New biological models are incorporating the realistic processes underlying biological responses to climate change and other human-caused disturbances. However, these more realistic models require ...detailed information, which is lacking for most species on Earth. Current monitoring efforts mainly document changes in biodiversity, rather than collecting the mechanistic data needed to predict future changes. We describe and prioritize the biological information needed to inform more realistic projections of species' responses to climate change. We also highlight how trait-based approaches and adaptive modeling can leverage sparse data to make broader predictions. We outline a global effort to collect the data necessary to better understand, anticipate, and reduce the damaging effects of climate change on biodiversity.
The effects of urban heat islands (UHIs) have a substantial bearing on the sustainability of cities and environs. This paper examines the efficacy of green and cool roofs as potential UHI mitigation ...strategies to make cities more resilient against UHI. We have employed the urbanized version of the Weather Research and Forecasting (uWRF) model at high (1 km) resolution with physically-based rooftop parameterization schemes (conventional, green and cool), a first-time application to the Chicago metropolitan area. We simulated a hot summer period (16-18 August 2013) and assessed (i) UHI reductions for different urban landuse with green/cool roofs, (ii) the interaction of lake breeze and UHI, and (iii) diurnal boundary layer dynamics. The performance of uWRF was evaluated using sensible heat flux and air temperature measurements from an urban mini-field campaign. The simulated roof surface energy balance captured the energy distribution with respective rooftop algorithms. Results showed that daytime roof temperature reduced and varied linearly with increasing green roof fractions, from less than 1 °C for the case of 25% green roof to ∼3 °C during peak daytime for 100% green roof. Diurnal transitions from land to lake breeze and vice versa had a substantial impact on the daytime cycle of roof surface UHI, which had a 3-4 hour lag in comparison to 2 m UHI. Green and cool roofs reduced horizontal and vertical wind speeds and affected lower atmosphere dynamics, including reduced vertical mixing, lower boundary layer depth, and weaker convective rolls. The lowered wind speeds and vertical mixing during daytime led to stagnation of air near the surface, potentially causing air quality issues. The selection of green and cool roofs for UHI mitigation should therefore carefully consider the competing feedbacks. The new results for regional land-lake circulations and boundary layer dynamics from this study may be extended to other urbanized areas, particularly to coastal areas.
Species distribution models (SDMs) have been criticized for involving assumptions that ignore or categorize many ecologically relevant factors such as dispersal ability and biotic interactions. ...Another potential source of model error is the assumption that species are ecologically uniform in their climatic tolerances across their range. Typically, SDMs treat a species as a single entity, although populations of many species differ due to local adaptation or other genetic differentiation. Not taking local adaptation into account may lead to incorrect range prediction and therefore misplaced conservation efforts. A constraint is that we often do not know the degree to which populations are locally adapted. Lacking experimental evidence, we still can evaluate niche differentiation within a speciesâ range to promote better conservation decisions. We explore possible conservation implications of making type I or type II errors in this context. For each of two species, we construct three separate MaxEnt models, one considering the species as a single population and two of disjunct populations. Principal component analyses and response curves indicate different climate characteristics in the current environments of the populations. Model projections into future climates indicate minimal overlap between areas predicted to be climatically suitable by the whole species vs. populationâbased models. We present a workflow for addressing uncertainty surrounding local adaptation in SDM application and illustrate the value of conducting populationâbased models to compare with wholeâspecies models. These comparisons might result in more cautious management actions when alternative range outcomes are considered.
Increases in thermal variability elevate metabolic rate due to Jensen's inequality, and increased metabolic rate decreases the fitness of dormant ectotherms by increasing consumption of stored energy ...reserves. Theory predicts that ectotherms should respond to increased thermal variability by lowering the thermal sensitivity of metabolism, which will reduce the impact of the warm portion of thermal variability. We examined the thermal sensitivity of metabolic rate of overwintering Erynnis propertius (Lepidoptera: Hesperiidae) larvae from a stable or variable environment reared in the laboratory in a reciprocal common garden design, and used these data to model energy use during the winters of 1973-2010 using meteorological data to predict the energetic outcomes of metabolic compensation and phenological shifts. Larvae that experienced variable temperatures had decreased thermal sensitivity of metabolic rate, and were larger than those reared at stable temperatures, which could partially compensate for the increased energetic demands. Even with depressed thermal sensitivity, the variable environment was more energy-demanding than the stable, with the majority of this demand occurring in autumn. Autumn phenology changes thus had disproportionate influence on energy consumption in variable environments, and variable-reared larvae were most susceptible to overwinter energy drain. Therefore the energetic impacts of the timing of entry into winter dormancy will strongly influence ectotherm fitness in northern temperate environments. We conclude that thermal variability drives the expression of metabolic suppression in this species; that phenological shifts will have a greater impact on ectotherms in variable thermal environments; and that E. propertius will be more sensitive to shifts in phenology in autumn than in spring. This suggests that increases in overwinter thermal variability and/or extended, warm autumns, will negatively impact all non-feeding dormant ectotherms which lack the ability to suppress their overwinter metabolic thermal sensitivity.

Scientific and societal unknowns make it difficult to predict how global environmental changes such as climate change and biological invasions will affect ecological systems. In the long term, these ...changes may have interacting effects and compound the uncertainty associated with each individual driver. Nonetheless, invasive species are likely to respond in ways that should be qualitatively predictable, and some of these responses will be distinct from those of native counterparts. We used the stages of invasion known as the "invasion pathway" to identify 5 nonexclusive consequences of climate change for invasive species: (1) altered transport and introduction mechanisms, (2) establishment of new invasive species, (3) altered impact of existing invasive species, (4) altered distribution of existing invasive species, and (5) altered effectiveness of control strategies. We then used these consequences to identify testable hypotheses about the responses of invasive species to climate change and provide suggestions for invasive-species management plans. The 5 consequences also emphasize the need for enhanced environmental monitoring and expanded coordination among entities involved in invasive-species management.
The thorough characterization of chitosan-cleaving enzymes is crucial to unveil structure-function relationships of this promising class of biomolecules for both, enzymatic fingerprinting analyses ...and to use the enzymes as biotechnological tools to produce tailor-made chitosans for diverse applications. Analyzing polymeric substrates as well as oligomeric products has been established as an effective way to understand the actions of enzymes, but it currently requires separate, rather laborious methods to obtain the full picture. Here, we present ultra high performance size exclusion chromatography coupled to refractive index and mass spectrometry detection (UHPSEC-RI-MS) as a straightforward method for the semi-quantitative analysis of chitosan oligomers of up to ten monomers in length. Additionally, the method allows to determine the average molecular weight of the remaining polymers and its distribution. By sampling live from an ongoing enzymatic reaction, UHPSEC-RI-MS offers the unique opportunity to analyze polymers and oligomers simultaneously-i.e., to monitor the molecular weight reduction of the polymeric substrate over the course of the digestion, while at the same time analyzing the emerging oligomeric products in a semi-quantitative manner. In this way, a single simple analysis yields detailed insights into an enzyme's action on a given substrate.
Hughes et al compare the utility of various statistical approaches for assessing the diversity of microbial communities. They argue that even without knowing the "truth," it is possible to rigorously ...compare relative diversity among communities. They also apply some of these diversity measures to microbial data sets and examine how the confidence of the measures changes with sample size.
The emerging field of climate-change adaptation has experienced a dramatic increase in attention as the impacts of climate change on biodiversity and ecosystems have become more evident. Preparing ...for and addressing these changes are now prominent themes in conservation and natural resource policy and practice. Adaptation increasingly is viewed as a way of managing change, rather than just maintaining existing conditions. There is also increasing recognition of the need not only to adjust management strategies in light of climate shifts, but to reassess and, as needed, modify underlying conservation goals. Major advances in the development of climate-adaptation principles, strategies, and planning processes have occurred over the past few years, although implementation of adaptation plans continues to lag. With ecosystems expected to undergo continuing climate-mediated changes for years to come, adaptation can best be thought of as an ongoing process, rather than as a fixed endpoint.
Chitosanases can be used to produce partially acetylated chitosan oligosaccharides (paCOS) for different applications, provided they are thoroughly characterized. However, recent studies indicate ...that the established classification system for chitosanases is too simplistic. Here, we apply a highly sensitive method for quantitatively sequencing paCOS to reassess the substrate specificities of the best-characterized class I-III chitosanases. The enzymes' abilities to cleave bonds at GlcNAc residues positioned at subsite (-1) or (+1), on which the classification system is based, vary especially when the substrates have different fractions of acetylation (F
). Conflicts with the recent classification are observed at higher F
, which were not investigated in prior specificity determinations. Initial analyses of pectin-degrading enzymes reveal that classifications of other polysaccharide-degrading enzymes should also be critically reassessed. Based on our results, we tentatively suggest a chitosanase classification system which is based on specificities and preferences of subsites (-2) to (+2).
