The energy reconstruction at KASCADE-Grande is based on a combination of the shower size and the total muon number, which are both estimated for each individual air shower event. We present ...investigations where we employed a second method to reconstruct the primary energy using S(500), which are the charged particle densities inferred with the KASCADE-Grande detector at a distance of 500 m from the shower axis. We considered the attenuation of inclined showers by applying the “Constant Intensity Cut” method and we employed a simulation-derived calibration to convert the recorded S(500) into primary energy. We observed a systematic shift in the S(500)-derived energy compared with previously reported results obtained using the standard reconstruction technique. However, a comparison of the two methods based on simulated and measured data showed that this shift only appeared in the measured data. Our investigations showed that this shift was caused mainly by the inadequate description of the shape of the lateral density distribution in the simulations.
Using data measured by the KASCADE-Grande air shower array, an upper limit to the flux of ultra-high energy gamma-rays in the primary cosmic-ray flux is determined. KASCADE-Grande measures the ...electromagnetic and muonic components for individual air showers in the energy range from 10 PeV up to 1 EeV. The analysis is performed by selecting air showers with low muon contents. A preliminary result on the 90% C.L. upper limit to the relative intensity of gamma-ray with respect to cosmic ray primaries is presented and compared with limits reported by other measurements.
Most porcine oocytes matured and fertilized in vitro fail to develop normally due to abnormal fertilization. The aim of this study was to determine the effect of cysteamine on pronuclear formation ...and developmental competence in pig embryos produced in vitro. Follicular oocytes were matured in vitro with or without cysteamine in Medium 199 supplemented with sodium pyruvate, FSH, LH, 17 beta-estradiol, antibiotics, and 25% porcine follicular fluid. Matured oocytes were then inseminated for 10 h. In the first experiment, pronuclear formation was assessed immediately after the insemination period. The addition of 50 millimole or 500 cysteamine to the maturation medium increased male pronuclear development, resulting in a higher proportion of oocytes with synchronously formed male and female pronuclei. The frequency of synchronous pronuclear formation was increased in monospermic oocytes from 10% in the control group to 43% (p 0.001) and 45% (p 0.001), respectively, and in polyspermic oocytes from 43% in the control group to 68% (p 0.05) and 75% (p 0.001), respectively. In the second experiment, development of the in-vitro produced embryos was assessed after 7 days of culture in vitro. The addition of 500 millimole cysteamine to the maturation medium increased the percentage of cleaving embryos developing to the 8-cell (37% vs. 16%; p 0.001), morula (19%vs.6%; p 0.001),and blastocyst (12% vs. 1%;p 0.001) stages. These results demonstrate that the addition of cysteamine to maturation medium increases synchronous pronuclear formation and normal embryonic development in porcine oocytes matured and fertilized in vitro

In Experiment 1, semen was collected from Merino rams (n = 3), sex-sorted, and then frozen at concentrations of 20, 40, or 80 × 10
6 sperm/mL in three forms of tris-citrate-glucose cryodiluent containing 5% (L-Cryo), 6% (M-Cryo), and 8% (H-Cryo) (v/v) glycerol. Motility, plasma membrane and acrosome integrity, and mitochondrial activity were assessed at 0, 2, 4, and 6 h post thaw. In Experiment 2, cleavage and blastocyst development rates were compared between non-sorted and sex-sorted sperm frozen at the aforementioned concentrations (in the cryodiluent most effective in Experiment 1). In Experiment 1, total motility between 0 and 6 h was similar for all sperm concentrations when frozen using L-Cryo. Mitochondrial activity was elevated in samples frozen in L-Cryo and M-Cryo at 0 h compared to those preserved in H-Cryo for all concentrations (P < 0.05). In Experiment 2, sex-sorted sperm with a higher pre-freeze concentration yielded a higher sperm concentration after preparation for IVF (8.57 ± 1.22 sperm/mL), compared to the lowest group (2.96 ± 0.18 sperm/mL; P < 0.05). There were no significant differences between non-sorted and sex-sorted sperm for rates of embryo cleavage or development. Therefore, sex-sorted sperm was effectively cryopreserved at a higher concentration than conventionally practiced. Although this yielded a higher sperm concentration for IVF, reduced insemination volume, and increased the number of potentially fertile gametes from which to select, fertilisation rate was not significantly improved.
The lipid content of porcine 1-cell stage embryos was reduced (delipated) through the use of micromanipulation to remove the lipid layer formed after centrifugation. Of 94 delipated embryos chilled ...to 4 degrees C for 1 h at the 1-cell or 2- to 4-cell stage 60 (64%) cleaved in culture with development to the morula-blastocyst stage whereas all of the control embryos lysed within 24 h. Significantly more embryos developed beyond the 8-cell stage when they were chilled at the 2- to 4-cell stage compared with chilling at the 1-cell stage (44%, 20 of 45 vs. 18%, 9 of 49). Fewer embryos developed after chilling if they were only partially rather than fully delipated. Developmental rates of partially delipated embryos to the 8-cell and blastocyst stages were 33% (13 of 40) and 8% (3 of 40) rates significantly (p 0.001 and 0.05) lower than the rate for fully delipated embryos (73%, 38 of 52 and 27%, 14 of 52 respectively). The in vitro developmental competence of the unchilled fully delipated embryos was comparable to that of intact zygotes (cleavage: 94%, 45 of 48 vs. 87%, 26 of 30; greater than or equal to blastocyst: 40%, 19 of 48 vs. 57%, 17 of 30). These data demonstrate that the sensitivity of porcine embryos to chilling is related to their high lipid content and that they can become tolerant to chilling if their lipid content is reduced

The KASCADE-Grande experiment operated at KIT from January 2004 to November 2012, measuring EAS generated by primary cosmic rays in the 1016–1018eV energy range. The experiment detected, for each ...single event, with a high resolution, the total number of charged particles (Nch) and of muons (Nμ).
In this contribution we present the latest results about:(i)The measurement of the all particle energy spectrum, discussing the influence of the hadronic interaction model used to derive the energy calibration of the experimental data.(ii)The energy spectra derived separating the events according to the Nμ/Nch ratio. This technique allowed us to unveil a steepening of the spectrum of heavy primaries at E~1016.92±0.04eV and a hardening of the spectrum of light primaries at E~1017.08±0.08eV.(iii)The elemental spectra (for five mass groups) obtained applying a detailed unfolding analysis technique.(iv)A search for large scale anisotropies.
A reliable ovarian stimulation protocol for marmosets is needed to enhance their use as a model for studying human and non-human primate oocyte biology. In this species, a standard dose of hCG did ...not effectively induce oocyte maturation in vivo. The objectives of this study were to characterize ovarian response to an FSH priming regimen in marmosets, given without or with a high dose of hCG, and to determine the meiotic and developmental competence of the oocytes isolated. Ovaries were removed from synchronized marmosets treated with FSH alone (50
IU/d for 6 d) or the same FSH treatment combined with a single injection of hCG (500
IU). Cumulus–oocyte complexes (COCs) were isolated from large (>1.5
mm) and small (0.7–1.5
mm) antral follicles. In vivo-matured oocytes were subsequently activated parthenogenetically or fertilized in vitro. Immature oocytes were subjected to in vitro maturation and then activated parthenogenetically. Treatment with FSH and hCG combined increased the number of expanded COCs from large antral follicles compared with FSH alone (23.5

S.E.M.). Approximately 90% of oocytes surrounded by expanded cumulus cells at the time of isolation were meiotically mature. A blastocyst formation rate of 47% was achieved following fertilization of in vivo-matured oocytes, whereas parthenogenetic activation failed to induce development to the blastocyst stage. The capacity of oocytes to complete meiosis in vitro and cleave was positively correlated with follicle diameter. A dramatic effect of follicle size on spindle formation was observed in oocytes that failed to complete meiosis in vitro. Using the combined FSH and hCG regimen described in this study, large numbers of in vivo matured marmoset oocytes could be reliably collected in a single cycle, making the marmoset a valuable model for studying oocyte maturation in human and non-human primates.

KCDC, the 'KASCADE Cosmic-ray Data Centre', is a web portal, where data of astroparticle physics experiments will be made available for the interested public. The KASCADE experiment, financed by ...public money, was a large-area detector for the measurement of high-energy cosmic rays via the detection of air showers. KASCADE and its extension KASCADE-Grande stopped finally the active data acquisition of all its components including the radio EAS experiment LOPES end of 2012 after more than 20 years of data taking. In a first release, with KCDC we provide to the public the measured and reconstructed parameters of more than 160 million air showers. In addition, KCDC provides the conceptional design, how the data can be treated and processed so that they are also usable outside the community of experts in the research field. Detailed educational examples make a use also possible for high-school students and early stage researchers.
KASCADE-Grande is an air-shower observatory devoted to the detection of cosmic rays with energies in the range of 1016 to 1018 eV. This energy region is of particular interest for the cosmic ray ...astrophysics, since it is the place where some models predict the existence of a transition from galactic to extragalactic origin of cosmic rays and the presence of a break in the flux of its heavy component. The detection of these features requires detailed and simultaneous measurements of the energy and composition of cosmic rays with sufficient statistics. These kinds of studies are possible for the first time in KASCADE-Grande due to the accurate measurements of several air-shower observables, i.e., the number of charged particles, electrons and muons in the shower, using the different detector systems of the observatory. In this contribution, a detailed look into the composition of 1016 - 1018 eV cosmic rays with KASCADE-Grande is presented.
