Strong coupling and the resultant mixing of light and matter states is an important asset for future quantum technologies. We demonstrate deterministic room temperature strong coupling of a ...mesoscopic colloidal quantum dot to a plasmonic nanoresonator at the apex of a scanning probe. Enormous Rabi splittings of up to 110 meV are accomplished by nanometer-precise positioning of the quantum dot with respect to the nanoresonator probe. We find that, in addition to a small mode volume of the nanoresonator, collective coherent coupling of quantum dot band-edge states and near-field proximity interaction are vital ingredients for the realization of near-field strong coupling of mesoscopic quantum dots. The broadband nature of the interaction paves the road toward ultrafast coherent manipulation of the coupled quantum dot-plasmon system under ambient conditions.
Middle infrared spectral results obtained in‐situ by the Hayabusa2 MARA instrument are generally in‐line with previous results and a new comparison with sample C0137 returned from asteroid (162173) ...Ryugu, being similar to an aqueously altered CI1 chondrite. The mid‐IR spectrum of the boulder on Ryugu measured by MARA is shallower around 9 μm compared to the laboratory spectrum of C0137. Here we show that discontinuous, fine dust deposits can partially explain the differences in the spectral data and remain in agreement with the temperature observations of the boulder by MARA if an opaque dust layer covers less than 3% of the field of view. Such a presence of dust covering a low porosity boulder was discounted by previous analysis of the mid‐infrared MARA data which did not consider a highly porous boulder as we do here.
Plain Language Summary
The Japanese Hayabusa2 space mission returned samples from asteroid Ryugu. Here we compare the spectra of the sample observed in the middle infrared (5–25 μm) with in‐situ observations in the part of the electromagnetic spectrum. We find differences between the spectra that can be explained by the presence of fine particles on the surface of Ryugu. Earlier works found that little to no dust can be present on Ryugu and we revisit these studies an derive an upper limit on the amount of dust consistent with the infrared observations. We find that less than 3% of the observed surface can be covered in an opaque dust layer which is likely concentrated in cracks and pores. Our results are consistent with the results of other studies that the amount of dust that a surface of an asteroid can retain decreases with its size.

The physical basis for how macromolecules regulate the onset of mineral formation in calcifying tissues is not well established. A popular conceptual model assumes the organic matrix provides a ...stereochemical match during cooperative organization of solute ions. In contrast, another uses simple binding assays to identify good promoters of nucleation. Here, we reconcile these two views and provide a mechanistic explanation for template-directed nucleation by correlating heterogeneous nucleation barriers with crystal–substrate-binding free energies. We first measure the kinetics of calcite nucleation onto model substrates that present different functional group chemistries (carboxyl, thiol, phosphate, and hydroxyl) and conformations (C11 and C16 chain lengths). We find rates are substrate-specific and obey predictions of classical nucleation theory at supersaturations that extend above the solubility of amorphous calcium carbonate. Analysis of the kinetic data shows the thermodynamic barrier to nucleation is reduced by minimizing the interfacial free energy of the system, γ. We then use dynamic force spectroscopy to independently measure calcite–substrate-binding free energies, Δ G b. Moreover, we show that within the classical theory of nucleation, γ and Δ G b should be linearly related. The results bear out this prediction and demonstrate that low-energy barriers to nucleation correlate with strong crystal–substrate binding. This relationship is general to all functional group chemistries and conformations. These findings provide a physical model that reconciles the long-standing concept of templated nucleation through stereochemical matching with the conventional wisdom that good binders are good nucleators. The alternative perspectives become internally consistent when viewed through the lens of crystal–substrate binding.
The brain's serotonergic system centrally regulates several physiological processes and its dysfunction has been implicated in the pathophysiology of several neuropsychiatric disorders. While in the ...past our understanding of serotonergic neurotransmission has come mainly from mouse models, the development of pluripotent stem cell and induced fibroblast-to-neuron (iN) transdifferentiation technologies has revolutionized our ability to generate human neurons in vitro. Utilizing these techniques and a novel lentiviral reporter for serotonergic neurons, we identified and overexpressed key transcription factors to successfully generate human serotonergic neurons. We found that overexpressing the transcription factors NKX2.2, FEV, GATA2 and LMX1B in combination with ASCL1 and NGN2 directly and efficiently generated serotonergic neurons from human fibroblasts. Induced serotonergic neurons (iSNs) showed increased expression of specific serotonergic genes that are known to be expressed in raphe nuclei. iSNs displayed spontaneous action potentials, released serotonin in vitro and functionally responded to selective serotonin reuptake inhibitors (SSRIs). Here, we demonstrate the efficient generation of functional human serotonergic neurons from human fibroblasts as a novel tool for studying human serotonergic neurotransmission in health and disease.
Calcified skeletons are produced within complex assemblages of proteins and polysaccharides whose roles in mineralization are not well understood. Here we quantify the kinetics of calcite nucleation ...onto a suite of high-purity polysaccharide (PS) substrates under controlled conditions. The energy barriers to nucleation are PS-specific by a systematic relationship to PS charge density and substrate structure that is rooted in minimization of the competing substrate–crystal and substrate–liquid interfacial energies. Chitosan presents a low-energy barrier to nucleation because its near-neutral charge favors formation of a substrate–crystal interface, thus reducing substrate interactions with water. Progressively higher barriers are measured for negatively charged alginates and heparin that favor contact with the solution over the formation of new substrate–crystal interfaces. The findings support a directing role for PS in biomineral formation and demonstrate that substrate–crystal interactions are one end-member in a larger continuum of competing forces that regulate heterogeneous crystal nucleation.

Deriving thermophysical properties such as thermal inertia from thermal infrared observations provides useful insights into the structure of the surface material on planetary bodies. The ...estimation of these properties is usually done by fitting temperature variations calculated by thermophysical models to infrared observations. For multiple free model parameters, traditional methods such as least-squares fitting or Markov chain Monte Carlo methods become computationally too expensive. Consequently, the simultaneous estimation of several thermophysical parameters, together with their corresponding uncertainties and correlations, is often not computationally feasible and the analysis is usually reduced to fitting one or two parameters. Data assimilation (DA) methods have been shown to be robust while sufficiently accurate and computationally affordable even for a large number of parameters. This paper will introduce a standard sequential DA method, the ensemble square root filter, for thermophysical modelling of asteroid surfaces. This method is used to re-analyse infrared observations of the MARA instrument, which measured the diurnal temperature variation of a single boulder on the surface of near-Earth asteroid (162173) Ryugu. The thermal inertia is estimated to be 295 ± 18 $\mathrm{J\, m^{-2}\, K^{-1}\, s^{-1/2}}$, while all five free parameters of the initial analysis are varied and estimated simultaneously. Based on this thermal inertia estimate the thermal conductivity of the boulder is estimated to be between 0.07 and 0.12,$\mathrm{W\, m^{-1}\, K^{-1}}$ and the porosity to be between 0.30 and 0.52. For the first time in thermophysical parameter derivation, correlations and uncertainties of all free model parameters are incorporated in the estimation procedure that is more than 5000 times more efficient than a comparable parameter sweep.
When light is brought to a standstill, its interaction with gain media increases dramatically due to a singularity in the density of optical states. Concurrently, stopped light engenders an inherent ...and cavity-free feedback mechanism, similar in effect to the feedback that has been demonstrated and exploited in large-scale disordered media and random lasers. Here we study the spatial, temporal and spectral signatures of lasing in planar gain-enhanced nanoplasmonic structures at near-infrared frequencies and show that the stopped-light feedback mechanism allows for nanolasing without a cavity. We reveal that in the absence of cavity-induced feedback, the subwavelength lasing mode forms dynamically as a phase-locked superposition of quasi dispersion-free waveguide modes. This mechanism proves remarkably robust against interface roughness and offers a new route towards nanolasing, the experimental realization of ultra-thin surface emitting lasers, and cavity-free active quantum plasmonics.
Amblyopia is a neurodevelopmental disorder of the visual system that is associated with disrupted binocular vision during early childhood. There is evidence that the effects of amblyopia extend ...beyond the primary visual cortex to regions of the dorsal and ventral extra-striate visual cortex involved in visual integration. Here, we review the current literature on global processing deficits in observers with either strabismic, anisometropic, or deprivation amblyopia. A range of global processing tasks have been used to investigate the extent of the cortical deficit in amblyopia including: global motion perception, global form perception, face perception, and biological motion. These tasks appear to be differentially affected by amblyopia. In general, observers with unilateral amblyopia appear to show deficits for local spatial processing and global tasks that require the segregation of signal from noise. In bilateral cases, the global processing deficits are exaggerated, and appear to extend to specialized perceptual systems such as those involved in face processing.
We tested the hypothesis that Crohn's disease (CD)-related genetic polymorphisms involved in host innate immunity are associated with shifts in human ileum-associated microbial composition in a ...cross-sectional analysis of human ileal samples. Sanger sequencing of the bacterial 16S ribosomal RNA (rRNA) gene and 454 sequencing of 16S rRNA gene hypervariable regions (V1-V3 and V3-V5), were conducted on macroscopically disease-unaffected ileal biopsies collected from 52 ileal CD, 58 ulcerative colitis and 60 control patients without inflammatory bowel diseases (IBD) undergoing initial surgical resection. These subjects also were genotyped for the three major NOD2 risk alleles (Leu1007fs, R708W, G908R) and the ATG16L1 risk allele (T300A). The samples were linked to clinical metadata, including body mass index, smoking status and Clostridia difficile infection. The sequences were classified into seven phyla/subphyla categories using the Naïve Bayesian Classifier of the Ribosome Database Project. Centered log ratio transformation of six predominant categories was included as the dependent variable in the permutation based MANCOVA for the overall composition with stepwise variable selection. Polymerase chain reaction (PCR) assays were conducted to measure the relative frequencies of the Clostridium coccoides - Eubacterium rectales group and the Faecalibacterium prausnitzii spp. Empiric logit transformations of the relative frequencies of these two microbial groups were included in permutation-based ANCOVA. Regardless of sequencing method, IBD phenotype, Clostridia difficile and NOD2 genotype were selected as associated (FDR ≤ 0.05) with shifts in overall microbial composition. IBD phenotype and NOD2 genotype were also selected as associated with shifts in the relative frequency of the C. coccoides--E. rectales group. IBD phenotype, smoking and IBD medications were selected as associated with shifts in the relative frequency of F. prausnitzii spp. These results indicate that the effects of genetic and environmental factors on IBD are mediated at least in part by the enteric microbiota.

The structure of the upper layer of a comet is a product of its surface activity. The Rosetta Lander Imaging System (ROLIS) on board Philae acquired close-range images of the Agilkia site during its ...descent onto comet 67P/Churyumov-Gerasimenko. These images reveal a photometrically uniform surface covered by regolith composed of debris and blocks ranging in size from centimeters to 5 meters. At the highest resolution of 1 centimeter per pixel, the surface appears granular, with no apparent deposits of unresolved sand-sized particles. The thickness of the regolith varies across the imaged field from 0 to 1 to 2 meters. The presence of aeolian-like features resembling wind tails hints at regolith mobilization and erosion processes. Modeling suggests that abrasion driven by airfall-induced particle "splashing" is responsible for the observed formations.
