Abstract
Stellar flares have been extensively studied in soft X-rays (SXRs) by basically every X-ray mission. Hard X-ray (HXR) emission from stellar superflares, however, have only been detected from ...a handful of objects over the past years. One very extreme event was the superflare from the young M-dwarf DG CVn binary star system, which triggered Swift/BAT as if it was a γ-ray burst. In this work, we estimate the expected γ-ray emission from DG CVn and the most extreme stellar flares by extrapolating from solar flares based on measured solar energetic particles (SEPs), as well as thermal and non-thermal emission properties. We find that ions are plausibly accelerated in stellar superflares to 100 GeV energies, and possibly up to TeV energies in the associated coronal mass ejections. The corresponding π0-decay γ-ray emission could be detectable from stellar superflares with ground-based γ-ray telescopes. On the other hand, the detection of γ-ray emission implies particle densities high enough that ions suffer significant losses due to inelastic proton–proton scattering. The next-generation Cherenkov Telescope Array (CTA) should be able to probe superflares from M dwarfs in the solar neighbourhood and constrain the energy in interacting cosmic rays and/or their maximum energy. The detection of γ-ray emission from stellar flares would open a new window for the study of stellar physics, the underlying physical processes in flares and their impact on habitability of planetary systems.

L-forms are "cell wall-deficient" bacteria which are able to grow as spheroplasts or protoplasts. They can be differentiated into four types depending on their ability to revert to the parental, ...cell-walled form and to the extent of their cell-wall modification. L-forms are significant in modern science because of their contributions to an improved understanding of principal questions and their interactions with eukaryotes. This review particularly focuses on research using stable protoplast-type L-forms which have contributed to a better understanding of the structural and functional organisation of the cytoplasmic membrane and of cell division. These L-forms, which have only a single surrounding bilayer membrane, also represent a unique expression system for production of recombinant proteins. A large proportion of L-form publications concern their putative role in human disease and its therapy, a topic which is discussed briefly. L-forms have also been used to form intracellular associations with plant cells and have been shown to elicit induced disease resistance offering a novel method for plant protection. The recent decline in active research on L-forms is a concern as knowledge and experience, as well as unique L-form strains which have been maintained for decades, are being lost.

The vertebrate kinetochore complex assembles at the centromere on α-satellite DNA. In humans, α-satellite DNA has a repeat length of 171 bp slightly longer than the DNA in the chromatosome containing ...the linker histone H1. The centromere-binding protein CENP-B binds specifically to α-satellite DNA with properties of a centromeric-linker histone. Here, we analysed if linker histone H1 is present at or excluded from centromeric chromatin by CENP-B. By immunostaining we detected the presence, but no enrichment or depletion of five different H1 subtypes at centromeric chromatin. The binding dynamics of H1 at centromeric sites were similar to that at other locations in the genome. These dynamics did not change in CENP-B depleted cells, suggesting that CENP-B and H1 co-exist in centromeric chromatin with no or little functional overlap. By bimolecular fluorescence complementation (BiFC) and Förster resonance energy transfer (FRET), we revealed that the linker histone H1 subtypes H1° and H1.2 bind to centromeric chromatin in interphase nuclei in direct neighbourhood to inner kinetochore proteins.
The Escherichia coli low-copy-number plasmid R1 contains a segregation machinery composed of parC, ParR and parM. The R1 centromere-like site parC contains two separate sets of repeats. By atomic ...force microscopy (AFM) we show here that ParR molecules bind to each of the 5-fold repeated iterons separately with the intervening sequence unbound by ParR. The two ParR protein complexes on parC do not complex with each other. ParR binds with a stoichiometry of about one ParR dimer per each single iteron. The measured DNA fragment lengths agreed with B-form DNA and each of the two parC 5-fold interon DNA stretches adopts a linear path in its complex with ParR. However, the overall parC/ParR complex with both iteron repeats bound by ParR forms an overall U-shaped structure: the DNA folds back on itself nearly completely, including an angle of ∼150°. Analysing linear DNA fragments, we never observed dimerized ParR complexes on one parC DNA molecule (intramolecular) nor a dimerization between ParR complexes bound to two different parC DNA molecules (intermolecular). This bacterial segrosome is compared to other bacterial segregation complexes. We speculate that partition complexes might have a similar overall structural organization and, at least in part, common functional properties.
Faithful chromatin segregation is mediated and controlled by the kinetochore protein network which assembles at centromeres. In this study, the neighbourhood relations of inner kinetochore and ...nucleosome-associated complex (NAC) proteins were analysed in living human interphase cells by acceptor photobleaching FRET. The data indicate that CENP-U is in close vicinity to CENP-I as well as to CENP-B and that CENP-M is close to CENP-T.
L-Glutamate is made with Corynebacterium glutamicum on a scale of more than 10(6) tons/year. Nevertheless, formation of this amino acid is enigmatic and there is very limited molecular information ...available to unravel the apparently complex conditions leading to L-glutamate efflux. Here, we report the isolation and overexpression of the genes involved in lipid synthesis: acp, fadD15, cma, cls, pgsA2, cdsA, gpsA, and plsC, and the inactivation of cma and cls. In addition, the consequences for phospholipid content, temperature sensitivity, as well as detergent-independent and detergent-dependent L-glutamate efflux were quantified. An in part strong alteration of the phospholipid composition was achieved; for instance, overexpression of fadD15 encoding an acylCoA ligase resulted in an increase of phosphatidyl inositol from 12.6 to 30.2%. All strains, except that overexpressing acp (acyl carrier protein), exhibited increased temperature sensitivity, with the strongest sensitivity present upon cls (cardiolipin synthetase) inactivation. As a consequence of the genetically modified lipid synthesis, L-glutamate efflux changed quite dramatically; for instance, overexpression of plsC (acylglycerolacyl transferase) resulted in a detergent-triggered increase of L-glutamate accumulation from 92 mM to 108 mM, whereas acp overexpression reduced the accumulation to 24 mM. With some of the overexpressed genes, substantial L-glutamate excretion even without detergent addition was obtained when the fermentation temperature was elevated. These data show that the chemical and physical properties of the cytoplasmic membrane are altered and suggest that this is a necessary precondition to achieve L-glutamate efflux.
The centromere sequence parC of Escherichia coli low-copy-number plasmid R1 consists of two sets of 11 bp iterated sequences. Here we analysed the intrinsic sequence-directed curvature of parC by its ...migration anomaly in polyacrylamide gels. The 159 bp long parC is strongly curved with anomaly values (k-factors) close to 2. The properties of the parC curvature agree with those of other curved DNA sequences. parC contains two regions of 5-fold repeated iterons separated by 39 bp. We modified 4 bp within this intermediate sequence so that we could analyse the two 5-fold repeated regions independently. The analysis shows that the two repeat regions are not independently curved parts of parC but that the overall curvature is a property of the whole fragment. Since the centromere sequence of an E.coli plasmid as well as eukaryotic centromere sequences show DNA curvature, we speculate that curvature might be a general property of centromeres.
Observations of astrophysical transients have brought many novel discoveries and provided new insights into the physical processes at work under extreme conditions in the Universe. Multi-wavelength ...and multi-messenger observations of variable objects require dedicated procedures and follow-up systems capable of digesting and reacting to external alerts to execute coordinated follow-up campaigns. The main functions of such follow-up systems are the processing, filtering, and ranking of the incoming alerts, the fully automated rapid execution of the observations according to an observation strategy tailored to the instrument, and real-time data analysis with feedback to the operators and other instruments. The High Energy Stereoscopic System (H.E.S.S.) has been searching for transient phenomena since its inauguration in 2003. In this paper, we describe the transients follow-up system of H.E.S.S. which became operational in 2016. The system allows H.E.S.S. to conduct a more versatile, optimised, and largely autonomous transient follow-up programme, combining all major functionalities in one systematic approach. We describe the design, central functionalities, and interfaces of the follow-up system in general and its three main components in detail: the Target of Opportunity (ToO) alert system, the data acquisition and central control system, and the real-time analysis. We highlight architectural decisions and features that enable fully automatic ToO follow-up and indicate key performance metrics of the subsystems. We discuss the system's capabilities and highlight the need for a fine-tuned interplay of the different subsystems in order to react quickly and reliably. Lessons learnt from the development, integration, and operation of the follow-up system are reviewed in light of new and large science infrastructures and associated challenges in this exciting new era of inter-operable astronomy.
