Amaranth has two morphological types described as red and green morphs. Previous studies have extensively characterised red morph amaranth regarding both morphological and chemical properties ...including antioxidant activity, antioxidant phytochemical profile, mineral content and proximate composition. However, there is scarce information concerning green morph amaranth. Hence, the present study evaluated 12 green morph genotypes for proximate composition, antioxidant activity, antioxidant pigments, minerals, and phytochemicals. Green morph amaranth was found to contain abundant carbohydrates, dietary fiber and protein. We found notable levels of inorganic minerals including potassium, calcium, magnesium, iron, manganese, copper and zinc. Antioxidant capacity quantified as free radical quenching capacity varied between 27 and 48 μg g
Trolox equivalents. We additionally quantified antioxidants, including total phenolics, total flavonoid equivalents and vitamin C, as well as the antioxidant pigments carotenoids, chlorophylls and betalains. These data indicated that four green morph genotypes could be considered as enriched in their antioxidant profiles. Green morph amaranth could be a potential source of nutritional components and antioxidant phytochemicals in the human diet providing opportunities to address mineral nutrient deficiencies and provide an antioxidant rich food.
Southern blight of tomato caused by Sclerotium rolfsii can cause severe plant mortality and yield losses. The use of rhizobacteria for the biological control of Southern blight disease is a potent ...alternative to chemical fungicides. Although rhizobacteria are prolific candidates, comprehensive reports regarding their use in tomato disease management are limited. The present study screened six rhizobacterial strains for antagonism against S. rolfsii in dual culture and culture filtrate assays. The selected promising strains were tested further for plant-growth-promoting and biocontrol potentials under in vitro, greenhouse, and field conditions. Of the six strains screened, Stenotrophomonas maltophilia PPB3 and Bacillus subtilis PPB9 showed the superior performance displaying the highest antagonism against S. rolfsii in dual culture (PPB3 88% and PPB9 71% inhibition), and culture filtrate assays (PPB3 53-100% and PPB9 54-100% inhibition at various concentrations). Oxalic acid produced by S. rolfsii was significantly inhibited by both rhizobacteria and supported their growth as a carbon source. The strains produced hydrogen cyanide, chitinases, siderophores, biofilm, and indole acetic acid. They showed the potential to solubilize phosphate and fix nitrogen. Seed treatment with S. maltophilia PPB3 and B. subtilis PPB9 improved seed germination and tomato seedling vigour. Significant increases in plant growth, chlorophyll contents, and N, P, and K concentrations were attained in bacterized plants compared to non-treated controls. The application of antagonists on container-grown seedlings in a greenhouse environment and field-grown tomato plants reduced symptoms of damping-off and Southern blight. The sclerotial counts decreased significantly in these soils. Bacteria-inoculated plants had a higher yield than those in the non-treated control. Bacteria colonized the entire roots, and their populations increased significantly in the protected plants. The results show the potential capabilities of S. maltophilia PPB3 and B. subtilis PPB9 for growth promotion, nutrient acquisition, and biocontrol of southern blight disease in tomatoes.

It is essential to construct a biofuel cell-based sensor and develop an effective strategy to detect glucose without any potentiostat circuitry in order to create a simple and miniaturized device. In ...this report, an enzymatic biofuel cell (EBFC) is fabricated by the facile design of an anode and cathode on a screen-printed carbon electrode (SPCE). To construct the anode, thionine and flavin adenine dinucleotide-dependent glucose dehydrogenase (FAD-GDH) are covalently immobilized via a crosslinker to make a cross-linked redox network. As a cathode, the Pt-free oxygen reduction carbon catalyst is employed alternative to the commonly used bilirubin oxidase. We proposed the importance of EBFC-based sensors through the connection of anode and cathode; they can identify a short-circuit current by means of applied zero external voltage, thereby capable of glucose detection without under the operation of the potentiostat. The result shows that the EBFC-based sensor could be able to detect based on a short-circuit current with a wide range of glucose concentrations from 0.28 to 30 mM. Further, an EBFC is employed as a one-compartment model energy harvester with a maximum power density of (36 ± 3) μW cm− 2 in sample volume 5 μL. In addition, the constructed EBFC-based sensor demonstrates that the physiological range of ascorbic acid and uric acid shows no significant effect on the short-circuit current generation. Moreover, this EBFC can be used as a sensor in artificial plasma without losing its performance and thereby used as a disposable test strip in real blood sample analysis.
Rapeseed (Brassica napus L.) is the world's third most important edible oilseed crop after soybean and palm. The clubroot disease caused by Plasmodiophora brassicae poses a significant risk and ...causes substantial yield losses in rapeseed. In this study, 13 endophytic fungal strains were isolated from the healthy roots of rapeseed (B. napus) grown in a clubroot-infested field and molecularly identified. Based on germination inhibition of resting spores of P. brassicae, two endophytic fungal antagonists, Trichoderma spp. ReTk1 and ReTv2 were selected to evaluate their potential for plant growth promotion and biocontrol of P. brassicae. The Trichoderma isolates were applied as a soil drench (1x10.sup.7 spore/g soil) to a planting mix and field soil, in which plants were grown under non-infested and P. brassicae-infested (2x10.sup.6 spore/g soil) conditions. The endophytic fungi were able to promote plant growth, significantly increasing shoot and root length, leaf diameter, and biomass production (shoots and root weight) both in the absence or presence of P. brassicae. The single and dual treatments with the endophytes were equally effective in significantly decreasing the root-hair infection, root index, and clubroot severity index. Both ReTk1 and ReTv2 inhibited the germination of resting spores of P. brassicae in root exudates. Moreover, the endophytic fungi colonized the roots of rapeseed extensively and possibly induced host resistance by up-regulated expression of defense-related genes involved in jasmonate (BnOPR2), ethylene (BnACO and BnSAM3), phenylpropanoid (BnOPCL and BnCCR), auxin (BnAAO1) and salicylic acid (BnPR2) pathways. Based on these findings, it is evident that the rapeseed root endophytes Trichoderma spp. ReTk1 and ReTv2 could suppress the gall formation on rapeseed roots via antibiosis, induced systemic resistance (ISR), and/or systemic acquired resistance (SAR). According to our knowledge, this is the first report of the endophytic Trichoderma spp. isolated from root tissues of healthy rapeseed plants (B. napus.), promoting plant growth and reducing clubroot severity.

Glucose level measurement is essential for the point-of-care diagnosis, primarily for persons with diabetes. A disposable electrochemical glucose sensor is constructed using flavin adenine ...dinucleotide-dependent glucose dehydrogenase (FAD-GDH) and redox mediator for electron transfer from the enzyme to the electrode surface. Ideally, a suitable mediator should have high water solubility, high kinetic constant, high stability, and redox potential between −0.2 and 0.1 V vs. Ag|AgCl|sat. KCl. We designed and synthesized two new quinone-based water-soluble mediators: quinoline-5,8-dione (QD) and isoquinoline-5,8-dione (IQD). The formal potentials for both QD and IQD at pH 7.0 were −0.07 V vs. Ag|AgCl|sat. KCl. The logarithms of the electron exchange rate constants (k2/(M−1 s−1)) between QD/IQD and FAD-GDH were 7.7 ± 0.1 and 7.4 ± 0.1 for QD and IQD, respectively, which are the highest value among the water-soluble mediators for FAD-GDH reported to date. Disposable amperometric glucose sensors were fabricated by dropping FAD-GDH and QD or IQD onto a test strip. The sensor achieved a linear response up to glucose concentrations of 55.5 mM. The linear response was obtained even when the mediator loading was low (0.5 nmol/strip); loading was only 0.2 mol% of glucose. The results proved that the response current was primarily controlled by glucose diffusion. In addition, the sensor using QD exhibited high stability over 3 months at room temperature.

Wheat (Triticum aestivum L.), a vital cereal, faces significant challenges from common root rot and spot blotch diseases caused by Bipolaris sorokiniana. This study aimed to explore the potential of ...plant growth promoting rhizobacteria (PGPR) to enhance wheat growth, reduce fertilizer input, and combat Bipolaris diseases. Two PGPR isolates, selected for their superior antagonistic properties, were identified as Stenotrophomonas koreensis RB11 and Bacillus amyloliquefaciens RB12. These PGPR strains displayed multiple plant growth promoting and biocontrol attributes, including phosphate solubilization, indole-3-acetic acid production, nitrogen fixation and antagonism against B. sorokiniana and other fungi. Wheat seed priming with the PGPR significantly improved germination, plant growth, nutrient content and biomass carbon accumulation in the rhizosphere soil. Importantly, the application of RB11 and RB12 allowed for a 25% and 50% reduction in nitrogen fertilizer usage, respectively, without compromising the yield. RB11 and RB12 also demonstrated potent inhibitory effects on B. sorokiniana conidial germination and significantly controlled common root rot and spot blotch in wheat, similar to those observed with the fungicide Protaf 250EC. Overall, this study underscores the multifaceted roles of S. koreensis RB11 and B. amyloliquefaciens RB12 in promoting wheat growth, reducing fertilizer inputs and effectively suppressing wheat pathogens. These findings contribute to the development of PGPR-based strategies for sustainable crop production and disease control.

The three isolates obtained from naturally infected plants were cross inoculated in hyacinth bean, okra and African-American marigold and they were pathogenic to these hosts. The optimum temperature and pH for its growth were 20°C and pH 5.0, respectively. Sclerotial development was favored at pH 5.0. Sucrose and mannitol were the best carbon sources to support hyphal growth, while glucose was the most favourable for sclerotial development. The hyacinth bean genotypes, HB-82 (Rupban Sheem) and HB-102 were found highly resistant, while HB-94 (Ashina) was moderate resistant to the fungus. Finally,
