The text is arranged in a pattern that mirrors Grumet's argument that women who teach make this passage between the so-called public and private worlds daily and that is also what we teach children ...to do. The chapters go back and forth between the experience of domesticity and the experience of teaching, between being with one's own children and being with the children of others, between being the child of one's own mother and the teacher of another mother's child, between feeling and form, family and colleagues. The first and last chapters address the familial relations that fall under the category of reproduction, a frame designed to emphasize the relations of reproduction and their importance to educational theory. The chapters closest to this margin are those that address women's work in schools, and the juxtaposition is chosen to accentuate the dialectical relation of our public and private meanings. The middle chapters are the ones most directly concerned with curriculum, that provisional ground that Grumet is naming as our mediating space, the place where we can heal. The fundamental argument of this text is that knowledge evolves in human relationships.
Identification and utilization of resistance to Phytophthora capsici could provide the basis for a viable management strategy against cucumber fruit rot, a persistent threat in cucumber (Cucumis ...sativus) production. Our objectives were to develop a method for testing detached, nonwounded, cucumber fruit for resistance to P. capsici, and to screen cucumber cultivars and plant introductions (collectively referred to as cultigens) for resistance. Four P. capsici isolates (differing in their sensitivity to the fungicide mefenoxam and compatibility type) were compared for their fruit infection capability in 1999 and 2000. No significant differences were found among isolates, and a single isolate was used for all subsequent screens. From 1999 to 2004, 480 cucumber cultigens were grown according to standard practices at Michigan State University research farms in four fields with no history of P. capsici. Commercially mature fruit were harvested, inoculated with P. capsici, and rated for lesion diameter, pathogen sporulation diameter, and density of pathogen sporulation. Although no fruit exhibited complete resistance to P. capsici, some cultigens exhibited limited pathogen sporulation. In the process of screening, it was observed that younger, smaller fruit were comparatively more susceptible than older, larger fruit. Replicated trials with hand-pollinated fruit showed that the transition from susceptible to more resistant appeared to coincide with the transition from the period of rapid fruit elongation to the period of increased fruit diameter. This is the first report using a nonwounded fruit screen to analyze cucumber resistance to P. capsici.

If teachers ...are to participate in the politics that determine curriculum and pedagogy, education programs must provide differentiated credentials that welcome adults into teaching and offer insight into the processes of political organizing and public speech.
Viral replication depends on compatible interactions between a virus and its host. For RNA viruses, the viral replicases (RNA-dependent RNA polymerases; RdRps) often associate with components of the ...host translational apparatus. To date, host factors interacting with potyvirus replicases have not been identified. The Potyviridae, which form the largest and most economically important plant virus family, have numerous similarities with the animal virus family, the Picornaviridae. Potyviruses have a single-stranded, plus sense genome; replication initiates at the viral-encoded, 3′ poly-(A) terminus. The yeast two-hybrid system was used to identify host plant proteins associating with the RdRp of zucchini yellow mosaic potyvirus (ZYMV). Several cDNA clones representing a single copy of a poly-(A) binding protein (PABP) gene were isolated from a cucumber (Cucumis sativus L.) leaf cDNA library. Deletion analysis indicated that the C-terminus of the PABP is necessary and sufficient for interaction with the RdRp. Full-length cucumber PABP cDNA was obtained using 5′ RACE; in vitro and Escherichia coli-expressed PABP bound to poly-(A)–Sepharose and ZYMY RdRp with or without the presence of poly-(A). This is the first report of an interaction between a viral replicase and PABP and may implicate a role for host PABP in the potyviral infection process.
A quick and inexpensive method has been demonstrated to remove polysaccharide contamination from plant DNA. Isolated plant genomic DNA with polysaccharide contaminants was dissolved in TE (10 mM ...Tris-HCl, pH 7.4, 1 mM EDTA) with NaCl ranging from 0.5-3.0 M, then precipitated with two volumes of ethanol. Most of the polysaccharides were removed effectively in a single high-salt precipitation at 1.0-2.5 M NaCl. At 3.0 M NaCl, the salt precipitated out of solution. Purified DNA was easily digested by either HindIII or EcoRI and was satisfactory as a template for PCR. The results show that high-salt precipitation effectively removed polysaccharides and their inhibitory effects on restriction enzyme and Taq polymerase activity.

Examining Sexton's pedagogy, with its "weird abundance" of tactics and strategies, Salvio argues that Sexton's use of the autobiographical "I" is as much a literary identity as a literal identity, one that can speak with great force to educators who recognize its vital role in the humanities classroom.
Phytophthora capsici causes severe losses in vegetable production, including many cucurbit crops. Our previous work showed that cucumber (Cucumis sativus) fruit are most susceptible to P. capsici ...when they are very young and rapidly elongating, but develop resistance as they approach full length at 10 to 12 days postpollination (DPP). In this study, fruit from seven additional cucurbit crops representing four species, melon (Cucumis melo), butternut squash (Cucurbita moschata), watermelon (Citrullus lanatus), and zucchini, yellow summer squash, acorn squash, and pumpkin (Cucurbita pepo), were tested for the effect of fruit development on susceptibility to P. capsici. Field-grown fruit of the different crops varied in overall susceptibility. Zucchini and yellow summer squash were the most susceptible, with the majority of fruit exhibiting water-soaking symptoms within 24 hours postinoculation. Fruit from all of the crops exhibited size-related decrease in susceptibility, but to varying degrees. Cucumber had the most pronounced effect. In infested fields, cucumber fruit were found to be most frequently infected at the blossom end. Comparison of the peduncle and blossom end showed a difference in susceptibility along the length of the fruit for cucumber, butternut squash, and zucchini. Greenhouse-grown, hand-pollinated pumpkin, acorn squash, and butternut squash showed an age-related decrease in susceptibility similar to field-grown fruit. For all of these fruit, a pronounced reduction in susceptibility accompanied the transition from the waxy green to green stage at approximately equal to 3 to 8 DPP.
Compared with other primary photosynthetic products (e.g. sucrose and starch), little is known about sugar alcohol metabolism, its regulation, and the manner in which it is integrated with other ...pathways. Mannose-6-phosphate reductase (M6PR) is a key enzyme that is involved in mannitol biosynthesis in celery (Apium graveolens L.). The M6PR gene was cloned from a leaf cDNA library, and clonal authenticity was established by assays of M6PR activity, western blots, and comparisons of the deduced amino acid sequence with a celery M6PR tryptic digestion product. Recombinant M6PR, purified from Escherichia coli, had specific activity, molecular mass, and kinetic characteristics indistinguishable from those of authentic celery M6PR. Sequence analyses showed M6PR to be a member of the aldo-keto reductase superfamily, which includes both animal and plant enzymes. The greatest sequence similarity was with aldose-6-phosphate reductase (EC 1.1.1.200), a key enzyme in sorbitol synthesis in Rosaceae. Developmental studies showed M6PR to be limited to green tissues and to be under tight transcriptional regulation during leaf initiation, expansion, and maturation. These data confirmed a close relationship between the development of photosynthetic capacity, mannitol synthesis, and M6PR activity
Background and purpose The New Zealand white rabbit subscapularis tendon passes under a bony arch to insert on the lesser tubercle of the humerus in a manner analogous to the supraspinatus tendon in ...humans. We assessed whether this unique anatomy may provide a new animal model of the shoulder to improve our understanding of rotator cuff pathology.
Methods The dimensions of the rotator cuff insertions (subscapularis, supraspinatus, and infraspinatus) were measured on 10 fresh frozen cadaveric New Zealand white rabbit shoulders. Mechanical testing was performed on 8 fresh frozen subscapularis insertions (4 matched pairs). Video analysis of the gait cycle was performed on 2 live animals.
Results The origins, insertions, and innervations of the rabbit rotator cuff musculature are analogous to those in humans. However, the rabbit acromion is a rudimentary structure with only the infraspinatus and teres minor muscles passing beneath. Furthermore, at the point where the infraspinatus passes under the arch, it is muscular rather than tendinous. The anterior aspect of the glenohumeral joint contains an additional bony tunnel with its boundaries being the tuberculum supraglenoidale laterally, the coracoideus process superiorly, the tuberculum infraglenoidale inferiorly, and the coracobrachialis muscle medially. The origin of the rabbit subscapularis muscle resides on the anterior scapula. The subscapularis tendon then traverses this bony tunnel prior to its insertion on the lesser tubercle of the humerus. Video analysis and anatomic dissections confirmed excursion of the subscapularis tendon within this bony tunnel throughout the gait cycle. The subscapularis footprint on the proximal humerus measured 6.8 mm (SD 0.29) × 2.5 mm (SD 0.17). Mechanical testing of the subscapularis tendon showed the stiffness to range from 57 to 117 N/mm (SD 23). Ultimate yield ranged from 88 to 215 N (SD 518). The elastic modulus of the rabbit tendon was 56 MPa. 6 of the 8 subscapularis tendons failed at the tendon mid-substance; the other 2 failed at the bony insertion.
Interpretation The unique anatomic architecture and the mechanical characteristics of the rabbit subscapularis muscle provide an opportunity to improve our understanding of rotator cuff pathology.
