The availability of multiple datasets comprising genome-scale RNAi viability screens in hundreds of diverse cancer cell lines presents new opportunities for understanding cancer vulnerabilities. ...Integrated analyses of these data to assess differential dependency across genes and cell lines are challenging due to confounding factors such as batch effects and variable screen quality, as well as difficulty assessing gene dependency on an absolute scale. To address these issues, we incorporated cell line screen-quality parameters and hierarchical Bayesian inference into DEMETER2, an analytical framework for analyzing RNAi screens ( https://depmap.org/R2-D2 ). This model substantially improves estimates of gene dependency across a range of performance measures, including identification of gold-standard essential genes and agreement with CRISPR/Cas9-based viability screens. It also allows us to integrate information across three large RNAi screening datasets, providing a unified resource representing the most extensive compilation of cancer cell line genetic dependencies to date.

Are there better ways to address community challenges than expending funds on international service-learning? In attempting to wed learning and service, are we are exploiting the "other" for new, or ...recycled, aims? As these questions attest, of all types of service-learning, international service-learning (ISL) most starkly illuminates the tensions between the liberatory and oppressive potentials of practice.This book explores the ramifications of realizing a new age of service-learning that pushes beyond single episodic course-based projects to rebalance student learning and community outcome priorities, and provides insight into what it looks like in its execution. In describing eleven international programs designed to achieve reciprocal, sustained relationships in which learning is co-created, the contributors reveal their struggles to change the balance of power relationships and move to a more transformative practice. Common themes are the developmental nature of this work; the recognition that it takes multiple attempts, often over years, for an individual or an institution to get this work even nearly right; that resolving the challenges of unequal resources, power, and privilege can never be completely erased; and that attention has to be given to the micro-level details.What emerge are seven guiding principles that define the nature of partnerships in liberatory practice, and that apply to all forms of service learning. They must be: strategic--linked to the mission and expertise of the institution; long-term; multifaceted--allowing both partners to play a multiplicity of roles; developmental in building capacities; contextualized in historic and cultural understanding; fully reciprocal; and create the potential for community-driven change.In addressing the problematic nature of ISL, and of service-learning in general, this book interrogates whether its experiences create the necessary conditions for the formation of individual values, convictions, and action; and whether their pivotal teaching and learning moments are indeed replicable and transferable across individual, institutional and even cultural contexts. Its conclusions and insights will be of intense interest to administrators and practitioners alike.
Increasing active travel (primarily walking and cycling) has been widely advocated for reducing obesity levels and achieving other population health benefits. However, the strength of evidence ...underpinning this strategy is unclear. This study aimed to assess the evidence that active travel has significant health benefits.
The study design was a systematic review of (i) non-randomised and randomised controlled trials, and (ii) prospective observational studies examining either (a) the effects of interventions to promote active travel or (b) the association between active travel and health outcomes. Reports of studies were identified by searching 11 electronic databases, websites, reference lists and papers identified by experts in the field. Prospective observational and intervention studies measuring any health outcome of active travel in the general population were included. Studies of patient groups were excluded.
Twenty-four studies from 12 countries were included, of which six were studies conducted with children. Five studies evaluated active travel interventions. Nineteen were prospective cohort studies which did not evaluate the impact of a specific intervention. No studies were identified with obesity as an outcome in adults; one of five prospective cohort studies in children found an association between obesity and active travel. Small positive effects on other health outcomes were found in five intervention studies, but these were all at risk of selection bias. Modest benefits for other health outcomes were identified in five prospective studies. There is suggestive evidence that active travel may have a positive effect on diabetes prevention, which may be an important area for future research.
Active travel may have positive effects on health outcomes, but there is little robust evidence to date of the effectiveness of active transport interventions for reducing obesity. Future evaluations of such interventions should include an assessment of their impacts on obesity and other health outcomes.

We present a uniform catalog of accurate distances to local molecular clouds informed by the Gaia DR2 data release. Our methodology builds on that of Schlafly et al. First, we infer the distance and ...extinction to stars along sightlines toward the clouds using optical and near-infrared photometry. When available, we incorporate knowledge of the stellar distances obtained from Gaia DR2 parallax measurements. We model these per-star distance-extinction estimates as being caused by a dust screen with a 2D morphology derived from Planck at an unknown distance, which we then fit for using a nested sampling algorithm. We provide updated distances to the Schlafly et al. sightlines toward the Dame et al. and Magnani et al. clouds, finding good agreement with the earlier work. For a subset of 27 clouds, we construct interactive pixelated distance maps to further study detailed cloud structure, and find several clouds which display clear distance gradients and/or are comprised of multiple components. We use these maps to determine robust average distances to these clouds. The characteristic combined uncertainty on our distances is 5%-6%, though this can be higher for clouds at greater distances, due to the limitations of our single-cloud model.
Coronal mass ejections (CMEs) were discovered in the early 1970s when space-borne coronagraphs revealed that eruptions of plasma are ejected from the Sun. Today, it is known that the Sun produces ...eruptive flares, filament eruptions, coronal mass ejections and failed eruptions; all thought to be due to a release of energy stored in the coronal magnetic field during its drastic reconfiguration. This review discusses the observations and physical mechanisms behind this eruptive activity, with a view to making an assessment of the current capability of forecasting these events for space weather risk and impact mitigation. Whilst a wealth of observations exist, and detailed models have been developed, there still exists a need to draw these approaches together. In particular more realistic models are encouraged in order to asses the full range of complexity of the solar atmosphere and the criteria for which an eruption is formed. From the observational side, a more detailed understanding of the role of photospheric flows and reconnection is needed in order to identify the evolutionary path that ultimately means a magnetic structure will erupt.
Primordial black holes (PBHs) can form in the radiation dominated early Universe from the collapse of large density perturbations produced by inflation. A power-law parametrization of the primordial ...power spectrum is often used to extrapolate from cosmological scales, where the amplitude of the perturbations is well measured by the cosmic microwave background and large scale structure observations, down to the small scales on which PBHs may form. We show that this typically leads to large errors in the amplitude of the fluctuations on small scales, and hence extremely inaccurate calculations of the abundance of PBHs are formed.
Although low vegetation productivity has been observed in karst regions, whether and how bedrock geochemistry contributes to the low karstic vegetation productivity remain unclear. In this study, we ...address this knowledge gap by exploring the importance of bedrock geochemistry on vegetation productivity based on a critical zone investigation across a typical karst region in Southwest China. We show silicon and calcium concentrations in bedrock are strongly correlated with the regolith water loss rate (RWLR), while RWLR can predict vegetation productivity more effectively than previous models. Furthermore, the analysis based on 12 selected karst regions worldwide further suggest that lithological regulation has the potential to obscure and distort the influence of climate change. Our study implies that bedrock geochemistry could exert effects on vegetation growth in karst regions and highlights that the critical role of bedrock geochemistry for the karst region should not be ignored in the earth system model.
ABSTRACT Inferences about the spatial density or phase-space structure of stellar populations in the Milky Way require a precise determination of the effective survey volume. The volume observed by ...surveys such as Gaia or near-infrared spectroscopic surveys, which have good coverage of the Galactic midplane region, is highly complex because of the abundant small-scale structure in the three-dimensional interstellar dust extinction. We introduce a novel framework for analyzing the importance of small-scale structure in the extinction. This formalism demonstrates that the spatially complex effect of extinction on the selection function of a pencil-beam or contiguous sky survey is equivalent to a low-pass filtering of the extinction-affected selection function with the smooth density field. We find that the angular resolution of current 3D extinction maps is sufficient for analyzing Gaia sub-samples of millions of stars. However, the current distance resolution is inadequate and needs to be improved by an order of magnitude, especially in the inner Galaxy. We also present a practical and efficient method for properly taking the effect of extinction into account in analyses of Galactic structure through an effective selection function. We illustrate its use with the selection function of red-clump stars in APOGEE using and comparing a variety of current 3D extinction maps.
Recent advances in silicon nanophotonics, including demonstrations of ultracompact modulators, germanium waveguide photodetectors and wavelength-division multiplexers, indicate the feasibility of ...on-chip optical interconnects integrated with multicore microprocessors. Studies have suggested that direct replacement of part or all of the electrical interconnect wiring with point-to-point optical links may not provide sufficient power savings to make this approach attractive to chip designers. However, if high-bandwidth optical signals can be switched and routed using an on-chip silicon nanophotonic interconnection network, significant performance gains can be expected. Here we show an ultracompact (40 × 12 µm2) wavelength-insensitive switch based on cascaded silicon microring resonators, which may bring this vision closer to reality by serving as a critical basic element for scalable on-chip optical networks. Fast (<2 ns) error-free (bit error rate < 1 × 10-12) switching of multiple (up to 9) 40-Gbit s-1 optical channels is demonstrated in a temperature-insensitive (±15 °C) device.
Abstract
We present a search for optical bursts from the repeating fast radio burst FRB 121102 using simultaneous observations with the high-speed optical camera ULTRASPEC on the 2.4-m Thai National ...Telescope and radio observations with the 100-m Effelsberg Radio Telescope. A total of 13 radio bursts were detected, but we found no evidence for corresponding optical bursts in our 70.7-ms frames. The 5σ upper limit to the optical flux density during our observations is 0.33 mJy at 767 nm. This gives an upper limit for the optical burst fluence of 0.046 Jy ms, which constrains the broad-band spectral index of the burst emission to α ≤ −0.2. Two of the radio pulses are separated by just 34 ms, which may represent an upper limit on a possible underlying periodicity (a rotation period typical of pulsars), or these pulses may have come from a single emission window that is a small fraction of a possible period.
