X-ray ptychography is a scanning variant of coherent diffractive imaging with the ability to image large fields of view at high resolution. It further allows imaging of non-isolated specimens and can ...produce quantitative mapping of the electron density distribution in 3D when combined with computed tomography. The method does not require imaging lenses, which makes it dose efficient and suitable to multi-keV X-rays, where efficient photon counting, pixelated detectors are available. Here we present the first highly resolved quantitative X-ray ptychographic tomography of an extended object yielding 16 nm isotropic 3D resolution recorded at 2 Å wavelength. This first-of-its-kind demonstration paves the way for ptychographic X-ray tomography to become a promising method for X-ray imaging of representative sample volumes at unmatched resolution, opening tremendous potential for characterizing samples in materials science and biology by filling the resolution gap between electron microscopy and other X-ray imaging techniques.
We perform attosecond time-resolved transient absorption spectroscopy around the first ionization threshold of helium and observe rapid oscillations of the absorption of the individual harmonics as a ...function of time delay with respect to a superimposed, moderately strong infrared laser field. The phase relation between the absorption modulation of individual harmonics gives direct evidence for the interference of transiently bound electronic wave packets as the mechanism behind the absorption modulation.
The C-Band All-Sky Survey (C-BASS) is an all-sky full-polarization survey at a frequency of 5 GHz, designed to provide complementary data to the all-sky surveys of WMAP and Planck, and future CMB ...B-mode polarization imaging surveys. The observing frequency has been chosen to provide a signal that is dominated by Galactic synchrotron emission, but suffers little from Faraday rotation, so that the measured polarization directions provide a good template for higher frequency observations, and carry direct information about the Galactic magnetic field. Telescopes in both northern and southern hemispheres with matched optical performance are used to provide all-sky coverage from a ground-based experiment. A continuous-comparison radiometer and a correlation polarimeter on each telescope provide stable imaging properties such that all angular scales from the instrument resolution of 45 arcmin up to full sky are accurately measured. The northern instrument has completed its survey and the southern instrument has started observing. We expect that C-BASS data will significantly improve the component separation analysis of Planck and other CMB data, and will provide important constraints on the properties of anomalous Galactic dust and the Galactic magnetic field.
Long intergenic non-coding RNA-Nucleotide Metabolism Regulator (lincNMR) is a long non-coding RNA (lncRNA) which is induced in hepatocellular carcinoma. Its depletion invokes a proliferation defect, ...triggers senescence and inhibits colony formation in liver, but also breast and lung cancer cells. Triple-label SILAC proteomics profiles reveal a deregulation of key cell cycle regulators in lincNMR-depleted cells like the key dNTP synthesizing enzymes RRM2, TYMS and TK1, implicating lincNMR in regulating nucleotide metabolism. LincNMR silencing decreases dNTP levels, while exogenous dNTPs rescues the proliferation defect induced by lincNMR depletion. In vivo RNA Antisense Purification (RAP-MS) identifies YBX1 as a direct interaction partner of lincNMR which regulates RRM2, TYMS and TK1 expression and binds to their promoter regions. In a Chick Chorioallantoic Membrane (CAM) in vivo model, lincNMR-depleted tumors are significantly smaller. In summary, we discover a lincRNA, lincNMR, which regulates tumor cell proliferation through a YBX1-RRM2-TYMS-TK1 axis governing nucleotide metabolism.
Since its commercial introduction three-quarters of a century ago, fluid catalytic cracking has been one of the most important conversion processes in the petroleum industry. In this process, porous ...composites composed of zeolite and clay crack the heavy fractions in crude oil into transportation fuel and petrochemical feedstocks. Yet, over time the catalytic activity of these composite particles decreases. Here, we report on ptychographic tomography, diffraction, and fluorescence tomography, as well as electron microscopy measurements, which elucidate the structural changes that lead to catalyst deactivation. In combination, these measurements reveal zeolite amorphization and distinct structural changes on the particle exterior as the driving forces behind catalyst deactivation. Amorphization of zeolites, in particular, close to the particle exterior, results in a reduction of catalytic capacity. A concretion of the outermost particle layer into a dense amorphous silica-alumina shell further reduces the mass transport to the active sites within the composite.Catalyst deactivation in fluid catalytic cracking processes is unavoidably associated with structural changes. Here, the authors visualize the deactivation of zeolite catalysts by ptychography and other imaging techniques, showing pronounced amorphization of the outer layer of the catalyst particles.
High-throughput three-dimensional cryogenic imaging of thick biological specimens is valuable for identifying biologically- or pathologically-relevant features of interest, especially for subsequent ...correlative studies. Unfortunately, high-resolution imaging techniques at cryogenic conditions often require sample reduction through sequential physical milling or sectioning for sufficient penetration to generate each image of the 3-D stack. This study represents the first demonstration of using ptychographic hard X-ray tomography at cryogenic temperatures for imaging thick biological tissue in a chemically-fixed, frozen-hydrated state without heavy metal staining and organic solvents. Applied to mammalian brain, this label-free cryogenic imaging method allows visualization of myelinated axons and sub-cellular features such as age-related pigmented cellular inclusions at a spatial resolution of ~100 nanometers and thicknesses approaching 100 microns. Because our approach does not require dehydration, staining or reduction of the sample, we introduce the possibility for subsequent analysis of the same tissue using orthogonal approaches that are expected to yield direct complementary insight to the biological features of interest.
A variational model for learning convolutional image atoms from corrupted and/or incomplete data is introduced and analyzed both in function space and numerically. Building on lifting and relaxation ...strategies, the proposed approach is convex and allows for simultaneous image reconstruction and atom learning in a general, inverse problems context. Further, motivated by an improved numerical performance, also a semi-convex variant is included in the analysis and the experiments of the paper. For both settings, fundamental analytical properties allowing in particular to ensure well-posedness and stability results for inverse problems are proven in a continuous setting. Exploiting convexity, globally optimal solutions are further computed numerically for applications with incomplete, noisy and blurry data and numerical results are shown.
We propose a variational model for artifact-free JPEG decompression. It is based on the minimization of the total variation over the convex set $U$ of all possible source images associated with given ...JPEG data. The general case where $U$ represents a pointwise restriction with respect to an $L^2$-orthonormal basis is considered. Analysis of the infinite dimensional model is presented, including the derivation of optimality conditions. A discretized version is solved using a primal-dual algorithm supplemented by a primal-dual gap-based stopping criterion. Experiments illustrate the effect of the model. Good reconstruction quality is obtained even for highly compressed images, while a graphics processing unit (GPU) implementation is shown to significantly reduce computation time, making the model suitable for real-time applications.
Protein hydroxylation affects protein stability, activity, and interactome, therefore contributing to various diseases including cancers. However, the transiency of the hydroxylation reaction hinders ...the identification of hydroxylase substrates. By developing an enzyme-substrate trapping strategy coupled with TAP-TAG or orthogonal GST- purification followed by mass spectrometry, we identify adenylosuccinate lyase (ADSL) as an EglN2 hydroxylase substrate in triple negative breast cancer (TNBC). ADSL expression is higher in TNBC than other breast cancer subtypes or normal breast tissues. ADSL knockout impairs TNBC cell proliferation and invasiveness in vitro and in vivo. An integrated transcriptomics and metabolomics analysis reveals that ADSL activates the oncogenic cMYC pathway by regulating cMYC protein level via a mechanism requiring ADSL proline 24 hydroxylation. Hydroxylation-proficient ADSL, by affecting adenosine levels, represses the expression of the long non-coding RNA MIR22HG, thus upregulating cMYC protein level. Our findings highlight the role of ADSL hydroxylation in controlling cMYC and TNBC tumorigenesis.
