vitro against human erythrocytes. Goat whey was skimmed, and crude protein extract (CPE) was obtained. Next, protein fractions (F) were obtained using ammonium sulphate precipitation method. The proteins were characterized by SDS-PAGE, two-dimensional electrophoresis and soluble protein measurements. No significant differences were observed in protein profile of CPE, F 30–60% and F 60–90%. The highest protein content was found in F 60–90% (0.41mgP/mL). All samples, except F 0–30% showed bacteriostatic activity against different bacterial strains. Only CPE at a concentration of 1000μg/mL was haemolytic against human erythrocytes. Oxidant activity against erythrocytes was not observed. Antioxidant activity was observed only for CPE. Cytotoxicity against C6 rat glioma cell line that was performed with CPE revealed tumour cell death>70% at concentrations of 0.05 and 0.1μg/mL. These results demonstrate at first time that CPE may be used as an antioxidant, bacteriostatic and cytotoxic compound against tumour cells.
