The clinical features and immune responses of asymptomatic individuals infected with severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) have not been well described. We studied 37 ...asymptomatic individuals in the Wanzhou District who were diagnosed with RT-PCR-confirmed SARS-CoV-2 infections but without any relevant clinical symptoms in the preceding 14 d and during hospitalization. Asymptomatic individuals were admitted to the government-designated Wanzhou People's Hospital for centralized isolation in accordance with policy
. The median duration of viral shedding in the asymptomatic group was 19 d (interquartile range (IQR), 15-26 d). The asymptomatic group had a significantly longer duration of viral shedding than the symptomatic group (log-rank P = 0.028). The virus-specific IgG levels in the asymptomatic group (median S/CO, 3.4; IQR, 1.6-10.7) were significantly lower (P = 0.005) relative to the symptomatic group (median S/CO, 20.5; IQR, 5.8-38.2) in the acute phase. Of asymptomatic individuals, 93.3% (28/30) and 81.1% (30/37) had reduction in IgG and neutralizing antibody levels, respectively, during the early convalescent phase, as compared to 96.8% (30/31) and 62.2% (23/37) of symptomatic patients. Forty percent of asymptomatic individuals became seronegative and 12.9% of the symptomatic group became negative for IgG in the early convalescent phase. In addition, asymptomatic individuals exhibited lower levels of 18 pro- and anti-inflammatory cytokines. These data suggest that asymptomatic individuals had a weaker immune response to SARS-CoV-2 infection. The reduction in IgG and neutralizing antibody levels in the early convalescent phase might have implications for immunity strategy and serological surveys.
With the emergence of computation-intensive vehicular applications, vehicular edge computing (VEC) offers a new paradigm to augment the capabilities of vehicles. In this article, we study the problem ...of dependency-aware task offloading and service caching in VEC, where each application can be divided into multiple tasks with task dependency, and vehicles can access the software-defined network (SDN) via roadside units (RSUs) to request edge servers to assist in processing tasks. Edge servers can selectively cache executed services for reuse by subsequent tasks to improve the offloading efficiency of the system. The offloading efficiency is defined as a weighted sum of the computation time of the task and the energy drained from the respective vehicle. To maximize the offloading efficiency, we formulate the tasks offloading and service caching problem as a mixed-integer non-linear programming (MINLP) problem and develop a semi-distributed algorithm based on dynamic programming to solve the optimization problem. Simulation results demonstrate that the proposed algorithm has higher offloading efficiency and a higher completion rate under application deadlines compared with similar offloading algorithms.
A patient's infectivity is determined by the presence of the virus in different body fluids, secretions, and excreta. The persistence and clearance of viral RNA from different specimens of patients ...with 2019 novel coronavirus disease (COVID-19) remain unclear. This study analyzed the clearance time and factors influencing 2019 novel coronavirus (2019-nCoV) RNA in different samples from patients with COVID-19, providing further evidence to improve the management of patients during convalescence.
The clinical data and laboratory test results of convalescent patients with COVID-19 who were admitted to from January 20, 2020 to February 10, 2020 were collected retrospectively. The reverse transcription polymerase chain reaction (RT-PCR) results for patients' oropharyngeal swab, stool, urine, and serum samples were collected and analyzed. Convalescent patients refer to recovered non-febrile patients without respiratory symptoms who had two successive (minimum 24 h sampling interval) negative RT-PCR results for viral RNA from oropharyngeal swabs. The effects of cluster of differentiation 4 (CD4)+ T lymphocytes, inflammatory indicators, and glucocorticoid treatment on viral nucleic acid clearance were analyzed.
In the 292 confirmed cases, 66 patients recovered after treatment and were included in our study. In total, 28 (42.4%) women and 38 men (57.6%) with a median age of 44.0 (34.0-62.0) years were analyzed. After in-hospital treatment, patients' inflammatory indicators decreased with improved clinical condition. The median time from the onset of symptoms to first negative RT-PCR results for oropharyngeal swabs in convalescent patients was 9.5 (6.0-11.0) days. By February 10, 2020, 11 convalescent patients (16.7%) still tested positive for viral RNA from stool specimens and the other 55 patients' stool specimens were negative for 2019-nCoV following a median duration of 11.0 (9.0-16.0) days after symptom onset. Among these 55 patients, 43 had a longer duration until stool specimens were negative for viral RNA than for throat swabs, with a median delay of 2.0 (1.0-4.0) days. Results for only four (6.9%) urine samples were positive for viral nucleic acid out of 58 cases; viral RNA was still present in three patients' urine specimens after throat swabs were negative. Using a multiple linear regression model (F = 2.669, P = 0.044, and adjusted R = 0.122), the analysis showed that the CD4+ T lymphocyte count may help predict the duration of viral RNA detection in patients' stools (t = -2.699, P = 0.010). The duration of viral RNA detection from oropharyngeal swabs and fecal samples in the glucocorticoid treatment group was longer than that in the non-glucocorticoid treatment group (15 days vs. 8.0 days, respectively; t = 2.550, P = 0.013) and the duration of viral RNA detection in fecal samples in the glucocorticoid treatment group was longer than that in the non-glucocorticoid treatment group (20 days vs. 11 days, respectively; t = 4.631, P < 0.001). There was no statistically significant difference in inflammatory indicators between patients with positive fecal viral RNA test results and those with negative results (P > 0.05).
In brief, as the clearance of viral RNA in patients' stools was delayed compared to that in oropharyngeal swabs, it is important to identify viral RNA in feces during convalescence. Because of the delayed clearance of viral RNA in the glucocorticoid treatment group, glucocorticoids are not recommended in the treatment of COVID-19, especially for mild disease. The duration of RNA detection may relate to host cell immunity.
We report acute antibody responses to SARS-CoV-2 in 285 patients with COVID-19. Within 19 days after symptom onset, 100% of patients tested positive for antiviral immunoglobulin-G (IgG). ...Seroconversion for IgG and IgM occurred simultaneously or sequentially. Both IgG and IgM titers plateaued within 6 days after seroconversion. Serological testing may be helpful for the diagnosis of suspected patients with negative RT-PCR results and for the identification of asymptomatic infections.
Abstract
Background
Coronavirus disease 2019 (COVID-19) is a global pandemic with no licensed vaccine or specific antiviral agents for therapy. Little is known about the longitudinal dynamics of ...severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2)–specific neutralizing antibodies (NAbs) in patients with COVID-19.
Methods
Blood samples (n = 173) were collected from 30 patients with COVID-19 over a 3-month period after symptom onset and analyzed for SARS-CoV-2–specific NAbs using the lentiviral pseudotype assay, coincident with the levels of IgG and proinflammatory cytokines.
Results
SARS-CoV-2–specific NAb titers were low for the first 7–10 days after symptom onset and increased after 2–3 weeks. The median peak time for NAbs was 33 days (interquartile range IQR, 24–59 days) after symptom onset. NAb titers in 93.3% (28/30) of the patients declined gradually over the 3-month study period, with a median decrease of 34.8% (IQR, 19.6–42.4%). NAb titers increased over time in parallel with the rise in immunoglobulin G (IgG) antibody levels, correlating well at week 3 (r = 0.41, P < .05). The NAb titers also demonstrated a significant positive correlation with levels of plasma proinflammatory cytokines, including stem cell factor (SCF), TNF-related apoptosis-inducing ligand (TRAIL), and macrophage colony-stimulating factor (M-CSF).
Conclusions
These data provide useful information regarding dynamic changes in NAbs in patients with COVID-19 during the acute and convalescent phases.
A wideband filtering patch antenna is investigated in this paper. The patch antenna is fed by an F-shaped probe, which consists of one upper and two lower horizontal arms, as well as a vertical arm ...that connects them. Owing to the novel excitation scheme, two probe modes along with a patch mode are simultaneously excited within the passband, producing a wide bandwidth of 21.3% with stable antenna gains and radiation patterns. Meanwhile, cross-coupling is constructed in the antenna, generating two symmetrical radiation nulls right at the two sides of the passband. Consequently, a compact filtering antenna with bandpass response and high selectivity is obtained, without utilizing any extra filtering circuit. This design is also extended to realize a reconfigurable filtering antenna. Two varactor diodes are embedded in the F-shaped probe to continuously tune the frequency of the operating band from 2.05 to 2.52 GHz. The antenna bandwidth can be flexibly tuned from 2.2% to 21.3%, and simultaneously good filtering performance is kept during the tuning of different states.
Abstract
Background
Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), a novel β-coronavirus, causes severe pneumonia and has spread throughout the globe rapidly. The disease associated ...with SARS-CoV-2 infection is named coronavirus disease 2019 (COVID-19). To date, real-time reverse-transcription polymerase chain reaction (RT-PCR) is the only test able to confirm this infection. However, the accuracy of RT-PCR depends on several factors; variations in these factors might significantly lower the sensitivity of detection.
Methods
In this study, we developed a peptide-based luminescent immunoassay that detected immunoglobulin (Ig)G and IgM. The assay cutoff value was determined by evaluating the sera from healthy and infected patients for pathogens other than SARS-CoV-2.
Results
To evaluate assay performance, we detected IgG and IgM in the sera from confirmed patients. The positive rate of IgG and IgM was 71.4% and 57.2%, respectively.
Conclusions
Therefore, combining our immunoassay with real-time RT-PCR might enhance the diagnostic accuracy of COVID-19.
A peptide-based magnetic chemiluminescence enzyme immunoassay for the detection of SARS-CoV-2 antibodies was developed; 71.4% (197 of 276) and 57.2% (158 of 276) of the COVID-19 inpatients were positive for IgG and IgM against SARS-CoV-2.
In order to satisfy the power thirst of communication devices in the imminent fifth generation era, wireless charging techniques have attracted much attention both from the academic and industrial ...communities. Although the inductive coupling and magnetic resonance-based charging techniques are indeed capable of supplying energy in a wireless manner, they tend to restrict the freedom of movement. By contrast, radio frequency (RF) signals are capable of supplying energy over distances, which are gradually inclining closer to our ultimate goal-charging anytime and anywhere. Furthermore, transmitters capable of emitting RF signals have been widely deployed, such as TV towers, cellular base stations, and Wi-Fi access points. This communication infrastructure may indeed be employed also for wireless energy transfer (WET). Therefore, no extra investment in dedicated WET infrastructure is required. However, allowing RF signal-based WET may impair the wireless information transfer (WIT) operating in the same spectrum. Hence, it is crucial to coordinate and balance WET and WIT for simultaneous wireless information and power transfer, which evolves to integrated data and energy communication networks (IDENs). To this end, a ubiquitous IDEN architecture is introduced by summarizing its natural heterogeneity and by synthesizing a diverse range of integrated WET and WIT scenarios. Then the inherent relationship between WET and WIT is revealed from an information theoretical perspective, which is followed by the critical appraisal of the hardware enabling techniques extracting energy from RF signals. Furthermore, the transceiver design, resource allocation, and user scheduling as well as networking aspects are elaborated on. In a nutshell, this treatise can be used as a handbook for researchers and engineers, who are interested in enriching their knowledge base of IDENs and in putting this vision into practice.
Non-alcoholic fatty liver disease (NAFLD) was a world-wide health burden. H3K27 acetylation, long non-coding RNA (lncRNA), and miRNA were all implicated in NAFLD regulation, yet the detailed ...regulatory mechanism was not well understood. LncRNA NEAT1, miR-212-5p, and GRIA3 expression were detected both in high fatty acid-treated hepatocytes cells and NAFLD patients. Lipid droplets were stained and analyzed by oil red O staining. Expression of fatty acid synthase (FASN), acetyl-CoA carboxylase (ACC), and GRIA3 was detected by qRT-PCR and western blot. RNA level of lncRNA NEAT1 and miR-212-5p was analyzed by qRT-PCR. The binding sequences of lncRNA NEAT1/miR-212-5p and miR-212-5p/GRIA3 were predicted bioinformatically and validated through luciferase assay. ChIP was performed to analyze H3K27 acetylation on the promoter of lncRNA NEAT1. LncRNA NEAT1 and GRIA3 was upregulated, while miR-212-5p was downregulated in NAFLD patients. FFA promoted lncRNA NEAT1 and GRIA3 expression while suppressing miR-212-5p and promoted lipid accumulation as indicated by increased oil red O staining and FAS and ACC expression. ChIP indicated enrichment of H3K27 on NEAT1 promoter. Inhibition of H3K27 acetylation suppressed lncRNA NEAT1 level. Luciferase results indicated direct interaction of NEAT1/miR-212-5p (which was confirmed by RIP) and miR-212-5p/GRIA3. LncRNA NEAT1 knockdown upregulated miR-212-5p level and inhibited FFA-induced lipid accumulation while suppressing GRIA3 expression. Such function was antagonized by miR-212-5p inhibition and GRIA3 knockdown counteracted with miR-212-5p inhibition. H3K27 acetylation was enriched within the promoter of lncRNA NEAT1 and promoted lncRNA NEAT1 transcription. LncRNA NEAT1 could then interact with miR-212-5p and suppress its cellular concentration.

Crop diseases are responsible for the major production reduction and economic losses in agricultural industry worldwide. Monitoring for health status of crops is critical to control the spread of diseases and implement effective management. This paper presents an in-field automatic wheat disease diagnosis system based on a weakly supervised deep learning framework, i.e. deep multiple instance learning, which achieves an integration of identification for wheat diseases and localization for disease areas with only image-level annotation for training images in wild conditions. Furthermore, a new in-field image dataset for wheat disease, Wheat Disease Database 2017 (WDD2017), is collected to verify the effectiveness of our system. Under two different architectures, i.e. VGG-FCN-VD16 and VGG-FCN-S, our system achieves the mean recognition accuracies of 97.95% and 95.12% respectively over 5-fold cross-validation on WDD2017, exceeding the results of 93.27% and 73.00% by two conventional CNN frameworks, i.e. VGG-CNN-VD16 and VGG-CNN-S. Experimental results demonstrate that the proposed system outperforms conventional CNN architectures on recognition accuracy under the same amount of parameters, meanwhile maintaining accurate localization for corresponding disease areas. Moreover, the proposed system has been packed into a real-time mobile app to provide support for agricultural disease diagnosis.
