Electrical transport measurements have been made on a series of trivalent-ion-doped tungsten bronzes MxWO3, with M = Y (0.05 ≤ x ≤ 0.12) or La (0.05 ≤ x ≤ 0.19), over the temperature range 2-300 K. ...The results are consistent with a metal-insulator transition (MIT) at a critical concentration xC 0.06, which corresponds to an electron concentration nC 3.3 × 1021 cm−3. The appearance of small concentrations of non-cubic phases for x xC does not have a significant impact on the evolution of the electronic properties of the trivalent bronzes in the low x range. Analysis of the transport results, and a comparison of the findings with those obtained by other workers for the sodium tungsten bronzes, suggest that electron-electron interaction effects play a significant role in inducing the MIT in this type of disordered system.
In recent years, pediatric emergency departments (PED) have seen an increase in presentations related to substance use among their adolescent patient population. We aimed to examine pediatric ...emergency medicine (PEM) physicians' knowledge, attitudes, and beliefs on caring for adolescents with substance use.
We conducted a cross-sectional online survey of PEM physicians through the American Academy of Pediatrics Pediatric Emergency Medicine Collaborative Research Committee (PEM-CRC) listserv. The 41-item survey contained the following domains: demographics, current protocols and education for managing adolescent substance use, and attitudes about treatment of substance use. We calculated descriptive statistics for each variable within the domains.
Of 177 respondents (38.2% response rate), 55.4% were female, 45.2% aged ≥ 50 years, 78% worked in a children's hospital, and 50.8% had > 15 years clinical practice. Overall, 77.8% reported caring for adolescents with a chief complaint related to non-opioid substance use and 26.0% opioid use at least once a month. Most (80.9%) reported feeling comfortable treating major medical complications of substance use, while less than half were comfortable treating withdrawal symptoms. 73% said that they were not interested in prescribing buprenorphine.
Among this national sample of PEM physicians, 3 of 4 physicians managed substance-related visits monthly, but 52% lacked comfort in managing withdrawal symptoms and 73.1% were not interested in prescribing buprenorphine. Almost all PEM physician identified substance use-related education is important but lacked access to faculty expertise or educational content. Expanded access to education and training for PEM physicians related to substance use is needed.
Background: Accumulating evidence indicates that the gut microbiota can synthesize neurotransmitters as well as impact host-derived neurotransmitter levels. In the past, it has been challenging to ...decipher which microbes influence neurotransmitters due to the complexity of the gut microbiota. Methods: To address whether a single microbe, Bifidobacterium dentium, could regulate important neurotransmitters, we examined Bifidobacteria genomes and explored neurotransmitter pathways in secreted cell-free supernatant using LC-MS/MS. To determine if B. dentium could impact neurotransmitters in vivo, we mono-associated germ-free mice with B. dentium ATCC 27678 and examined fecal and brain neurotransmitter concentrations. Results: We found that B. dentium possessed the enzymatic machinery to generate γ-aminobutyric acid (GABA) from glutamate, glutamine, and succinate. Consistent with the genome analysis, we found that B. dentium secreted GABA in a fully defined microbial media and elevated fecal GABA in B. dentium mono-associated mice compared to germ-free controls. We also examined the tyrosine/dopamine pathway and found that B. dentium could synthesize tyrosine, but could not generate L-dopa, dopamine, norepinephrine, or epinephrine. In vivo, we found that B. dentium mono-associated mice had elevated levels of tyrosine in the feces and brain. Conclusions: These data indicate that B. dentium can contribute to in vivo neurotransmitter regulation.
The instrumentation for a pathfinder mission towards a possible large scale neutrino telescope named “STRings for Absorption length in Water” (STRAW) is presented in terms of design and performance. ...In June 2018 STRAW was deployed at the Cascadia Basin site operated by Ocean Networks Canada and has been collecting data since then. At a depth of about 2600 meters, the two STRAW 120 meters tall mooring lines are instrumented by three “Precision Optical Calibration Modules” (POCAM) and five Digital Optical Sensors (sDOM). The main objectives of STRAW are the measurement of light extinction in different wavelength bands and bioluminescence at Cascadia Basin. We describe the instrumentation deployed in the Pacific Ocean and show some data from the first measurements.
The vestibular-ocular reflex (VOR) integrates the vestibular and ocular systems to maintain gaze during head motion. This reflex is often negatively affected following sport-related concussion. ...Objective measures of gaze stability, a function mediated by the VOR, such as the computerized dynamic visual acuity test (DVAT) and gaze stabilization test (GST), may have utility in concussion management. However, normative data specific to sport, sex, or concussion history have not been established in collegiate athletes.
The objective of this study was to establish normative values for the DVAT and GST in collegiate athletes and explore the effect of sport, sex, and concussion history on VOR assessments.
The DVAT and GST were completed by 124 collegiate athletes (72 male, 52 female, mean±SD, age: 19.71±1.74 years, height: 173.99±13.97 cm, weight: 80.06±26.52 kg) recruited from Division-I athletic teams (football, soccer and cheerleading). The DVAT and GST were performed in the rightward and leftward directions during a single session in a standardized environment. Normative values for DVAT and GST measures were expressed as percentiles. Non-parametric statistics were used to compare differences between groups based on sex, sport, and concussion history. Alpha was set a-priori at 0.05.
Overall, the median LogMAR unit for 124 athletes completing the DVAT was 0 (IQR = 0.17) for both leftward and rightward. The median velocities achieved on the GST were 145 °/sec and 150 °/sec (IQR = 45 and 40) for the leftward and rightward directions respectively. Significant differences were observed between sports (p = 0.001-0.17) for the GST with cheerleading demonstrating higher velocities than the other sports. However, no significant differences were identified based on sex (p≥0.09) or history of concussion (p≥0.15).
Normative estimates for the DVAT and GST may assist in the clinical interpretation of outcomes when used in post-concussion evaluation for collegiate athletes. Although sex and previous concussion history had no effect on the DVAT or GST, performance on these measures may be influenced by type of sport. Sport-related differences in the GST may reflect VOR adaptations based on individual sport-specific demands.

Large-scale flood events often show spatial correlation in neighbouring basins, and thus can affect adjacent basins simultaneously, as well as result in superposition of different flood peaks. Such ...flood events therefore need to be addressed with large-scale modelling approaches to capture these processes. Many approaches currently in place are based on either a hydrologic or a hydrodynamic model. However, the resulting lack of interaction between hydrology and hydrodynamics, for instance, by implementing groundwater infiltration on inundated floodplains, can hamper modelled inundation and discharge results where such interactions are important. In this study, the global hydrologic model PCR-GLOBWB at 30 arcmin spatial resolution was one-directionally and spatially coupled with the hydrodynamic model Delft 3D Flexible Mesh (FM) for the Amazon River basin at a grid-by-grid basis and at a daily time step. The use of a flexible unstructured mesh allows for fine-scale representation of channels and floodplains, while preserving a coarser spatial resolution for less flood-prone areas, thus not unnecessarily increasing computational costs. In addition, we assessed the difference between a 1-D channel/2-D floodplain and a 2-D schematization in Delft 3D FM. Validating modelled discharge results shows that coupling PCR-GLOBWB to a hydrodynamic routing scheme generally increases model performance compared to using a hydrodynamic or hydrologic model only for all validation parameters applied. Closer examination shows that the 1-D/2-D schematization outperforms 2-D for r2 and root mean square error (RMSE) whilst having a lower Kling–Gupta efficiency (KGE). We also found that spatial coupling has the significant advantage of a better representation of inundation at smaller streams throughout the model domain. A validation of simulated inundation extent revealed that only those set-ups incorporating 1-D channels are capable of representing inundations for reaches below the spatial resolution of the 2-D mesh. Implementing 1-D channels is therefore particularly of advantage for large-scale inundation models, as they are often built upon remotely sensed surface elevation data which often enclose a strong vertical bias, hampering downstream connectivity. Since only a one-directional coupling approach was tested, and therefore important feedback processes are not incorporated, simulated discharge and inundation extent for both coupled set-ups is generally overpredicted. Hence, it will be the subsequent step to extend it to a two-directional coupling scheme to obtain a closed feedback loop between hydrologic and hydrodynamic processes. The current findings demonstrating the potential of one-directionally and spatially coupled models to obtain improved discharge estimates form an important step towards a large-scale inundation model with a full dynamic coupling between hydrology and hydrodynamics.
Abstract Ankle dorsiflexion range of motion (DROM) is often a point of emphasis during the rehabilitation of lower extremity pathologies. With the growing popularity of weight-bearing DROM ...assessments, several versions of the weight-bearing lunge (WBLT) test have been developed and numerous reliability studies have been conducted. The purpose of this systematic review was to critically appraise and synthesize the studies which examined the reliability and responsiveness of the WBLT to assess DROM. A systematic search of PubMed and EBSCO Host databases from inception to September 2014 was conducted to identify studies whose primary aim was assessing the reliability of the WBLT. The Quality Appraisal of Reliability Studies assessment tool was utilized to determine the quality of included studies. Relative reliability was examined through intraclass correlation coefficients (ICC) and responsiveness was evaluated through minimal detectable change (MDC). A total of 12 studies met the eligibility criteria and were included. Nine included studies assessed inter-clinician reliability and 12 included studies assessed intra-clinician reliability. There was strong evidence that inter-clinician reliability (ICC = 0.80–0.99) as well as intra-clinician reliability (ICC = 0.65–0.99) of the WBLT is good. Additionally, average MDC scores of 4.6° or 1.6 cm for inter-clinician and 4.7° or 1.9 cm for intra-clinician were found, indicating the minimal change in DROM needed to be outside the error of the WBLT. This systematic review determined that the WBLT, regardless of method, can be used clinically to assess DROM as it provides consistent results between one or more clinicians and demonstrates reasonable responsiveness.
Despite advances in allogeneic stem cell transplantation, BCR-ABL-positive acute lymphoblastic leukaemia (ALL) remains a high-risk disease, necessitating the development of novel treatment ...strategies. As the known oncomir, miR-17~92, is regulated by BCR-ABL fusion in chronic myeloid leukaemia, we investigated its role in BCR-ABL translocated ALL. miR-17~92-encoded miRNAs were significantly less abundant in BCR-ABL-positive as compared to -negative ALL-cells and overexpression of miR-17~19b triggered apoptosis in a BCR-ABL-dependent manner. Stable isotope labelling of amino acids in culture (SILAC) followed by liquid chromatography and mass spectroscopy (LC-MS) identified several apoptosis-related proteins including Bcl2 as potential targets of miR-17~19b. We validated Bcl2 as a direct target of this miRNA cluster in mice and humans, and, similar to miR-17~19b overexpression, Bcl2-specific RNAi strongly induced apoptosis in BCR-ABL-positive cells. Furthermore, BCR-ABL-positive human ALL cell lines were more sensitive to pharmacological BCL2 inhibition than negative ones. Finally, in a xenograft model using patient-derived leukaemic blasts, real-time, in vivo imaging confirmed pharmacological inhibition of BCL2 as a new therapeutic strategy in BCR-ABL-positive ALL. These data demonstrate the role of miR-17~92 in regulation of apoptosis, and identify BCL2 as a therapeutic target of particular relevance in BCR-ABL-positive ALL.
There is an enormous need to make better use of the ever increasing wealth of publicly available genomic information and to utilize the tremendous progress in computational approaches in the life ...sciences. Transcriptional regulation of protein-coding genes is a major mechanism of controlling cellular functions. However, the myriad of transcription factors potentially controlling transcription of any given gene makes it often difficult to quickly identify the biological relevant transcription factors. Here, we report on the identification of Hnf4a as a major transcription factor of the so far unstudied DnaJ heat shock protein family (Hsp40) member C22 (Dnajc22). We propose an approach utilizing recent advances in computational biology and the wealth of publicly available genomic information guiding the identification of potential transcription factor candidates together with wet-lab experiments validating computational models. More specifically, the combined use of co-expression analyses based on self-organizing maps with sequence-based transcription factor binding prediction led to the identification of Hnf4a as the potential transcriptional regulator for Dnajc22 which was further corroborated using publicly available datasets on Hnf4a. Following this procedure, we determined its functional binding site in the murine Dnajc22 locus using ChIP-qPCR and luciferase assays and verified this regulatory loop in fruitfly, zebrafish, and humans.
