Introduction
Long-acting somatostatin analogs (SSAs) are most widely used to treat growth hormone (GH)-secreting pituitary adenoma. However, approximately 30 % of treated patients show resistance to ...SSAs, which may be associated with the reduction of somatostatin receptor subtype 2 (SSTR2) mRNA and protein expression.
Materials and methods
The present study used immunohistochemistry to detect the expression of SSTR2 and SSTR5 in twenty human GH-secreting adenoma samples treated with SSAs and seven normal pituitary samples.
Results
The staining intensities of SSTR2 and SSTR5 were stronger in most adenoma samples than in normal pituitary. The expression of SSTR2 tended to be lower in the SSA non-responder group than in responders. A search of the Bioinformatics data bank and the miRCURY™ LNA array confirmed miR-185 as the putative mircoRNA (miRNA) regulating the expression of SSTR2. An in vitro study using Dual Luciferase reporter assay demonstrated that miR-185 likely targets the 3′-UTR of SSTR2 mRNA in the rat pituitary adenoma GH3 cell line. MiR-185 also downregulated or upregulated the expression of SSTR2 mRNA and SSTR2 protein, following transfection with miR-185 mimics or inhibitors, respectively.
Conclusion
MiR-185 enhanced the cell proliferation and inhibited the apoptosis of GH3 cells.
To explore the characteristics of natural foci of hemorrhagic fever with renal syndrome (HFRS) in Gansu Province.
The information of HFRS case data and rodent density monitoring data from 2012 to ...2022 in Gansu Province were collected and epidemiological methods were used to analyze and investigate the characteristics of the epidemic focus.
A total of 869 cases of HFRS were reported, and four patients died from 2012 to 2022. The annual incidence rate is between 0.05 per 100 000 and 1.21 per 100 000. The cases were mainly distributed in the eastern, southeast, southern, and south of the central region of Gansu Province. Most cases were distributed between age 20-60, and the sex ratio was 1.85∶1 (564∶305). Most cases were farmers (61.80%, 537/869), herdsmen (19.79%,172/869) and students (6.33%, 55/869). In a wild rat-type epidemic focus, the incidence peak was from November to January of the following year. The natural rodent hosts of HFRS were

The cracked chevron-notched Brazilian disc (CCNBD) was proposed by the International Society for Rock Mechanics (ISRM) to test the mode I (opening mode) fracture toughness of rock. The test method ...has been vigorously discussed and debated, despite being the subject of intensive research for decades. The minimum (critical) dimensionless stress intensity factors affiliated with the formula for calculating the fracture toughness using CCNBD specimens with different geometric parameters remain elusive and complex. The matter cannot be resolved by simply replacing the diameter in the original formula with the radius, as claimed by several authors. In this paper, the formula is fundamentally improved, as wide-ranging minimum dimensionless stress intensity factors pertaining to diversified CCNBD geometries are recalibrated by three-dimensional finite element analysis, and an expression with tabulated coefficients is obtained through curve-fitting the data obtained from the numerical calibration. The present results are shown to be more accurate than those in the literature. Furthermore, the importance of the reasonability of the results is highlighted; a comprehensive comparison of different values shows that the upper bounds of minimum stress intensity factors are violated by the above claim. The confusion resulting from the claim is, thus, clarified conclusively.
To analyze the differentially phosphorylated proteins in DENV-2-infected human umbilical venous endothelial cells (HUVECs) and explore the possible pathogenic mechanism of DENV-2 infection.
The total ...proteins were extracted from DENV-2-infected HUVECs and blank control HUVEC using SDT lysis method. The phosphorylated proteins were qualitatively and quantitatively analyzed using tandem mass spectrometry (TMT). The identified differentially phosphorylated proteins were analyzed by bioinformatics analyses such as subcellular localization analysis, GO enrichment analysis, KEGG pathway analysis and protein-protein interaction (PPI) analysis. Western blotting was used to detect the expressions of phosphorylated Jun, map2k2 and AKT1 proteins in DENV-2-infected HUVECs.
A total of 2918 modified peptides on 1385 different proteins were detected, and among them 1346 were significantly upregulated (FC > 1.2,
< 0.05) and 1572 were significantly downregulated (FC < 0.83,
< 0.05). A total of 49 phosphorylated conserved moti
The minimum dimensionless stress intensity factor of cracked chevron notched Brazilian disc specimen is the coefficient in the crucial formula for calculating the fracture toughness using the method ...suggested by the International Society for Rock Mechanics. This coefficient can be conveniently obtained by the analytical method based on the straight through crack assumption proposed by Munz et al. (1980) 17, so it is very easy to determine the upper bound on the coefficient for any cracked chevron notched Brazilian disc specimen. Furthermore, the upper bound is explained by comparing the load bearing capacity of a cracked chevron notched Brazilian disc with that of the counterpart cracked straight through Brazilian disc, this comparison identifies that the upper bound concept is inherent with sound physical significance, so violation of this upper bound for the stress intensity factor is unreasonable.
Using the Cluster data during the period from January to April between 2001 and 2006, we find an observation of solar wind entry due to magnetic reconnection occurred in the terrestrial high‐latitude ...magnetospheric lobes, tailward of the cusps under northward interplanetary magnetic field (IMF). Occurrence rate of solar wind entry events in this study is of the same order as that for the Cluster orbital interval from August to October between the years of 2002 and 2004 as reported by Shi et al. (2013). In this paper, we further study the role of the IMF Bx and By components in the control of solar wind plasma entry based on the investigations of different magnetic dipole tilt variations between our database and Shi et al. (2013). This study shows that the asymmetry distribution of solar wind entry events in the northern and southern lobes could be caused by the variation of magnetic dipole tilt, which could influence the locations of the reconnection site on the high‐latitude lobe magnetopause. On the other hand, IMF Bx can also affect the solar wind plasma entry rate, which is also consistent with previous results. Therefore, we conclude that the “north‐south asymmetry” of solar wind entry events in the lobes could be the combined result of magnetic dipole tilt and IMF Bx. In addition, the IMF By component can influence the entry events in conjunction with the variation of IMF Bx component, which is in line with the Parker Spiral of the IMF.
Key Points
The solar wind entry rate into the lobes is the same order in different seasons
The “north‐south asymmetry” of SW entry is caused by both magnetic dipole tilt and IMF Bx
The IMF By (with Bx) affects event occurrence, as expected from Parker Spiral
