The subcortical white matter of the adult human brain harbors a pool of glial progenitor cells. These cells can be isolated by fluorescence-activated cell sorting (FACS) after either transfection ...with green fluorescent protein (GFP) under the control of the CNP2 promoter, or A2B5-targeted immunotagging. Although these cells give rise largely to oligodendrocytes, in low-density culture we observed that some also generated neurons. We thus asked whether these nominally glial progenitors might include multipotential progenitor cells capable of neurogenesis. We found that adult human white-matter progenitor cells (WMPCs) could be passaged as neurospheres in vitro and that these cells generated functionally competent neurons and glia both in vitro and after xenograft to the fetal rat brain. WMPCs were able to produce neurons after their initial isolation and did not require in vitro expansion or reprogramming to do so. These experiments indicate that an abundant pool of mitotically competent neurogenic progenitor cells resides in the adult human white matter.

The 7 year data set of the Milagro TeV observatory contains 2.2 x 10(11) events of which most are due to hadronic cosmic rays. These data are searched for evidence of intermediate scale structure. ...Excess emission on angular scales of approximately 10 degrees has been found in two localized regions of unknown origin with greater than 12sigma significance. Both regions are inconsistent with pure gamma-ray emission with high confidence. One of the regions has a different energy spectrum than the isotropic cosmic-ray flux at a level of 4.6sigma, and it is consistent with hard spectrum protons with an exponential cutoff, with the most significant excess at approximately 10 TeV. Potential causes of these excesses are explored, but no compelling explanations are found.
In a randomized trial of bortezomib, cyclophosphamide, and dexamethasone as compared with the same therapy plus daratumumab, patients with light-chain amyloidosis who received daratumumab had a ...higher frequency of hematologic complete response than those who did not (53.3% vs. 18.1%). Deaths were most commonly due to cardiac failure.
Results are presented of a harmonic analysis of the large-scale cosmic-ray (CR) anisotropy as observed by the Milagro observatory. We show a two-dimensional display of the sidereal anisotropy ...projections in right ascension (R.A.) generated by the fitting of three harmonics to 18 separate declination bands. The Milagro observatory is a water Cherenkov detector located in the Jemez mountains near Los Alamos, New Mexico. With a high duty cycle and large field of view, Milagro is an excellent instrument for measuring this anisotropy with high sensitivity at TeV energies. The analysis is conducted using a seven-year data sample consisting of more than 95 billion events, the largest such data set in existence. We observe an anisotropy with a magnitude around 0.1% for CRs with a median energy of 6 TeV. The dominant feature is a deficit region of depth (2.49 - 0.02 stat. - 0.09 sys.) X10-3 in the direction of the Galactic north pole centered at 189 deg R.A. We observe a steady increase in the magnitude of the signal over seven years.
Abstract
Submesoscale stirring contributes to the cascade of tracer variance from large to small scales. Multiple nested surveys in the summer Sargasso Sea with tow-yo and autonomous platforms ...captured submesoscale water-mass variability in the seasonal pycnocline at 20–60-m depths. To filter out internal waves that dominate dynamic signals on these scales, spectra for salinity anomalies on isopycnals were formed. Salinity-gradient spectra are approximately flat with slopes of −0.2 ± 0.2 over horizontal wavelengths of 0.03–10 km. While the two to three realizations presented here might be biased, more representative measurements in the literature are consistent with a nearly flat submesoscale passive tracer gradient spectrum for horizontal wavelengths in excess of 1 km. A review of mechanisms that could be responsible for a flat passive tracer gradient spectrum rules out (i) quasigeostrophic eddy stirring, (ii) atmospheric forcing through a relict submesoscale winter mixed layer structure or nocturnal mixed layer deepening, (iii) a downscale vortical-mode cascade, and (iv) horizontal diffusion because of shear dispersion of diapycnal mixing. Internal-wave horizontal strain appears to be able to explain horizontal wavenumbers of 0.1–7 cycles per kilometer (cpkm) but not the highest resolved wavenumbers (7–30 cpkm). Submesoscale subduction cannot be ruled out at these depths, though previous observations observe a flat spectrum well below subduction depths, so this seems unlikely. Primitive equation numerical modeling suggests that nonquasigeostrophic subinertial horizontal stirring can produce a flat spectrum. The last need not be limited to mode-one interior or surface Rossby wavenumbers of quasigeostrophic theory but may have a broaderband spectrum extending to smaller horizontal scales associated with frontogenesis and frontal instabilities as well as internal waves.

Monoclonal B-cell lymphocytosis (MBL) is a hematologic condition wherein small B-cell clones can be detected in the blood of asymptomatic individuals. Most MBL have an immunophenotype similar to ...chronic lymphocytic leukemia (CLL), and 'CLL-like' MBL is a precursor to CLL. We used flow cytometry to identify MBL from unaffected members of CLL kindreds. We identified 101 MBL cases from 622 study subjects; of these, 82 individuals with MBL were further characterized. In all, 91 unique MBL clones were detected: 73 CLL-like MBL (CD5(+)CD20(dim)sIg(dim)), 11 atypical MBL (CD5(+)CD20(+)sIg(+)) and 7 CD5(neg) MBL (CD5(neg)CD20(+)sIg(neg)). Extended immunophenotypic characterization of these MBL subtypes was performed, and significant differences in cell surface expression of CD23, CD49d, CD79b and FMC-7 were observed among the groups. Markers of risk in CLL such as CD38, ZAP70 and CD49d were infrequently expressed in CLL-like MBL, but were expressed in the majority of atypical MBL. Interphase cytogenetics was performed in 35 MBL cases, and del 13q14 was most common (22/30 CLL-like MBL cases). Gene expression analysis using oligonucleotide arrays was performed on seven CLL-like MBL, and showed activation of B-cell receptor associated pathways. Our findings underscore the diversity of MBL subtypes and further clarify the relationship between MBL and other lymphoproliferative disorders.

Diffuse -ray emission produced by the interaction of cosmic-ray particles with matter and radiation in the Galaxy can be used to probe the distribution of cosmic rays and their sources in different ...regions of the Galaxy. With its large field of view and long observation time, the Milagro Gamma Ray Observatory is an ideal instrument for surveying large regions of the northern hemisphere sky and for detecting diffuse -ray emission at very high energies. Here the spatial distribution and the flux of the diffuse -ray emission in the TeV energy range with a median energy of 15 TeV for Galactic longitude between 30 degree and 110 degree and between 136 degree and 216 degree and for Galactic latitude between -10 degree and 10 degree are determined. The measured fluxes are consistent with predictions of the GALPROP model everywhere, except for the Cygnus region. For the Cygnus region, the flux is twice the predicted value. This excess can be explained by the presence of active cosmic-ray sources accelerating hadrons, which interact with the local dense interstellar medium and produce gamma rays through pion decay.
